[Different expressions of bFGF gene in three kinds of skeletal muscles].

OBJECTIVE: To explore the distribution characteristics of bFGF gene, bFGF protein and receptor in rat skeletal muscles. METHODS: bFGF gene, and bFGF protein and receptor in rat gastrocnemius muscles were localized by ABC immunohistochemical method, in situ hybridization and a special staining methods. RESULTS: Based on the results from the special staining, the fibers in rat gastrocnemius muscle could be divided into three types: type I, II a and II b. The positive expression of bFGF gene was only found in the cytoplasm of type I and type II a fibers. The immunohistochemical localization showed that bFGF protein and its receptor were found in the connective parts of muscle endomysium in fibers of type I and type II a. CONCLUSION: This is the first study to identify the different expressions of bFGF gene, bFGF protein and its receptor in rat skeletal muscles. The results will help to understand the action and the relationship between bFGF autocrine and the embryo development of skeletal muscle fibers.

[Effect of exogenous p16 gene on the growth of wild-type p53 human lung adenocarcinoma cells].

OBJECTIVE: To study the effect of p16 gene on human adenocarcinoma cell lines and the posibility of exogenous p16 gene for gene therapy of lung cancer. METHODS: A p16 gene recombinant expression vector was constructed by cloning p16 cDNA into the pcDNA3 vector at the sites of BamH1 and Xho I, and was transferred by electroporation into p16 gene deficient human lung adenocarcinoma cell lines (A549 and H460) which expressed wild-type p53 gene. The p16 mRNA and protein expression of A549-p16 and H460-p16 cell was detected by Northern blots and immunocytochemistry, respectively. The cell growth properties and cell cycle pattern were aassessed by flow cytometry. RESULTS: The growth and colony forming efficiency of A549-p16 and H460-p16 cells were significantly inhibited compared with the controls. Flow cytometry showed A549-p16 and H460-p16 cells were arrested in G1 phase of cell cycle, and the percentage of cells in S phase and G2/M phase was decreased significantly. Tumorigenicity of the A549-p16 and H460-p16 cells in nude mice was greatly inhibited. CONCLUSION: The results show that human p16 gene can suppress the malignant phenotype of wild-type p53 human lung adenocarcinoma cell lines (A549 and H460). p16 gene can play a role in the gene therapy of human lung adenocarcinoma.

[Clinical observation on effect of yiqi huoxue decoction in comprehensive treatment on advanced stage of esophageal cancer].

OBJECTIVE: To observe the effect of Yiqi Huoxue (Replenishing Qi and activating blood circulation, YQHX) decoction in patients with advanced stage esophageal cancer treated with radiotherapy (RT) and intervention chemotherapy (ICT). METHODS: In comparing 31 patients who were treated with RT and ICT (Group A), and 31 patients treated with RT and ICT plus YQHX decoction (Group B). RESULTS: Immediate effective rate were 48.4% in Group A and 64.5% in Group B (P > 0.05). The function of bone marrow in Group B was obviously higher than that in Group A. The 1 year metastasis rate in Group A was higher than that in Group B, but the long-term survival rate of Group B was obviously higher than that in Group A (P < 0.05). CONCLUSION: YQHX decoction could reduce the inhibition of bone marrow caused by radiotherapy and intervention chemotherapy, lower the metastasis rate, prolong the long-term survival rate and improve the quality of life.

[Surgical treatment of the functional obstruction of eustachian tube of middle-aged and aging people].

OBJECTIVE: To expore the effect and the way of palatosapingeus tendon-shortening surgery to treat the functional obstruction of eustachian tube of middle-aged and aging people. METHOD: Seven cases of palatosapingeus tendon-shortening surgery were analyzed retrospectively. RESULT: After operations the symptoms of all cases were relieved in a week. The tympanogram returned to type "A". The audiogram showed that the airbone gap disappeared. The shape of tympanic membrance returned to normal state. None of them recured in one-three year following up. CONCLUSION: The effect of palatosapingeus tendon-shortening surgery is satisfactory so long as its surgical indication is correctly selected.

[Migration behavior of low-mass anions in capillary electrophoresis].

