[Homologous amelogenin gene test of archaeological samples].

OBJECTIVE: Based on the sequence differences of Amelogenin homologous gene in the X and Y chromosomes, a pair of specific primers was designed to identify the sex of archaeological samples. METHODS: Ancient DNA fragments were extracted from the bones and teeth of sacrificial slaves with an improved method that combines phenol-chloroform extraction, silicon dioxide adsorption with ultrafiltration concentration. The polyacrylamide gel electrophoresis (PAGE) was used to detect PCR products. RESULTS: Seven in sixteen samples from eight graves showed positive results and the targeted segments were visible: a male with two bands of 106bp (Amel-X) and 112 bp (Amel-Y), while a female with only one band of 106 bp (Amel-X). Ancient DNA analyzing results from tooth samples are more marked than that from bones. CONCLUSION: The improved extraction method is more effective for ancient DNA extraction, which reduced the PCR inhibitors and lowered experimental costs. The sex determination technology based on Amelogenin homologous gene is an important and feasible method in the molecular archaeological research.

Impact of environmental factors on the toxicity of Bacillus thuringiensis var. israelensis IPS82 to Chironomus kiiensis.

To determine the feasibility of microbiological control of chironomid larvae in water sources for the city of Shenzhen, China, the toxicity characteristics of Bacillus thuringiensis var. israelensis (Bti) IPS82 on Chironomus kiiensis Tokunaga were studied. Tests on fermentation products of IPS82 showed good correlations between toxicity, cell density, dissolved oxygen, and the spore-forming phase. In this study, bioassays were carried out with different stage larvae. Results showed that the LC50s (24 h postexposure) were 8.2, 15.2, 24.7, and 38.6 mg/liter for the 1st, 2nd, 3rd, and 4th instars, respectively. Tests on environmental factors influencing toxicity of Bti to C. kiiensis showed that sunlight is the most important factor, shortening the half-life of Bti from 21 days in dark to 10 days under sunlight. Temperature variations (15-30 degrees C) caused no impact on toxicity, but a 16% increase in larval mortality was observed at 35 degrees C. The toxicity of IPS82 was greatest at a pH of 7. In field trials, dosages above 100 mg/liter were effective in the control of C. kiiensis. Our study indicated that it is feasible to use Bti to control C. kiiensis in city source water.

[Study on genetic diversity of wild quail in China with microsatellite DNA markers].

Genetic diversity of domestic quail and two wild quail species distributed in China, wild Japanese quail and wild common quail,was studied by using microsatellite DNA markers. According to the comparison of corresponding genetic index in the three quail populations, such as polymorphism information content (PIC), mean heterozygosity (H) and fixation index etc, wild common quail possessed rich genetic diversity of 4.67 alleles per locus. Its value of PIC and H were the highest, 0.5732 and 0.6621, respectively. Meanwhile, domestic quail had the lowest value, 0.5467 and 0.5933, respectively. Wild Japanese quail had little difference in genetic diversity with domestic quail. In addition,from analyses of fuzzy cluster based on standard genetic distance,the similarity relation matrix coefficients between wild Japanese quail and domestic quail was 0.937, and that between wild common quail and domestic quail was 0.783. All these results showed that wild Japanese quail was closer to the domestic quail in phylogenetic relationship than wild common quail. These results at the molecular level further proved the thesis that domestic quail is originated from wild Japanese quail.

Study of the determination and pharmacokinetics of Compound Danshen Dripping Pills in human serum by column switching liquid chromatography electrospray ion trap mass spectrometry.

Compound Danshen Dripping Pill (CDDP) is an important drug widely used for the treatment of cardiovascular diseases. An active component Danshensu (DS) of CDDP was separated by reversed-phase high performance liquid chromatography using column-switching system and analyzed by electrospray mass spectrometry. With this validated assay the pharmacokinetics of CDDP was studied in 10 healthy volunteers after a single oral administration of 250 mg. After trichloroacetic acid precipitation of serum proteins, the analytes were preconcentrated and black-flushed on a reversed-phase column for separation using a switching valve. The analytes were ionized using negative electrospray ionization (ESI) mode. The precursor ion of m/z 196.6 was used to quantify DS in serum. The linear calibration curve ranged from 1.25 to 175 microg/mL. The limit of quantification (LOQ) for DS was 0.15 microg/mL. The intra-day and inter-day precision (R.S.D.) was less than 7.4 and 7.9%, respectively.

