[Experiment research on moxibustion treating infected EHFV rat].

In this experiment, we established a rat model of EHFV infected by abdominal cavity, and observed the effect of moxibustion on the changes of antigens, antibodies, neural transmitters in blood and tissue on 60 weanling Wistar rats with EHF. The rats were divided into three groups: normal control group (N. group), EHF control group (C. group) and EHF with moxibustion group (M. group) at random. Moxibustion was given at "Shenshu" for 30 minutes daily, 7 days. The 14th day after infection of EHFV, blood and lungs of the rats were fetched. The results obtained are as follows: 1. The effect value of EHF antibodies dilution: in C. group, it is 1:267 +/- 82 (n = 8), in M.group, it is 1:336 +/- 176 (n = 10), there is a significantly different of specific antibodies between C.group and M.group. The results show that moxibustion had an effect of enhancing the function of immune system in rats with EHF. 2. The examination rate of EHFV antigens in lungs, in C.group, it is 62.5% (n = 8), in M.group, it is 49.4% (n = 16). The results show that moxibustion may enhance the immune function and the effect of eliminating virus of lymphocytes in lungs. 3. The value of BUN in serum: in N.group, it is 15.3 +/- 2.50 (n = 10), in C.group, it is 24.63 +/- 7.24 (n = 10), and in M.group, it is 16.36 +/- 4.59 (n = 26). There is a significantly different between C.group and N.group or M.group (P less than 0.001). There is no significantly different between N.group and M.group (P greater than 0.05).(ABSTRACT TRUNCATED AT 250 WORDS)

[Studies on the toxicity of a new synthetic drug SC1001-aminum].

SC1001A is a new sedative, hypnotic and anti-epileptic drug. Screening for possible mutagenicity of the agent consisted of a battery scheme of short-term mutagenic tests with different hereditary detecting end points. Three assays were with in the scheme: Ames test (for detecting gene mutation in vitro), micronucleus test (for detecting chromosomal aberration in vivo) and CHL (Chinese hamster lung cell line) test (for detecting chromosomal aberration in vitro). In addition, a routine teratogenicity study on SC1001A was carried out in mice. They were given daily at 3-dose levels (exposed to up to 33% of the LD50 by gastric incubation from the 6th through the 15th day of gestation. SC1001A gave uniformly negative results in all three mutagenic assay systems. Results of the teratogenic experiment showed that only the pregnant rate in bred mice in the moderate dose group and the mean maternal body weight gain of the dams during gestation in the low dose group were significantly lower than those in the corresponding control group. A clear dose-response relationship was not demonstrated. All dose levels of SC1001A used caused no adverse effects on the number of survival fetuses per litter, growth or development of the fetal mice. No malformations in the external appearance, of the internal organs and of the skeletal systems in fetal mice were observed. The above-mentioned results indicated that SC1001A induced neither gene mutation nor chromosomal damage both in vitro and in vivo. There was no evidence of teratogenesis. Therefore, it is estimated that SC1001A is probably comparatively safe as a pharmaceutical product for human beings in usual dosage.

[The anti-hemorrhagic shock of acupuncture on neiguan and its effect on the cardial pump function and the blood viscosity].

The purpose of this paper is to study and analyse the effects of the cardial pump function and the blood viscosity on rabbits with hemorrhagic shock by puncturing Neiguan. In the experiment 30 rabbits were divided into the electroacupuncture (EA) group and the control group, each group was 15 rabbits. The cardial pump function, the blood pressure and the blood viscosity were recorded during normalize, shock, and EA. The results were as following. 1. The blood pressure of the rabbits which were in hemorrhagic shock was raised by needling Neiguan. MAP was raised from 43 +/- 8 to 87 +/- 15 mmHg (p less than 0.001). 2. The cardial pump function were reinforced by needling Neiguan. SV was increased from 0.41 +/- 0.46 (before EA) to 0.73 +/- 0.12 ml (p less than 0.0001). CO, SI, CI. and WL were improved in a certain degree with difference significant. In the EA group the each index of the cardial pump function was higher than that of the control group. There was statistical significant between the two groups. 3. Needling Neiguan, the blood viscosity was tending to normality. The blood plasma viscosity was increased. There was significant difference (p less than 0.05). These results suggest that needling Neiguan may protect the cardial pump function, raise the blood pressure, play a positive role during anti hemorrhagic shock.

[The effect of acupuncture on the changes of ANP and AII in plasma of rabbits with hemorrhagic shock].

In this work, we measured the changes of atrial natriuretic polypeptide, (ANP) and angiotensin (AII) in plasma of normal and hemorrhagic shock rabbits by the radioimmunoassay (PIA). Meanwhile, we observed the effect of acupuncture on the changes of ANP and AII in plasma of hemorrhagic shock rabbits. The results showed that the contents of ANP and AII in plasma increased when the hemorrhagic shock extended. Acupunctured at "Neiguan" can anti-shock by increasing blood pressure and can decrease the ANP and AII in plasma. Shock may promote the secretion and reduce the disintegration of ANP. Acupuncture can anti-shock by increasing blood pressure and correcting the disturbance of secretion and metabolism of ANP and AII during shock.

Physical growth of Chinese children from birth to 6 years old in urban and rural areas in 1985.

During 1985 a survey of the physical growth of 175,290 Chinese children less than 7 years in age was undertaken in the urban and rural areas of ten provinces. The results of weight, height, and head circumference are similar to those generally found as regards the differences between boys and girls. The children in rural areas were lighter and shorter. From the age of 3-6 months onwards the weights and heights of all groups were less than international standards. Nutrition is probably the major factor in these differences in growth.

