Pilot study of allele-specific multi-InDel markers for the detection of extremely unbalanced DNA mixtures.

Mixtures are common in forensic casework, and they represent one of the most challenging types of biological evidence. Traditional short tandem repeat analyses are often associated with limitations when dealing with extremely unbalanced mixtures because alleles from minor contributors can easily be masked by those of major contributors. Consequently, researchers have developed new technologies and methods for improving the analysis of mixtures, spanning upstream DNA extraction and downstream software analysis. Among these, strategies combining allele-specific amplification with compound markers have drawn particular interest because of their ability to selectively detect minor contributors in complex mixtures. In this study, we screened multi-InDels across the entire genome, designed allele-specific primers compatible with the capillary electrophoresis platform, and further explored their potential in unbalanced DNA mixtures and cell-free fetal DNA (cffDNA). Ultimately, a set comprising 10 multi-InDels was developed, and this included two groups of primers that separately amplified the long alleles (L primer set) and short alleles (S primer set). The results demonstrated that each primer pair could detect the minor component at a 1:1000 mixture ratio, whereas the L and S primer sets successfully detected the minor contributors at mixture ratios of 1:200 and 1:500, respectively. Furthermore, in the cffDNA analysis, 60 of 78 informative markers were successfully detected, with the complete detection of all informative markers achieved in 18 mother-child reference pairs. Overall, allele-specific amplification-based multi-InDel markers enabled the sensitive detection of minor contributors, providing a potential strategy for the analysis of unbalanced two-person mixtures.

[Sequence analysis and genetic polymorphisms of D4S1647 and D6S2414 loci in Chinese Han, Mongolian and Tibetan populations].

OBJECTIVE: To investigate the polymorphisms of two short tandem repeats (STR) loci D4S1647 and D6S2414 in Chinese Han, Mongolian and Tibetan populations. METHODS: Blood and saliva samples were collected from 308 unrelated individuals in the three populations. DNA was extracted by chelex-100 method and amplified by the polymerase chain reaction (PCR). The PCR products were analyzed by nondenaturing polyacrylamide gel electrophoresis and silver staining. RESULTS: The genotype distributions of D4S1647 and D6S2414 loci were in accordance with Hardy-Weinberg equilibrium. The frequencies of alleles showed no significant differences among the three populations. Analyses of Han families indicated that the alleles agreed with Mendelian inheritance law. CONCLUSION: This study yielded information on the genetic polymorphisms of the two STR loci in Chinese Han, Mongolian and Tibetan populations.

[Associations between aldosterone synthase gene polymorphisms and left ventricular structure and function in Chinese normal Han People].

OBJECTIVE: The aim of the present study was to investigate the association between a polymorphism of the aldosterone synthase gene CYP11B2 and left ventricular structure and function in Chinese Han people. METHODS: One handred and ten persons aged 20 to 49 years were studies by echocardiography to assess left ventricular size, mass and function. The subjects were genotyped with the use of PCR-RFLP. RESULTS: Three genotypes for aldosterone synthase at position -334 in the transcriptional regulatory region were found: TT, TC and CC. The distribution of sex and arterial blood pressure was homogeneous in the aldosterone synthase genotype groups. No significant associations of echocardiographic LV dimensions, mass and function with a specific aldosterone synthase genotype were noted. CONCLUSION: The above data are not in favor of a significant association between the polymorphisms of aldosterone synthase gene CYP11B2 and the LV structure and function in Chinese normal Han people.

[A method for rapid and early diagnosis of trisomy 21 using molecular techniques].

OBJECTIVE: Using molecular techniques, we typed 2 short tandem repeat (STR) loci on 21 chromosome to establish the method for rapid and accurate diagnosis of trisomy 21. METHODS: Genomic DNA samples from 50 individuals diagnosed previously by karyotype as trisomy 21 and 40 children with severe mental retardation (IQ < 50) suspected of trisomy 21 were analyzed for 2 short tandem repeat loci on 21 chromosome, D21S1435 and D21S2055. Typing was carried out after polymerase chain reaction (PCR) and silver staining. The trisomy was identified by the number of alleles: 3 alleles bands whose density is same, two alleles bands with one obvious higher density compared to the other and one allele band whose density is three times than the normal control. RESULTS: All of the complete trisomy 21 were detected by this method; the parental source was easily determined. CONCLUSION: This method for diagnosing trisomy 21 is rapid and accurate.
