Tryptamine and homotryptamine-based sulfonamides as potent and selective inhibitors of 15-lipoxygenase.

A series of inhibitors of mammalian 15-lipoxygenase based on tryptamine and homotryptamine scaffolds is described. Compounds with aryl substituents at C-2 of the indole core of tryptamine and homotryptamine sulfonamides (e.g., 37a-p) proved to be potent inhibitors of the isolated enzyme. Selected compounds also demonstrated desirable inhibition selectivities over isozymes 5- and P-12-LO.

The R-enantiomer of the nonsteroidal antiinflammatory drug etodolac binds retinoid X receptor and induces tumor-selective apoptosis.

Prostate cancer is often slowly progressive, and it can be difficult to treat with conventional cytotoxic drugs. Nonsteroidal antiinflammatory drugs inhibit the development of prostate cancer, but the mechanism of chemoprevention is unknown. Here, we show that the R-enantiomer of the nonsteroidal antiinflammatory drug etodolac inhibited tumor development and metastasis in the transgenic mouse adenocarcinoma of the prostate (TRAMP) model, by selective induction of apoptosis in the tumor cells. This proapoptotic effect was associated with loss of the retinoid X receptor (RXRalpha) protein in the adenocarcinoma cells, but not in normal prostatic epithelium. R-etodolac specifically bound recombinant RXRalpha, inhibited RXRalpha transcriptional activity, and induced its degradation by a ubiquitin and proteasome-dependent pathway. The apoptotic effect of R-etodolac could be controlled by manipulating cellular RXRalpha levels. These results document that pharmacologic antagonism of RXRalpha transactivation is achievable and can have profound inhibitory effects in cancer development.

Effect of GRP75/mthsp70/PBP74/mortalin overexpression on intracellular ATP level, mitochondrial membrane potential and ROS accumulation following glucose deprivation in PC12 cells.

Glucose regulated protein 75 (GRP75) is an important molecular chaperon belonged to the heat shock protein (HSP) family. To evaluate the effect of GRP75 overexpression on PC12 cells under glucose deprivation, cell viability and mitochondrial function of GRP75-overexpressing PC12 cells and the vector transfected control PC12 cells were monitored during glucose deprivation. Upon exposure to glucose deprivation, GRP75-overexpressing PC12 cells exhibited more moderate cell damage than control PC12 cells. Both of the two groups of cells showed a decreased ATP level following an early increase in the condition of glucose deprivation, and the mitochondrial potential were also reduced in the similar manner in the two groups of cells. Control PC12 cells showed an immediate and rapid increase in ROS accumulation after the onset of GD treatment, and this accumulation was slowed and reduced in GRP75-overexpressing PC12 cells. These findings suggested that GRP75 could inhibit the ROS accumulation, and it may be associated with the cytoprotective effect of GRP75 overexpression upon glucose deprivation.

A bent infusion cannula for vitreous surgery.

PURPOSE: To modify an infusion cannula for vitreous surgery and decrease cannula-related complications. DESIGN: New surgical instrument. METHODS: A conventional 20-gauge infusion cannula was modified by forming a 135-degree angle at the joint of the flange between the cannula adapter and the needle. The sloped plane of the needle tip was opposite the bent adapter. The cannula was routinely fastened with a 7-0 absorbable suture after insertion into the eye. RESULTS: We used the modified infusion cannula in 50 consecutive phakic eyes (48 patients). Intraoperatively, the cannula remained attached to the globe surface; it did not tilt anteriorly and damage the lens when touched inadvertently. No suprachoroidal or subretinal infusion was observed. Neither the lens nor the retina was damaged. There were no cannula-related retinal detachments or breaks during follow-up. CONCLUSIONS: The modified infusion cannula is more effective and safer during pars plana vitrectomy in phakic eyes compared with a conventional cannula.

The neocarzinostatin biosynthetic gene cluster from Streptomyces carzinostaticus ATCC 15944 involving two iterative type I polyketide synthases.

