The AIDS disease of CD4C/HIV transgenic mice shows impaired germinal centers and autoantibodies and develops in the absence of IFN-gamma and IL-6.

The mechanisms responsible for degeneration of germinal centers (GC) and follicular dendritic cell (FDC) networks during progression to AIDS remain elusive. Here, we show that CD4(+) T cells from CD4C/HIV-1 Tg mice, which develop a severe AIDS-like disease, express low levels of CD40 ligand. Accordingly, GC formation, FDC networks, and immunoglobulin isotype switching are impaired in these animals. However, Tg B cells respond to in vitro CD40 stimulation. Total serum IgG levels are reduced in Tg mice, whereas total IgM levels are increased with a significant amount showing DNA specificity. IFN-gamma- and IL-6-deficient CD4C/HIV Tg mice also develop the AIDS-like disease and produce auto-Ab. Thus, CD4C/HIV Tg mice have immune dysfunction accompanied by autoimmune responses.

The pathogenicity of human immunodeficiency virus (HIV) type 1 Nef in CD4C/HIV transgenic mice is abolished by mutation of its SH3-binding domain, and disease development is delayed in the absence of Hck.

The human immunodeficiency virus type 1 (HIV-1) Nef protein is an important determinant of AIDS pathogenesis. We have previously reported that HIV-1 Nef is responsible for the induction of a severe AIDS-like disease in CD4C/HIV transgenic (Tg) mice. To understand the molecular mechanisms of this Nef-induced disease, we generated Tg mice expressing a mutated Nef protein in which the SH3 ligand-binding domain (P(72)XXP(75)XXP(78)) was mutated to A(72)XXA(75)XXQ(78). This mutation completely abolished the pathogenic potential of Nef, although a partial downregulation of the CD4 cell surface expression was still observed in these Tg mice. We also studied whether Hck, one of the effectors previously found to bind to this PXXP motif of Nef, was involved in disease development. Breeding of Tg mice expressing wild-type Nef on an hck(-/-) (knockout) background did not abolish any of the pathological phenotypes. However, the latency of disease development was prolonged. These data indicate that an intact PXXP domain is essential for inducing an AIDS-like disease in CD4C/HIV Tg mice and suggest that interaction of a cellular effector(s) with this domain is required for the induction of this multiorgan disease. Our findings indicate that Hck is an important, but not an essential, effector of Nef and suggest that another factor(s), yet to be identified, may be more critical for disease development.

Efficacy of continuous tracheal gas insufflation in spontaneously breathing canine with acute lung injury.

OBJECTIVE: To investigate the effects of continuous tracheal gas insufflation (CTGI) combined with biphasic intermittent positive airway pressure (BIPAP) ventilation on dogs with spontaneous breathing. METHODS: Eight canine models with oleic acid induced lung injury and spontaneous breathing were ventilated in a random order by Evita 2 (Drager Inc., Germany) in modes of BIPAP (BIPAP group) and BIPAP with CTGI flow rate of 3, 6 and 9 L/min (T3, T6 and T9 groups), respectively. The setting parameters of BIPAP were fiction of inspired oxygen 60%, inspiratory to expiratory ratio 1:1, respiratory rate 20 and positive end expiratory pressure 5 cm H2O. Arterial and mixed venous blood gas, lung mechanics, systemic and pulmonary hemodynamics status were monitored at the same level of PaCO2 obtained by adjusting peak inspiratory pressure of BIPAP. RESULTS: Peak inspiratory pressure in the T6 group (14 +/- 4 cm H2O) and in the T9 group (11 +/- 3 cm H2O) were significantly lower than that of BIPAP (20 +/- 5 cm H2O, P < 0.01), but there was no significant difference among the T3, T6 and T9 groups or between the T3 and BIPAP groups. PaO2, mean artery blood pressure, mean pulmonary artery pressure, pulmonary artery wedge pressure, cardiac output, oxygen delivery and oxygen consumption all remained unchanged in four different conditions. CONCLUSIONS: Using BIPAP combined with CTGI does not cause asynchrony between ventilator and spontaneous breathing, but significantly decreases airway pressure with no influence on hemodynamics and oxygenation. Therefore, BIPAP with CTGI may be a useful support technique, especially in cases where the airway pressure should be limited.

