[Bone marrow stem cells transplantation improve locomotive function of dystrophin/utrophin gene double knock-out mice].

OBJECTIVE: Study the improvement of locomotive faculty of dystrophin/utropin gene double knock-out mice (dko mice) by transplanting bone marrow stem cells. METHODS: The bone marrow stem cells of C57BL/6 mice (4- to 5-weeks age) were cultured in vitro for three days, before transplanted intravenously (1.0 x 10(7) for each) into 11 dko mice (7- to 8-weeks age). The dko mice were irridiated with 7Gy gamma-ray before transplantation. 8-9 weeks after transplantation, the locomotroy function, electromyography items and expression of dystrophin in transplanted mice and controls were observed. RESULTS: 8-9 weeks after transplantation, the dropping times of hauling wire were 3.09 +/- 2.47, compared with that of the control dko mice(16.78 +/- 3.60), there are distinct differences. About electromyography items, the duration of active potential and amplitude of maxim contractions were (4.99 +/- 1.62) ms and(2872 +/- 1474.33) microV, compare with those of control dko mice(3.69 +/- 0.40) ms and(1210.0 +/- 551.0) microV, respectively, about 7% fibers of the muscle tissue of transplanted dko mice expressed dystrophin protein. CONCLUSIONS: 8-9 weeks after transplanted with homology bone marrow stem cells, the locomotive function and electromyography items of transplanted dko mice were obviously improved, and about 7% muscle tissue fibers of the mice expressing dystrophin protein were observed. It suggested that there is an ideal prospect for DMD therapy with bone marrow stem cells transplantation.

[Study on the gene knockout model mice of Duchenne muscular dystrophy].

OBJECTIVE: With a view to further experimental studies on Duchenne muscular dystrophy (DMD), we investigated the motor function, serum creatine kinase (CK) level and pathological characteristics of muscular tissue in C57, mdx, dystrophin/utrophin gene double knockout (dko)mice. METHODS: Gene identification was performed for the filial generation of heterozygote of utrophin gene knockout of mdx mice for acquiring mdx and dko mice. The motor function, serum CK level, and the pathology of muscular tissue of C57, mdx and dko mice were compared. RESULTS: There were significant differences in all detecting aspects between C57 mouse and dko mouse. In the aspects of serum CK level and pathology of muscular tissue, there were differences between C57 mouse and mdx mouse; however, no significant difference in motor function was observed between them. CONCLUSION: Compared with mdx mouse, dko mouse is in a more serious illness state which is nearer to the natural state of DMD. Therefore, dko mouse is a more ideal model for studying the praxiology of DMD.

[Analysis of short tandem repeat polymorphism in a female patient with Duchenne muscular dystrophy].

OBJECTIVE: To understand the mechanism of Duchenne/Becker muscular dystrophy (DMD) in a female patient. METHODS: A multiplex PCR (mPCR) protocol was applied to detect the dystrophin gene of the female DMD patient and her family members, whose haplotypes were analyzed in light of short tandem repeat polymorphism (STR) of five microsatellite markers (located in 5' terminus and introns 44, 45, 49, and 50). RESULTS: No deletion was detected in the affected female patient and her affected son. Examination of the female DMD patient's STR haplotypes identified in the female patient and her affected son the same haplotype inherited from her unaffected mother, who was a likely germinal mosaicism. The female patient has different haplotype from her affected son and her mother as a deletion was identified in the intron 45 of the affected son, and the female patient and her mother were probably heterozygous for this deletion. CONCLUSION: STR haplotype may not be responsible for the pathogenesis of DMD in the female patient and her affected son.

[Effects of the antibacterial peptide cecropins from Chinese oak silkworm, Antheraea pernyi on 1, 2-dimethylhydrazine-induced colon carcinogenesis in rats].

OBJECTIVE: To gain insight into the putative anticancer effect of the antibacterial peptides, cecropins, from Chinese oak silkworm, Antheraea pernyi, on the cancer cells and 1,2-dimethylhydrazine (DMH)-induced colon carcinogenesis in rats. METHODS: Growth inhibitory effect of the cecropins on normal human gastric epithelial cell line (GES-1) and human colon adenocarcinoma cell line (LS-174T) was observed using a microculture tetrazolium (MTT) colorimetric methods. Male Wistar rats were divided into 4 groups. Group1 was given on a weekly basis cecropins from Antheraea pernyi (3,000 Ua/ml) by gavage at 2 doses of 10 ml/kg body weight and exposed to subcutaneous injection of DMH at the dose of 20 mg/kg body weight. Group 2 was received weekly DMH only. Group 3 was given the cecropins by gavage at the same dose as in group 1. Group 4 was weekly exposed to subcutaneous injection of EDTA (1 mmol/L). All treatments were completed in a course of 18 weeks and the experiment was finished at week 33. RESULTS: MTT assay showed selective cytotoxic activity of the cecropins against the human colon adenocarcinoma cells line. The viability of the cancer cells was about 54% and 100% for the normal cells. There was a significantly lower incidence of large intestinal tumors in rats gavaged with cecropins (65%, P<0.01), but the tumor burden (tumors/tumor-bearing animal) and tumor mass index were comparable between the groups (P>0.05). CONCLUSION: The cecropins possess effective anti-tumor activity with no cytotoxicity against normal eukaryotic cells, and impede the neoplastic process in murine large intestines.

