Anisotropy and corotation of galactic cosmic rays.

The intensity of Galactic cosmic rays is nearly isotropic because of the influence of magnetic fields in the Milky Way. Here, we present two-dimensional high-precision anisotropy measurement for energies from a few to several hundred teraelectronvolts (TeV), using the large data sample of the Tibet Air Shower Arrays. Besides revealing finer details of the known anisotropies, a new component of Galactic cosmic ray anisotropy in sidereal time is uncovered around the Cygnus region direction. For cosmic-ray energies up to a few hundred TeV, all components of anisotropies fade away, showing a corotation of Galactic cosmic rays with the local Galactic magnetic environment. These results have broad implications for a comprehensive understanding of cosmic rays, supernovae, magnetic fields, and heliospheric and Galactic dynamic environments.

The contribution from distant dust sources to the atmospheric particulate matter loadings at XiAn, China during spring.

Mass concentration data for PM(10) (particulate matter, PM, less than 10 mum) combined with an air mass back-trajectory clustering technique, a potential source contribution function (PSCF) model, and a concentration-weighted trajectory (CWT) method were used to evaluate the transport pathways and sources of XiAn PM(10) in spring 2001 to 2003. Three dust source areas: "Northwesterly Sources," "Northerly Sources," and a "Loess Plateau Source" and an anthropogenic "Southerly Source" contributing to the high particulate matter concentrations at XiAn were identified using these methods. The CWT method provided more compelling information on dust sources than the PSCF model, but there are clear advantages to using multiple interpretive tools. A comparison of the major dust transport pathways shows differences for XiAn versus Beijing, with "Northwesterly Sources" more important for XiAn and arid and semi-arid regions in Mongolia more important for Beijing.

Combined amino acid mutations occurring in the envelope closely correlate with pathogenicity of EIAV.

The Chinese equine infectious anemia virus (EIAV) donkey-leukocyte attenuated vaccine (DLV) provides a unique natural model system to study the attenuation mechanism and immunological control of lentivirus replication. Critical consensus mutations were identified between virulent Chinese EIAV strains and vaccine strains. Based on a full-length infectious clone of EIAV vaccine strain pLGFD3, two molecular clones, mFD5-4-7 and mFD7-2-11, were successfully constructed, in which 4 and 6 critical consensus mutations in the env gene of the vaccine strain were point-mutated to the wild-type sequence, respectively by an overlap PCR mutagenesis strategy. The infectivity, virulence, and pathogenesis of the constructed clones were investigated in vitro using a reverse transcriptase assay, an indirect immunofluorescence assay, observation of cytopathogenic effect, and virion observation as well as in vivo by inoculation of animals with the resulting infectious clones. The pathogenic symptoms in horses inoculated with mFD7-2-11 were more severe than those inoculated with mFD5-4-7, whereas no pathogenic symptoms were detected in animals inoculated with their parental clone pLGFD3 strain. The results indicate that the consensus mutation residues of the env region involved in this study play significant roles in the virulence and pathogenicity of EIAV. This will contribute to the elucidation of the attenuating and protective mechanisms of the Chinese EIAV vaccine.

Spectroscopic analysis of iron-oxide minerals in aerosol particles from northern China.

Diffuse reflectance spectrometry was used to study iron-oxide minerals and to investigate the reflectance characteristics of eolian dust collected during the spring of 2001 and 2002 on bulk filters from three sites in northern China. The first derivatives of the reflectance spectra were consistent with signals from two iron-oxide minerals, hematite and goethite, at wavelengths of 565 and 435 nm, respectively, and these values varied with the iron concentrations in the samples. The percent reflectances for the yellow, orange and red bands increased with the iron concentrations and with the first derivative values representing hematite and goethite while those for violet, blue and green bands decreased correspondingly. The results show that iron-oxide minerals play an important role in determining the aerosol particles' color and reflectance properties. Moreover, the relative amounts of the two iron-oxides in Asian dust apparently differ from those in African dust, suggesting that the iron-oxides may provide another tool for tracing the origins of eolian dust on a global scale.

An approach for isolating high-molecular-weight glutenin subunit genes using monoclonal antibodies.