[A study on the pathogenesis of Streptococcus mitis exotoxin].

OBJECTIVE: To study the isolation, purification and pathogenesis of Streptococcus mitis pyrogenic exotoxin causing toxic shock syndrome. METHODS: Streptococcus mitis isolated from patients' throat were shaking cultivated. After being centrifuged, the supernatant fluid of the culture was precipitated with 20%, 40%, 60%, 80% (NH(4))(2)SO(4) respectively and the fast protein liquid chromatography(FPLC) was used for the final step of purification. Rabbits receiving subcutaneous injection with respective purified proteins were monitored daily for fever. The ability of the purified proteins to enhance the susceptibility of the rabbits to lethal Escherichia coli endotoxin shock is recorded, when the endotoxin was injected intravenously 4 hours after administration of 10 microg Streptococcus mitis exotoxin. RESULTS: Only the protein precipitated by 20% (NH(4))(2)SO(4) (molecular weight is 34,000) from culture supernatant fluid was pyrogenic for rabbits (average temperature increase near 1 degrees C), and it can also cause the proliferation of rabbit splenocytes (mitogenicity). All the animals receiving subcutaneous injection of exotoxin containing purified proteins precipitated with higher concentrations of (NH(4))(2)SO(4) died within 16 approximately 29 hours after intravenous injection of the Escherichia coli endotoxin, demonstrating the enhanced susceptibility of the animals to lethal endotoxin shock. The control rabbits displayed none of these effects. CONCLUSION: Streptococcus mitis exotoxin is a novel streptococcal pyrogenic exotoxin.

[The effects of extrinsic positive end-expiratory pressure on work of breathing in patients with chronic obstructive pulmonary disease].