The biosynthetic gene cluster for the enediyne antitumor antibiotic neocarzinostatin (NCS) was localized to 130 kb continuous DNA from Streptomyces carzinostaticus ATCC15944 and confirmed by gene inactivation. DNA sequence analysis of 92 kb of the cloned region revealed 68 open reading frames (ORFs), 47 of which were determined to constitute the NCS cluster. Sequence analysis of the genes within the NCS cluster suggested dNDP-D-mannose as a precursor for the deoxy aminosugar, revealed two distinct type I polyketide synthases (PKSs), and supported a convergent model for NCS chromophore biosynthesis from the deoxy aminosugar, naphthoic acid, and enediyne core building blocks. These findings shed light into deoxysugar biosynthesis, further support the iterative type I PKS paradigm for enediyne core biosynthesis, and unveil a mechanism for microbial polycyclic aromatic polyketide biosynthesis by an iterative type I PKS.

Purification and characterization of bovine adrenal cytochrome b561 expressed in insect and yeast cell systems.

Bovine adrenal chromaffin granule cytochrome (cyt) b561 is a transmembrane hemoprotein that plays a key role in transporting reducing equivalents from ascorbate to dopamine-beta-hydroxylase for catecholamine synthesis. We have developed procedures for expression and purification of functional bovine adrenal cyt b561 in insect and yeast cell systems. The bovine cyt b561 coding sequence, with or without a hexahistidine-tag sequence at the C-terminus, was cloned into the pVL1392 transfer vector under the control of the polyhedrin promoter to generate recombinant baculovirus for protein expression in Sf9 insect cells (approximately 0.5 mg detergent-solubilized cyt b561/L culture). For the yeast system, the cyt b561 cDNA was modified with a hexahistidine-tag sequence at the C-terminus, and inserted into the pPICZB vector under the control of the alcohol oxidase promoter. The recombinant plasmid was transformed into Pichia pastoris GS115 competent cells to give methanol-inducible cyt b561 expression (approximately 0.7 mg detergent-solubilized cyt b561/L culture). Recombinant His-tagged cyt b561 expressed in Sf9 or Pichia cells was readily solubilized from membrane fractions with dodecyl maltoside and purified to electrophoretic homogeneity by one-step chromatography on Ni-NTA affinity resin. The purified recombinant cytochrome from both systems had a heme to protein ratio close to two and was fully functional, as judged by comparison with the spectroscopic and kinetic parameters of the endogenous cytochrome from chromaffin granules. A novel procedure for isolation of chromaffin granule membranes was developed to utilize frozen adrenal glands instead of fresh tissue.

Antiinflammatory and analgesic activities of the tissue culture of Saussurea involucrata.

The tissue culture of Saussurea involucrata (TCSauI) KAR. et KIR. was studied to determine its antiinflammatory and analgesic activities in experimental animals. Similar to wild S. involucrata, TCSauI at doses of 75-300 mg/kg i.g. for 7 d markedly inhibited hindpaw edema induced by carrageenin in rats, ear edema induced by dimethylbenzene, and increased capillary permeability in the mouse abdominal cavity induced by acetic acid. Moreover, TCSauI had inhibitory activities against the writhing reaction induced by acetic acid and the hot plate reaction in mice. The present study provided evidence first that TCSauI has antiinflammatory and analgesic activities, suggesting the potential of the tissue culture technique to substitute for wild S. involucrata in the pharmaceutical industry.

[Dermatomyositis/polymyositis associated with head and neck malignancy].

OBJECTIVE: To observe the relation between dermatomyositis/polymyositis and head-neck malignancy. METHOD: All 136 cases of dermatomyositis/polymyositis admitted to PUMCH during 10 years (1994-2003) were studied. The clinical features of 6 patients with an associated head-neck malignancy were analyzed. RESULT: There were 17 cases of malignancy in 136 patients with dermatomyositis/polymyositis, of which, 6 cases (6/17, 35.29%) were found to have head-neck malignancy: 4 cases of nasopharyngeal carcinoma, 1 case of non-Hodgkin lymphoma and 1 case of hypopharynx carcinoma. CONCLUSION: Nasopharyngeal carcinoma represents the first neoplasm among head-neck malignancy associated with dermatomyositis/polymyositis. Nasal endoscopic and/or fibrolaryngoscopic search for potential head-neck malignancy should be performed in every patient with dermatomyositis/polymyositis.

