Isolation of three dormancy QTLs as Mendelian factors in rice.

Seed dormancy is a key adaptive trait under polygenic control in many plants. We introduced the chromosomal regions containing the dormancy QTLs qSD1, qSD7-1, and qSD12 from an accession of weedy rice into a nondormant genetic background to examine component genetic effects and their interactions with time of afterripening (DAR). A BC4F2 plant, which was heterozygous for the three loci, was selected to develop the BC4F3 population. Single point analysis detected only qSD7-1 and qSD12 (R2 = 38-72%) at 10, 30, and 50 DAR in the population. However, multiple linear regression analysis detected genetic effects of the three QTLs and their trigenic epistasis, an environmental effect of DAR (E), and interactions of E with qSD12 and with the qSD1 x qSD7-1 and qSD7-1 x qSD12 epistases. The linear model demonstrates that QTL main effects varied with DAR, and that some epistasis or epistasis-by-DAR interactions partially counteract the main effects. The three QTLs were isolated as single Mendelian factors from the BC4F3 population and estimated for component genic effects based on the BC4F4 populations. Isolation improved estimation of the qSD1 effect and confirmed the major effect of qSD12. The qSD1 and qSD12 loci displayed a gene-additive effect. The qSD7-1, which was further narrowed to a chromosomal region encompassing the red pericarp color gene Rc, displayed gene additive and dominant effects.

A simple procedure for elimination of fungal contamination for enumeration of iron-oxidizing bacteria from bioleaching matrix of sewage sludge.

Enumeration of iron-oxidizing bacteria from the bioleaching matrix of sewage sludge is always confronted by fungal contamination. The objective of the present study was to find a reliable and simple method to remove fungal growth and to shorten the incubation time for facilitating enumeration of the iron-oxidizers from complex sludge samples. The results demonstrated that filtering the sludge sample through sterile No. 5 Whatman filter paper before serial dilution was effective in eliminating the fungal growth on agarose media. Both the counts and the incubation time required for enumeration were highly dependent on the medium pH, with maximum counts at pH 2.5-2.75. Medium prepared at pH values outside of this range led to lower counts and a longer lag time for colony formation. However, the introduction of heterotrophic microorganisms into the solid medium did not show further improvement in enumeration efficiency and shortening of the incubation period. By incorporating the optimal conditions obtained, the incubation time could be reduced to 7 and 10 d for pure cultures and sludge samples, respectively.

Bioleaching of heavy metals from anaerobically digested sewage sludge using FeS2 as an energy source.

The effect of using FeS2 as an energy source, on the bioleaching of heavy metals (Zn, Cr, Cu, Pb and Ni) and nutrients (nitrogen and phosphorus) from anaerobically digested sludge using isolated indigenous iron-oxidizing bacteria was investigated in this paper. Addition of FeS2 in the range of 0.5-4.0 g l(-1) accelerated the acidification of sludge and raised the oxidation-reduction potential of sludge medium with an inoculation of 15% (v/v) of active bacteria, thus resulting in an overall increase in metal dissolution efficiency. After 16 days of bioleaching at 28 degrees C and an initial pH of 3.0, up to 99% of Zn, 65% of Cr, 74% of Cu, 58% of Pb and 84% of Ni can be removed from the sludge. In contrast, only 94% of Zn, 12% of Cr, 21% of Cu, 32% of Pb and 38% of Ni were leached out in the control without inoculation of iron-oxidizing bacteria and the addition of FeS2. Less than 15% of nitrogen and 6% of phosphorous were lost after 16 days of bioleaching when using FeS2 as the energy source. Comparing to 39% and 45% loss respectively for these two nutrients when using FeSO4.7H2O as the energy source, FeS2 appears to be a more suitable energy source for preserving nutrients in sludge while removing heavy metals from sludge.

Characterization of an indigenous iron-oxidizing bacterium and its effectiveness in bioleaching heavy metals from anaerobically digested sewage sludge.

The objective of the present study was to isolate the indigenous iron-oxidizing bacterium and compare its effectiveness in bioleaching of heavy metals from fresh anaerobically digested sludge and aged sludge which had undergone a storage period in a sludge holding tank. An acidophilic iron-oxidizing bacterium Acidithiobacillus ferrooxidans strain ANYL-1 was successfully isolated from the sludge collected from a wastewater treatment plant at Yuen Long district in Hong Kong. It was a Gram negative, non-motile rod shaped bacterium which used ferrous iron, elemental sulfur or thiosulfate as energy source, but did not utilize tetrathionate or glucose as energy source. The optimal temperature and pH for its growth and iron oxidation were 30-35 degrees C and pH 2.0-2.5, respectively. When it was used in the bioleaching of anaerobically digested sewage sludge, an inhibition on metal solubilization was observed in fresh sludge except for Zn whose dissolution was solely a chemical process. Compared to the 3 and 4 days required for solubilization of Cu and Cr respectively from the sludge sample collected after the sludge holding tank (Sludge SHT), 6 days were required to bioleach Cu and Cr from fresh sludge (Sludge AD). The fresh sewage sludge posed an unfavorable condition for bioleaching of heavy metals from anaerobically digested sludge as reflected by the prolonged bioleaching time. Therefore, further studies were needed to understand the inhibitory effects in the fresh anaerobically digested sludge and develop measures to remove it in order to improve the heavy metal bioleaching efficiency.

