Immunohistochemical expression of progesterone and estrogen receptors in meningiomas.

In this study we present results of investigations of progesterone and estrogen receptors in most frequent, WHO grade I histological types of meningiomas (meningothelial, fibrous, and transitional) and in atypical--WHO grade II variant of these tumors. Samples from 64 tumors were examined. The cohort consisted of 46 WHO grade I (21 transitional, 13 fibrous and 12 meningothelial histologic variants) and of 18 atypical meningiomas. Apart from immunohistochemical examination of progesterone and estrogen receptors, MIB 1 labeling index was estimated. Positive immunoreaction for progesterone receptors was found in 100% meningothelial, 95% transitional, 46% fibrous and 78% atypical variant of meningiomas. Intensity of immunoreaction was stronger in grade I than in grade II tumors. Immunoexpression of estrogen receptors was found in 48% of the investigated meningiomas. No correlation was stated between WHO grade I and grade II tumors, and between meningothelial, transitional and fibrous variants of the neoplasms.

Expression of vascular endothelial growth factor (VEGF) in vulvar squamous cancer and VIN.

Angiogenesis plays an important role both in progression of solid tumors and in metastasizing. An invasive growth of a neoplasm is mainly connected with appearing of blood vessels within a tumor. Inhibition of angiogenesis in solid neoplasms may deter both tumor growth and metastases. New treatment strategies based on suppressing of angiogenesis and selective damaging of neoplastic blood vessels may prove to be as efficient as those based on direct destruction of neoplastic cells. One of important angiogenic factors is vascular endothelial growth factor (VEGF), which is produced by neoplastic cells and shows high promitotic activity almost entirely for endothelial cells (paracrine activity). We decided to investigate VEGF expression in precancerous lesions as well as in squamous cancers of vulva. Our material included 31 cases of vulvar squamous cancer, 28 cases of VIN (vulvar intraepithelial neoplasia) III, 10 VIN II cases and 12 VIN I cases. A diagnosis was established according to WHO criteria on the ground of post-operative histopathological examination complemented with proliferation index estimated by the use of MIB-1 antibody. Immunohistochemical examinations were performed on paraffin-embedded material, using MIB-1 antibody (Immunotech), VEGF antibody (Santa Cruz), Goat serum Normal (DAKO), DAKO StreptAB-Complex/HRP Duet, Mouse/Rabbit DAKO DAB Chromogen Tablets, TBS (Sigma). Positive cytoplasmic expression of anti-VEGF polyclonal antibody (diffuse and/or focal and of various intensity) was observed in almost all samples from precancerous and cancerous lesions. The expression was especially strong and diffuse in all cancer cases; in cases of VIN it was mainly focal and weak.

Cystic nephroma in adults. A report of two cases and review of the literature.

We present two cases of cystic nephroma in a 55-year old and a 61-year old women. In both patients the results of ultrasound and clinical examinations were not characteristic enough to establish the precise preoperative diagnosis. Due to the age of the patients and the location of the lesions, possibility of clear cell carcinoma with cystic changes was considered. However, microscopic examination of postoperative specimens revealed benign nature of the tumors.

Complement factors C1q and C3b in brains with Creutzfeldt-Jakob disease.

Neuronal loss in Creutzfeldt-Jakob disease (CJD) is due to apoptosis and autophagy, however the apoptotic cell death may be related to the influence of cytokines. The role of complement system in induction of this process was recently postulated. In our study we examined the immunoreactivity for "early activation" complement factors C1q and C3b in the brains of the patients with CJD. We showed the positive immunoreactivity for C1q and C3b in neurons mainly in areas of spongiform change, in Purkinje cells, molecular layer and granular layer of cerebellum, and in prion protein positive plaques. The evident positive reactivity of two complement factors may suggest the role of complement system in neurodegeneration in CJD.

CD44 expression in human meningiomas: an immunohistochemical analysis.

CD44 molecules play an important role in cell adhesion, interaction between cells and extracellular matrix and cellular invasive growth potential. The aim of the study was to evaluate CD44 expression in atypical and benign meningiomas. The tumor specimens were obtained from 61 patients (27 males and 34 females; aged 19 - 78). The analysed material included 10 atypical meningiomas (G2), and following subtypes of benign meningiomas (G1): 21 transitional, 17 meningothelial and 13 fibroplastic. Paraffin tissue sections were immunostained using CD44 antibody (DAKO). The expression of CD44 was multifocal and diffuse. The strong expression was observed in atypical meningiomas, no expression in fibroplastic meningiomas and moderate expression in the other subtypes of benign meningiomas. Statistically significant differences were revealed between the expression of CD44 in G2 and G1 tumors. The results of the study support the thesis on the role of mutated CD44 molecule in the invasive growth potential of neoplastic cells.

[Neurilemmoma of the neck--case report].

Neurilemmoma is a mild tumor growing from the Schwann's cells of the external part of the nerve trunk. Authors, in their works describe a case of a 32-year-old man with a neck tumor on its right side. The tumor was identified as the neurilemmoma on the grounds of a postoperative histopathological examination. Thin-needle biopsy that was made twice during the history of the patient that lasted two years did not reveal the character of the tumor.

[Angiosarcoma of the temporal bone].

A case of 55-yrs old female was reported, in whom angiosarcoma of temporal bone had been diagnosed. A mode of treatment was surgery including partial petrosectomy and labyrinthectomy. Radiotherapy was not given because of a possibility of further cerebrospinal liquid leakage after dura mater rupture, which was seen during first operation. Local recurrence had developed about 2 months after surgery and a patient died 10 months after the time of diagnosis. It was stressed that angiosarcoma is a very rare neoplasm and in our case it was presented as few--symptomatic lesion despite of it's high extent in temporal bone. A review of literature concerning angiosarcoma was also reported.

The morphological analysis of vasculature and angiogenic potential in meningiomas: immunoexpression of CD31 and VEGF antibodies.

Meningiomas, despite being the mostly well-differentiated tumours, are quite often characterised by invasive growth and recurrences. As this fact may be caused by proangiogenic factors, we have investigated immunoexpression of CD31 and vascular endothelial growth factor (VEGF) in these tumours. The materials included 10 atypical meningiomas (G2), and the following subtypes of benign meningiomas (G1): 21 transitional, 17 meningothelial and 13 fibrous. Paraffin sections were immunostained with CD31 (DAKO) and VEGF (Santa Cruz) antibodies. The highest number of blood vessels was revealed in atypical meningiomas and much lower (statistically significant difference) in benign ones. Diffuse or focal expression of VEGF of various intensity was shown both in atypical and benign types of tumours with no statistically significant differences between both groups of tumours and between subtypes of benign meningiomas.

[Primary leptomeningeal sarcomatosis. Case report].

Primary intracranial sarcomas are very rare and aggressive tumors. A case of a 40-year-old woman with primary leptomeningeal sarcomatosis is presented. Initially her clinical symptoms resembled those of a pseudo tumor cerebri. Neuroimaging did not allow establishing a diagnosis of leptomeninges neoplastic infiltration. The patient died 8 months since the onset of symptoms. A neuropathological examination revealed diffuse thickening of leptomeninges. Microscopically a widespread sarcomatous infiltration of the subarachnoid space was shown, as well as a neoplastic infiltration of the choroid plexus and of some perivascular spaces in the cerebral cortex. On the grounds of a review of the literature diagnostic problems, differential diagnosis and clinical course of primary leptomeningeal sarcomatosis are discussed.
