Computerized image analysis of smooth muscle fibers in normal Chinese men and impotence patients.

Smooth muscle fibers are considered very important in penile erection and relaxation. The reduction of intracavernosus smooth muscle fibers can provoke impotence. The percentage of smooth muscle fibers in corpus cavernosum specimens was studied quantitatively in normal men and impotence patients by means of computerized image analysis. The percentage of smooth muscle fibers in the specimens of 14 normal men, ranged from 41% to 61% (average 50%) and in 12 impotence patients, it was 22% to 46% (average 31%). A decrease of smooth muscle fibers and an increase of collagen fibers were noted in impotence patients.

Detection of HPV-16 DNA in cervical carcinoma by paraffin section in situ hybridization.

Thirty-eight cervical carcinomas and one cervical condyloma were examined for the presence of human papillomavirus type 16 (HPV-16) DNA using 3H-dcTP-labelled HPV-16 DNA probe in paraffin section in situ hybridization and dot blot hybridization (Tm-17 degrees C). The results showed that HPV-16 DNA positive rate in our series was about 74.4% (29/39) as determined by paraffin section in situ hybridization and about 71.8% (28/29) by dot blot hybridization. There was no significant difference between the two methods. We thus confirmed that paraffin section in situ hybridization is an informative, reliable, and sensitive method for the diagnosis of cervical HPV infection.

[Effect of alpha-difluoromethylornithine on sporogony of Plasmodium yoelii].

The effect of alpha-difluoromethylornithine (DFMO) on the sporogony of P yoelii was studied by electron microscopy. DFMO sugar solution was fed to female mosquitoes. Scanning electron microscopy (SEM) revealed that the oocysts were smaller and the surface of oocysts was uneven as compared with control. Transmission electron microscopy (TEM) showed that cytoplasm of the affected oocysts contained vacuoles and the multilamellate structures increased. The number of matrix cavitations and deformations of mitochondria increased. Nuclear membrane was damaged. The number of autophagocytic vacuoles increased. The membrane system was markedly affected.

[Effect of Poly I:C on the sporozoite invasion and the development of exoerythrocytic forms of Plasmodium yoelii yoelii in rats].

When Wistar rats were given intravenously with Poly I:C 5 mg/kg 18 h before sporozoite inoculation, the density of exoerythrocytic forms (EEF) in the rat liver 42 h after inoculation was markedly decreased, the sporozoite viability being only 5-12% of that of the controls. However, the size and maturation rate of EEF were all normal. When Poly I:C was given 2 h after sporozoite inoculation, the sporozoite viability and the size and the maturation rate of EEF were similar to those of the controls. When the sporozoites were inoculated with rat serum containing interferon for 40 min before inoculation, no detrimental effect on the sporozoite invasion and the development of EEF could be found. These findings suggested that the target of antimalarial action of interferon might not be the EEF itself. It is possible that interferon could minimize the sporozoite number by enhancing the phagocytosis and the biocidal activity of macrophages, especially the Kupffer cells.

[Prediction of ovulation].

Prediction of ovulation was carried out by B-ultrasonography, BBT graph, cervical mucus and appearance of mittelschmerz in 45 normally menstruation women for a total of 72 cycles. LH peak values were also determined in 20 cases (20 cycles). The results were as follows: 1. The relation-ship between each index and ovulation: (1) The ultrasound image of follicle: average daily follicular growth during the 4-5 day before ovulation was 2.5 +/- 2.1 mm (means +/- s). Average diameter of the biggest follicles one day before ovulation was 20.6 +/- 2.0 mm. (2) The change of cervical mucus: the opening of external os and the secretion of mucus with ferning became most typical one day before the ovulation. (3) BBT graph: in a majority of cycles, ovulation occurred at the first day of the initial rise in temperature. (4) Mittelschmerz: occurred in 35% of cycles and appeared within 24 hours before ovulation. (5) LH peak value: ovulation occurred 24 to 48 hours after the first significant rise of LH, and within 24 hours after LH peak value. 2. The relation-ship between indices: in the majority of cases, when we employ the ultrasound image of follicular rupture as a signs of ovulation, LH peak value, follicle of biggest diameter, maximum volume of cervical mucus, the lowest point of BBT and mittelschmerz appear on the same day, namely, one day before ovulation. The authors suggest that ultrasonography used combination with other parameters provides a feasible and relatively accurate and reliable method for the prediction of ovulation in clinical practice.

[A comparison of the efficiency of density gradient and low-speed centrifugation methods for isolating Plasmodium sporozoites].

P. yoelii sporozoites(Sp) in Anopheles stephensi were first isolated with low-speed centrifugation and the Sp suspension was subsequently purified with the density gradient centrifugation. The recovery rates of Sp by the latter method is about 50 approximately 75% of that by the former, but few debris could be found in the Sp suspension and the infectivity of the Sp was not weakened as compared with the Sp obtained by low-speed method. Sp would retain partial infectivity, when its suspension was maintained in medium 199 at 4 degrees C for 48 hrs. When rats and mice were infected with these Sp, pathological changes of hepatocytes such as cloudy swelling or fatty degeneration, which had been evidenced to be induced by mosquito tissues, would be absent or present in the slightest degree.

[Clinical application of intrauterine insemination].

Nineteen infertile couples were treated with intrauterine insemination (IUI) using husband's semen. Three women with asthenospermic husbands, one with unknown cause and one treated with a combination of induced ovulation and IUI conceived. Three healthy infants have been delivered up to now. The indications for IUI, the importance of proper preparation of semen, the technique of insemination and prediction of ovulation in IUI are discussed.

[Application of monoclonal antibody OC 125 in gynecological oncology].

This study was undertaken to determine whether the measurement of CA125 could effect an early diagnosis and a method for monitoring the course of gynecologic tumors. CA 125 in serum of 195 patients, including 15 apparently healthy women; 39 benign gynecologic tumors; 2 borderline ovarian tumors; 139 malignancies were measured by CENTOCOR cancer antigen 125 kits. Diagnosis of all patients was confirmed by pathology. None of the healthy women; 10% of benign tumors; 78% of epithelial ovarian cancers; 31% of endometrial adenocarcinomas and one out of five adenocarcinoma of uterine cervix had CA 125 level over 65 U/ml. In addition, 23 cases of ovarian cancer were monitored serially up to 9 months. In more than 80% of these patients, CA 125 levels were correlated with the regression or progression of the disease. The significance of this assay for early diagnosis and monitoring the course of ovarian cancer is discussed. It is considered that CA 125 is a promising and useful antigenic marker for monitoring the course of ovarian cancers.