The dependence of effective mobility on ionic equivalent conductivity for 33 anions was studied using 5 mmol/L sodium chromate and 0.5 mmol/L tetradecyltrimethylammonium bromide (TTAB) as background electrolyte. The relationship was developed between effective mobility mueff and carbon number of mono-carboxylic acid and dicarboxylic acid homologues. The results showed that the effective mobility of anions increased with increasing ionic equivalent conductivity. The 1/mueff was linearly related to the carbon number of mono-carboxylic acid (C2-C7) and dicarboxylic acid(C2-C10). The correlation coefficient was more than 0.997.

The effects of the Rho-kinase inhibitor Y-27632 on arachidonic acid-, GTPgammaS-, and phorbol ester-induced Ca2+-sensitization of smooth muscle.

The effects of the Rho-kinase inhibitor, Y-27632 [1] on Ca2+-sensitization of force induced by arachidonic acid (AA), phorbol 12,13-dibutyrate (PDBu), GTPgammaS, and by the stable thromboxane analog, 9,11-dideoxy-9alpha,11alpha-methanoepoxy-PGF2alpha (U-46619), were determined in alpha-toxin-permeabilized smooth muscles. Y-27632 relaxed (up to 99%) Ca2+-sensitization by GTPgammaS (10 microM) and U46619 (1 microM), but not by PDBu (20 microM), and reduced GTPgammaS-induced myosin light chain (MLC20) phosphorylation from 28% to 17% (P=0.002). GTPgammaS-induced force sensitization was inhibited by Y-27632 more potently when the inhibitor was added during the plateau of force than prior to stimulation. In alpha-toxin-permeabilized smooth muscle, Y-27632 inhibited AA (50 microM)-induced Ca2+-sensitization of force (by 66 +/- 1.3%) and reduced MLC20 phosphorylation. In contrast, Y-27632 did not relax force Ca2+-sensitized by AA in smooth muscle permeabilized with Triton X-100. We conclude that (i) AA induces Ca2+-sensitization through dual mechanisms, one mediated by Rho-kinase (or a related kinase), and (ii) Rho-kinase is not required for phorbol ester-induced Ca2+-sensitization.

Randomised placebo-controlled trial of use of topical recombinant bovine basic fibroblast growth factor for second-degree burns.

BACKGROUND: Wound healing is a dynamic process that could be accelerated by growth factors. We investigated the effect of recombinant bovine basic fibroblast growth factor (rbFGF) on burn healing in a randomised placebo-controlled trial. METHODS: We recruited 600 patients with superficial or deep second-degree burns. Patients received 150 AU/cm2 daily topical rbFGF (n=300) or placebo (n=300) plus vehicle. We assessed healing by photography, punch-biopsy, and clinical examination. FINDINGS: All patients treated with rbFGF had faster granulation tissue formation and epidermal regeneration than those in the placebo group. Superficial and deep second-degree burns treated with rbFGF healed in a mean of 9.9 (SD 2.5) days and 17.0 (4.6) days, respectively, compared with 12.4 (2.7) and 21.2 (4.9) days (p=0.0008 and p=0.0003, respectively). No adverse effects were seen locally or systemically with rbFGF. INTERPRETATION: rbFGF effectively decreased healing time and improved healing quality. Clinical benefits would be shorter hospital stays and the patient's skin quickly becoming available for harvesting and grafting.

[DNA sequencing of human ciliary neurotropic factor gene by silver staining method].

OBJECTIVE: To introduce a non-radioactive protocol for DNA sequence analysis which employs a silver staining procedure METHODS: DNA template is amplified by Taq DNA polymerase to generate sequence ladder, No labeled nucleotide or primer is involved in the thermal-cycling sequencing reaction. A sensitive silver staining procedure is employed to visualize bands in sequencing gel. RESULTS: This silver staining method is used for DNA sequence analysis of human cilliary neurotrophic factor (hCNTF) gene. The resulted band resolution is comparable to radioactive sequencing method, and data can be obtain within less than 2 hours after sequencing reaction. CONCLUSION: Silver staining is a rapid, inexpensive and reliable method for DNA sequence analysis.

