A frequent Y chromosome b2/b3 subdeletion shows strong association with male infertility in Han-Chinese population.

BACKGROUND: Azoospermia factor c (AZFc) subdeletions were reported to be significant risk factors for spermatogenesis. In this study, we assessed the occurrence of classical AZF deletions and AZFc subdeletions and their impact on male infertility in a Han-Chinese population. METHODS: This study analysed a population of 699 subjects, including 451 idiopathic infertile patients with a range of fertility disorders and 248 fertile controls, using a retrospective design. Deletions were identified by multiplex PCR. RESULTS: The prevalence and phenotypes of the classical AZF deletions were similar to previous studies. Subdeletions of the AZFc region in patients showed similar overall frequencies in all sperm concentration categories of gr/gr (7.0%) and b2/b3 (8.9%). For controls, these subdeletions were also found with a prevalence of gr/gr (7.7%) and b2/b3 (3.2%). b1/b3 deletions were not found either in the patients or in the controls. CONCLUSION: Our data showed a higher frequency of deletion events in this Han-Chinese population than in populations elsewhere in the world. The classical AZF deletions were the primary genetic factors for spermatogenic failure, while no significant association was found for AZFc subdeletions with sperm concentration. However, the b2/b3 subdeletion was significantly associated with idiopathic male infertility (odds ratio, 2.93; 95% confidence interval 1.34-6.39) (P = 0.005), indicating a potential impairment of male fertility.

Theoretical limit of the minimal magnetization switching field and the optimal field pulse for Stoner particles.

The theoretical limit of the minimal magnetization switching field and the optimal field pulse design for uniaxial Stoner particles are investigated. Two results are obtained. One is the existence of a theoretical limit of the smallest magnetic field out of all possible designs. It is shown that the limit is proportional to the damping constant in the weak damping regime and approaches the Stoner-Wohlfarth (SW) limit at large damping. For a realistic damping constant, this limit is more than 10 times smaller than that of so-called precessional magnetization reversal under a noncollinear static field. The other is on the optimal field pulse design: if the magnitude of a magnetic field does not change, but its direction can vary during a reversal process, there is an optimal design that gives the shortest switching time. The switching time depends on the field magnitude, damping constant, and magnetic anisotropy.

Detection of human chromosomal abnormalities using a new technique combining 4',6-diamidino-2-phenyl-indole staining and image analysis.

Chromosomal abnormalities are associated with a variety of diseases. We have developed a new technique for detecting chromosomal abnormalities, and the technique combines conventional 4',6-diamidino-2-phenyl-indole staining (DAPI) with image analysis. The image analysis consists of two simple steps: deconvolution and three-dimensional reconstruction. The technique has been reported for analyzing plant chromosomes but has not been applied to analyze human chromosomes yet. To test the technique, we analyzed five translocations: 46,XX,t(3;21)(12;18), 46,XX,t(11;22), 46,XY,t(7;22), 46,XY,t(11;18), and 46,XY,t(3;7). The results showed that the karyotype of the 46,XX,t(3;21)(12;18) was 46,XX,t(3;21)(q11.1;p13),t(12;18) (q21.2;q23), and the karyotypes of the 46,XX,t(11;22), 46,XY,t(7;22), 46,XY,t(11;18), and 46,XY,t(3;7) were 46,XX,t(11;22)(q23;q12.1); 46,XY,t(7;22)(q32;q13.2); 46,XY,t(11;18)(q13.3;q23), and 46,XY,t(3;7)(q22.1;p13), respectively. The identity of derivative chromosomes involved in the translocations was verified by chromosome painting as well as FISH analyses with centromere probes. The new technique has two advantages: the procedure is simple and convenient, and the results are accurate. The technique has the potential to be used in cytogenetic studies and clinical diagnosis of human diseases in the future.

Increase of glutathione in mine population of Sedum alfredii: a Zn hyperaccumulator and Pb accumulator.

Phytochelatins (PCs) have been induced in a large range of plant species, but their role in heavy metal tolerance is unclear. Sedum alfredii is a new zinc (Zn) hyperaccumulator and lead (Pb) accumulator found in an old Pb/Zn mine in the Zhejiang Province of China. Until now, the mechanisms of its hyperaccumulation/accumulation and tolerance were poorly understood. The aim of this work was to investigate whether PCs were differentially produced in mine populations of S. alfredii compared with a non-mine control of the same species. The results showed that plants from the mine site were more tolerant to increasing Zn and Pb concentrations than those from the control site. No PCs and cysteine (Cys) were detected by pre-column derivatization with HPLC fluorescence in any tissues of two populations at any treatment, which in turn indicated they were not responsible for Zn and Pb tolerance in the mine population. Instead, Zn and Pb treatments resulted in the increase of glutathione (GSH) for both populations in a tissue-dependent manner. Significant increases were observed in leaf, stem and root tissues of plants grown on the mine site. The results suggest that GSH, rather man PCs, may be involved in Zn and Pb transport, hyperaccumulation/accumulation and tolerance in mine population of S. alfredii.

