[The role of cell junction in hypothermic impairment of tissue].

OBJECTIVE: To investigate the role of cell junction in the hypothermal impairment of tissue without any cryoprotectant. METHODS: Epithelial MDCK cell line which is able to form well-defined cell junctions after confluency, and fibroblast V79 cell line which is unable to be used for graded freezing experiments, either in suspension, single tough attached to flasks or as monolayers on flasks. Then the viability was assessed by WST-1 and MT, respectively. RESULTS: The results of viability of the two types of cells assessed by WST-1 were well-fitted with that of MTT. The recovery of the two types of cells in suspension was higher than that of single tough group and monolayer group. The single tough group of MDCK had a higher recovery than did the monolayer group. At the same time, such tendency was not observed on V79 cells. As compared with V79 monolayers, lower recovery was found on MDCK monolayers. CONCLUSIONS: The junction between cells and the adhesion between cell and matrix have important roles in the hypothermic impairment of tissue. Their existence affect the recovery of cells under hypothermia. The cytoskeleton system may contribute to the impairment caused by junctions.

Alkali and alkaline-earth metal ketyl complexes: isolation, structural diversity, and hydrogenation/protonation reactions

The use of hexamethylphosphoric triamide (HMPA) as a stabilizing ligand allowed successful isolation of a series of structurally characterizable alkali metal and calcium ketyl complexes. Reaction of lithium and sodium with one equivalent of fluorenone and reaction of sodium with one equivalent of benzophenone in THF, followed by addition of two equivalents of HMPA, yielded the corresponding ketyl complexes 1, 2, and 11, respectively, as microketyl-bridged dimers. If one equivalent of HMPA was used in the reaction of sodium with fluorenone, a further aggregated complex, the mu3-ketyl-bridged tetramer 3, was isolated, whereas analogous reaction of benzophenone with sodium afforded the trimeric ketyl complex 13, rather than a simple benzophenone analogue of 3. In the reaction of potassium with fluorenone, the use of two equivalents of HMPA gave the tetramer 4, rather than a dimeric complex analogous to 1 or 2. Compared to the tetrameric sodium complex 3, there is an extra HMPA ligand that bridges two of the four K atoms in 4. When 0.5 equiv of HMPA was used in the above reaction, complex 5, a THF-bridged analogue of 4, was isolated. In the absence of HMPA, the reaction of sodium with an excess of fluorenone yielded the tetrameric ketyl complex 6, in which two of the four Na atoms are each terminally coordinated by a fluorenone ligand, and the other two Na atoms are coordinated by a THF ligand. Two bridging THF ligands are also observed in 6. Reaction of 1,2-bis(biphenyl-2,2'-diyl)ethane-1,2-diol (7) with two equivalents of LiN(SiMe3)2 or NaN(SiMe3)2 in the presence of four equivalents of HMPA easily afforded 1 or 2, respectively, via C-C bond cleavage of a 1,2-diolate intermediate. The reaction of calcium with two equivalents of fluorenone or benzophenone in the presence of HMPA gave the corresponding complexes that bear two independent ketyl ligands per metal ion. In the presence of 3 or four equivalents of HMPA, the fluorenone ketyl complex was isolated in a six-coordinate octahedral form (10), while the benzophenone ketyl complex was obtained as a five-coordinate trigonal bipyramid (13). The radical carbon atoms in both benzophenone ketyl and fluorenone ketyl complexes are still in an sp2-hybrid state. However, in contrast with the planar configuration of the whole fluorenone ketyl unit, the radical carbon atom in a benzophenone ketyl species is not coplanar with any of the phenyl groups; this explains why benzophenone ketyl is more reactive than fluorenone ketyl. Hydrolysis of 2 or 11 with 2N HCI yielded the corresponding pinacol-coupling product, while treatment of 2 or 11 with 2-propanol, followed by hydrolysis, gave the pairs fluorenone and fluorenol or benzophenone and benzhydrol, respectively. A possible mechanism for these reactions is proposed.

