Synthesis and structure-activity relationships of new benzodioxinic lactones as potential anticancer drugs.

A set of disubstituted tetracyclic lactones has been synthesized and tested for potential antitumor activity. Several of them possess a noticeable cytotoxicity against L1210 and HT-29 colon cells in vitro. Relationships between chain nature and biological properties were sought. Lactones with a pentyl or hexyl substituent at C-11 are the most active ones. The introduction of a functional group at the side chain of C-11 modified the potency; carboxylic acid and primary amine decreased the cytotoxicity, whereas a cyano group increased the activity. An extensive structure-activity relationship study of these derivatives, including carbon homologues and bioisosteres has been performed. The synthesis and cytotoxicity of these compounds are discussed. Two lactones are recognized as potential lead compounds.

A singular noninterpenetrating coordination polymer with the Pt3O4 structure containing naked [Na+]4 units.

The homoleptic low-spin complex [Fe(L)3]2+ where L is the bisbidentate ligand 1,10-phenanthroline-5,6-dione, coordinates Na+ ions via exo-oriented dione groups defining a three-dimensional cationic network {[Fe(L)3]4Na3}11+}n with Pt3O4 topology. The large volume generated by the network is filled with 11 perchlorate ions, 7 "NaClO4" ionic pairs, and 9 H2O molecules. Singular [Na+]4 units, in which the Na+ ions are practically uncoordinated, are formed.

Crystal structure and DNA interaction of the facial-type rac-tris[2-(aminomethyl)benzimidazole]cobalt(III) complex.

rac-[Co(ambi)3]Cl3.3H2O corresponds to a tris-chelate complex bearing 2-(aminomethyl)benzimidazole (ambi) ligands bound to the Co(III) ion in a facial-type fashion. The high metal q/r ratio and electronic delocalization between coordination heterocyclic functionalities can provide powerful tools to minimize DNA condensation, which arises from the neutralization of the negatively charged sugar phosphate backbone in the presence of multivalent [Co(ambi)3]3+.

Influence of the position of substituents in the cytotoxic activity of trans platinum complexes with hydroxymethyl pyridines.

The synthesis and chemical characterization of two trans platinum complexes, (1) trans-[PtCl(2)NH(3)(2-hydroxymethylpyridine)] and (2) trans-[PtCl(2)NH(3)(3-hydroxymethylpyridine)], are described. The structures and chemical behaviour of these compounds have been compared to those of their isomer (3) trans-[PtCl(2)NH(3)(4-hydroxymethylpyridine)] previously studied. X-ray structures of all of them were solved and some interesting differences were found. The values of the dihedral angle (85 degrees , 57 degrees and 42 degrees for 1, 2 and 3, respectively) demonstrate how important is the position of substituent from a structural point of view. Studies of circular dichroism (CD), electrophoretic mobility (EM) in agarose gel and atomic force microscopy (AFM) showed differences in the modifications caused by the three complexes on DNA. Studies of antiproliferative activity of complexes 1 and 2 against cell tumour lines (HL-60) and apoptosis assays have also been carried out, showing that 1 as well as 2 are far less active than the previously described complex 3 (IC(50)=19; 19 and 3 microM, respectively). This fact probes that slight modifications on the drug's design may generate significant differences in the final antitumour activity by modifying the DNA-drug adducts, performance of resistance mechanisms and all the factors that play a fundamental role in Pt complexes' cytotoxicity.

Isomeric distribution and catalyzed isomerization of cobalt(III) complexes with pentadentate macrocyclic ligands. importance of hydrogen bonding.

We have investigated the isomeric distribution and rearrangement of complexes of the type [CoXLn]2+,3+ (where X = Cl-, OH-, H2O, and Ln represents a pentadentate 13-, 14-, and 15-membered tetra-aza or diaza-dithia (N4 or N2S2) macrocycle bearing a pendant primary amine). The preparative procedures for chloro complexes produced almost exclusively kinetically preferred cis isomers (where the pendant primary amine is cis to the chloro ligand) that can be separated by careful cation-exchange chromatography. For L13 and L14 the so-called cis-V isomer is isolated as the kinetic product, and for L15 the cis-VI form (an N-based diastereomer) is the preferred, while for the L14(S) complex both cis-V and trans-I forms are obtained. All these complexes rearrange to form stable trans isomers in which the pendent primary amine is trans to the monodentate aqua or hydroxo ligand, depending on pH and the workup procedure. In total 11 different complexes have been studied. From these, two different trans isomers of [CoClL14(S)]2+ have been characterized crystallographically for the first time in addition to a new structure of cis-V-[CoClL14(S)]2+; all were isolated as their chloride perchlorate salts. Two additional isomers have been identified and characterized by NMR as reaction intermediates. The remaining seven forms correspond to the complexes already known, produced in preparative procedures. The kinetic, thermal, and baric activation parameters for all the isomerization reactions have been determined and involve large activation enthalpies and positive volumes of activation. Activation entropies indicate a very important degree of hydrogen bonding in the reactivity of the complexes, confirmed by density functional theory studies on the stability of the different isomeric forms. The isomerization processes are not simple and even some unstable intermediates have been detected and characterized as part of the above-mentioned 11 forms of the complexes. A common reaction mechanism for the isomerization reactions has been proposed for all the complexes derived from the observed kinetic and solution behavior.

