Comparison of the mutagenic properties of 8-oxo-7,8-dihydro-2'-deoxyadenosine and 8-oxo-7,8-dihydro-2'-deoxyguanosine DNA lesions in mammalian cells.

The comparative mutagenicity of 8-oxo-7,8-dihydro-2'-deoxyadenosine (8-oxodA) and 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxodG) was explored using simian kidney (COS-7) cells. Oligodeoxynucleotides ¿5'-TCCTCCT- G(1)X(2)CCTCTC or 5'-TCCTCCTX(1)G(2)CCTCTC (X = dA, dG, 8-oxodA or 8-oxodG) containing 8-oxodA or 8-oxodG positioned within codon 60 or 61 of the non-coding strand of human c-Ha-ras1 gene were inserted into a single-stranded phagemid shuttle vector. The vector was replicated in COS-7 cells and the progeny plasmids were used to transform Escherichia coli DH10B. The transformants were analyzed by oligodeoxynucleotide hybridization and DNA sequence analysis to establish the mutation frequency and specificity. When 8-oxodA was positioned at X(1), targeted A(oxo)-->C transversions were detected; the mutation frequency was 1.2%. When 8-oxodA was positioned at X(2), one targeted mutant among 416 colonies screened (an A(oxo)-->G transition) was detected. Thus, the mutation frequency and spectrum of 8-oxodA depend on the sequence context of the lesion. The mutation frequency of 8-oxodG at X(1) and X(2) was 5.2 and 6.8%, respectively. G(oxo)-->T transversions dominated the spectrum, accompanied by small numbers of G(oxo)-->A transitions and G(oxo)-->C transversions. We conclude that 8-oxodA has mutagenic potential in mammalian cells, generating A-->C transversions. However, when tested under similar conditions, the mutation frequency of 8-oxodA is at least four times lower than that of 8-oxodG.

Role of reactive oxygen species in acetylcholine-induced preconditioning in cardiomyocytes.

We examined the ability of ACh to mimic ischemic preconditioning in cardiomyocytes and the role of ATP-sensitive potassium (KATP) channels and mitochondrial reactive oxygen species (ROS) in mediating this effect. Chick embryonic ventricular myocytes were studied in a flow-through chamber while flow rate, pH, PO2, and PCO2 were controlled. Cell viability was quantified with propidium iodide (5 microM), and production of ROS was measured using 2', 7'-dichlorofluorescin diacetate. Data were expressed as means +/- SE. Preconditioning with 10 min of ischemia followed by 10 min of reoxygenation or 10 min of ACh (1 mM) followed by a drug-free period before 1 h of ischemia and 3 h of reoxygenation reduced cell death to the same extent [preconditioning 19 +/- 2% (n = 6, P < 0.05) ACh 21 +/- 5% (n = 6, P < 0.05) vs controls 42 +/- 5% (n = 9)]. Like preconditioning, ACh increased ROS production threefold before ischemia [0.60 +/- 0.16 (n = 7, P < 0.05) vs. controls, 0.16 +/- 0. 03 (n = 6); arbitrary units]. Protection and increased ROS production during ACh preconditioning were abolished with 5-hydroxydecanoate (5-HD, 100 microM), a selective mitochondrial K(ATP) channel antagonist, and the thiol reductant 2-mercaptopropionyl glycine (2-MPG, 1 mM), an antioxidant [cell death: 5-HD+ACh 37 +/- 7% (n = 5), 2-MPG+ACh 47 +/- 6% (n = 6); ROS signals: 5-HD+ACh 0.09 +/- 0.03 (n = 5), 2-MPG+ACh 0.01 +/- 0.04 (n = 4)]. In addition, ACh-induced ROS signaling was blocked by the mitochondrial site III electron transport inhibitor myxothiazol (0.02 +/- 0.07, n = 5). These results demonstrate that activation of mitochondrial K(ATP) channels and increased ROS production from mitochondria are important intracellular signals that participate in ACh-induced preconditioning in cardiomyocytes.

Efficacy of recombinant methioninase in combination with cisplatin on human colon tumors in nude mice.

