The role of the cytoplasmic domains of individual subunits of the acetylcholine receptor in 43 kDa protein-induced clustering in COS cells.

The 43 kDa protein, a cytoplasmic peripheral membrane protein, is closely associated with the acetylcholine receptor (AChR) at the neuromuscular junction, where it is thought to anchor the receptor in the postsynaptic membrane. We have used the 43 kDa protein-induced clustering of AChRs that occurs when both proteins are transiently expressed in COS cells to investigate which parts of the AChR might interact with the 43 kDa protein. By constructing chimeric subunits, we showed that the cytoplasmic domains of neither the epsilon nor delta subunits are required for 43 kDa protein-induced clustering. Systematic mutational analysis of the long cytoplasmic loops of the alpha and beta subunits showed that most of the loops can be altered without affecting the ability of the AChR to be clustered; in each case, however, one or more sequences could not be tested, because mutation in these regions prevented AChR assembly. Our results suggest either that these regions are involved in clustering or that the 43 kDa protein can interact with multiple, alternative sites on the cytoplasmic surface of the AChR. Our experiments also show that the postulated sites of tyrosine phosphorylation in the beta subunit and of serine phosphorylation in the alpha subunit can be mutated without affecting 43 kDa protein-induced AChR clustering.

Appearance of new receptive fields in rat dorsal horn neurons following noxious stimulation of skeletal muscle: a model for referral of muscle pain?

To test the hypothesis that painful stimuli to skeletal muscle lead to a widespread unmasking of synaptic connections in dorsal horn neurons, intramuscular injections of bradykinin (BKN) were made outside the receptive fields (RFs) of these cells in the rat. Following BKN injections, new RFs all of which were located in the deep tissues and had high mechanical thresholds appeared in 9 out of 21 (42.8%) nociceptive dorsal horn neurons which originally had a single RF in deep tissues or in the skin. The appearance of new RFs may lead to a mislocation of the source of pain if in fact the impulse activity of a nociceptive dorsal horn neuron contains information on the site of the stimulus.

Differential effects of cutaneous and deep application of inflammatory irritant on mechanoreceptive field properties of trigeminal brain stem nociceptive neurons.

1. The aim of this study was to determine whether there are differential neuroplastic effects of cutaneous and deep stimuli on the mechanoreceptive field (RF) properties of nociceptive brain stem neurons recorded in trigeminal (V) subnucleus caudalis. 2. A total of 31 caudalis neurons were functionally identified in anesthetized rats as nociceptive neurons receiving convergent cutaneous and deep afferent inputs, and the effects of the small-fiber excitant and inflammatory irritant mustard oil applied to either tongue muscle or facial skin were tested on their cutaneous and deep RFs. 3. The expression of neuroplasticity on the cutaneous and deep RFs of the neurons was found to depend on the site of application of this irritant. Mustard oil injected into the tongue muscle produced a significant and reversible expansion of both the cutaneous and deep RFs. In contrast, application of mustard oil to facial skin resulted in a significant and reversible expansion of the cutaneous RF, but no deep RF expansion occurred. 4. These findings suggest that considerable functional neuroplasticity of the cutaneous and deep RF properties of V nociceptive neurons can be induced by a peripheral inflammatory irritant and that deep inputs are especially effective in evoking these neuroplastic changes.

The properties of an HBV surface antigen protein carrying the binding site for the receptor of hepatocytes--its formation of surface antigen particles and secretion from discrete cell lines.

A human hepatitis B virus (HBV) gene, which encodes the major surface antigen protein (S protein) carrying the hepatocyte receptor-binding site, was constructed with site-directed mutagenesis and in vitro recombination. When expressed in monkey kidney cell line COS-M6, this gene product (S309 protein) formed surface antigen (HBsAg) particles and secreted from the cells. It was stable within the cells and in the culture medium and could be immunoprecipitated with antisera directed against plasma-derived HBsAg or synthetic preS1 polypeptide. Isopycnic CsCl gradient centrifugation showed that the density of S309 protein particles (1.25 g/ml) was slightly higher than that of S protein particles. The S309 protein was readily secretable from hepatoma cell lines, and the amount secreted was comparable to that of the S protein. By contrast, only about 10% of the S309 protein was secreted from COS-M6 cells, and its appearance in culture medium was delayed. The efficiency of the secretion of the S309 protein can be improved when it is coexpressed with the S protein.

