Simultaneous quantification of CTN986 and its deglycosylation products in rat serum using liquid chromatography/tandem mass spectrometry.

A quantitative method for the simultaneous determination of CTN986, a flavonol triglycoside, and its two deglycosylation products rutin and hirsutin in rat serum was developed and validated for the investigation of the pharmacokinetics of CTN986. Analytes were isolated from the serum samples (200 microL) prior to analysis by liquid chromatography/tandem mass spectrometry (LC/MS/MS) using C(18) solid-phase extraction, and were separated on a Zorbax C(8) reversed-phase column with an isocratic mobile phase consisting of methanol/isopropanol/water/formic acid (20:10:70:0.1, v/v/v/v). The protonated analytes generated in the positive ion mode were monitored through multiple reaction monitoring in an eletrospray ionization source. Calibration was performed by internal standardization with CTN987, a flavonoid structurally similar to CTN986, and regression curves were constructed ranging from 2 to 1000 ng/mL in 200 microL serum samples. The intra- and inter-day precision values were below 11% and accuracy was between -2.37 and 1.4% for all quality control samples. This quantitation method was successfully applied to pharmacokinetic studies of CTN986 in rats following oral and intravenous administration. Rutin and hirsutin were not detected in rat serum.

Layer-by-layer self-assembly of multilayer zirconia nanoparticles on silica spheres for HPLC packings.

A novel zirconia-based HPLC packing material, ZrO2/SiO2, which consists of micrometer-sized silica spheres as core and nanometer-sized zirconia particles as surface coating, was prepared by a layer-by-layer self-assembly technique. The material exhibits favorable characteristics for HPLC applications, including high surface area and pore volume, good pore structure, narrow particle size, and pore size distribution. Not only the support ZrO2/SiO2 but also the stationary-phase C18 bonded ZrO2/SiO2 exhibits excellent chemical stability. In addition, good permeability was observed for both of them. High specific area surface and good permeability of ZrO2/SiO2 permit a high loading amount of chiral polymer on it and greatly improved the enantioselectivity and resolution for some chiral separations.

Direct optical resolution of the enantiomers of novel chiral tetrahedral metal clusters by HPLC on a cellulose tris-(3,5-dimethylphenylcarbamate) stationary phase.

The enantioseparation of seven novel chiral transition metal tetrahedral clusters has been achieved for the first time on cellulose derivatized with tris(3,5-dimethylphenylcarbamate) (CDMPC) as chiral stationary phase (CSP) and hexane containing different alcohols as modifiers as mobile phases. The effect of mobile-phase composition on enantioselectivity was studied, and the effect of structural variation of the solutes on their enantioseparation was also investigated. It was found that both the metal in the tetrahedral core and the ligand coordinated to the atom in the tetrahedral core had significant effects on the chromatographic behavior of the analytes.

Enantioseparation of novel chiral heterometal tetrahedral clusters by high-performance liquid chromatography.

A series of novel chiral heterometal tetrahedral clusters have firstly been separated on cellulose tris-(3,5-dimethylphenylcarbamate) stationary phase by high-performance liquid chromatography using n-hexane as the mobile phase containing different alcohols as modifiers. The effects of concentration and structures of alcohols in the mobile phases and structures of chiral heterometal tetrahedral clusters on enantioseparation were investigated. The results showed that the concentration and structures of alcohols had large effects on enantioseparation. It also was found that both the metal in the tetrahedral core and the ligand coordinated to the atom in tetrahedral core had significant effects on their chromatographic behavior.

[Comparative study between montmorillonite and Smecta on anti-dysentery effect].

OBJECTIVE: To compare the efficiency of montmorillonite (Mengtuoshi) and Smecta. METHODS: The effects on the dysentery mice induced by MgSO4, rhubarb powder and castor oil, were observed by given respectively 15.6%, 31.2% Mengtuoshi and 31.2% Smecta. And the effects on the over-contracted rabbit intestines were also observed. RESULTS: Both Mengtuoshi and Smecta could relieve the over pushing speeding of stomach and intestine, and reduce the times of dysentery. CONCLUSION: The anti-dysentery effect of Mengtuoshi was similar to Smecta from France and may be used as good anti-dysentery drug.