The present treatment of colon cancer is based on 5-fluorouracil (5-FU). Despite promising results of combining leucovorin or levamisole with 5-FU, the 5-year survival rate of patients with advanced colon cancer has not increased significantly. Colon tumors in vitro have been shown previously to have an elevated requirement for methionine, suggesting a new therapeutic target. In this study, targeting the methionine dependence of colon tumors is effected by recombinant methioninase (rMETase), alone and in combination with cisplatin (CDDP). In vitro results demonstrated that CDDP and rMETase act synergistically on the human colon cancer cell line SW 620, with a combination index (CI) of 0.45, as well as on the human colon cancer cell line Colo 205 with a CI of 0.7. Human colon cancer lines HCT 15, HT 29, Colo 205, and SW 620 growing in nude mice were treated with rMETase to determine an effective dose for depletion of tumor methionine. rMETase at 15 units/g/day for 5 days depleted tumor methionine in all four tumor types to approximately 30% of untreated control. rMETase alone arrested growth of HCT 15 and HT29 in nude mice for 1 week after treatment termination. Colo 205 and SW 620 were partially arrested by rMETase. However, CDDP in combination with rMETase resulted in tumor regression of Colo 205 and growth arrest of SW 620 in nude mice. The ratio of the treated:control group (T:C) tumor weights for Colo 205 was 8% when CDDP was given on day-5, followed by treatment on days 5-9 with rMETase. This treatment schedule resulted in two of the six animals having no detectable tumor when the experiment was terminated on day 16. SW620 was resistant to CDDP alone and only partially sensitive to rMETase alone. However, when SW 620 was treated with rMETase from days-5 to -9 and CDDP on day-5, tumor growth was arrested. The results demonstrate that rMETase used simultaneously in combination with CDDP had significant antitumor efficacy in colon cancer in vitro and in vivo. The data suggest a novel and promising therapeutic approach by targeting the elevated methionine dependence of colon cancer.

[Primary experience in inframammary crease reconstruction].

OBJECTIVE: To improve the result of augmentation mammaplasty. METHODS: The authors selected 15 patients whose primary breast volume was less than 70 ml. During augmentation mammaplasty on these patients, the inframammary creases were reconstructed by lifting and fixation under the pectorales major. The paper elaborated on the principle, design and method of the operation. RESULTS: Postoperative follow-up for 6 to 12 months showed good results. CONCLUSION: This method is simple, of rational design and leaving no scar on the skin.

[Clinical observation of hysteroscopic electric resection: an analysis of 36 patients with abnormal uterine bleeding].

OBJECTIVE: To assess the efficacy and influences of hysteroscopic electrosurgery in treating abnormal uterine bleeding (AUB). METHODS: 36 women with AUB received hysteroscopic transcervical resection of the endometrium or intrauterine lesions with a continuous flow resectoscope. RESULTS: The complications rate was 8.3%. 32 patients were followed for 4 to 12 months. Menstrual symptoms ameliorated in 82.4% of the patients. 5.9% of the women became amenorrhoeic. 58.8% had only spotting, 17.6% recovered normal period. 6 out of the 8 cases with dysmenorrhea had relieved their pain. Satisfactory rate of the procedure was 76.5%. Two patients received hysterectomy later for recurrent uterine bleeding. CONCLUSION: Hysteroscopic electrosurgery is an effective procedure in treating abnormal uterine bleeding. But its long-term efficacy is yet to be identified. When improving the technique and basic knowledge, the indications should be given appropriatly.

[DNA content and cell cycle phase analysis in uterine sarcomas and its clinical significance].

OBJECTIVE: To investigate the relationship among DNA content, cell cycle phase analysis with the clinical-pathological features and prognosis of uterine sarcomas. METHODS: DNA content and cell cycle phase analysis were performed on 75 paraffin-embedded specimens of uterine sarcomas using flow cytometry. RESULTS: Both the aneuploidy rate and DNA index (DI) in specimens from advanced stage and poorly-differentiated cases were significantly higher than those in specimens from early stage and well-differentiated cases (P < 0.05). S-phase fraction (SPF) and proliferation index (PI) were significantly higher in advanced stage than those in early stage (P < 0.05). The 5-year suvival rates for DNA aneuploid and diploid patients were 27% and 76%, respectively (P < 0.01). The 5-year suvival rates for patients with a SPF > 20% and those with SPF < or = 20% were 10% and 56% respectively (P < 0.01). CONCLUSION: DI, SPF, PI and clinical stage are the most important factors affecting the prognosis.

[Vitreous surgery for intraocular lens dislocated into vitreous cavity].