Two novel cell surface antigens on small cell lung carcinoma defined by mouse monoclonal antibodies NE-25 and PE-35.

Two mouse monoclonal antibodies, NE-25 and PE-35, defining novel cell surface antigens of small cell lung carcinoma (SCLC) were produced. The molecular weight of NE-25 and PE-35 antigens estimated by radioimmunoprecipitation was 25,000 and 35,000, respectively. NE-25 antigen was expressed on the majority of cell lines and tumor specimens of SCLC among lung carcinoma. These NE-25-positive cell lines showed typical growth morphology as SCLC classic lines and expressed high levels of neuroendocrine biomarkers, such as aromatic L-amino acid decarboxylase, while NE-25 antigen-negative lines lacked apparent neuroendocrine properties. This antigen was expressed also on a subset of neoplastic cells with (neuro)endocrine properties, including pulmonary carcinoid, and on various tumors of nervous tissues, such as neuroblastoma. Among the normal cells, Kulchitski cells of lung, thyroid gland, adrenal gland, Langerhans islet, and nervous tissues were positive. Thus, the expression of NE-25 antigen is closely associated with the neural and/or (neuro)endocrine differentiation state. On the contrary, PE-35 antigen was present on four major types of lung carcinomas as well as on squamous cell carcinoma and adenocarcinomas of various tissues, but it was absent from nervous tissue tumors. Thus, PE-35 antibody showed a "pan-epithelial" reactivity. Analysis by NE-25 and PE-35 antibodies provided evidence for the heterogeneities of SCLC by demonstrating four surface phenotypes, with the NE-25+/PE-35+ phenotype being most common. In addition, the results supported the current understanding that various histological types of lung carcinoma, including SCLC, are derived from a stem cell of the bronchial epithelium.

Rapid and genomic biological responses are mediated by different shapes of the agonist steroid hormone, 1alpha,25(OH)2vitamin D3.

The hormone 1alpha,25(OH)2vitamin D3 (1,25-D) produces biological responses via both genomic and rapid mechanisms. The genomic responses are linked to a nuclear receptor, while the rapid responses are believed to utilize other signal transduction pathways that are likely linked to a putative cell membrane receptor for 1,25-D. The natural seco-steroid, 1,25-D, is capable of facile rotation about its 6,7 single carbon bond to permit generation of a continuum of potential ligand shapes extending from the 6-s-cis (6C) to the 6-s-trans (6T). To identify the shape of the conformer(s) that can serve as agonists for the genomic and rapid responses, we synthesized two families of analogs that were locked in either the 6T or 6C conformation. We found that 6T-locked analogs were inactive or significantly less active than 1,25-D in both rapid responses (transcaltachia or the rapid stimulation of intestinal Ca2+ absorption in perfused chick intestine, stimulation of whole cell chloride currents in osteoblastic ROS 17/2.8 cells, and stimulation of phosphorylation of mitogen-activated protein kinase in promyelocytic NB4 leukemic cells) and in genomic responses (induction of osteocalcin in human MG-63 osteoblastic cells). For genomic responses, the 6C-locked analogs bound poorly to the nuclear receptor and were much less potent than 1,25-D. In contrast, the 6C-locked analogs were potent agonists of the three rapid responses studied and had activities equivalent to 1,25-D. These results demonstrate that the signal transduction pathways that support rapid and genomic responses can discriminate between different shapes of the conformationally flexible 1,25-D.

Daily torpor and energy expenditure in Sminthopsis macroura: interactions between food and water availability and temperature.

Endothermy allows maintenance of a constant internal thermal milieu for optimal physiological functions but results in high energy expenditure and water loss. Since torpor can reduce both expenditure of energy and loss of water and thus reduces food and water requirements, we determined how different food and water regimes affect torpor occurrence and patterns in the dasyurid marsupial Sminthopsis macroura at ambient temperatures of 18 degrees C (well below the thermoneutral zone) and 28 degrees C (close to the thermoneutral zone). At 18 degrees C, torpor was more frequent and more pronounced than at 28 degrees C. Withdrawal of food reduced the average daily metabolic rate by 20%; withdrawal of water alone had little effect. The average daily metabolic rate of individuals displaying torpor was 20% lower than that of individuals remaining normothermic. At 28 degrees C, the average daily metabolic rate under food restriction was 85% of that with food available ad lib. However, this reduction of average daily metabolic rate at 28 degrees C seems due not to the use of torpor but mainly to a reduction of the resting metabolic rate. The results suggest that at low temperature, torpor is used to adjust energy expenditure to availability of food, whereas at high temperature, reductions of resting metabolic rate are employed.

Changes of superoxide dismutase, glutathione perioxidase and lipid peroxides in the brain of mice preconditioned by hypoxia.

We have developed an animal model of hypoxic preconditioning and assumed that oxygen radicals and their endogenous scavenging enzymes may play an important role in the preconditioning. To test this hypothesis, activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and the content of lipid peroxides (LPO) were measured during the preconditioning. Compared with unpreconditioned control animals, in animals exposed to hypoxia only once, the activities of SOD and GSH-Px in whole brain were found to be significantly decreased while the LPO content in the hippocampus significantly increased. However, those in animals exposed to 4 runs of hypoxia tended to return to control levels and were lower than those in animals exposed to 1 and 2 runs of hypoxia. Our results indicate that the oxygen radicals and their specific scavenging enzymes seem to be involved in the development of tolerance to hypoxia.