[Biomarkers in the studies on chemoprevention of colorectal cancer].

The present study was to investigate the chemopreventive effects of tea on colorectal cancer with a series of biomarkers. The results showed that the number of silver-stained nucleolar organizer dots per nucleus(AgNORs), labeling index(LI) of proliferating cell nuclear antigen(PCNA) of intestinal mucosa, and the expression of ras-p21 protein were significantly reduced in the tea-treated groups(P < 0.01) as compared with the positive control group. Furthermore, tea and tea pigments inhibited the expression of Bcl-2 protein and induced the expression of Bax protein(P < 0.05 or P < 0.01). The study provided evidence supporting that PCNA-LI, AgNORs, Bcl-2, Bax and ras-p21 protein could be used as effective biomarkers for colorectal carcinogenesis in human chemopreventive trials.

[Effects of tea on aberrant crypt foci and colorectal tumors in rats].

The present study was designed to investigate the chemopreventive effects of green tea and tea pigments (the main component of black tea) on 1,2-dimethylhydrazine (DMH)-induced rat colorectal carcinogenesis. Compared with the positive control group, green tea and tea pigments groups had less numbers of aberrant crypt foci (ACF) (P < 0.01) at the end of week 16. At the end of week 32, all rats in the positive control group developed colorectal tumors with an average of 2.6 tumors per rat, and a mean volume of 294.7 mm3 per tumor, while in the groups drinking green tea and tea pigments, the average numbers of colorectal tumors per rat were only 47.1% and 43.1% of controls respectively, and the mean tumor volume was inhibited by 77.1% and 68.1% respectively. It was concluded that tea pigments had a chemopreventive effect on colorectal tumor and ACF formation can be used as a useful intermediate end point to study chemopreventive effects on colorectal cancer.

Heparin-free hemodialysis in the treatment of hypernatremia in severely burned patients.

BACKGROUND: Hypernatremia in severely burned patients is associated with high morbidity and mortality rates. As the causes of hypernatremia in major burn patients are still not clear, hemodialysis is the method of choice for the treatment. While hemodialysis is effective for the control of hypernatremia, it can cause bleeding complications that may be fatal for burn patients with extensive wounds and potential gastro-intestinal mucosal damage. CLINICAL DATA: In the present study heparin-free hemodialysis in which the heparin is firmly absorbed to the haemofiltration membrane, hemophan, dispensed with systemic use of heparin. In two extensively burned patients with burn area of 100% TBSA and 98% TBSA respectively and hypernatremia with serum sodium concentration as high as 169 and 158 mmol/l respectively, heparin-free hemodialysis was performed five times and three times each. RESULTS: Hypernatremia was satisfactorily corrected with no interference to the coagulation system in the two patients as indicated by clinical observation and biochemical analysis. The patient with burn area of 98% TBSA survived and the patient with burn area of 100% TBSA died of wound coverage failure 6 weeks after injury because of non-availability of autograft. CONCLUSION: Heparin-free hemodialysis is an effective and safe method in the treatment of hypernatremia in extensively burned patients.

A multi-element ICP-MS survey method as an alternative to the heavy metals limit test for pharmaceutical materials.

A multi-element inductively coupled plasma-mass spectrometry (ICP-MS) survey method has been demonstrated as an alternative to the antiquated 'heavy metals limit test' prescribed by United States Pharmacopoeia (USP), European Pharmacopoeia (EP), and British Pharmacopoeia (BP), for drug substances, intermediates, and raw materials. The survey method is simple, fast, sensitive, semi-quantitative to quantitative, and includes all the elements which can be analyzed by atomic spectroscopy.

Antimicrobial peptides protect coho salmon from Vibrio anguillarum infections.