OBJECTIVE: To study the effect of extrinsic positive end-expiratory pressure(PEEPe) on work of breathing in chronic obstructive pulmonary disease (COPD) patients and its mechanism. METHODS: 10 ventilated patients [4 males and 6 females with mean age of (70.1 +/- 5.5) years] with exacerbation of COPD were admitted into the study. All the patients' cardiopulmonary functions were stable, and they could receive pressure support ventilation (PSV). Patients with sever heart dysfunction, hepatic dysfunction, renal dysfunction or fever were ruled out. Each patient had an esophagus balloon pressure sensor inserted, and the esophagus balloon was properly positioned using "occlusion test". Flow sensor was placed at the end of tracheal tube opening. Then esophagus balloon pressure sensor and flow sensor were connected with Bicore CP-100 pulmonary monitor. Dynamic intrinsic positive end-expiratory pressure (PEEPi, dyn), difference of esophagus pressure (dPes), work of breathing patient (WOBp), work of breathing ventilator (WOBv), pressure time product (PTP), tidal volume (Vt), respiratory rate (RR), minute ventilation (MV), peak inspiratory flow rate (PIFR), peak expiratory flow rate (PEFR), expiratory airway resistance (Rawe), fraction of inspiration (Ti/Ttot) and so on were measured. PEEPi,dyn was measured by esophagus balloon technique. Electrocardiogram and noninvasive blood pressure modes were added to HP M1165A monitor. Heart rate (HR) and blood pressure (BP) were also measured. Arterial blood was analyzed to measure pH, partial pressure of oxygen in arterial blood (PaO(2)), partial pressure of carbon dioxide in arterial blood (PaCO(2)) and saturation of arterial blood oxygen (SaO(2)). At first, PEEPi, dyn was measured when PEEPe was zero, which was called PEEPi, dynz and was set as baseline. Then PEEPe was set randomly, which was 0%, 40%, 60%, 80% and 100% of PEEPi, dynz respectively. Other parameters remained stable. All the indexes were measured 30 minutes after the level of PEEPe was changed. RESULTS: WOBp decreased as PEEPe increased, from (1.08 +/- 0.49) J/L of PEEPe-0% of PEEPi,dynz to (0.57 +/- 0.32) J/L of PEEPe-100% of PEEPi, dynz, and significant difference was found between each level of PEEPe and the baseline (P < 0.01). But we didn't find significant difference between PEEPe-80% of PEEPi,dynz and PEEPe-100% of PEEPi, dynz. WOBv increased significantly as PEEPe increased, from (1.50 +/- 0.47) J/L of PEEPe-0% of PEEPi, dynz to (1.82 +/- 0.56) J/L of PEEPe-80% of PEEPi, dynz and (1.85 +/- 0.48) J/L of PEEPe-100% of PEEPi,dynz(P < 0.01). The total WOB (WOBp + WOBv) remained steady. PTP trended to decrease, from (153.6 +/- 76.8) cm H(2)O x s x m(-1) of PEEPe-0% of PEEPi, dynz to (86.9 +/- 45.9) cm H(2)O x s x m(-1) of PEEPe-100% of PEEPi, dynz; the difference of PTP between PEEPe-40% of PEEPi,dynz and PEEPe-0% of PEEPi, dynz was significant(P < 0.05), the differences between other levels of PEEPe and PEEPe-0% of PEEPi, dynz were very significant (P < 0.01), but no significant difference was found between PEEPe-80% of PEEPi, dynz and PEEPe-100% of PEEPi, dynz. PEEPi,dyn also decreased significantly as PEEPe increased, from (7.70 +/- 1.42) cm H(2)O of PEEPe-0% of PEEPi, dynz to (2.10 +/- 0.57) cm H(2)O of PEEPe-100% of PEEPi, dynz, very significant differences were found between each level of PEEPe and the baseline (P < 0.01). Significant positive linear correlation was found between DeltaWOBp and DeltaPEEPi, dyn were(r = 0.609, P < 0.01, n = 10). dPes decreased as PEEPe increased, from (15.20 +/- 4.16) cm H(2)O of PEEPe-0% of PEEPi, dynz to (8.20 +/- 4.13) cm H(2)O of PEEPe-100% of PEEPi, dynz, and very significant differences were found between each level of PEEPe and the baseline (P < 0.01). But the difference between PEEPe-80% of PEEPi, dynz and PEEPe-100% of PEEPi, dynz wasn't significant, even dPes of PEEPe-100% of PEEPi, dynz was higher than that of PEEPe-80% of PEEPi, dynz. Vt didn't change markedly. RR, MV and f/Vt increased slightly. Both PIFR and PEFR didn't change markedly. At the same time, HR, BP and blood gas analysis were investigated, but no significant change was found. CONCLUSION: PEEPe can reduce WOBp significantly and improve the coordination between the patient and ventilator. It is shown that decrement of pressure difference between alveolar and central airway leads to decrement of WOBp.

[Further study on cyclin D1 expression and function in human nasopharyngeal carcinoma cell line].

OBJECTIVE: In the present study, we tried to further elucidate the expression and function of cyclin D1 in human nasopharyngeal carcinoma (NPC) cell line. METHODS: Western blot was used in the analysis of D-type cyclin expression profile. The distribution of this protein was determined in the double parameter flow cytometry (FCM). The function of cyclin D1 in nasopharyngeal carcinoma cell line was determined in the antisense-oligonucleotied-cyclin D1 and antibody mediated knock-out experiments. RESULTS: The immunoblot analysis of two NPC cell lines showed that the spectra of D-type in NPC cell lines founded their expressions in D1, D2, and D3. It also suggested that the overexpression of cyclin D1 could lead to deregulation of G1/S control. An additional double parameter FCM showed a characteristic variation in HNE1 which was consistent with the cell-cycle oscillation and the peak level expressed in G0/G1 phase and the lowest level in the S-phase. Two experimental approaches aimed at specific functional knock-out of cyclin D1 were employed; the cyclin D1-ASPODN experiment showed an inhibition of cyclin D1 expression at mRNA level and a down-regulated cyclin D1 partial inhibition of progression into S phase, resulting in cell growth arrest while the electroporation of neutralizing antibodies demonstrated a specific delay of S-phase entry in HNE1 and the requirement for the cyclin D1 in cell cycle progression of HNE1 cell line in regulation of G1/S. CONCLUSION: These data reveal that the cell-cycle regulatory function of cyclin D1 is essential for cell cycle progression in the G1 of nasopharyngeal carcinoma cell line and suggest that cyclin D1 play an important role in NPC carcinogenesis.