[No association of -1438G/A polymorphism in promoter region of 5-HT2A receptor gene with antipsychotic agent-induced weight gain].

OBJECTIVE: To investigate whether the -1438G/A polymorphism in the promoter region of 5-HTR2A gene associates with the weight gain following antipsychotic agents (APS) acute treatment in schizophrenic patients. METHODS: Eighty-four Chinese Han patients with schizophrenia at the first onset were recruited from among 70 nuclear families. The polymorphism of 5-HTR2A gene was determined with PCR-RFLP technique. Body weight was measured in the patients on admission after 10 weeks of treatment with risperidone or chlorpromazine. RESULTS: There were no statistically significant differences in the distribution frequencies of genotype (chi2: 0.172, v1, P > 0.05) and allele (chi2: 0.121, v1, P > 0.05) of -1438G/A polymorphism of 5-HTR2A gene between subgroups (weight gain >or= 7% or < 7%). Likewise, there was no significant difference in weight gain between genotype groups. By means of transmission disequilibrium test and quantitative transmission disequilibrium test, no significant association between the -1438G/A polymorphism of 5-HTR2A gene and weight gain was observed. CONCLUSION: 5-HTR2A gene -1438G/A polymorphism was probably not associated with APS-induced weight gain in Chinese Han patients with schizophrenia in this study.

[Effect of glutamine on the expression of grp75 in PC12 cells].

Glutamine (Gln), a conditionally essential amino acid, can be a potential enhancer of the heat stress response. And glucose-regulated protein 75(grp75) is a member of the hsp family. To evaluate the effect of glutamine on the expression of grp75, PC12 cell was cultured with DMEM, glucose-free DMEM, DMEM within Gln, and glucose-free DMEM within Gln, and the expression of grp75 was detected by immunocytochemistry and western blot, the mRNA level of grp75 in the cells was detected by reverse transcriptase-polymerase chain reaction (RT-PCR). Data indicated that Gln can upregulated the expression of grp75 in PC12 cell line, and the effect is more significantly in PC12 cell which was glucose deprivation than the normal cell. To investigate the effect of Gln on PC12 cells under glucose deprivation, MTT method was used to monitor the cell viability after Gln treatment for the cell under glucose depriving. The experiments results showed that glutamine at a concentration range of 0.2-40 mmol/L significantly enhanced the cell viability in glucose-free DMEM. And the protective effect to grp75 low-expresssion on PC12 cells is markedly decreased.

External approach microsurgery of retinal dialysis.

PURPOSE: To explore the effect of external approach microsurgery in retinal dialysis. METHODS: Consecutive 30 eyes of 28 patients with retinal dialysis were enrolled for this study. The progresses of the external approach microsurgery were following. Under the surgical microscopy, the preplacement of mattress sutures for buckling and/or encircling following retrobulbar anesthesia and scleral exposure, draining subretinal fluid, the cryotherapy of retinal breaks, checking the position of breaks on scleral buckle and gases injection were performed in turn. RESULTS: After drainage of subretinal fluid, with scleral depression cryotherapy reaction around breaks could be observed clearly under the microscopy. All breaks were located on anterior slope of the buckle. Intraoperative complications were mild subretinal hemorrhage at drainage site and corneal epithelium exfoliation in 3 eyes, respectively. Postoperative complications were mainly secondary glaucoma and retinal redetachment. The one-operation reattachmentl rate was 96.7% (29 eyes), and the final reattachment rate was 100% after one eye had a second external approach microsurgery. The postoperative vision acuity (VA) was significantly better than the preoperative VA (chi(2)= 9.529, P< 0.01). CONCLUSION: External approach microsurgery has favourable effect on the surgery of retinal dialysis.

Characterization of the heme environment in Arabidopsis thaliana fatty acid alpha-dioxygenase-1.