Combined effects of diamines and carboxylate bridges on structural and magnetic properties of a series of polynuclear copper(II) complexes with 1,1-cyclobutanedicarboxylic acid.

Six new copper(II) complexes of formula [Cu(mu-cbdca)(H2O)]n (1) (cbdca = cyclobutanedicarboxylate), [Cu2(mu-cbdca)2(mu-bipy)2]n (2) (bipy = 4,4'-bipyridine), [Cu(mu-cbdca)(mu-bpe)]n (3) (bpe = 1,2-bis(4-pyridyl)ethane), [Cu(mu-cbdca)(bpy)]2 (4) (bpy = 2,2'-bipyridine), [Cu(terpy)(ClO4)]2(mu-cbdca).H2O (5) (terpy = 2,2':6',2' '-terpyridine), and [Cu(cbdca)(phen) (H2O)].2H2O (6) (phen = 1,10-phenanthroline) were obtained and structurally characterized by X-ray crystallography. Complex 1 is a two-dimensional network with a carboxylate bridging ligand in syn-anti (equatorial-equatorial) coordination mode. Complexes 2 and 3 are formed by chains through syn-anti (equatorial-apical) carboxylate bridges, linked to one another by the corresponding amine giving two-dimensional nets. Complexes 4 and 5 are dinuclear, with the copper ions linked by two oxo (from two different carboxylate) bridging ligands in 4 and with only one carboxylate showing the unusual bis-unidentate mode in complex 5. Complex 6 is mononuclear, with the carboxylate linked to copper(II) in a chelated form. Intermolecular hydrogen bonds and pi-pi stacking interactions build an extended two-dimensional network. Magnetic susceptibility measurements of complexes 1-5 in the temperature range 2-300 K show the occurrence of weak ferromagnetic coupling for 1 and 4 (J = 4.76 and 4.44 cm(-1), respectively) and very weak antiferromagnetic coupling for 2, 3, and 5 (J = -0.94, -0.67, and -1.61 cm(-1), respectively). Structural features and magnetic values are compared with those reported for the similar copper(II) malonate and phenylmalonate complexes.

Synthesis, crystal structure, and magnetic studies of one-dimensional cyano-bridged Ln3+-Cr3+ complexes with bpy as a blocking ligand.

The reaction of Ln(NO3)3.aq with K3[Cr(CN)6] and 2,2'-bipyridine (bpy) in a water/ethanol solution led to two families of complexes: 4 one-dimensional (1D) complexes of the formula trans-[Cr(CN)4(mu-CN)2Ln(H2O)3(bpy)2]n.4nH2O.3.5nbpy (Ln3+ = La, Ce, Pr, and Nd) and 10 1D complexes of the formula trans-[Cr(CN)4(mu-CN)2Ln(H2O)4(bpy)]n.3.5nH2O.1.5nbpy (Ln3+ = Nd, Sm, Eu, Tb, Dy, Ho, Er, Tm, Yb, and Lu). The structures for the fourteen complexes [LaCr]n (1), [CeCr]n (2), [PrCr]n (3), [NdCr]n (4), [NdCr]n (4'), [SmCr]n (5), [EuCr]n (6), [TbCr]n (7), [DyCr]n (8), [HoCr]n (9), [ErCr]n (10), [TmCr]n (11), [YbCr]n (12), and [LuCr]n (13) have been solved. Complexes 1-4 crystallize in the orthorhombic space group Pbam and are isomorphous; complexes 4'-13 crystallize in the triclinic space group PI and are isomorphous. The X-ray structural characterization of complexes 1-4 shows the presence of a discrete decameric water cluster built around a cyclic hexameric core stabilized by the solid-state structure, which represents another new mode of association of water molecules. The Ln3+-Cr3+ magnetic interaction is negligible in 6 and 12, antiferromagnetic in 2, 4', 7, 8, 9, 10, and 11, and unresolved for 3. The complex 5 is a ferrimagnet because its magnetic studies suggest the onset of a very weak ferromagnetic three-dimensional ordering.