High-molecular-weight glutenin subunits (HMW-GSs) play an important role in the breadmaking quality of wheat flour. In China, cultivars such as Triticum aestivum 'Xiaoyan No. 6' carrying the 1Bx14 and 1By15 glutenin subunits usually have attributes that result in high-quality bread and noodles. HMW-GS 1Bx14 and 1By15 were isolated by preparative sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) and used as an antigen to immunize BALB/c mice. A resulting monoclonal antibody belonging to the IgG1 subclass was shown to bind to all HMW-GSs of Triticum aestivum cultivars, but did not bind to other storage proteins of wheat seeds in a Western blot analysis. After screening a complementary DNA expression library from immature seeds of 'Xiaoyan No. 6' using the monoclonal antibody, the HMW-GS 1By15 gene was isolated and fully sequenced. The deduced amino acid sequence showed an extra stretch of 15 amino acid repeats consisting of a hexapeptide and a nonapeptide in the repetitive domain of this y-type HMW subunit. Bacterial expression of a modified 1By15 gene, in which the coding sequence for the signal peptide was removed and a BamHI site eliminated, gave rise to a protein with mobility identical to that of HMW-GSs extracted from seeds of 'Xiaoyan No. 6' via SDS-PAGE. This approach for isolating genes using specific monoclonal antibody against HMW-GS genes is a good alternative to the extensively used polymerase chain reaction (PCR) technology based on sequence homology of HMW-GSs in wheat and its relatives.

Molecular and biochemical characterization of puroindoline a and b alleles in Chinese landraces and historical cultivars.

Kernel hardness that is conditioned by puroindoline genes has a profound effect on milling, baking and end-use quality of bread wheat. In this study, 219 landraces and 166 historical cultivars from China and 12 introduced wheats were investigated for their kernel hardness and puroindoline alleles, using molecular and biochemical markers. The results indicated that frequencies of soft, mixed and hard genotypes were 42.7, 24.3, and 33.0%, respectively, in Chinese landraces and 45.2, 13.9, and 40.9% in historical cultivars. The frequencies of PINA null, Pinb-D1b and Pinb-D1p genotypes were 43.8, 12.3, and 39.7%, respectively, in hard wheat of landraces, while 48.5, 36.8, and 14.7%, respectively, in historical hard wheats. A new Pinb-D1 allele, designated Pinb-D1t, was identified in two landraces, Guangtouxianmai and Hongmai from the Guizhou province, with the characterization of a glycine to arginine substitution at position 47 in the coding region of Pinb gene. Surprisingly, a new Pina-D1 allele, designated Pina-D1m, was detected in the landrace Hongheshang, from the Jiangsu province, with the characterization of a proline to serine substitution at position 35 in the coding region of Pina gene; it was the first novel mutation found in bread wheat, resulting in a hard endosperm with PINA expression. Among the PINA null genotypes, an allele designed as Pina-D1l, was detected in five landraces with a cytosine deletion at position 265 in Pina locus; while another novel Pina-D1 allele, designed as Pina-D1n, was identified in six landraces, with the characterization of an amino acid change from tryptophan-43 to a 'stop' codon in the coding region of Pina gene. The study of puroindoline polymorphism in Chinese wheat germplasm could provide useful information for the further understanding of the molecular basis of kernel hardness in bread wheat.

Application of support vector machine (SVM) for prediction toxic activity of different data sets.

As a new method, support vector machine (SVM) were applied for prediction of toxicity of different data sets compared with other two common methods, multiple linear regression (MLR) and RBFNN. Quantitative structure-activity relationships (QSAR) models based on calculated molecular descriptors have been clearly established. Among them, SVM model gave the highest q(2) and correlation coefficient R. It indicates that the SVM performed better generalization ability than the MLR and RBFNN methods, especially in the test set and the whole data set. This eventually leads to better generalization than neural networks, which implement the empirical risk minimization principle and may not converge to global solutions. We would expect SVM method as a powerful tool for the prediction of molecular properties.

Study of probabilistic neural networks to classify the active compounds in medicinal plants.

Probabilistic neural networks (PNNs) were utilized for the classifications of 102 active compounds from diverse medicinal plants with anticancer activity against human rhinopharyngocele cell line KB. Molecular descriptors calculated from structure alone were used to represent molecular structures. A subset of the calculated descriptors selected using factor correlation analysis and forward stepwise regression was used to construct the prediction models. Linear discriminant analysis (LDA) was also utilized to construct the classification model to compare the results with those obtained by PNNs. The accuracy of the training set, the cross-validation set, and the test set given by PNNs and LDA were 100, 92.3, 90.9% and 71.8, 92.3, 54.5%, respectively, which indicated that the results obtained by PNNs agree well with the experimental values of these compounds and also revealed the superiority of PNNs over LDA approach for the classification of anticancer activities of compounds. The models built in this work would be of potential help in the design of novel and more potent anticancer agents.