Plant alpha-dioxygenases (PADOX) are hemoproteins in the myeloperoxidase family. We have used a variety of spectroscopic, mutagenic, and kinetic approaches to characterize the heme environment in Arabidopsis thaliana PADOX-1. Recombinant PADOX-1 purified to homogeneity contained 1 mol of heme bound tightly but noncovalently per protein monomer. Electronic absorbance, electron paramagnetic resonance, and magnetic circular dichroism spectra showed a high spin ferric heme that could be reduced to the ferrous state by dithionite. Cyanide bound relatively weakly in the ferric PADOX-1 heme vicinity (K(d) approximately 10 mm) but did not shift the heme to the low spin state. Cyanide was a very strong inhibitor of the fatty acid oxygenase activity (K(i) approximately 5 microm) and increased the K(m) value for oxygen but not that for fatty acid. Spectroscopic analyses indicated that carbon monoxide, azide, imidazole, and a variety of substituted imidazoles did not bind appreciably in the ferric PADOX-1 heme vicinity. Substitution of His-163 and His-389 with cysteine, glutamine, tyrosine, or methionine resulted in variable degrees of perturbation of the heme absorbance spectrum and oxygenase activity, consistent with His-389 serving as the proximal heme ligand and indicating that the heme has a functional role in catalysis. Overall, A. thaliana PADOX-1 resembles a b-type cytochrome, although with much more restricted access to the distal face of the heme than seen in most other myeloperoxidase family members, explaining the previously puzzling lack of peroxidase activity in the plant protein. PADOX-1 is unusual in that it has a high affinity, inhibitory cyanide-binding site distinct from the distal heme face and the fatty acid site.

Conversion of Bcl-2 from protector to killer by interaction with nuclear orphan receptor Nur77/TR3.

The Bcl-2 family proteins are key regulators of apoptosis in human diseases and cancers. Though known to block apoptosis, Bcl-2 promotes cell death through an undefined mechanism. Here, we show that Bcl-2 interacts with orphan nuclear receptor Nur77 (also known as TR3), which is required for cancer cell apoptosis induced by many antineoplastic agents. The interaction is mediated by the N-terminal loop region of Bcl-2 and is required for Nur77 mitochondrial localization and apoptosis. Nur77 binding induces a Bcl-2 conformational change that exposes its BH3 domain, resulting in conversion of Bcl-2 from a protector to a killer. These findings establish the coupling of Nur77 nuclear receptor with the Bcl-2 apoptotic machinery and demonstrate that Bcl-2 can manifest opposing phenotypes, induced by interactions with proteins such as Nur77, suggesting novel strategies for regulating apoptosis in cancer and other diseases.

Identification of Tyr504 as an alternative tyrosyl radical site in human prostaglandin H synthase-2.

Hydroperoxides induce formation of a tyrosyl radical on Tyr385 in prostaglandin H synthase (PGHS). The Tyr385 radical initiates hydrogen abstraction from arachidonic acid, thereby mechanistically connecting the peroxidase and cyclooxygenase activities. In both PGHS isoforms the tyrosyl radical undergoes a time-dependent transition from a wide doublet to a wide singlet species; pretreatment with cyclooxygenase inhibitors results in a third type of signal, a narrow singlet [Tsai, A.-L.; Kulmacz, R. J. (2000) Prost. Lipid Med. 62, 231-254]. These transitions have been interpreted as resulting from Tyr385 ring rotation, but could also be due to radical migration from Tyr385 to another tyrosine residue. PATHWAYS analysis of PGHS crystal structures identified four tyrosine residues with favorable predicted electronic coupling: residues 148, 348, 404, and 504 (ovine PGHS-1 numbering). We expressed recombinant PGHS-2 proteins containing single Tyr --> Phe mutations at the target residues, a quadruple mutant with all four tyrosines mutated, and a quintuple mutant, which also contains a Y385F mutation. All mutants bind heme and display appreciable peroxidase activity, and with the exception of the quintuple mutant, all retain cyclooxygenase activity, indicating that neither of the active sites is significantly perturbed. Reaction of the Y148F, Y348F, and Y404F mutants with EtOOH generates a wide singlet EPR signal similar to that of native PGHS-2. However, reaction of the Y504F and the quadruple mutants with peroxide yields persistent wide doublets, and the quintuple mutant is EPR silent. Nimesulide pretreatment of Y504F and the quadruple mutant results in an abnormally small amount of wide doublet signal, with no narrow singlet being formed. Therefore, the formation of an alternative tyrosine radical on Tyr504 probably accounts for the transition from a wide doublet to a wide singlet in native PGHS-2 and for formation of a narrow singlet in complexes of PGHS-2 with cyclooxygenase inhibitors.