Three-dimensional correction of scoliosis using TSRH instrumentation.

OBJECTIVE: To evaluate the results of TSRH instrumentation in the correction of coronal, sagittal and rotational deformity of scoliosis. METHODS: From January 1998 to December 1999, thirty-two consecutive patients (6 males, 26 females)with scoliosis underwent anterior or posterior spinal instrumentation and fusion using TSRH instrumentation. Of these cases, 21 were idiopathic scoliosis and 11 were congenital scoliosis. The average age at surgery was 16.4 years (range, 11 approximately 45 years). The mean Cobb angle at surgery was 71.2 degrees range, 44 degrees approximately 125 degrees) in the coronal plane, and 49. degrees range, 16 degrees aprroximately 67degrees in the sagittal plane. Rotational deformity (Nash-Moe) ranged from I to III degree. Preoperative apical translation averaged 4.8 cm (range, 3 approximately 9 cm). RESULTS: The average follow-up duration was 13.3 months (range, 10 approximately 24 months). At the final follow-up, the mean Cobb angle in the coronal plane was 26.6 (range, 10 degrees approximately 73 degrees), with a 63.8% of improvement. Sagittal alignment was well maintained with a mean Cobb angle of 28 degrees (range, 10 degrees approximatelky 45 degrees). The average correction of rotation of the apical vertebra was I degree. The average apical translation was 1.6 cm (range, 0.5 approximately 5.0 cm) representing a correction rate of 66, 7%. Complication was noted in two cases with an incidence of 3.1%, one case had superficial infection and the other one had lower hook dislocation. There was no neurologic deficit and pseudoarthrodesis in this series. CONCLUSION: TSRH instrumentation is an effective and convenient three-dimensional correction system with a lower rate of complication, which can not only correct the coronal and rotational deformity, but maintain the sagittal alignment as well.

[Study on the relationships between leptin levels and weights of mothers and infants and the relationships of cord serum leptin to C-peptide, insulin and insulin like growth factor-II].

OBJECTIVE: To determine the relationships between serum leptin levels and maternal weights in late pregnancy and cord blood leptin levels to birth-weights, C-peptide, insulin and insulin like growth factor (IGF-II). METHODS: Fifty normal pregnant women at 37-38 weeks and their newborns were studied, and 29 non-pregnant women were set as control. Venous blood was taken from women and from the cord at delivery. Blood leptin and cord blood C-peptide, insulin, and IGF-II were measured by radio-immunoassay. RESULTS: The average leptin level in maternal sera was (13.62 +/- 3.68) micrograms/L, significantly higher than that in the control (6.60 +/- 3.04) micrograms/L and that in cord blood (8.05 +/- 4.61) micrograms/L. Maternal leptin levels were significantly correlated with maternal weights and body mass index (BMI. r = 0.33, 0.35, P < 0.05), but not with infant birth-weights (r = 0.10, P > 0.05). Cord blood leptin levels were significantly correlated with birth-weights and BMI (r = 0.54, 0.49, P < 0.001) but have no correlation with maternal leptin levels (r = 0.19, P > 0.05). Significant difference of the cord leptin levels was not seen between the males and females. The cord blood C-peptide was (0.86 +/- 0.35) microgram/L, insulin (8.49 +/- 4.76) mU/L and IGF-II (0.218 +/- 0.076) microgram/T. Cord leptin levels were correlated with C-peptide levels (r = 0.37, P < 0.05), but not with insulin and IGF-II levels (r = 0.19, -0.14, P > 0.05). CONCLUSIONS: Maternal leptin levels in late pregnancy were significantly higher than those in normal non-pregnant women and positively correlated with maternal weights and BMI. Cord blood leptin levels were positively correlated with birth-weights and BMI of the newborns. The leptin levels of cord blood were correlated with those of C-peptide but not insulin and IGF-II.