Effects of a novel piperazine derivative (CGP 29030A) on nociceptive dorsal horn neurons in the rat.

In anaesthetized rats and cats, the effects of the piperazine derivative CGP 29030A on the discharges of functionally identified dorsal horn neurons and gamma-motoneurons was studied. The compound exhibited a marked inhibitory action on presumably nociceptive dorsal horn neurons that processed input from high-threshold mechanosensitive receptors in the skin and deep somatic tissues (muscle, tendon, joint). Significant effects were obtained at a dose of 10 mg/kg p.o. and higher; the onset of action was fast (approximately 10 min). In contrast to presumably nociceptive dorsal horn neurons, cells having input from low-threshold mechanosensitive receptors in the skin and deep tissues were not affected by the compound. Thus CGP 29030A has a quite specific action on dorsal horn neurons that probably mediate pain. Systemic effects on the blood pressure and on the activity of sympathetic efferent fibres did not occur. The lack of effect of CGP 29030A on nociceptive neurons in spinalized animals suggests that the site of action is not the segmental spinal cord but the supraspinal CNS. gamma-motoneurons supplying the gastrocnemius-soleus muscle were likewise inhibited by the compound but at a higher dose (equal to and exceeding 30 mg/kg). The data show that CGP 29030A is a compound with marked and specific analgesic action. As it also inhibits gamma-motoneurons, CGP 29030A may be of benefit in the treatment of painful disorders which are complicated by increased motor activity (cramp, spasm).

An HBV large surface antigen protein which can be secreted from mammalian cells.

The N-terminal 54 base pairs (encoding amino acid residues 2-19) within the preS1 region of the human hepatitis B virus surface antigen gene were deleted by site-directed mutagenesis. Unlike the wild type large surface antigen protein, when this mutated gene was expressed in monkey kidney cell line COS-M6, the protein product (S301 protein) could be secreted from the cells. Moreover, the inhibition of the secretion of the major surface antigen protein by this altered large surface antigen protein was greatly reduced, suggesting that the deleted region contained a retention sequence which prevented the secretion of the large surface antigen. However, the coexpression of the major S protein was essential for the secretion of the S301 protein. When coexpressed, the secretion of these two proteins was synchronous. Like the wild type large surface antigen protein, the S301 protein could be translocated into endoplasmic reticulum and glycosylated after its synthesis in COS cells. The S301 protein was thermostable and proteinase-resistant. It also retained the antigenicity of the large S and major S proteins. Given the fact that the S301 protein is readily secretable, stable and identical to the large S protein in terms of their antigenicity, it may be developed into a new generation of recombinant vaccine for the prevention of viral hepatitis.

Extracellular domains mediating epsilon subunit interactions of muscle acetylcholine receptor.

Ligand-gated ion channels, a major class of cell-surface proteins, have a pseudosymmetric structure with five highly homologous subunits arranged around a central ion pore. The correct assembly of each channel, whose subunit composition varies with cell type and stage of development, requires specific recognition between the subunits. Assembly of the pentameric form of the acetylcholine receptor from adult muscle (AChR; alpha 2 beta epsilon delta) proceeds by a stepwise pathway starting with the formation of the heterodimers, alpha epsilon and alpha delta. The heterodimers than associate with the beta subunit and with each other to form the complete receptor. We have now determined which parts of the subunits mediate the interactions during assembly of the adult form of the receptor from mouse muscle by using a chimaeric subunit in which the N-terminal and C-terminal extracellular domains are derived from the epsilon subunit with the remainder from the beta subunit. The epsilon and beta subunits were chosen because the epsilon subunit forms a heterodimer with the alpha subunit in the pathway for assembly of the receptor, whereas the beta subunit does not. The epsilon beta chimera can substitute for the epsilon but not the beta subunit in the oligomeric receptor, indicating that the alpha subunit specifically recognizes an extracellular domain of the epsilon subunit.

The C-terminal half of the preS1 region is essential for the secretion of human hepatitis B virus large S protein devoid of the N-terminal retention sequence.