Photodynamic therapy of human undifferentiated thyroid carcinoma-bearing nude mice using topical 5-aminolevulinic acid.

OBJECTIVE: We determined and evaluated the effect of photodynamic therapy (PDT) using topical 5-aminolevulinic acid (ALA) cream in treating human undifferentiated thyroid carcinoma (UTC)-bearing nude mice. BACKGROUND DATA: UTC constitutes almost 10% of thyroid cancers and shows a very poor response to chemotherapy. Today PDT with topical ALA is successfully used for the treatment of a variety of neoplastic and non-neoplastic diseases. MATERIALS AND METHODS: UTC tumor was implanted in the right flank of the nude mice after anesthesia. When the actual tumor size reached about 345 mm3, all mice were divided randomly into a treatment group (n = 10) and a control group (n = 10). ALA cream was applied to tumors in treated mice and left on for 6 has a retention interval; then the tumors were irradiated by a 633-nm diode laser with an incident dose of 60 J/cm2. The tumor volumes were measured 7, 14, and 21 days after PDT in both groups. The percentage of tumor growth delay after PDT was calculated. All mice were sacrificed and the tumor weights were determined 21 days after PDT. RESULTS: Mean tumor volumes on 21 days after PDT were 1141.19 +/- 153.74 mm3 in the treated group and 2370.75 +/- 300.42 mm3 in the control group. The percentage of tumor growth delay was -48.13 +/- 13.76 7 days after PDT, -54.9 +/- 8.83 14 days after PDT, and -51.11 +/- 8.95 21 days after PDT. Mean tumor weights 21 days after PDT were 1.78 +/- 0.32 g in the treated group and 3.60 +/- 0.60 g in the control group. There were significant differences (p < 0.001) in mean tumor volume and weight between the treated and control groups after PDT. CONCLUSIONS: PDT is effective in delaying the growth of UTC-bearing nude mice using topical ALA cream and laser light with inappropriate parameters. ALA cream as a topically applied photosensitizer was effective, safe, and convenient in PDT.

Observation by an air-shower array in Tibet of the multi-TeV cosmic-ray anisotropy due to terrestrial orbital motion around the Sun.

We report on the solar diurnal variation of the galactic cosmic-ray intensity observed by the Tibet III air shower array during the period from 1999 to 2003. In the higher-energy event samples (12 and 6.2 TeV), the variations are fairly consistent with the Compton-Getting anisotropy due to the terrestrial orbital motion around the Sun, while the variation in the lower-energy event sample (4.0 TeV) is inconsistent with this anisotropy. This suggests an additional anisotropy superposed at the multi-TeV energies, e.g., the solar modulation effect. This is the highest-precision measurement of the Compton-Getting anisotropy ever made.

Formation of thermal vacancies on the Si sublattice of the intermetallic compound MoSi2.

For a detailed understanding of high-temperature processes in complex solids the identification of the sublattice on which thermal defects are formed is of basic interest. Theoretical studies in intermetallic compounds favor a particular sublattice for thermal vacancy formation. In the present study we detect in ordered MoSi2 thermal vacancies with a low formation enthalpy of H(F)(V)=(1.6+/-0.1) eV, and we succeed in showing by experimental and theoretical efforts that they are preferentially formed on the Si sublattice. By these data self-diffusion in MoSi2 can be understood.

The comparison of effects between pulsed and CW lasers on wound healing.