Asymmetry of brain functional activation: fMRI study under language and music stimulation.

OBJECTIVE: To determine the asymmetry of the human brain functional activation. METHODS: With the help of GE Signa Horizon MRI system, 14 cases of right-handed volunteers were examined and the blood oxygenation level dependent method was used. The T1-weighted images were obtained with spin echo pulse sequence and the functional imaging (T2*-weighted) was performed using a single shot echo planar imaging pulse sequence. Data analysis was done with Sun Sparc Workstation and by the method of student t test or correlation analysis. RESULTS: Most of activation areas were in the left hemisphere under language stimulation, while they were in the right side under music stimulation. Besides, a few brain areas in the contralateral cerebral cortex were also activated under both stimulations. CONCLUSION: The present study supported the hypothesis of the asymmetry of brain functional activation and many brain areas of the cerebral cortex as well as both hemispheres worked in coordination. In addition, it also proved that fMRI is a feasible method in the study of human brain in vivo.

[Detection of the asymptomatic infection by human papillomavirus in pregnant women and neonates].

OBJECTIVE: To investigate the state of human papillomavirus (HPV) infection in similar average-aged pregnant women of different gestational periods, in the puerperium and neonates. METHODS: Polymerase chain reaction (PCR) assay was employed to detect HPV-6, 11, 16 and 18 DNA in 30 pregnant women in the first trimester, 42 in the second and 31 in the third (who were followed up to their puerperium), and 30 non-pregnant women asking for intrauterine device in our out-patient clinic were taken as controls. Average age in the four groups showed no significant difference (P > 0.05). Samples from cervical, vaginal exfoliated cells, maternal peripheral blood and nasopharyngeal secretion of the newborns were examined respectively. RESULTS: (1) In the first trimester, HPV-DNA was detected in the cervical, vaginal exfoliated cells of 5 cases and in the maternal peripheral blood of 7 cases. (2) In the second trimester, HPV-DNA was detected in the cervical, vaginal exfoliated cells of 12 cases and in the maternal peripheral blood of 11 cases. (3) In the third trimester, HPV-DNA was detected in the cervical, vaginal exfoliated cells of 23 cases and in the maternal peripheral blood of 18 cases. (4) In the puerperium, HPV-DNA was detected in samples of cervical, vaginal exfoliated cells of 8 cases and maternal peripheral blood of 7 cases. (5) In the control group, HPV-DNA was detected in the cervical, vaginal exfoliated cells of 8 cases and in the maternal peripheral blood of 6 cases. (6) Consecutive examinations were carried out in 31 pregnant women from the third trimester, through labor to 6 weeks of postpartum. HPV-DNA was positive in the cervical, vaginal samples of 17, 21 and 8 cases, respectively, according to the perinatal periods, and in the maternal peripheral blood of 14, 13 and 7 cases, respectively. The result through the above gestational stages was fluctuated in the cervical, vaginal samples of 6 cases and in the maternal peripheral blood of 7 cases. (7) Successive examinations in infants at time of birth, 48-72 h and 6 weeks after birth showed positive HPV-DNA in the nasopharyngeal secretion of 13, 6 cases and 1 case with respect to the examining periods. (8) The positive cases were mainly infected by HPV-16, 18. CONCLUSIONS: (1) Infective rate of HPV is statistically significant in the third trimester, but no significant difference exists among the first trimester, the second trimester, the puerperium or the non-pregnancies. (2) Examining consecutively, the HPV positive rate is found to be decreased after delivery, the positive expression of HPV during the gestational periods exhibited fluctuation. (3) Infective rate of HPV in the neonatal nasopharyngeal specimens tends to decrease with time after delivery.

