Nuclear resonance scattering of synchrotron radiation by 40K

We have observed the excitation of the 29.83 keV nuclear level of the radioactive 40K nuclide, which level is not populated by any radioactive source, by synchrotron radiation for the first time. The absolute energy of the level of 29.834+/-0.011 keV and the half-life of 4.13+/-0.12 ns were obtained from our measurements. Our success in observing the nuclear excitation of radioactive 40K in a KCl powder sample shows the possibility for study of the local vibrational states and the electronic states of potassium in various areas of research.

Expression of the fusion protein recombinant human granulocyte-macrophage colony stimulating factor and leukemia inhibitory factor in a baculovirus vector system.

A fusion gene coding human granulocyte-macrophage colony stimulating factor (GM-CSF) and leukemia inhibitory factor (LIF) cDNAs was inserted into the transfer vector pSXIVVI+ X3 with the control of Syn and XIV promoters. The Sf9 cells (Spodoptera frugiperda) were co-transfected with the recombinant plasmid and TnNPV DNA (Trichoplusia ni nuclear polyhedrosis virus DNA). The fusion protein recombinant human granulocyte-macrophage colony stimulating factor (GM-CSF) and leukemia inhibitory factor (LIF) could be synthesized in cells infected with recombinant virus at a level of about 23% of their total cellular protein. Activity analysis of the fusion protein in infected cells revealed that it exhibited the dual activities of GM-CSF and LIF. Western blot analysis of the expressed fusion protein in infected larvae showed that the virus-mediated fusion protein, with a molecular weight of approximately 35 kDa, is confirmed with immunoreactivity.

Dynamics and stability analysis of the growth and astaxanthin production system of Haematococcus pluvialis.

This paper investigated high cell density cultivation of Haematococcus pluvialis for astaxanthin production in 3.7-L bioreactors. A biomass concentration of 2.74 g L-1and an astaxanthin yield of 64.4 mg L-1 were obtained. Based on the experimental results, a new and simple dynamic model is proposed, differing from Monod kinetics, to describe cell growth, product formation and substrate consumption. Good agreement was found between the model predictions and experimental data. The model revealed that there was cell growth inhibition on product formation and product feedback compensation for substrate consumption, but no substrate inhibition or product inhibition of cell growth. Stability analysis demonstrated that no multiplicity of steady states was observed; the unique positive steady state was locally asymptotically stable; and the effect of dilution rate on steady states was greater than that of the initial substrate concentration.