OBJECTIVE: In order to evaluate the effects of pulsed continuous wave (CW) laser and detect the role of wound healing in rats using both pulsed and CW 635-nm low-level laser therapy (LLLT), a pilot study was undertaken. BACKGROUND DATA: Some acceleration effects of wound healing on animals were found after treatment using various lasers with CW. There are other reports, however, using pulsed CW laser to evaluate the effects of wound healing in rats. MATERIALS AND METHODS: An elliptic wound was created aseptically with a scalpel on the shaved back of the rats after anesthesia. The rats treated were restrained in a Plexiglas cage without anesthesia during the laser irradiation period. An Erchonia pulse laser (635 nm) was used in the experiment. The laser beam was delivered through an expander. The percentage of relative wound healing was calculated. RESULTS: The percentage of relative wound healing was 4.32 in 100 Hz, 3.21 in 200 Hz, 3.83 in 300 Hz, 2.22 in 400 Hz, 1.73 in 500 Hz and 4.81 in CW. CONCLUSION: LLLT using pulsed, CW laser at the appropriate dosimetry and frequency can provide acceleration in wound healing in rats. The 100-Hz frequency had a better effect than other pulse frequencies used in the study. The effects of treatment using CW laser was higher than pulse frequency. The frequency of pulsed CW laser was not found to increase wound healing in rats compared with normal CW laser, as reported in our previous studies.

Analysis of lewis antigens on cell surface and alpha1,3 fucosyltransferase subtypes in H7721 human hepatocarcinoma cells.

The expressions of Lewis antigens on H7721 human hepatocarcinoma cells were detected with monoclonal antibodies and flow-cytometry. It was found that H7721 mainly expressed SLex, and a small amount of SDLex, but Lex and SLea was negligible. The monoclonal antibody of SLex (KM93) significantly blocked the adhesion of H7721 cells to human umbilical vein epithelial cells, as well as cell migration and invasion, but the blocking effect of SDLex antibody (FH6) was not statistically significant. The expressions of five subtypes of alpha1,3fucosyltransferases (alpha1,3FucTs), the enzyme responsible for the fucosylation step in Lewis antigen synthesis, were also studied using real-time RT-PCR. The expression of FucT mRNAs were FucT-IV > FucT-III > FucT-VI > or = FucT-VII > FucT-IX. FucT-VI is supposed to be the main enzyme responsible for the synthesis SLex and SDLex. FucT-VII and III may also participate in SLex synthesis. The absence of FucT-IX expression and FucT-III not being the rate-limiting enzyme for SLea synthesis may be the reasons for the negligible expressions of Lex and SLea on the cell surface, respectively.

Field assessment of recombinant Schistosoma japonicum 26 kDa glutathione S-transferase in Chinese water buffaloes.

We have shown previously that anti-fecundity immunity can be induced experimentally against recombinant 26 kDa glutathione S-transferase (reSjc26GST) in Chinese water buffaloes (Bos buffelus), important reservoir hosts for Schistosoma japonicum in China. In the field study described here, we immunized buffaloes with reSjc26GST to induce protective immunity against S. japonicum and to evaluate its effectiveness in controlling schistosomiasis japonica. We selected two villages as test and control groups in inside-embankment areas endemic for schistosomiasis japonica. The buffaloes in the test village were vaccinated with reSjc26GST, whereas those in the control village were not. The indicators of the effect of the vaccine included the generation of specific IgG antibodies in the vaccinated buffaloes, changes in the prevalence and infection intensity in buffaloes and village children, changes in the density of infected snails, and changes in the infectivity of water bodies (assessed by sentinel mice) in transmission areas adjacent to both villages. Twenty months after vaccination, the infection rate of buffaloes in the test village was decreased by 60.4% (from an initial prevalence of 13.5% to 5.4%), and 67.9% when compared with that in the control village (initial prevalence of 16.7%). However, the infection rate in village children remained unchanged. The density of infected snails decreased by 71.4%, from 0.0049/0.11 m2 to 0.0014/0.11m2 in the high transmission area outside the embankment in the test village. There was no change in the infectivity of the water body transmission areas between the test and control villages. The levels of specific antibodies to reSjc26GST showed a continuous increase after vaccination. These results indicate that protective immunity was induced and maintained in buffaloes after vaccination with reSjc26GST. The vaccine could thus play a significant role in reducing S. japonicum transmission caused by water buffaloes in the Lake region of China.

An estimation of the minimum number of SSR alleles needed to reveal genetic relationships in wheat varieties. I. Information from large-scale planted varieties and cornerstone breeding parents in Chinese wheat improvement and production.