Fish losses from infectious diseases are a significant problem in aquaculture worldwide. Therefore, we investigated the ability of cationic antimicrobial peptides to protect against infection caused by the fish pathogen Vibrio anguillarum. To identify effective peptides for fish, the MICs of certain antimicrobial peptides against fish pathogens were determined in vitro. Two of the most effective antimicrobial peptides, CEME, a cecropin-melittin hybrid peptide, and pleurocidin amide, a C-terminally amidated form of the natural flounder peptide, were selected for in vivo studies. A single intraperitoneal injection of CEME did not affect mortality rates in juvenile coho salmon infected with V. anguillarum, the causative agent of vibriosis. Therefore, the peptides were delivered continuously using miniosmotic pumps placed in the peritoneal cavity. Twelve days after pump implantation, the fish received intraperitoneal injections of V. anguillarum at a dose that would kill 50 to 90% of the population. Fish receiving 200 microg of CEME per day survived longer and had significantly lower accumulated mortalities (13%) than the control groups (50 to 58%). Fish receiving pleurocidin amide at 250 microg per day also survived longer and had significantly lower accumulated mortalities (5%) than the control groups (67 to 75%). This clearly shows the potential for antimicrobial peptides to protect fish against infections and indicates that the strategy of overexpressing the peptides in transgenic fish may provide a method of decreasing bacterial disease problems.

Molecular cytogenetic analysis of intergeneric chromosomal translocations between wheat (Triticum aestivum L.) and Dasypyrum villosum arising from tissue culture.

Fluorescence in situ hybridization (FISH) was applied with total genomic DNA extracted from Dasypyrum villosum (L.) Candargy as a probe to characterize chromosome translocations arising from tissue culture in hybrids of Triticum aestivum x (T. durum - D. villosum, amphiploid). Chromosome translocations between wheat and D. villosum occurred in callus cells at an average frequency of 1.9%. Translocations existed not only in callus cells but also in regenerants. Three plants with translocation chromosomes were characterized among 66 regenerants of T. aestivum 'Chinese Spring' x 'TH1W' and 'NPFP' x 'TH1'. One of them proved to be a reciprocal translocation with an exchange of about one third of a wheat chromosome arm with about one half of a chromosome arm of D. villosum. The breakpoints of the other two translocations were located at, or near centromeres. The results are similar for both callus cells and regenerants and provide further evidence that translocations take place in tissue culture. Other structural chromosomal changes, for example, fragments, telocentrics, dicentromeres, and deletions, as well as numerical alterations including aneuploidy and polyploidy were recorded both in callus cells and regenerants.

[Isolation and verification of Triticum aestivum-Eremopyrum orientale addition lines and substitution lines by GISH].

Eremopyrum is a potential genus in wheat improvement. To breed T. aestivum-Er. orientale additional and substitutional lines, chromosome number of 96 BC2F3 individuals were accounted. Fifteen plants with 2n = 43 and eight plants with 2n = 44 were checked out. GISH results of the individuals with 43 chromosomes showed that they blonged to three different monosomic additions. Two disomic additions, one double-monosomic addition, one trible-monosomic addition and one monosomic addition were found in the 2n = 44 individuals. One double-monosomic substitution and one monosomic substitution were obtained. We also proved that it is relatively easier to obtain addition lines in the selfing population than to obtain double-monosomic additions.

[Estimation of biolistic transformation effect by transient expression of C1-R regulatory genes of anthocyanin biosynthesis].

In the present study, the transient expression of maize C1-R regulatory genes of anthocyanin biosynthesis in maize calli, wheat immature embryos, rice calli and tobacco leaves were described. C1-R regulatory genes could activate anthocyanin biosynthesis in vivo and the pigmented cells were visible. For major crops, including maize, wheat and rice, which were transformed mainly by biolistic method, C1-R genes were novel visible markers, and the expression was easier to detect than GUS gene. And these genes also worked well in transient expression in biolistic transformation system of tobacco leaves.

[Tissue culture induced translocation conferring powdery mildew resistance between wheat and Dasypyrum villosum and its marker-assisted selection].