A large surface protein of human hepatitis B virus was expressed from a mutated S gene in which both the reported retention sequence (aa 2-19) and the C-terminal half of the preS1 region (aa 66-117) were deleted. This retention sequence-free protein was not secreted from the monkey kidney cell line COS-M6 in transient expression assays. When this large S protein was overexpressed in the presence of the major S protein, the secretion of the latter was severely inhibited. Moreover, the presence of S protein in large abundance did not facilitate the secretion of the mutated large S protein at all, indicating that it was subject to more profound retention than the wild-type large S protein. The findings that, like the wild-type large S protein, this altered protein retained some of the properties of its C-terminal S domain, and that the preS1 region was still on its surface suggested that there was no extensive alteration of the polypeptide folding. The interpretation is favored that the C-terminal half of the preS1 region plays a crucial role in the secretion of the large S protein when its N-terminal retention sequence is not present.

The effects of intracerebroventricular injection of naloxone, phentolamine and methysergide on the transmission of nociceptive signals in rat dorsal horn neurons with convergent cutaneous-deep input.

In anaesthetized rats, recordings were made from nociceptive dorsal horn neurons with convergent input from the skin and deep somatic tissues. The results of a previous study have shown that in these neurons the input from deep nociceptors is subjected to a much stronger tonic descending inhibition than is the input from cutaneous nociceptors. The aim of the present study was to find out whether at supraspinal levels opioidergic, adrenergic, or serotoninergic transmitters are involved in this quite specific inhibition of deep nociception. Injections of naloxone, phentolamine, and methysergide into the third ventricle showed that only naloxone is capable of abolishing the tonic inhibition of the deep nociceptive input to spinal neurons. The input from cutaneous nociceptors to the same cells was largely unaffected by naloxone. Thus the effects of intracerebroventricular injection of naloxone resembled those obtained with a spinal cold block in a previous study; with the exception that the increase in background activity--which is prominent during cold block--was missing after the injection of naloxone. The present results demonstrate that the tonic descending inhibition of the deep nociception operates with opioidergic synapses at the supraspinal level. In contrast, supraspinal adrenergic and serotoninergic mechanisms do not appear to contribute to the tonic inhibition. The data confirm and extend previous results which suggested that a particular portion of the descending antinociceptive system may act mainly on the input from deep nociceptors. Pharmacologically, this particular portion seems to be opioidergic in nature.

Assembly of the mammalian muscle acetylcholine receptor in transfected COS cells.

We have investigated the mechanisms of assembly and transport to the cell surface of the mouse muscle nicotinic acetylcholine receptor (AChR) in transiently transfected COS cells. In cells transfected with all four subunit cDNAs, AChR was expressed on the surface with properties resembling those seen in mouse muscle cells (Gu, Y., A. F. Franco, Jr., P.D. Gardner, J. B. Lansman, J. R. Forsayeth, and Z. W. Hall. 1990. Neuron. 5:147-157). When incomplete combinations of AChR subunits were expressed, surface binding of 125I-alpha-bungarotoxin was not detected except in the case of alpha beta gamma which expressed less than 15% of that seen with all four subunits. Immunoprecipitation and sucrose gradient sedimentation experiments showed that in cells expressing pairs of subunits, alpha delta and alpha gamma heterodimers were formed, but alpha beta was not. When three subunits were expressed, alpha delta beta and alpha gamma beta complexes were formed. Variation of the ratios of the four subunit cDNAs used in the transfection mixture showed that surface AChR expression was decreased by high concentrations of delta or gamma cDNAs in a mutually competitive manner. High expression of delta or gamma subunits also each inhibited formation of a heterodimer with alpha and the other subunit. These results are consistent with a defined pathway for AChR assembly in which alpha delta and alpha gamma heterodimers are formed first, followed by association with the beta subunit and with each other to form the complete AChR.

Expression from herpesvirus promoters does not relieve the intron requirement for cytoplasmic accumulation of human beta-globin mRNA.

Expression plasmids were constructed in which the human beta-globin gene or a variant of it precisely lacking its two introns was transcribed from its own promoter, the herpes simplex virus type 1 thymidine kinase (HSV-tk) promoter, or the immediate early promoter of human cytomegalovirus (CMV-IE). Forty two hours after transfection of these plasmids into monkey kidney cells, nuclear and cytoplasmic RNA were isolated. Quantitative S1 nuclease mapping and primer extension analysis were used to determine the relative abundances, cellular locations, and leader sizes of the accumulated beta-globin RNAs. Whereas transcripts of all of the intron-containing genes accumulated in the cytoplasm to high levels, transcripts of their cDNA variants were neither stably maintained in the nucleus nor accumulated in the cytoplasm, irrespective of the promoter from which transcription was driven. We conclude that the intron requirement for cytoplasmic accumulation of beta-globin RNA can not be circumvented by synthesis from either the promoter of the intronless HSV-tk gene or the CMV-IE promoter.