Effects of interactions between cadmium and zinc on phytochelatin and glutathione production in wheat (Triticum aestivum L.).

It has been proposed that phytochelatins (PCs) act as a biomarker for the evaluation of metal toxicity. Little attention has been paid to the effects on metal combinations and glutathione (GSH), the most abundant cellular thiol. In the present study the effects of interactions between cadmium (Cd) and zinc (Zn) on PC and GSH production were examined in wheat tissue over 14 days' exposure. The results showed that the presence of Zn alleviated Cd toxicity, accompanied by a reduction of Cd uptake. Cd and Zn exposure increased PC-SH levels in concentration-, tissue- and time-dependent manners. Of the two metals, Cd was more effective than Zn in PC-SH production. Interactions of Cd and Zn with respect to PC-SH production may be synergistic or inhibitory, strongly depending on duration of exposure and concentration of the metal combinations. Cd also stimulated GSH production in concentration-, tissue- and time-dependent manners, whereas Zn had no significant effects on GSH levels. Compared to the presence of Cd alone, the presence of Zn reduced GSH levels in a tissue-dependent manner over the growth period. The results of the study suggest that metal interactions should be highly considered in the application of PCs and GSH as potential biomarkers for the evaluation of metal toxicity, as most metal-polluted natural environments are contaminated with more than one metal.

Effects of exogenous organic chelators on phytochelatins production and its relationship with cadmium toxicity in wheat (Triticum aestivum L.) under cadmium stress.

Phytochelatins (PCs) have been proposed as a potential biomarker for metal toxicity. In this study, cadmium (Cd) toxicity, PCs production and their relationship in wheat under Cd stress were examined using various exogenous organic chelator-buffered nutrient solutions. Single Cd stress produced strong toxic effects, as indicated by decreases of growth parameters, high level of lipid peroxidation in leaf and overproduction of PCs in root. Exogenous organic chelators with proper dose more or less reduced Cd toxicity by increasing growth parameters and decreasing lipid peroxidation in leaves. Of organic chelators (EDTA, DTPA, citric acid, malic acid and oxalic acid), EDTA was the most effective in decreasing Cd toxicity in plants, followed by DTPA and citric acid. Simultaneously, the concentrations of Cd-induced PCs in roots decreased, and the greatest decrease was caused by application of EDTA and DTPA. Linearly positive relationships were observed between Cd toxicity and root PCs concentrations under the influences of organic chelators, particularly EDTA, DTPA and citric acid. Furthermore, present results provide stronger evidence that PCs synthesis in plant cells was related to free Cd ion concentrations, not total Cd, and demonstrate that the levels of PCs production in plants correlated well with toxic effects caused by the bioavailable Cd levels.

Retrograde cerebral perfusion of oxygenated, compacted red blood cells attenuates brain damage after hypothermia circulation arrest of rat.

BACKGROUND: It was proved that higher haematocrit (Hct) might improve the function of brain after hypothermia circulation arrest (HCA). In the present study we established a new rat HCA model and investigated whether retrograde cerebral perfusion of oxygenated, compacted red blood cells (RBC) could attenuate brain injury after HCA. METHODS: A new rat HCA model was developed and rats were randomly distributed into three groups: HCA group, HCA combined with retrograde cerebral perfusion of oxygenated, compacted red blood cell group (HCArcp group), and sham operation group (sham op. group). Animals both in the HCA group and in the HCArcp group underwent HCA 90 min at 18 degrees C. Brain damage after HCA was evaluated with light microscopy and electron microscopy. Immunohistochemistry and RT-PCR techniques were used to measured the different expressions of the C-Fos, Bcl-2, Bax mRNA and protein among the groups. Additionally we measured the wet/dry ratio of the brain in order to evaluate the oedema degree after HCA. RESULTS: The new HCA model of rat we developed was comparable to the clinical setting not only in terms of the intubation, anaesthesia method and materials employed but also in terms of the priming volume in relation to body weight. The number of injured neurones in the hippocampus CA1 and parietal cortex, but not in the thalamus of the HCA group, was significantly greater than that of the HCArcp group (P<0.05). The mean score of mitochondrion of the hippocampus CA1 in the HCA group was significantly higher than in the HCArcp group (P<0.05). The expression of C-Fos, Bax mRNA and protein in the hippocampus CA1 and/or parietal cortex area was higher in the HCA group than in the HCArcp group (P<0.05). Expression of the mRNA and protein of Bcl-2 was higher in the HCArcp group than in the HCA group (P<0.05). The degree of oedema after HCA between the HCA group and HCArcp group had no significant difference (P>0.05). CONCLUSIONS: We established a new rat model of HCA comparable to the clinical setting. Retrograde cerebral perfusion of oxygenated, compacted RBC is a simple, effective, and safe method to protect the brain during HCA. Adjusting the gene expression in relation to apoptosis might contribute to the neuroprotective effects of a retrograde cerebral infusion of oxygenated, compacted RBC.