Glutamate oxaloacetate transaminase (GOT) electrophoretic analyses were performed in 175 regenerants arising from immature embryos of crosses between wheat (Triticum aestivum L.) and 6D/6V substitution stocks. The GOT-V2 coding specific enzyme band was absent in two regenerants, designated 98R149 and 98R159 respectively, originated from cross of Yi 4095 and 6D/6V substitution stock c.v. RW15. Pm21 gene linked SCARs (Sequence Characterized Amplified Regions) analysis indicated that 6VS chromosome arms existed in 98R149 and 98R159. Fluorescence in situ hybridization with total genomic DNA extracted from Dasypyrum villosum (L.) as a probe confirmed the occurrence of translocation between 6V chromosome and a wheat one in the two regenerants mentioned above. 98R149 and 98R159 were immune to powdery mildew (Erysiphe graminisDC. f. sp. tritici) inoculation with mix races collected from Hebei Province. The results of the present paper added another feasible example of useful translocations via tissue culture.

[High efficient intergeneric chromosomal translocations between wheat (Triticum aestivum L.) and Dasypyrum villosum arising from tissue culture and irradiation].

Intergeneric chromosomal translocations were discernable both in callus cells and in regenerants arising from crosses between Triticum aestivum and T. durum-Dasypyrum villosum amphiploid c.v. TH1 and TH1W by means of fluorescence in situ hybridization. There were not only reciprocal translocations, but small fragment translocations. The results proved again the feasibility of creating intergeneric translocations via tissue culture. Irradiation facilitated numerical and structural chromosome changes in callus cells. The frequency of translocations was as high as 7.4 percent in irradiated callus cells. Callus age had an important impact on numerical and structural chromosome abnormalities. During a given time of culture, the frequency of unchanged cells was declined, while those cells with chromosome losses were inclined. The duration of culture had not significant effects on cells with chromosome gains. As structural chromosome changes is concerned, the duration of culture predominantly increased the frequency of cells with telocentric chromosomes. The chromosomal changes occurred at the initiation period of tissue culture. A number of cells which were doubled their chromosome numbers (2n = 84) were observed at a certain frequency in the period of tissue culture. These cells, however, disappeared in the successive culture.

[The genetic behavior of an Agropyron intermedium chromosome conferring BYDV resistance in wheat background].

Based on the chromosome origin identification of an Agropyron intermedium chromosome with BYDV resistance in Yi4212, a BYDV-resistant substitution line derived from the hybrids between 77-5433 and Zhong5, its ability compensating the lost wheat chromosomes and its transmission rate were investigated. The results indicated that the BYDV-resistant chromosomes originated from Zhong 5 could compensate the wheat chromosomes of homotologous group 2, 5 and 7. The chromosome conferring resistance to BYDV showed the intendance to substitute 2D rather than 2A, 2B wheat chromosomes in disomic substitution lines. In (77-5433/Zhong 5) selfing F2 population, 10 kinds of chromosome constitutions, including 9 expected and 1 unexpected kinds, and much chromosome number variation and little chromosome structure variation were observed. The transmission rate of the BYDV-resistant chromosome and the gametes carrying this chromosome were 56.3% and 33.0% respectively, both are lower than the expected ratio 75.0% and 50.0%. Some reasons were given to explain the related results, and chromosome in situ hybridization was an efficient and accurate way in studying the alien chromosome behavior in wheat background.

[Fluorescence in situ hybridization applied to the meiotic analysis and spontaneous chromosome translocation in the pollen mother cells of hybrids of Triticum-Haynaldia].

Fluorescence in situ hybridization was applied to the meiotic analysis and chromosome pairing in the pollen mother cells of hybrids of Triticum-Haynaldia. The results indicated that most (90.5%-93.2%) of the PMCs of two Triticum-Haynaldia hybrids contained two univalents involving one Haynaldia chromosome at metaphase I, and only 1 of 320 PMCs with homologous pairing between one wheat chromosome and one Haynaldia chromosome was observed in the hybrid of Guinong 22 x common wheat Chinese Spring, the frequency was 0.3%. The frequencies of the PMCs with mis-dividing chromosome ranged from 32.7% to 37.5% at anaphase I and from 20.5% to 24.4% at anaphase II in the hybrids between Triticum-Haynaldia. The frequency of Haynaldia chromatin translocating into Triticum chromosome was about 0.7% at anaphase I in the hybrid of Guinong 22 x common wheat Chinese Spring and was 0.8%-1.7% at anaphase II in two wheat-Haynaldia hybrids. Otherwise, the chromosome morphology involving Haynaldia chromatin was observed at preleptotene I, and the frequencies of chromatin nucleus at tetrad stage and at microspore stage were investigated. The time of chromosome translocation in meiosis and the transmitting frequencies of Haynaldia chromosome in wheat background were discussed.

