A simple analysis system for the estimation of recombination efficiency using fluorescence-activated cell sorting.

A simple and accurate analysis system for the estimation of recombination efficiency in vivo using fluorescence-activated cell sorting (FACS) was designed and was subsequently used to compare the efficiency of recombination related to different spacer mutants. F(3) and F(5) mutant sequences were used for Flpe-mediated cassette exchange, and m2 and lox2272 mutant sequences were used for Cre-mediated cassette exchange due to their high incompatibility with wild-type sequences. The incompatibilities with wild-type were almost the same between mutant sequences. However, the recombination efficiencies were different. F(3) and m2 could mediate more efficient recombination than F(5) and lox2272, respectively. These results are consistent with the fact that the sequence of spacer region affects not only the reactivity upon wild-type sequence but also the recombination efficiency. It was also confirmed that the recombination process was mediated in a site-specific manner through PCR analysis using different sizes of exchange cassettes. In this experiment, the feasibility of the FACS analysis system for the estimation of recombination efficiency was verified. This system should be readily applicable for estimating recombination efficiency of other various mutant candidates, which will contribute to more precise and efficient site-specific recombination.

Optimized structured treatment interruption for HIV therapy and its performance analysis on controllability.

This paper presents dynamic programming therapy to reduce medication and establish long-term immune response against HIV-infection. Understanding HIV-related immune system control enables better HIV therapy without using full-treatments. Discrete regimen and continuous regimen characteristics are compared. Controllability of HIV-related immune system is analyzed for better understanding of optimal control in HIV therapy. Using optimal control provides more effective therapy than the full treatment without interruption in terms of controllability analysis. Case studies indicated the proposed therapy induces long-term nonprogression while preserving high CD4 T-helper cell count and low virus load in HIV-infected patients.

A survey of Enterobius vermicularis infection among children on western and southern coastal islands of the Republic of Korea.

To determine the status of Enterobius vermicularis infection among children living on western and southern coastal islands of the Republic of Korea, children (3-10 years) in 39 kindergartens and primary schools were examined using the cello-tape anal swab method, during July and August 2000. Of 1,661 children examined, 307 (18.5%) were found to be positive for E. vermicularis eggs. The highest infection rate (59.3%) was found in a kindergarten and a branch school of Shinyang primary school on Chujado, Jeju-do (Province). Remarkable differences in egg positive rates were observed in different localities. The egg positive rate for boys (21.3%) was significantly higher than that of girls (15.4%) (P = 0.02). However, positive rates were not significantly dependent on age. The results of the present study show that E. vermicularis infection is highly prevalent among pre-school and primary school children living on the western and southern coastal islands of the Republic of Korea, and indicate the need for efforts to control this infection.