[Determination of lead and cadmium concentration in children's toys].

Soluble lead and cadmium in 24 kinds of children's toys were determined by atomic absorption spectrophotometer. The results showed that the concentration of soluble lead and cadmium in the toys which had a paint coating were respectively from 110.3 mg/kg to 5,156.0 mg/kg and from 0.34 mg/kg to 35.50 mg/kg, but soluble lead and cadmium concentration in the toys which did not have a paint coating were very low. This study suggested that the children's exposure to lead and cadmium was mainly from the paint coating of toys, and that the important problem was too high concentration soluble lead in the colouring and varnishing substances. In preventing children's exposure to low lead and cadmium level, the hazards from toys containing soluble lead should be paid attention.

Somatotopical arrangement of rat spinal dorsal horn cells processing input from deep tissues.

Extracellular recordings were made from 29 dorsal horn neurones which could be driven by mechanical stimulation of deep tissues, in spinal segment L4 of rats. The recording sites were marked by iontophoresis of pontamine sky blue. According to the location of the receptive fields the cells were classified as deep neurones (12 cells), which had input from deep tissues only, and as cutaneo-deep neurones (17 cells), which had receptive fields in both skin and deep tissues. The deep cells showed a correlation between the recording sites in the dorsal horn and the location of the receptive fields on the hind-limb: the more distal a deep receptive field was situated on the hind-limb the more medial was the location of the recording site in the dorsal horn. In cutaneo-deep neurones the correlation was statistically significant for the deep receptive fields but not for the cutaneous ones.

Response properties and descending control of rat dorsal horn neurons with deep receptive fields.

The study was designed to obtain information on the spinal processing of input from receptors in deep somatic tissues (muscle, tendon, joint). In anaesthetized rats, the impulse activity of single dorsal horn cells was recorded extracellularly. In a pilot series, the proportion of neurons responding to mechanical stimulation of deep tissues was determined: 46.7% had receptive fields in the skin only, 35.5% could only be driven from deep tissues (deep cells), and 17.7% possessed a convergent input from both skin and deep tissues (cutaneous-deep cells). In each category, neurons with low and high mechanical thresholds were encountered. Experiments employing a reversible cold block of the spinal cord showed that deep cells with high threshold were subject to a stronger descending inhibition than low-threshold deep cells. In cutaneous-deep neurons the input combination high-threshold cutaneous and high-threshold deep was the most frequent one (48.7% of the cutaneous-deep cells). In these presumably nociceptive cells the descending inhibition had a differential action in that the input from deep tissues was more strongly affected than was the cutaneous input to the same neuron. The recording sites of the neurons with deep input were located in the superficial dorsal horn and in and around lamina V. The results suggest that in the rat a considerable proportion of dorsal horn cells receives input from deep nociceptors and that this input is controlled by descending pathways in a rather selective way.

[Relation of left ventricular compliance and heart size].

53 patients with myocardiopathy were studied to determine the left ventricular compliance (LVC) with apexcardiography and the heart size with radiography. Relation between LVC and heart size was analysed by using linear correlation. In cardiomyopathy LVC decreased and correlated with the size of heart. It was concluded that the size of heart was one of factors which influenced LVC. The decrease of LVC may be related with the length of myocyte, the geometric shape of heart and histopathologic alteration.

[Dynamic changes in depolarization of action potentials by high frequency stimulation in guinea pig papillary muscles].

The effect of high frequency stimulation on rate-dependent depression of depolarization of action potentials was studied with computer and standard microelectrode techniques in guinea pig papillary muscles. After increasing driving frequency from 0.5 to 6 Hz, decreases in Vmax, APA and OS were found. Changes of upstroke velocity during phase 0 between driving frequency 6 and 0.5 Hz could be described as a parabola. The rate-dependent depression of depolarization was exaggerated by tetrodotoxin (TTX), particularly during the accelerating period of phase 0. A progressive decline in Vmax, which could be fitted by a power function curve, was observed after changing driving frequency from 1 to 5 Hz. TTX significantly influenced the decline in Vmax and the regression coefficients in fitted equations.