Enhanced heavy metal bioleaching efficiencies from anaerobically digested sewage sludge with coinoculation of Acidithiobacillus ferrooxidans ANYL-1 and Blastoschizomyces capitatus Y5.

Prolonged bioleaching period was required to remove heavy metals from anaerobically digested sewage sludge in the presence of low molecular weight organic acids. The purpose of the present study was therefore to enhance metal solubilization efficiencies through introducing organic acid-degrading microorganisms into this artificial bioleaching system. An acetic and propionic acid-degrading yeast Blastoschizomyces capitatus Y5 was successfully isolated from a local Yuen Long sewage sludge and it could achieve optimum growth in synthetic liquid media containing 2000 mg l(-1) acetic acid or 1000 mg l(-1) propionic acid. When it was inoculated simultaneously with Acidithiobacillus ferrooxidans ANYL-1 into anaerobically digested sewage sludge, which contained 648 mg l(-1) acetic acid and 731 mg l(-1) propionic acid, both acids were completely decomposed within 24 hours. As a result, ferrous iron oxidation was greatly improved, resulting in enhanced metal solubilization. Compared with the 8, 10 and 12 days required for maximum solubilization of Zn, Cu and Cr for the control sludge, the bioleaching period was significantly shortened to 4, 5 and 8 days respectively for sludge receiving co-inoculation.

[The measurement of intra-cellular AOX in recombinant Pichia pastoris].

The activities of intracellular alcohol oxidase(AOX) in recombinant P. pastoris expressing Pro-UK were determined by a self-designed dissolved oxygen measuring equipment. The enzyme vitality and specific enzyme vitality were defined nd the condition for detecting the enzyme vitality was also established. The experimental results showed that with a certain quantity of biomass in a phosphate buffer containing methanol, the consuming rate of dissolved-oxygen reflected the enzyme vitality of intracellular AOX. It was also found that the pH of the buffer could be very freely between 4.7 and 7.4 and the suitable optical dersity of cell concentration at 600 nm was between 0.5 and 2.0. Furthermore, the values of qo2max and Km of AOX versus oxygen consumption, which were 0.409 s-1 and 0.16 respectively, were calculated. It is a simple and sensitive and feasible method for quick measuring of AOX.

Implication of DNA-dependent protein kinase in an early, essential, local phosphorylation event during end-joining of DNA double-strand breaks in vitro.

Previous work with Xenopus egg extracts suggested that a wortmannin-sensitive protein phosphorylation event precedes both the removal of modified termini from DNA double-strand break ends and the joining of unmodified ends. To assess the possible role of DNA-dependent protein kinase in effecting this phosphorylation, both DNA end-joining and DNA-stimulated phosphorylation were examined in the presence of various inhibitors. Linear but not supercoiled DNA stimulated the phosphorylation of several endogenous proteins in the extracts, including species of approximately 48, 87, and 96 kDa. This phosphorylation was selectively suppressed by the kinase inhibitors wortmannin, dimethylaminopurine, and LY294002, with a dose response that in each case paralleled the inhibition of DNA end-joining. If wortmannin was added while the end-joining reaction was in progress, end-joining of DNA already present in the reaction continued for some time, but newly added DNA was not joined or processed at all. Ends with 3'-hydroxyl termini were joined much faster than those with 3'-phosphoglycolate termini, although both were equally effective in stimulating protein phosphorylation. The results support a role for DNA-dependent protein kinase in regulating end-joining in vitro, and suggest that at least one of the necessary phosphorylations involves a protein bound at or near the DNA end to be joined. In contrast, nuclear extracts from human cells joined double-strand breaks with normal but not modified termini, and the joining was unaffected by kinase inhibitors, suggesting that the dominant mechanism of end-joining in these extracts did not involve DNA-PK.

End-joining of free radical-mediated DNA double-strand breaks in vitro is blocked by the kinase inhibitor wortmannin at a step preceding removal of damaged 3' termini.