OBJECTIVE: To study the way to manage intraocular lenses (IOLs) dislocated into vitreous cavity. METHODS: Three methods of vitreoretinal surgery were applied for 15 cases with severe dislocation of IOL into vitreous cavity: vitreoretinal surgery combined with simple IOL removal in 7 cases (simple group), vitreoretinal surgery combined with liquid perfluorocarbon (PFCL) applied for IOL removal in 3 cases and vitreoretinal surgery combined with IOL removal and fixation of IOL at ciliary sulcus in 5 cases. RESULTS: The dislocated IOLs were removed successfully in all cases. The preoperative visual acuities were light perception to hand movement. Postoperatively, the patients were followed for 3 months to 3 years. In the cases with ciliary sulcus fixation of IOL, all their visual acuities were above 20/100 and the best visual acuity was 20/25. In the 10 cases without fixation of IOL, the corrected visual acuities were 20/1000 to 20/50. The complications were retinal detachment in 1 case, slight vitreous hemorrhage in 2 cases and temporary uveitis in 3 cases. CONCLUSION: Vitreous surgery is the best way for the treatment of a case with IOL severely dislocated into the vitreous cavity, and such an IOL should be removed. Visual acuity can be improved significantly by the fixation of IOL at ciliary sulcus.

[Retreatment after excimer laser photorefractive keratectomy].

OBJECTIVE: To evaluate the efficacy and safety of reoperation of residual myopia because of regression, undercorrection with or without coexisting severe corneal haze after excimer laser photorefractive keratectomy (PRK). METHODS: A VISK 20/20 excimer laser was used to treat myopia of -1.00 to -16.50 diopters (spherical equivalent) at corneal plane. 35 patients (51 eyes) who had residual myopia because of regression, undercorrection with or without coexisting severe corneal haze after initial PRK were retreated by using PRK and phototherapeutic keratectomy. RESULTS: At 12 months after retreatment, the rates of uncorrected visual acuity more than or equal to 10/20 or 20/20 were 82.6% and 39.1% respectively, 100.0% and 50.0% in myopia up to -6.00 D, 73.3% and 33.3% in myopia of -6.25 to -16.50 D respectively. 86.3% of eyes were within +/- 1.00 diopters of desired emmetropia. Corneal haze after retreatment was not greater than that observed after initial procedures. CONCLUSIONS: The residual myopia because of regression, undercorrection with or without severe corneal haze after initial PRK can be successfully retreated. Although the therapeutic result of retreatment of PRK is less successful than that of the initial procedure, retreatment appears to enhance it.

[Changes of serum cardiac troponin T in patients undergoing cardiac surgery].

We detected the changes of serum cardiac troponin T(cTn T) content and serum cardiac enzyme activities in 15 patients with cardiopulmonary bypass during cardiac operation. The central venous blood samples were collected to measure the serum concentration of cTn T by enzyme immunoassay methods and activities of lactate dehydrogenase(LDH), creatine kinase(CK), and creatine kinase isoenzyme(CK-MB) by biochemical methods. The content of cTn T and activities of LDH, CK, and CK-MB increased significantly after cardiac surgery. There was positive correlation between cTn T and aortic cross clamping time (r = 0.6014, P < 0.05), but there was no correlation between CK-MB and aortic cross clamping time (r = 0.4712, P > 0.05). We conclude that serum cTn T and CK-MB can monitor cardiac ischemic reperfusion injury and can evaluate myocardial protection methods during cardiac operation, but cTn T is better than CK-MB.

[Effect of desflurane on quantitative electroencephalogram and hemodynamics].

To evaluate the feasibility of monitoring depth of desflurane anesthesia by quantitative electroencephalogram(QEEG) and hemodynamics, eight ASA I-II patients were studied. Anesthesia was induced by propofol and vecuronium, after tracheal intubation and the lungs were ventilated with desflurane in oxygen. The results showed that desflurane significantly reduced bispectral index(BI), 95% spectral edge frequency(SEF) and median frequency(MF) in a negative linear dose-related manner. When the end-tidal desflurane concentration reached 1.5 MAC, mean artery pressure(MAP) was decreased significantly, and heart rate(HR) increased by 14.5% (but there was no statistic significance), compared with pre-anesthesia. The results suggest that QEEG can be used to monitor depth of desflurane anesthesia, but MAP and HR are not the ideal monitoring indices during desflurane anesthesia.

[Changes in lipoprotein (a) and the relation between lipoprotein (a) and endothelin in patients during upper abdominal surgery].