Prevalence of a newly identified SEN virus in China.

AIM: To establish nested-PCR methods for the detection of SENV-D and SENV-H and to investigate the epidemiology of SEN virus in China. METHODS: According to published gene sequences, primers from the conserved region were designed. Then, 135 samples from healthy voluntary blood donors and 242 samples from patients with various forms of liver disease were detected by nested-PCR of SENV-D/H. Some PCR products were cloned and sequenced. RESULTS: By sequencing, the specificity of genotype-specific PCR was confirmed. SENV-D/H DNA was detected in 31% of the blood donors, which was higher than those in America and Italy (2%), and in Japan and Taiwan (15-20%). The prevalence of SENV-D/H viremia was significantly higher in patients with hepatitis B and hepatitis C than in blood donors (59-85% vs 31%, P<0.05). The prevalence among patients with non-A-E hepatitis was significantly higher than among blood donors (68% vs 31%, P<0.01), and equivalent to that among patients with hepatitis B and C. CONCLUSION: Nested-PCR with genotype-specific primers could serve as a useful SENV screening assay. SENV has the same transmission modes as HBV and HCV. The high prevalence in patients with non-A-E hepatitis may attribute to the transmission modes, and SENV may not serve as the causative agents.

Chromatographic behaviors of proteins on cation-exchange column.

A weak cation-exchanger (XIDACE-WCX) has been synthesized by the indirect method. The chromatographic characteristics of the synthesized packing was studied in detail. The standard protein mixture and lysozyme from egg white were separated with the prepared chromatographic column. The chromatographic thermodynamics of proteins was studied in a wide temperature range. Thermodynamic parameters standard enthalpy change (deltaH0) and standard entropy change (deltaS0) and compensation temperature (beta) at protein denaturation were determined in the chromatographic system. By using obtained deltaS0, the conformational change of proteins was judged in the chromatographic process. The linear relationship between deltaH0 and deltaS0 can be used to identify the identity of the protein retention mechanism in the weak cation-exchange chromatography. The interaction between weak cation-exchanger and metal ions was investigated. Several metal chelate columns were prepared. The effects of introducing metal ion into the naked column on protein retention and the retention mechanism of proteins in the metal chalet affinity chromatography were discussed.

[Stomatal and nonstomatal limitations of photosynthesis in mung bean leaves under the combination of enhanced UV-B radiation and NaCl stress].

Stomatal and nonstomatal limitations of photosynthesis in mung bean (Phaseolus radiatus L.) leaves under the combination of 0.35 W/m(2) UV-B radiation and 0.4% NaCl stress were studied. Separated or combined treatments of enhanced UV-B radiation and NaCl stress all resulted in a decrease in net photosynthetic rate, stomatal conductance, photosynthetic ability, efficiency of CO(2) carboxylation and Rubisco content, and the extent of those decreases enhanced obviously with the increase of treatment days. Intercellular CO(2)concentration tended to decrease in short period, followed by a gradual increase and to be higher than the control on the seventh day after treatment under NaCl stress alone, while a tendency of gradual increase was observed under enhanced UV-B radiation alone or two stresses combined and the concentration was higher than the control after three day of treatments. By contrast, the tendency of stomatal limitation value was just reverse to the intercellular CO(2) concentration, but the stomatal limitation value was always higher than control under all stress conditions (did not include that on the fifth day under the combined stresses). As compared with an individual stress the effect of combined stress on the above parameters was more serious. These results indicate that under all stress conditions the inhibition of photosynthesis in mung bean leaves is the results of both stomatal and nonstomatal limitations. And the stomatal limitation is dominant in short period, nonstomatal limitation becomes the dominant one in longer period. The decrease in Rubisco content leads to nonstomatal limitation of photosynthesis under all stresses.

Reduction of Rubisco amount by UV-B radiation is related to increased H2O2 content in leaves of mung bean seedlings.