Effects of water-soluble fractions of diesel oil on the antioxidant defenses of the goldfish, Carassius auratus.

Larval goldfish Carassius auratus were exposed to 0.05 and 0.1 mg/L No. 20 diesel oil, widely used as transportation power in China, for 2, 4, 7, 10, 15, 25, and 40 days, while one control group was designated for each exposure group. Some fish after 25 days of exposure were transferred to diluted water until the 40th day. Hepatic antioxidant defense parameters of fish, including contents of reduced glutathione and glutathione disulfide and activities of superoxide dismutase, catalase, glutathione reductase, selenium-dependent glutathione peroxidase, and glutathione S-transferase, were determined and compared to control values. All the results indicated that the antioxidant responses of the fish to the two concentrations of oil exposure were similar on the whole. The possible defense mechanisms of fish and prospective early biomarkers for the evaluation of an oil-dominant contaminated aquatic ecosystem are discussed. In addition, after fish were removed from oil exposure, the recovery status of these antioxidant indices was explored.

Study on the relation between occupational fenvalerate exposure and spermatozoa DNA damage of pesticide factory workers.

AIMS: To determine sperm nuclear DNA integrity and to investigate the relation between fenvalerate (FE) exposure and spermatozoa DNA damage. METHODS: Sperm DNA fragmentation was detected by a modified alkaline single cell gel electrophoresis (Comet) assay and a terminal deoxynucleotidyl transferase mediated dUTP nick end labelling (TUNEL) assay. The olive tail moment (OTM) and percentage tail DNA were measured by the Comet assay, and cell positive percentage was measured by the TUNEL assay for DNA damage evaluation. RESULTS: The DNA integrity of spermatozoa of external and internal control groups were both significantly greater than that of the FE exposed group. The median value of tail DNA percentage in the exposure group was 11.30, which was significantly higher than 5.60 in the internal control group and 5.10 in the external control group. The median value of OTM was 3.80 in the exposure group, significantly higher than 1.50 in the internal control group and 2.00 in the external control group. Mean cell positive was 31.2% in the exposure group, significantly higher than 17.4% in the internal control and 19.6% in the external control groups. Cell positive (%) was significantly correlated with tail DNA percentage and with OTM of whole subjects (n = 63). CONCLUSIONS: Results showed that occupational FE exposure is associated with an increase in sperm DNA damage. A combination of the Comet and TUNEL assays would offer more comprehensive information for a better understanding of sperm DNA damage, and the biological significance of sperm DNA damage in sperm function and male infertility.

Molecular cytogenetic characterization of a familial balanced reciprocal translocation t(11;18) (q13.3; q23) associated with pregnancy wastage.

A new male patient associated with a pregnancy wastage was detected in China. Cytogenetic analyses including G-banding, chromosome painting and observation of synaptonemal complexes (SCs) demonstrated that the pregnancy wastage was associated with a balanced reciprocal translocation t(11;18) (q13.3; q23). The proband was the carrier of the translocation and his karyotype was 46,XY,t(11;18)(11pter-->11q13.3:: 18q23-->18qter; 18pter-->18q23::11q13.3-->11qter). The pedigree was analyzed based on a G-banded karyotype of the nine familial members. The translocation chromosomes came from the proband's mother. The result of the SC observation in the proband showed that each of the spermatocytes displayed one quadrivalent during their pachytene stages. In the quadrivalents, there existed homologous and nonhomologous synapses and the latter occurred widely during early, middle and late pachytene stages. The reasons and genetic basis of the pregnancy wastage are discussed.

A capsule review of recent studies on the application of mass spectrometry in the analysis of Chinese medicinal herbs.

Chinese herbal medicine is gaining increasing popularity worldwide as an alternative approach to the development of pharmaceuticals in therapeutic applications. Chemical characterization and compositional analysis of Chinese medicines provide the necessary scientific basis for the discovery and development of new drugs of natural origin. Applications of mass spectrometry in the analysis of Chinese herbal medicines have been growing rapidly in recent years owing to the rapid technical advances and increasing availability of the instrumentation. This paper reviews the current status of how different mass spectrometric techniques are being used to support research studies of Chinese medicines. The focus is on crude herbal medicines and their derived products. The review is not meant to be exhaustive, but rather to provide a general overview of the various research activities in this rapidly expanding field. In the discussion of specific herbs, the emphasis is placed on ginseng and Danshen, two of the herbs for which active experimental work is on-going in the authors' laboratories. Other selected herbs will be discussed only briefly, aiming primarily to illustrate the current status of research in the area.