Screening for mt-DNA mutations in optic neuritis of unknown cause.

OBJECTIVE: To investigate mitochondrial DNA (mt-DNA) mutations in optic neuritis of unknown cause (ONUC) and to assess the practical value of mt-DNA mutation detection in etiologically and differentially diagnosing ONUC. METHODS: Thirty patients with ONUC were screened for mt-DNA mutations of nt11778, nt3460 and nt15257 by using SSCP, mutation-specific primer PCR and sequencing. RESULTS: mt-DNA mutations were found in twelve of thirty ONUC patients. All of the mutations were at nt11778 position, but no one at nt3460 and nt15257. CONCLUSIONS: Forty percent (12/30) of ONUC patients were caused by an mt-DNA mutation. Combined with other routine measures, screening for mt-DNA mutations in ONUC patients is of great significance in diagnosing ONUC etiologically and differentially.

Comparison of computer-photoscreening with non-cycloplegic retinoscopy for amblyopiogenic risk factors in children.

OBJECTIVE: To determine the sensitivity and specificity of using a computer-photoscreener and non-cycloplegic retinoscopy in the detection of amblyopiogenic factors in nine to fifty months old infants and children. METHODS: Three hundred children, nine to fifty months old, were screened with the computer-photoscreener and non-cycloplegic retinoscopy. With a blinded standardized clinical assessment as the standard, an overall comparison of the sensitivity of and specificity results obtained with the two techniques was made. Photoscreen images on the computer monitor screen were reviewed and analyzed immediately by two independent observers for indicators of amblyopiogenic risk factors. Simultaneously, the results were compared to the findings of a full ophthalmologic examination. RESULTS: The computer-photoscreener revealed a sensitivity of 94.6% and specificity of 90.1%, and the non-cycloplegic retinocopy revealed a sensitivity of 85.7% and specificity of 81.0% for the detection of amblyopiogenic risk factors, including hyperopia (+2.75 D or more), myopia (-1.50 D or more), astigmatism (1.75 D or more), anisometropia (2.00 D or more), ocular misalignment (5 degrees or more), and media opacity (1.5 mm or more). CONCLUSIONS: The computer-photoscreener offers an opportunity to identify problems that limit vision, and could provide a feasible and sufficiently reliable screening technique in infants and preschool children to be screened successfully for amblyopiogenic risk factors.

[Periodontal regeneration by application of the porous beta-TCP/BMP artificial bone to Class II furcation defects].

OBJECTIVE: To study the regeneration of periodontal tissue after the application of beta-TCP/BMP artificial bone to class II furcation defects. METHODS: Periodontal defects were produced surgically around premolar teeth in 4 dogs. The flap surgery was performed and the root surfaces were curetted to remove the periodontal ligament and the cementum. A synthetic bioabsorble barrier manufactured from poly DL-lactide was used. The defects were divided into 3 groups (beta-TCP/BMP + PDLLA, PDLLA, control). 12 weeks after surgery, the animals were sacrificed and the teeth with their supporting periodontium were processed for light microscopic evaluation. RESULTS: The definitive evidences of regeneration were founded both in BMP/GTR group and GTR group, but the BMP/GTR group exhibited more new bone formation, cementum deposition occupying the majority of the furcation defects than the other two groups. CONCLUSION: It was concluded that the beta-TCP/BMP artificial bone can be used in GTR technique to promote periodontal regeneration.