Both mammalian cells and Xenopus eggs possess activities for the joining of nonhomologous DNA ends, and such activities may play a major role in double-strand break repair. In order to dissect the biochemical processing of breaks with oxidatively modified ends, vectors containing various site-specific double-strand breaks with 3'-phosphoglycolate termini were constructed and treated with Xenopus egg extracts. These vectors were rejoined by the extracts at rates 30-100 times slower than comparable 3'-hydroxyl vectors. Vectors with blunt or cohesive 3'-phosphoglycolate ends yielded single repair products corresponding to simple phosphoglycolate removal followed by ligation, while a vector with mismatched ends was also rejoined but yielded a mixture of products. Addition of the kinase inhibitors wortmannin and dimethylaminopurine not only blocked rejoining, but also suppressed phosphoglycolate removal, implying an early, essential, kinase-dependent restriction point in the pathway. The results suggest that double-strand breaks with oxidatively modified ends are repaired in Xenopus eggs by a highly conservative and stringently regulated end-joining pathway, in which all biochemical processing of the breaks is contingent on both end alignment and a specific phosphorylation event. Several lines of indirect evidence suggest DNA-dependent protein kinase as a likely candidate for effecting this phosphorylation.

Construction of a vector containing a site-specific DNA double-strand break with 3'-phosphoglycolate termini and analysis of the products of end-joining in CV-1 cells.

Previous studies have shown that linearized SV40-based shuttle vectors transfected into mammalian cells are efficiently recircularized by an error-prone end-joining pathway. To determine whether and with what specificity free radical-mediated double-strand breaks are rejoined by this pathway, a structural mimic of such a break was introduced at a specific site in an SV40-based shuttle vector, by ligating purified 3'-phosphoglycolate-terminated oligonucleotides into 3' recessed ends generated in the linearized vector. These terminally blocked linear vectors were efficiently repaired and replicated when transfected into simian CV-1 cells. Sequencing across the repair joints in progeny plasmid indicated that, for a blunt-ended vector, the most frequent mechanism of rejoining was splicing at a terminal 4-base homology; however, a significant fraction of the joints retained all bases from both ends of the break, consistent with a mechanism involving simple 3'-phosphoglycolate removal, followed by blunt-end ligation. For the analogous 3'-hydroxyl terminated break, the fraction of simple blunt-end ligations was considerably higher. For a phosphoglycolate-terminated vector with cohesive ends the most frequent repair mechanism was simple ligation of the annealed cohesive ends, presumably preceded by phosphoglycolate removal. For all these substrates, the remaining repair joints showed small or large deletions from one or both of the ends, usually with apparent annealing at short (1-4-base) homologies. The results suggest that while breaks with 3'-phosphoglycolates can be repaired, these blocked termini represent a significant barrier to DNA end-joining, and can significantly alter its specificity. The presence of cohesive ends appears to improve markedly the fidelity of rejoining for terminally blocked double-strand breaks.

Bringing basic health care to the rural poor.

In the 1960s and 1970s China provided a good model for financing and delivering rural health services in developing countries. Since the implementation of economic reforms which began in the 1980s, however, health services in poor areas have been showing signs of declining, with fewer people able to pay for the services they need. This article examines major problems of access to basic health care faced by the poor, and proposes a strategy for improving health service delivery in the poor areas.

[Effect of anti-opioid peptide sera on oxytocin-induced enhancement of electroacupuncture analgesia].

The effects of intracerebroventricular injection of Anti-opioid peptide sera on oxytocin-induced enhancement of electroacupuncture (EA) analgesia were observed in this study. It was found that injection of anti-beta-endorphin serum (AEPS) alone could attenuate EA analgesia in rats. Injection of AEPS prior to intraventricular injection of oxytocin could not block the enhancement of EA analgesia by oxytocin. The antidynorphin A1-13 serum (ADYNS) alone could also reduce the EA analgesia, whereas injection of ADYNS prior to injection of oxytocin could potentiate the enhancement of EA analgesia by oxytocin. However, neither the anti-methionine enkephalin serum nor the anti-leucine enkephalin exerted any effect on the enhancement of EA analgesia by oxytocin. The above results showed that the dynorphin attenuate oxytocin-induced enhancement of EA analgesia, but Beta-endorphin and enkephalin do not affect this role of oxytocin. These data suggest the enhancement of EA analgesia by oxytocin is not dependent upon the endogenous pioid peptides in brain.

Modeling the effect of insurance on health expenditures in the People's Republic of China.

A pilot study was undertaken in two rural counties in Sichuan Province to determine the feasibility of offering health insurance to peasant families. Models of per capital inpatient and outpatient health care expenditures were developed using self-reported utilization from a survey of 880 households, supplemented by cost and utilization data from the providers in the counties. Expenditures at a facility were modeled as a function of level of insurance in three parts: (1) as the product of the probability of any use, (2) the expected number of visits given any use, and (3) the cost per visit at the facility. Output from the model for representative insurance plans is presented.

Changes in health conditions in the Huainan coal mine in the past three decades.

Health conditions in the Huainan Coal Mine have improved greatly since 1950. In particular, the work and living environments, the underground dust concentration, the water supply and waste disposal systems, and the nutritional value of miners' food have become much better. Thus the rates of occupational, infectious, and parasitic diseases and nutritional deficiency diseases have decreased considerably among miners. The decreases in the prevalence of anthracosilicosis and hookworm have been especially striking.