Ischemia and reperfusion impair the gene expression of endogenous basic fibroblast growth factor (bFGF) in rat skeletal muscles.

Our previous studies showed that the amount of endogenous basic fibroblast growth factor (bFGF) was reduced after ischemia and reperfusion insult. One of the mechanisms involved in the decrease of endogenous bFGF is the increased destruction of this growth factor associated with oxygen free radical activation and inflammation. We hypothesized that the wounding also impairs the secretion of bFGF and examined the bFGF gene expression in skeletal muscles after ischemia and reperfusion insult. In this study, a rat leg ischemia (4 h) and reperfusion (24 h) injury model was prepared and the in situ hybridization method and reverse transcriptase polymerase chain reaction technique (RT-PCR) were used to evaluate the bFGF gene expression and its localization in control (normal) and injured rat skeletal muscles. The results showed that the bFGF mRNA expression was localized in the cytoplasm in control skeletal muscle, especially at the periphery inside the cells. According to the intensity of the stain, four main classes of fibers could be identified: strongly, moderately, weakly, and negatively stained fibers. Based on the positive stain, about 82% of the total fibers examined were positive for bFGF mRNA stain. In ischemic or ischemic and reperfused rat skeletal muscles, the localization of bFGF mRNA expression was similar to that in normal skeletal muscles, but only 52% in ischemic muscles and 22% in ischemic and reperfused muscles had positive bFGF mRNA staining. RT-PCR confirmed a significant decrease in bFGF mRNA expression in ischemic and reperfused rat skeletal muscles. These results suggest that the acute ischemia and reperfusion not only induce the destruction of endogenous bFGF molecule, which is stored at the extracellular matrix of the fibers, but also downregulate the bFGF gene expression. The simultaneous dysregulation of endogenous bFGF gene expression and decreased synthesis of bFGF protein suggest a possible role of this growth factor in delayed wound healing.

Factors controlling activity of zirconia-titania for photocatalytic oxidation of ethylene.

Small photocatalytic devices were developed to remove ethylene from closed plant growth units flown in space. The devices utilized sol-gel-derived catalyst pellets of zirconia-titania. This study was undertaken to understand the significance of different factors on the photocatalytic activity of the catalyst. Increasing reaction temperatures and decreasing humidity of the air significantly increased oxidation of ethylene. The quantity of ethylene oxidized per unit time increased linearly with increasing flow rates, and increasing concentrations of ethylene. Zirconia-titania pellet size and heel depth had little effect on oxidation of ethylene. Platinizing the zirconia-titania significantly increased ethylene oxidation. The catalyst was found to absorb large quantities of water when the humidity of the air stream was elevated and this greatly decreased catalytic activity.

New imidazole alkaloids and zinc complexes from the micronesian sponge Leucetta cf. chagosensis.

Four new imidazole alkaloids, 2-5, along with the known isonaamidine B (1) were isolated from a Pacific sponge, Leucetta cf. chagosensis, collected from Yap, Federated States of Micronesia. Among these, 4 and 5 are zinc complexes derived from isonaamidine B and isonaamidine D. The structures of the new compounds were elucidated from spectral data. Isonaamidine D (3) showed weakly antifungal activity against Aspergillus niger with MIC = 100 micrograms/mL.

Agelasines H and I, 9-methyladenine-containing diterpenoids from an Agelas sponge.

Two new diterpenes possessing a 9-methyladeninium substituent, agelasines H (5) and I (6), along with five related known ones, 1-4 and 7, were isolated from a sponge, Agelas sp. The structures of the new compounds were determined from spectral data. Compounds 2-4, 6, and 7 exhibited varying degrees of antimicrobial activity.

New diketopiperazines from the sponge Dysidea chlorea.

Twelve new polychlorinated diketopiperazines (7-18), along with six known ones (1-6) and a known sterol (19), were isolated from the sponge Dysidea chlorea, collected from Yap, Federated States of Micronesia. The structures of dysamides I-T (7-18) were elucidated by spectroscopic methods, and one (8) was confirmed by chemical conversion. The stereochemistry of the dysamides is discussed.

New cyclic peptides from the ascidian Lissoclinum patella.