To understand the role of H(2)O(2) in the damaging effect of UV-B radiation on ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) content, the time courses of changes in Rubisco and H(2)O(2) contents under UV-B treatment were measured in the primary leaves of two mung bean cultivars (Phaseolus raditus L. cv. Zhonglv-1 and Qindou-20). A 0.4 W/m(2) UV-B treatment caused a drop of the former to about 1/3 and a 30%-40% rise in H(2)O(2) content in a week in both cultivars but with Qindou-20 showing stronger responses. Such higher rate of increase in proteolytic activity under UV-B treatment implies that UV-B lowers Rubisco by activating proteolytic enzymes which hydrolizes Rubisco. Scavenging of H(2)O(2) by exogenously added ascorbic acid (AsA) significantly inhibited the UV-B-induced decrease in Rubisco amount and increases in H(2)O(2) level and proteolytic activity. It is shown that H(2)O(2) participates in the UV-B-induced reduction of Rubisco amount. It is suggested that H(2)O(2) stimulates the degradation of Rubisco by activation of proteolytic systems.

Experimental study of anti-tumor effects of polysaccharides from Angelica sinensis.

AIM: To investigate the in vivo anti-tumor effects of total polysaccharide (AP-0) isolated from Angelica sinensis (Oliv.) Diels (Danggui) on mice and the in vitro inhibitory effects of AP-0 and the sub-constituents (AP-1, AP-2 and AP-3) separated from AP-0 on invasion and metastasis of human hepatocellular carcinoma. METHODS: Three kinds of murine tumor models in vivo, sarcoma 180 (S180), leukemia L1210 and Ehrlich ascitic cancer (EAC) were employed to investigate the anti-tumor effects of AP-0. For each kind of tumor model, three experimental groups were respectively given AP-0 at doses of 30, 100 and 300 mg/kg by ip once a day for 10 days. Positive control groups were respectively given Cy at a dose of 30 mg/kg for S180 and leukemia L1210, and 5-FU at a dose of 20 mg/kg for EAC. On d 11, mice bearing S180 were sacrificed and the masses of tumors, spleens and thymus weighed. The average living days of mice bearing EAC and of mice bearing L1210 were observed, and the rates of life prolongation of each treatment were calculated, respectively. The inhibitory effects of APs on hepatoma invasion and metastasis in vitro were investigated by employing human hepatocellular carcinoma cell line (HHCC) with the Matrigel invasion chamber, adhesion to extracellular matrix and chemotatic migration tests, respectively. RESULTS: AP-0 had no obviously inhibitory effect on the growth of S180, but it could significantly decrease the thymus weights of the mice bearing S180. AP-0 could significantly reduce the production of ascitic liquids and prolong the life of mice bearing EAC. AP-0 could also increase the survival time of mice bearing L1210. AP-0 and AP-2 had significantly inhibitory effects on the invasion of HHCC into the Matrigel reconstituted basement membrane with the inhibitory rates of 56.4 % and 68.3 %, respectively. AP-0, AP-1, AP-2 and AP-3 could influence the adhesion of HHCC to extracellular matrix proteins (Matrigel and fibronectin) at different degrees, among them only AP-3 had significant blocking effect on the adhesion of HHCC to fibronectin with an inhibitory rate of 30.3 %. AP-0, AP-1 and AP-3 could partially inhibit the chemotactic migration abilities of HHCC. CONCLUSION: The experimental findings suggest that the total polysaccharide of Angelica sinensis (Oliv.) Diels (Chinese Danggui) possesses anti-tumor effects on experimental tumor models in vivo and inhibitory effects on invasion and metastasis of hepatocellular carcinoma cells in vitro.

Identification and expression analysis of a full-length cDNA encoding a Kandelia candel tonoplast intrinsic protein.