[Lung injury caused by passive smoking and its effects on cytokines in rats].

OBJECTIVE: A rat model with chronic bronchitis was replicated by passive inhalation of cigarette smoking fume to study its long-term effects on lung injury and nitric oxide (NO), interluekin-6 (IL-6), interleukin-8 (IL-8). METHODS: Levels of nitrogen dioxide (NO(2)) and nitrogen trioxide (NO(3)) were measured with spectrophotometry in rats indicating their level of nitric oxide (NO). Levels of IL-6 and IL-8 were determined by enzyme-linked immunosorbent assay (ELISA). RESULTS: Levels of NO in serum, bronchial alveolar lavage fluid (BALF) and lung tissue in the smoking group were significantly lower than those in the normal controls (P < 0.01). But, levels of IL-6 and IL-8 were higher in the smoking group than those in the controls. CONCLUSION: Long-term passive smoking could cause injury of lung tissue to certain extent, reduction in secretion of NO in endothelial cells and damage to pulmonary vessels.

[Prediction of peritoneal fluid pancreatic amylase to histopathological change and prognosis of acute experimental necrotic pancreatitis of monkey].

OBJECTIVE: To observe the significance of peritoneal fluid pancreatic amylase in predicting the degree of histopathological change and prognosis of acute necrotic pancreatitis (ANP) of monkey. METHODS: Injection of the mixture of bile and trypsin into the pancreatic duct set up ANP monkey model. RESULTS: The severity of ANP depended on the injection rate and volume, in correlation to the peritoneal fluid pancreatic amylase (PF Amylase) in the early stage of ANP. With the rate (3-13 ml/min) and volume (0.1-1.0 ml/kg) of injection increase, the PF Amylase in the early stage of the ANP significantly went up. Meanwhile, the survival time was negatively correlated to the PF Amylase in the early stage (0.5 hour) of ANP (t = -9.06 InX + 129.85, r = -0.88, P < 0.01). CONCLUSIONS: The PF Amylase in the early stage of ANP can effectively predict the severity of ANP and prognosis.

The operative treatment of spinal fracture-dislocation without neurologic deficits.

OBJECTIVE: To evaluate the results of operative treatment of spinal fracture-dislocation without neurologic deficits. METHODS: Eighteen patients with spinal fracture-dislocation were neurologically intact at the time of injury, and all were treated operatively. The fracture sites were: 8 cases in cervical spine, 3 cases in thoracic spine, and 7 cases in lumbar spine. Eight patients with cervical injuries had variant degrees of forward slide and kyphotic deformity. Of the 10 thoracic and lumbar fractures, one had lateral dislocation, 4 cases with kyphotic deformities, 5 cases with spinal canal compromise averaged 50% (ranging from 40% to 70%). RESULTS: The average period of follow-up was 4.4 years with a range of 11 months to 13 years. All the patients returned to full-time work. No patient developed neurologic deterioration. Kyphotic deformity was corrected in the 4 cases, and no progressive kyphosis was noted. There was no operation-related complication. The averaged post-operative hospitalization time was 13 days. CONCLUSION: Despite the rare incidence of spinal fracture-dislocation without neurologic deficits, we suggested that kind of fracture be considered unstable fracture because of its potential risk of delayed neurologic deterioration and kyphotic deformity, and be treated operatively to restore the sagittal alignment and the stability of the spine.

Imaging the functioning human brain.

One of the most exciting methodological advances for brain research field arises in functional brain imaging, which enables us to localize and characterize neural activity and biochemical events in the living human brain. Recently developed event-related functional MRI makes it possible to visualize the brain activity associated with cognitive processes with the temporal resolution of the hemodynamic response. In addition, the high sensitivity and selectivity of positron-emission tomography allow us to probe the neurochemical processes at the molecular level. Positron-emission tomography also has been applied to investigate the effects of therapeutic drugs as well as the effects of drugs of abuse.

