[Verrucous carcinoma (carcinoma cuniculatum)--clinicopathologic study on 10 cases].

10 cases of verrucous carcinoma occurring in different sites were studied clinicopathologically. Clinically, the tumor grew slowly and presented as an ulcerous or cauliflower-like mass. The tumor, existing in the skin, had openings like the sinus tracts on the surface from which foul, grey and greasy material oozed out when pressured. There was no exudation if the lesion grew in the mucosa. Histologically, the tumor was composed of mature and well-differentiated squamous cells showing papilloma-like growth upwards, their epidermal rete showing spheroid expansions downwards. Prickle cell masses invaded into the soft tissue in the deeper layer. In view of the tumors having location either in the skin or mucosa with or without effusion, similar histologic features and biologic behavior, we suggest that the verrucous carcinoma and carcinoma cuniculatum be two different variants of the disease. Finally, its diagnosis, differential diagnosis and treatment are discussed with a review of the literature.

[Analysis of trace elements by proton induced X-ray emission (PIXE) in serum of patients with liver cancer and normal subjects].

The levels of trace elements in serum of the patients with liver cancer and the normal subjects were determined by the PIXE technique. The significant increase of serum copper level (SCL) and the decrease of serum zinc level (SZL) in the patients with liver cancer, as compared to those of normal, were observed. Cu/Zn ratio in the patients with liver cancer was significantly higher than that of the normal (P less than 0.01). In the Cartesian coordinate graph of SCL + SZL, the liver cancer patients were separated from the normal by a line joining the intersection of abscissa and ordinate and the point of the sum of the mean value and the standard deviation of the Cu/Zn ratio of the normal subjects. The authors believe that the serum Cu/Zn ratio is likely a supplementary target in the diagnosis and prognosis of liver cancer.

Establishment of a model of transplantable myelocytic leukemia (L801) in LACA mice.

A transplantable myelocytic leukemia model of LACA mice, designated by the name of L801, was established by intravenous injection of spleen cell suspension from mice with radiation-induced myelocytic leukemia into mice of the same strain. Until now, for more than three years, the L801 has maintained stable and rapid growth and has been reproduced for over 130 serial passages. The incidence of leukemia in inoculated animals was approximately 100% and mean survival time was 10.9 +/- 2.1 days. The L801 is of myelocytic type which has been determined by cytological, cytochemical, pathological and ultrastructural observations. Its karyotype was hypodiploid, characterized by modal number of 39, loss of Y chromosome and an abnormal huge marker chromosome. The cell cycle duration of the L801 was 16 h. C-type viral particles were observed under the electron-microscope. The L801 was sensitive, to varying extents, to various anti-tumor agents. We presume that the L801 is a useful tool in studies on mechanism of leukemogenesis, anti-tumor agent screening and treatment of experimental tumors.

Site-directed isotope labeling and FTIR spectroscopy: assignment of tyrosine bands in the bR-->M difference spectrum of bacteriorhodopsin.

Fourier transform infrared difference spectroscopy has been used extensively to probe structural changes in bacteriorthodopsin and other retinal proteins. However, the absence of a general method to assign bands to individual chemical groups in a protein has limited the application of this technique. While site-directed mutagenesis has been successful in special cases for such assignments, in general, this approach induces perturbations in the structure and function of the protein, thereby preventing unambiguous band assignments. A new approach has recently been reported (Sonar et al., Nature Struct. Biol. 1 (1994) 512-517) which involves cell-free expression of bacteriorhodopsin and site-directed isotope labeling (SDIL). We have now used this method to re-examine bands assigned in the bR-->M difference spectrum to tyrosine residues. Our results show that out of 11 tyrosines in bR, only Tyr 185 is structurally active. This work further demonstrates the power of SDIL and FTIR to probe conformational changes at the level of individual amino acid residues in proteins.

Dimorphic fungus characteristic of fumonisin-producing strains of Fusarium moniliforme from Zhejiang.

Fusarium moniliforme and its fumonisins have been shown to be carcinogenic in lab animals and have been linked to high incidences of human esophageal cancer. In this study we report the dimorphic fungus characteristic of fumonisin-producing strains of F. moniliforme from foodstuffs in Zhejiang, China. All of the twenty strains of F. moniliforme shown produce fumonisin B1 475.9-6322.2 micrograms/g in corn medium. These strains of F. moniliforme form yeast-like colonies in Sabouraud's agar plates contained 9% NaCl at 37 degrees C incubator and shows mostly budding reproduction. In blood agar plates these strains of F. moniliforme appear grass-green haemolytic reactions. This is the first report that yeast-like growth, dimorphic pathogenic fungus feature is found in F. moniliforme. These results suggest that it is also important to program epidemiological surveys of F. moniliforme as a primary pathogenic fungus, while proceeding to produce mycotoxins of F. moniliforme in food hygiene.