Four new cyclic peptides, patellamide G (2) and ulithiacyclamides E-G (3-5), along with the known patellamides A-C (6-8) and ulithiacyclamide B (9), were isolated from the ascidian Lissoclinum patella collected in Pohnpei, Federated States of Micronesia. The planar structures of these peptides were determined from 1D and 2D 1H and 13C NMR spectra. The absolute stereochemistries of the amino acid units, except for cysteine, were assigned by chiral GC analysis of N(O)-trifluoroacetyl isopropyl ester derivatives of amino acids obtained by acid hydrolysis of the intact and ozonized peptides. The structures of ulithiacyclamides E-G (3-5) were confirmed by chemical conversion. Patellamides B (7) and C (8) exhibited in vitro modulation of multidrug resistance in CEM/VBL100 cells.

Expression and immunolocalisation of the snowdrop lectin, GNA in transgenic rice plants.

Transgenic rice (Oryza sativa L.) plants generated through particle bombardment expressed high levels of an insecticidal protein (the snowdrop lectin, GNA) directed against sap-sucking insects. Engineered plants expressed GNA either constitutively or in a tissue specific manner, depending on the nature of the promoter used to drive expression of the gene. We used specific antibodies raised against GNA to localize its expression in phloem tissue in plants engineered with the rice sucrose synthase promoter driving GNA expression. We report here molecular, biochemical and immunological analyses for fifteen independently-derived transformants out of more than 200 plants we generated.

Intravenous nitroglycerin for rapid uterine relaxation.

BACKGROUND: To investigate the effect of nitroglycerin in vitro and in vivo on human uterine contractile activity. METHODS: In vitro myometrial strips were obtained from six pregnant women at term who underwent elective cesarean section. The biopsies were mounted in tissue baths. After spontaneous or oxytocin-induced activity had been accomplished, nitroglycerin in various concentrations was added to the baths and the effects were continuously registered. In vivo, in an open study nitroglycerin was administered as a bolus injection of 100-200 micrograms intravenously to 32 women at cesarean section when uterine relaxation was urgently needed; to 22 other women after vaginal delivery for facilitation of manual removal of retained placentas, and to one patient at vaginal delivery of premature twins. RESULTS: In vitro nitroglycerin induced a dose-dependent inhibition of spontaneous as well as oxytocin-induced myometrial contractile activity. Complete muscular relaxation was obtained at a concentration of 25-50 micrograms/ml. In vivo all patients had rapid effective uterine relaxation after intravenous injection of 100-200 micrograms nitroglycerin. CONCLUSION: Nitroglycerin administered intravenously seems to be a rapid and effective uterine muscle relaxant agent without overt adverse effects on mother or fetus.

Expression of snowdrop lectin (GNA) in transgenic rice plants confers resistance to rice brown planthopper.

Snowdrop lectin (Galanthus nivalis agglutinin; GNA) has been shown previously to be toxic towards rice brown planthopper (Nilaparvata lugens; BPH) when administered in artificial diet. BPH feeds by phloem abstraction, and causes 'hopper burn', as well as being an important virus vector. To evaluate the potential of the gna gene to confer resistance towards BPH, transgenic rice (Oryza sativa L.) plants were produced, containing the gna gene in constructs where its expression was driven by a phloem-specific promoter (from the rice sucrose synthase RSs1 gene) and by a constitutive promoter (from the maize ubiquitin ubi1 gene). PCR and Southern analyses on DNA from these plants confirmed their transgenic status, and that the transgenes were transmitted to progeny after self-fertilization. Western blot analyses revealed expression of GNA at levels of up to 2.0% of total protein in some of the transgenic plants. GNA expression driven by the RSs1 promoter was tissue-specific, as shown by immunohistochemical localization of the protein in the non-lignified vascular tissue of transgenic plants. Insect bioassays and feeding studies showed that GNA expressed in the transgenic rice plants decreased survival and overall fecundity (production of offspring) of the insects, retarded insect development, and had a deterrent effect on BPH feeding. gna is the first transgene to exhibit insecticidal activity towards sap-sucking insects in an important cereal crop plant.