Contribution of the -455G/A polymorphism at the beta-fibrinogen gene to erythrocyte aggregation in patients with coronary artery disease.

BACKGROUND: A high level of red blood cell (RBC) aggregation has been consistently found in patients with coronary artery disease (CAD) in case-control studies. Plasma fibrinogen has been shown to promote RBC aggregability. The purpose of this study was to investigate the influence of the genetic variability of the beta-fibrinogen gene on RBC aggregation in patients with CAD. METHODS AND RESULTS: The genotype of the beta-fibrinogen gene locus was determined by polymerase chain reaction using the restriction enzyme HaeIII for a G to A substitution at position -455 upstream from the transcriptional start site in 135 French Canadians with premature CAD (age: 51+/-7 years). Indices measuring the RBC aggregation kinetics (S10) and shear resistance of the aggregates (gammaS) were obtained by laser reflectometry. Patients were separated into groups by using the medians of S10 and gammaS. Using chi2 analyses, the distribution of the -455GG, -455GA, and -455AA genotypes in the groups with high levels of S10 (0.43, 0.49, and 0.08) and gammaS (0.45, 0.49, and 0.06) were found to be significantly distinct from those in the groups with low levels of S10 (0.67, 0.27, and 0.06; p<0.05) and gammaS (0.70, 0.23, and 0.07; p<0.01). High levels of RBC aggregation were closely associated with the rare -455A allele. Multivariate linear regression analyses showed that S10 was positively correlated with the linear combination of the fibrinogen concentration, age, and the -455G/A genotype (adjusted r = 0.63, p<0.0001). Fibrinogen and age were positive determinants, and HDL-cholesterol was a negative predictor of gammaS (adjusted r = 0.51, p<0.0001). CONCLUSION: These findings support the hypothesis that RBC hyperaggregation in premature CAD may be associated with the beta-fibrinogen -455G/A polymorphism. This association may be explained by a change in the concentration and/or the functional properties of the fibrinogen protein.

Amplified fragment length polymorphism analysis on D5S436 locus and its application to linkage analysis in gene diagnosis of asthma.

OBJECTIVES: To detect the polymorphism of D5S436 locus, and to carry out an amplified fragment length polymorphism (Amp-FLP) genotyping linkage analysis in asthma families. METHODS: Allele and genotype frequencies of the high polymorphic D5S436 locus were determined in 92 unrelated Chinese individuals by using polymerase chain reaction (PCR). The amplified fragments were separated by denaturing polyacrylamide gel electrophoresis and silver staining techniques. RESULTS: Eight alleles and 22 genotypes were observed in this population. The heterozygosity was 78.26%. The polymorphism information content (PIC) was 0.76. Amp-FLP genotyping linkage analyses were carried out in four asthma families, demonstrating that the locus was inherited according to Mendel's law and had a clear result. CONCLUSION: The PIC of D5S436 is high in Chinese individuals, so it could be used as a genetic marker of asthma in Chinese and might be useful in the gene diagnosis of asthma.

The theory of "expelling pathogens regardless of hard stool", a significant contribution of Wu Youke.

Though there are famous theories of "assorted Qi", "membranous origin", the contribution of Wu Youke's Wen yi lun (On Warm Pestilence) of the theory of " expelling pathogens regardless of hard stool" (EPRHS) is even more important. The main points of this theory includes: it is essential to expel pathogens at an early stage, while for attacking therapy, expelling pathogens with rhubarb is essential. EPRHS elucidates the significance of expelling pathogens in attacking therapy theoretically, and elevates the efficacy clinically with profound influence in later ages. In the past 2 decades, the early application of rhubarb to elevate the therapeutic effect for the treatment of pneumonia, epidemic meningitis, and Bencephalitis even in the absence of hard constipated stool marks the significance of Wu's such theory.