The genetic relationships of 43 wheat varieties were analyzed with SSR markers. The materials employed included 14 cornerstone breeding parents used before 1980 and another 29 other large-scale planted varieties currently in use in China. A total of 501 different alleles were amplified, including 166 alleles of the A genome, 174 of the B genome and 161 of the D genome. Data obtained were used to estimate genetic similarity using the DICE coefficient, and dendrograms were constructed using the UPGMA method. The dendrogram with 501 alleles successfully differentiated all the cornerstone breeding parents and the large-scale planted varieties, and the dendogram tree was basically consistent with the pedigrees of these varieties. The correlation coefficient between the genetic distance matrix of 501 alleles and that of 450 was 0.99. Correlation coefficients among random samples of alleles suggested that 350 to 400 alleles were needed to detect genetic relationships among common wheat varieties. Correlation coefficients of a genetic similarity matrix based on 580, and those of 501 and 400, random alleles were 0.96 and 0.94, respectively. However, there were marked differences between the matrix based on the 501 alleles and those based on markers located on the A-, B- or D-genome independently. The correlation coefficients between the genetic distance matrix of 501 alleles and alleles within A, B or D genomes were 0.77, 0.76 and 0.67. The estimation of genetic similarity should be based on data from all genomes rather than any one genome.

Normal and SV40 transfected human peritoneal mesothelial cells produce IL-6 and IL-8: implication for gynaecological disease.

High levels of interleukin-6 (IL-6) and interleukin-8 (IL-8) have been demonstrated in the peritoneal fluid of benign and malignant gynaecological disease. Peritoneal monocytes and macrophages, endometrial cells, endometrial and peritoneal stromal cells and tumour cells produce these cytokines in vitro. To investigate whether normal human peritoneal mesothelial cells (HPMC) produce IL-6 and IL-8, HPMC were isolated from omental biopsies. Primary HPMC (P-HPMC) were transfected with pSV3-neo encoding SV40 large T antigen (T-HPMC) to generate sufficient cells. T-HPMC preserved the characteristics of P-HPMC as assessed by phase contrast microscopy, electron microscopy, immunocytochemistry and flow cytometry (FACS) analysis. T-HPMC retained a stable phenotype up to passage 14-19, whereas P-HPMC proliferated poorly and became senescent by passage 4-6. T-HPMC and P-HPMC constitutively expressed IL-6 and IL-8 at both protein and mRNA level. IL-6 and IL-8 production was stimulated by recombinant human interleukin-1beta (hIL-1beta) or human tumour necrosis factor-alpha (hTNF-alpha) alone in a dose-dependent manner. Moreover, hIL-1beta or hTNF-alpha up-regulated IL-6 and IL-8 gene expression as determined by competitive PCR. In contrast, human interferon-gamma (hIFN-gamma) or lipopolysaccharide (LPS) showed no effect. These data indicate that (1) T-HPMC lines mimic the morphological and functional features of P-HPMC, (2) P-HPMC and T-HPMC constituitively produce IL-6 and IL-8, which is enhanced by hIL-1beta and hTNF-alpha and (3) HPMC in vivo may participate in the pathogenesis of benign and malignant gynaecological disease.

The value of MG7 antigen in predicting cancerous change in dysplastic gastric mucosa.

The aim of this study was to ascertain whether MG7Ag is a useful predictor of evolution of gastric dysplasia to carcinoma. A total of 1090 patients with confirmed dysplasia were stained immunohistochemically with MG7 monoclonal antibody by the ABC method. A prospective follow-up study was undertaken on 19 patients with MG7Ag positive staining and 16 with MG7 negative staining over a period of 10-78 months. The expression of MG7Ag was also compared in another two groups by conducting retrospective studies. One group showed an evolution into gastric cancer over 2-4 years, the other did not. Quantitative analysis of MG7Ag expression was carried out on the last two groups. The receiver operating characteristic curve and Youden index were used to assess the best critical value for MG7Ag. MG7Ag was found positive in 456/1090 cases (41.8%) with dysplasia. Prospective follow-up of 35 patients showed that 6/19 patients with MG7Ag positive staining developed gastric cancer, but there were no carcinomatous changes in 16 patients with MG7 negative staining. The results of MG7Ag expression in 72 cases with retrospective follow-up showed there were 24 with positive immunostaining among 34 cancerous cases (70.6%), and only 7 in 38 non-cancerous cases (18.4%) (p<0.01). Image analysis showed that an average MG7Ag density index ++0.19 could be regarded as the critical value for high risk of gastric mucosa with dysplasia evolving to cancer. Positive MG7Ag expression in gastric mucosa of patients with dysplasia, especially in cases with a density index ++0.19, was an indicator of high risk of malignant change.