OBJECTIVE: We observed the changes in both serum lipoprotein(a) [Lp(a)] and plasma endothelin (ET) during upper abdominal surgery under intravenous combined with inhalation anesthesia and we also analyzed their correlativity. METHODS: Fifteen patients, scheduled for upper abdominal surgery, were anesthetized with total does of fentanal 8-12 micrograms.kg-1 and inhalation of 1-2 MAC enflurane. Serum Lp(a) and plasma ET at pre-anesthesia, pre-operation, 1 h from the start of operation and 30 min after operation were determined. RESULTS: The average concentrations of Lp(a) and ET were increased at 1 h from the start of operation compared with pre-anesthesia, but had no statistical significance. Lp(a) and ET were significantly higher at 30 min after operation than pre-anesthesia(P < 0.05). CONCLUSION: Lp(a) and ET have a close relation in advancing cardio-cerebral blood vessel diseases.

[A model of closed epidural balloon inflation brain injury in rats].

OBJECTIVES: To establish an animal model of closed brain epidural occupancy trauma in rats. METHODS: Sixty-eight Wistar rats were randomly divided into 7 groups: normal control group(Group A), sham-operated group(Group B), and 5 experimental groups. The last ones included: Group C in which water was injected 0.05 ml into balloon and kept for 1 hour; Group D injected 0.05 ml and kept for 3 hours; Group E injected 0.1 ml and kept for 1 hour; Group F injected 0.1 ml and kept for 2 hours. Group B, C, D were kept maintaining and observing for two days after operation, respectively. Group E was divided into EI (maintaining for two days) and EII (for three days). RESULTS: Of all the experimental groups, the mortality rate during acute phase was 100% in Group F, 28% in Group E, 50% in Group D, and 0% in Group C. Four rats died during the raising-up period in Group E. Both ICP and cerebral water content were increased in 46 survived rats, and nerve functions were damaged in all the experimental groups. Pathologic changes read as follows: Hemorrhage was found in damage-area; injury and dead cells were seen in compressed region. These changes became worse with time prolonged and injury degree developed. CONCLUSIONS: The method is simple, reproducible and economical, well showing the pathological classification of epidural occupancy trauma.

[Detection of Epstein-Barr viral genome in tumor cells of Warthin's tumor of parotid gland].

OBJECTIVE: To evaluate the relations between Warthin's tumors of salivary gland and Epstein-Barr virus (EBV). METHODS: 75 cases of salivary gland Warthin's tumor and 20 cases of normal salivary gland tissue were examined for to detect EBV genoma by using the polymerase chain reaction (PCR) of DNA, 62 cases of single Warthin's tumor and 13 cases of multiple/bilateral Warthin's tumor's. RESULTS: EBV DNA was detected in 13 cases of single Wathin's tumors, 9 cases of multiple/bilateral Warthin's tumor and 3 cases of normal salivary gland. CONCLUSION: There was a close relationship between EBV and multiple/bilateral Warthin's tumors.

[Supercritical-CO2 fluid extraction of fatty oils from Perilla frutescens seeds and pharmacological study].

Fatty oils from the seeds of Perilla frutescens were extracted by supercritical-CO2 fluid or petroleum ether and its quality was analyzed by GC-MS method. Pharmacological action of lowering blood-lipid for the oils was studied. The results showed that the technology of supercritical-CO2 extraction of the oils of P. frutescens seeds is superior than that of petroleum ether and the oil from SFE-CO2 have the action of lowering blood-lipid with low toxicity.

[Studies on the supercritical-CO2 fluid extraction and quality evaluation of ginger oils].

Essential oils from the dried rhizomes of Zingiber officinale were extracted by supercritical-CO2 fluid(SFE-CO2) and traditional water still distillation (WSD) methods, and the SFE extraction technology and quality (GC-MS, etc.) of the oils were studied. The results showed that the SFE-CO2 contains 49 constituents, such as 6-paradol, etc, the ginger peppery component of them gets to 22.90%.

[Clinical study on keratoplasty with corneal cryopreservation].

PURPOSE: To evaluate the clinical application value of the corneal cryopreservation in keratonosus. METHODS: 32 patients with keratonosus treated by corneal transplantation were clinically observed. RESULTS: In 32 keratonosus cases, 59.37% (19 cases) of the grafts remained transparant; in 24 corneal ulcer preforation cases, 45.83% (11 cases) of the grafts were transparent, all eyeballs(100%) were remained. CONCLUSION: Corneal cryopreservation can be a practical, effective method in the treatment of keratonosus in district hospitals.