Metallic phase in quantum Hall systems due to inter-Landau-band mixing.

The electronic eigenstates of a quantum Hall (QH) system are chiral states. Strong inter-Landau-band mixings among these states can occur when the bandwidth is comparable to the spacing of two adjacent Landau bands. We show that mixing of localized states with opposite chirality can delocalize electronic states. Based on numerical results, we propose the existence of a metallic phase between two adjacent QH phases and between a QH phase and the insulating phase. This result is consistent with nonscaling behaviors observed in recent experiments on a quantum Hall liquid-to-insulator transition.

Cancer mortality among workers exposed to amphibole-free chrysotile asbestos.

The issue of whether exposure to chrysotile asbestos alone, without contamination from amphibole asbestos, causes lung cancer and mesothelioma was investigated in a 25-year longitudinal study (1972-1996) in Chongqin, China. The study cohort comprised 515 male asbestos plant workers exposed to chrysotile only; the control cohort included 650 non-dust-exposed workers. The results of analysis in which the proportional hazards model was used indicated that mortality due to all causes, all cancers, and lung cancer was related to asbestos exposure; the relative risks, adjusted for age and smoking, were 2.9, 4.3, and 6.6, respectively. Fiber concentrations in the raw material section and the textile section of the plant were 7.6 and 4.5 fibers/ml, respectively. Because of differences between the study and control plants, the authors also compared various sections of the asbestos plant that had different levels of dust exposure. The adjusted relative risk of lung cancer was 8.1 for workers exposed to high versus low levels of asbestos. Two cases of malignant mesothelioma, one pleural and the other peritoneal, were found in the asbestos cohort. These results suggest that heavy exposure to pure chrysotile asbestos alone, with negligible amphibole contamination, can cause lung cancer and malignant mesothelioma in exposed workers.

Nonclassical behavior in the capacitance of a nanojunction.

The capacitance of a nanojunction formed by a scanning tunneling microscope (STM) tip and a two-dimensional gold cluster was measured through the single electron tunneling spectroscopy of a double-barrier tunnel junction. By decreasing the STM tip-cluster separation, it was observed that the capacitance first increases and then decreases at short separation. This characteristic clearly deviates from the classical behavior and provides evidence for potential quantum effects on the capacitance.

Predicted maximal oxygen uptake in normal Hong Kong Chinese schoolchildren and those with respiratory diseases.

To assess the maximum oxygen uptake (V'O2 max) of Hong Kong Chinese children and to explore its association with respiratory illnesses, we conducted the Multistage Fitness Test (MFT), a 20-m shuttle run test, in 1,427 schoolchildren aged between 8-12 years. Information on respiratory symptoms in the previous year, cumulative respiratory illnesses, and habitual physical activities were collected. Spirometry was carried out to derive forced expiratory volume in 1 sec (FEV1). The V'O2max was 30.3 mL x kg( - 1) x min( - 1) for boys, and 28.6-mL x kg( - 1) x min( - 1) for girls; these values were low by Western standards. V'O2max was significantly reduced in children with asthma ( - 1.3 mL x kg( - 1) x min( - 1)) and bronchitis ( - 0.7.mL x kg( - 1) x min( - 1)) when adjusted for other covariates. A significant decrease in FEV1 was also observed in these children. The correlation between V'O2max and FEV1 was significant but weak. Habitual physical activity was an independent predictor of V'O2max, but not of FEV1. The low V'O2 max in Hong Kong children may be explained in part by ethnic differences and possibly a low level of physical activity.

Optical rubbery ormosils sensor for the detection of ammonia.

Rubbery ormosil films with immobilized aminofluorescein (AF) were investigated to develop an optochemical sensor for the determination of ammonia in water. The gel precursors with tetramethoxysilane (TMOS) and dimethyldimethoxysilane (DiMeDMOS) were deposited on glass supports, and characterized in terms of response to pH, and to dissolved ammonia at constant pH. After preconditioning the sensing film was stable for 6 months. The detection limit for ammonia in water was 0.2 microg mL(-1) (S/N 2), the response being linearly dependent on concentration in the range of 0.5 to 80 microg mL(-1) ammonia. The response time was less than 5 min. The effects of sodium chloride concentration, temperature, and coexisting metal ions and compounds were investigated.