[Regulation of Fas expression in gastric epithelium: one of the auto-immune mechanisms of Helicobacter pylori pathogenesis].

OBJECTIVE: To investigate the pathogenic mechanism of Helicobacter pylori (H.pylori)-mediated gastric epithelial cell damage. METHODS: Fas expressions in gastric epithelial cell lines and freshly isolated gastric epithelial cells with or without H.pylori infection were evaluated by flow cytometry. The modulation of Fas expression in gastric epithelial cells by H.pylori or by Th1 cytokines present in H.pylori-infected gastric mucosa was assessed in vitro. The function of Fas expressed by the gastric epithelial cells to induce cell apoptosis was determined by enzyme-linked immunosorbent assay (ELISA) after incubation of the cells with anti-Fas antibody. RESULTS: All of the three gastric epithelial cell lines, AGS, N87 and kato-III, expressed detectable Fas protein when examined by flow cytometry. The percentage of positive cells and the amount of Fas protein on cell surface were larger in freshly isolated gastric epithelial cells with H.pylori infection than in uninfected cells (P<0.05). H. pylori alone or in combination with Th1 cytokines (IFN-gamma and TNF-alpha) significantly increased the expression of Fas in gastric epithelial cell lines in vitro. After incubation with IgM anti-Fas mAb, Fas-bearing gastric epithelial cells underwent apoptosis, and this effect of Fas was enhanced by IFN-gamma. CONCLUSION: Th1 cells accumulated in the gastric mucosa during H.pylori infection is involved in the damage of gastric epithelium through the modulation of Fas/Fas ligand interaction.

Solution structure of the viral receptor domain of Tva and its implications in viral entry.

Tva is the cellular receptor for subgroup A avian sarcoma and leukosis virus (ASLV-A). The viral receptor function of Tva is determined by a 40-residue, cysteine-rich motif called the LDL-A module. Here we report the solution structure of the LDL-A module of Tva, determined by nuclear magnetic resonance (NMR) spectroscopy. Although the carboxyl terminus of the Tva LDL-A module has a structure similar to those of other reported LDL-A modules, the amino terminus adopts a different conformation. The LDL-A module of Tva does not contain the signature antiparallel beta-sheet observed in other LDL-A modules, and it is more flexible than other reported LDL-A modules. The LDL-A structure of Tva provides mechanistic insights into how a simple viral receptor functions in retrovirus entry. The side chains of H38 and W48 of Tva, which have been identified as viral contact residues by mutational analysis, are solvent exposed, suggesting that they are directly involved in EnvA binding. However, the side chain of L34, another potential viral contact residue identified previously, is buried inside of the module and forms the hydrophobic core with other residues. Thus L34 likely stabilizes the Tva structure but is not a viral interaction determinant. In addition, we propose that the flexible amino-terminal region of Tva plays an important role in determining specificity in the Tva-EnvA interaction.

Coronary bypass revascularization with radial artery and internal mammary artery grafts.

OBJECTIVE: To evaluate radial artery (RA) and internal mammary artery (IMA) grafts in coronary artery bypass and the use of color Doppler ultrasound in the peri-operative evaluation of IMA and radial-ulnar collateral circulation. METHODS: From June 1998 to June 2000, sixty cases of coronary bypass revascularization with RA and IMA were performed. Preoperatively, the radial-ulnar collateral circulation was evaluated with the modified Allen's test, color Doppler ultrasound and noninvasive oxygen saturation measurement. The IMA lumen and blood flow were measured at the first intercostal space with color Doppler ultrasound preoperatively and postoperatively. RESULTS: One patient (1.7%) died of serious cardiac arrhythmia on the fourth postoperative day. There were no arterial graft harvest related complications. Before harvesting, the ulnar artery blood flow was 30.78 +/- 9.71 ml/min, and it increased to 43.36 +/- 13.98 ml/min (40.87% increase, P < 0.01) after the operation. Compared with the baseline, there was no obvious change of IMA blood flow postoperatively (P > 0.05), but the systolic/diastolic flow ratio markedly decreased from 8.57 +/- 3.98 ml/min to 3.41 +/- 4.87 ml/min (P < 0.01). CONCLUSIONS: Arterial grafts can be safely used for coronary bypass revascularization with good results. The ulnar artery blood flow can increase compensatively after RA harvesting. The diastolic blood flow of grafted IMA markedly increased postoperatively. Color Doppler ultrasound was very helpful both in evaluating the radial-ulnar collateral circulation before RA harvesting and in assessing the patency of the grafted IMA after coronary artery bypass grafting (CABG).

[Construction and expression of recombinant human thymosin alpha1 fusion gene in Pichia pastoris].

The human Thymosin alpha1 (hTalpha1) gene was synthesized according to the optimal codons of Pichia pastoris and was fused in 5' terminal of ribosomal protein (RP) gene using over lapping polymerase chain reaction. The fusion gene was inserted into expression vector of pPIC3. 5K and was transformed into HIS4 mutant strain GS115 by electroporation. Both SDS-PAGE and Western blot indicated that this fusion protein was expressed. The expression level was about 25mg/L.