Soil salinity is an important issue, as most crop plants are low in salt tolerance. Salt tolerance, a complex, multifactorial, and multigenic process, has been known to be a quantitative trait. The identification of the salt stress responsive genes or salt tolerance genes is essential for the breeding programs. Most recent efforts have been focused on the products of structural genes (transport proteins, ion channels, enzymes of solute synthesis) while little attention were paid to the regulatory aspects of these proteins. Since the first aquaporin gene from plants was cloned and functionally expressed in 1993, there has been a growing interest in the molecular biology of MIPs (membrane intrinsic proteins) and their bearing on the biophysics of water flow across plant membranes. In the last decades, studies on Mangroves, a special kind of wood plants, grow in high-salt and flooding conditions have been concentrated almost exclusively on their physiological and ecological characteristics. Kandelia candel, one of the dominant species of mangroves along the Chinese coast, lacks salt glands or salt hairs used for removal of excess salt in other mangroves. This makes K. candel a perfect model to study the molecular mechanism of salt tolerance in mangrove plants. Using cDNA RDA, a cDNA-specific modification of genomic representational difference analysis, a series of salt responsive genes of Kandelia candel were cloned. Among these gene fragments, a 183 bp fragment (termed as SRGKC1) encoding a tonoplast intrinsic protein (TIP) in Kandelia candel (KCTIP1) was identified. Based on the sequence of SRGKC1, two gene specific primers were designed, and the 3' and 5' end of the KCTIP1 gene were obtained using the SMART RACE cDNA Amplification Kit. RACE products were purified from low-melting agarose, and sequenced directly with GSPs as the sequencing primers. A 500-bp fragment corresponding to the 3'end of this gene was obtained using the GSP1 primer, and a 690 bp fragment corresponding to the 5' end of this gene was obtained using the GSP2 primer. Two primers that flank the putative open reading frame (ORF) were designed to obtain the cDNA containing the complete ORF by RACE PCR reaction. The full-length cDNA of KCTIP1, containing a 756 bp open reading frame (ORF), was approximately 1.1 kb; the start codon was located at the nucleotides of 99-101 and stop codon at the nucleotides of 855-857 followed by a poly (A) tail. The KCTIP1 cDNA sequence in this research was released in GenBank with accession number AF521135. Using ExPASy Proteomics tools provided by EMBL, the isoelectric point and MWt of KCTIP1 are estimated as 5.77 and 26.3 kD respectively. Transmembrane prediction analysis revealed the deduced KCTIP1 protein sequence contains six transmembrane regions at amino acid residues of 20 - 42, 57 - 79, 86 - 108, 113 - 135, 142 - 164 and 217 - 239. Two highly conserved asparagine-proline-alanine (NPA) motifs were located at 85 - 87 and 199 - 201 amino acid residues respectively. KCTIP1 is also predicted to contain the Cys residue (Cys 118) that are shown to confer Hg-sensitivity in Arabidopsis gamma-TIP and delta-TIP. Similarity analysis showed that KCTIP1 shared 77% - 79% amino acid sequence identity with the TIPs from Vitis berlandieri, Brassica oleracea and Arabidopsis thaliana. Expression analyses indicated that KCTIP1 had different expression among species of Mangroves. Expressions of KCTIP1 in Kandelia candel, Rhizophora apoculata and Ceriops tagal were suppressed by salt, and were insensitive to salt stress in unknown species of Mangroves. Previous studied showed that salt conditions might result in large and rapid changes in extracellular water potential and serious disturbance to the cytoplasm. In order to compensate for this imbalance, the relative contribution of water channels to flow across the root could thus vary. K. candel is a species that is native to intertial zone of tropical and subtropical coast and is well-adapted to salt conditions. The coordinated down-regulation of aquaporins in this plant may decrease membrane water permeability and thus increase the cellular water conserva- tion during periods of salt stress. The results reported here are consistent with the postulated roles for tonoplast water channels in regulating the hydraulic permeability of the vacuolar membranes and in adjusting the water homeostasis of the protoplasm under various physiological conditions. The identification of KCTIP1 as one of salt-responsive genes implies that intracellular osmotic equilibration is a part of salt-tolerant mechanisms in Mangroves.

[The methods of identifying differences in mRNA expression].

Identifying changes in gene expression is of prime interest in molecular biology. A variety of methods have been developed for analyzing differentially expressed genes. Among these techniques, differential display reverse transcription PCR (DDRT-PCR), representational difference analysis (RDA) and suppression subtractive hybridization (SSH), which were based on differentially screening and subtractive hybridization technology, are applied most widely. Gene chips (gene microarrays) for analyzing gene expression patterns on larger-scale had also been developed. Here we present a review of these methods.