Sellmeier's equation and the expression of the thermal refractive-index coefficient for a Nd0.007Gd0.993VO4 crystal.

The principal refractive indices of a Nd0.007Gd0.993VO4 crystal for wavelengths of 0.488, 0.6328, 1.064, 1.0795, and 1.3414 microm under the temperature range of 20-170 degrees C are measured by the autocollimation method, and the Sellmeier's equation and the expression of the thermal refractive-index coefficient are then obtained. The reliability of these results is checked by comparing the calculated values with the measured values for the measured wavelengths and the temperature region, as well as with the published values by Studenikin et al. [Quantum Electron. 25, 1162-1165 (1995)] for wavelengths of 0.491, 0.546, 0.578, 0.632, 0.808, and 1.062 microm at 20 degrees C. In addition, the birefringence and the thermal coefficient of the birefringence were obtained by measured results. The results show that the birefringence is larger than that of a YVO4 crystal and that the thermal coefficient of birefringence is less than that of a YVO4 crystal. Therefore it can be expected that the GdVO4 crystal is not only an excellent laser crystal but also an excellent birefringent crystal used in a modern optical transmission system as a passive fiber-optic component.

Aminoimidazoles as bioisosteres of acylguanidines: novel, potent, selective and orally bioavailable inhibitors of the sodium hydrogen exchanger isoform-1.

Inhibition of the sodium hydrogen exchanger isoform-1 (NHE-1) has been shown to limit damage to the myocardium under ischemic conditions in animals. While most known NHE-1 inhibitors are acylguanidines, this report describes the design and synthesis of a series of heterocyclic inhibitors of NHE-1 including aminoimidazoles with undiminished in vitro activity and oral bioavailability.

The effects of ankle bracing on motion of the knee and the hip joint during trunk rotation tasks.

BACKGROUND: The use of prophylactic ankle braces is common during athletic activities since the ankle is one of the most commonly injured joints. Past studies have focused on the effects of ankle braces on ankle movement restriction, preventing injuries, proprioception, balance and athletic performance. However, the influence of ankle restriction on other joints has not been studied. The constraint of ankle movement may lead to an increased loading on the knee joint, which could be a potential risk of knee injuries during athletic activities. The primary goal of the current study was to determine quantitatively the effect of an ankle brace on the knee axial rotation during two different trunk turning tasks. METHODS: Ten healthy subjects performed trunk turning movements while standing on one leg: turning sideways to catch a ball and turning sideways to touch a target with the shoulder. The tasks were performed with and without an ankle brace worn on the supporting leg. The trunk axial rotation in reference to the floor and three dimensional joint angular motions of the ankle, knee and hip were determined. FINDINGS: The use of an ankle brace resulted in reduced trunk axial rotation during the ball catching tasks, and increased knee axial rotation during the target touching tasks. INTERPRETATION: The results of this study showed that the effect of the ankle brace on the knee axial rotation depended on the context of the tasks performed. Under situations that required forceful trunk turning movement while standing on a single leg, the ankle braces may cause an increase in the knee axial rotation indicating higher risk of knee injury.

Sex difference in psychological behavior changes induced by long-term social isolation in mice.

Social isolation can induce psychological behavior changes. It is interesting to know whether there is sex difference in responding to social isolation or not. The present study compared the behavior difference between male and female mice isolated for 1-4 months. The results showed that the isolated male mice had higher accounts of locomotor activity than the isolated female and group-housed ones. Both isolated male and female mice spent shorter time in the dark box than the group-housed mice in the light/dark test, and isolated male mice spent less time in the closed arms than isolated female and group-housed mice when isolated for 2, 3 and 4 months in the elevated plus-maze test. These results suggest that isolation induce an anxiolytic-like effect. The immobile time in the forced swimming test was shortened in male mice isolated for 1 and 2 months. Both isolated male and female mice showed shorter time in pentobarbital-induced loss of righting reflex and less body weight gain. These results demonstrated that there was a sex difference in psychological behavior changes in mice undergoing social isolation and the male mice were more easily affected by isolation.