Copper(II) hexaaza macrocyclic binuclear complexes obtained from the reaction of their copper(I) derivates and molecular dioxygen.

Density functional theory (DFT) calculations have been carried out for a series of Cu(I) complexes bearing N-hexadentate macrocyclic dinucleating ligands and for their corresponding peroxo species (1c-8c) generated by their interaction with molecular O2. For complexes 1c-7c, it has been found that the side-on peroxodicopper(II) is the favored structure with regard to the bis(mu-oxo)dicopper(III). For those complexes, the singlet state has also been shown to be more stable than the triplet state. In the case of 8c, the most favored structure is the trans-1,2-peroxodicopper(II) because of the para substitution and the steric encumbrance produced by the methylation of the N atoms. Cu(II) complexes 4e, 5e, and 8e have been obtained by O2 oxidation of their corresponding Cu(I) complexes and structurally and magnetically characterized. X-ray single-crystal structures for those complexes have been solved, and they show three completely different types of Cu(II)2 structures: (a) For 4e, the Cu(II) centers are bridged by a phenolate group and an external hydroxide ligand. The phenolate group is generated from the evolution of 4c via intramolecular arene hydroxylation. (b) For 5e, the two Cu(II) centers are bridged by two hydroxide ligands. (c) For the 8e case, the Cu(II) centers are ligated to terminally bound hydroxide ligands, rare because of its tendency to bridge. The evolution of complexes 1c-8c toward their oxidized species has also been rationalized by DFT calculations based mainly on their structure and electrophilicity. The structural diversity of the oxidized species is also responsible for a variety of magnetic behavior: (a) strong antiferromagnetic (AF) coupling with J = -482.0 cm(-1) (g = 2.30; rho = 0.032; R = 5.6 x 10(-3)) for 4e; (b) AF coupling with J = -286.3 cm(-1) (g = 2.07; rho = 0.064; R = 2.6 x 10(-3)) for 5e; (c) an uncoupled Cu(II)2 complex for 8e.

Highly diastereoselective one-pot synthesis of spiro[cyclopenta[a]indene-2,2'-indene]diones from 1-indanones and aromatic aldehydes.

Treatment of 1-indanones with aromatic aldehydes and NaOEt in THF affords complex spiropolycyclic compounds through a four-component reaction in which two molecules of each starting compound are combined with formation of four new carbon-carbon bonds, leading to the elaboration of a new five-membered ring that bears five contiguous stereogenic centers with a well-defined relative configuration. Different amounts of a minor epimer of the main product are also formed. The presence of methoxy substituents in the indanone component and the use of aldehydes derived from pi-excedent heterocycles make the dimerization step a slower transformation. In these cases, better yields of spirodimers are obtained starting from the preformed enones. The reaction seems to take place by cross-aldol condensation, dehydration, and dimerization of the thus formed enones. The molecular mechanism of the dimerization reaction of enone 5g has been studied using DFT methods at the B3LYP/6-31G level. The dimerization takes place through a process involving a Michael addition of a carbanion, obtained by deprotonation of 5g at the 3-position, to a second molecule of 5g, followed by an intramolecular Michael addition in the corresponding intermediate. The final protonation of the resulting anion accounts for the formation of the cis-fused pentacyclic system.

Morpholine-2,5-dione.

In the title compound, C4H5NO3, the morpholine ring adopts a boat conformation that is distorted towards twist-boat, the boat ends being the two Csp3 atoms of the ring. The molecular packing is stabilized by the establishment of strong intermolecular NH...OC hydrogen bonds, which give rise to centrosymmetric dimers, and a network of weak CH2...OC hydrogen bonds, where each dimer interacts with eight neighbouring morpholinedione rings.

Complementarity and countercomplementarity in polynuclear copper(II) complexes with R2NCH2CH(OH)CH2NR2 (R = H, CH3): crystal structures and magnetic study.

The syntheses, structural characterization and magnetic behavior of five new copper(II) polynuclear compounds with formulae [Cu4(mu2-CH3COO)2(mu-bdmap)2(micro(1,5)-dca)2(dca)2(H2O)2] 1, [Cu2(mu2-CH3COO)(mu-bdap)(mu(1,1,5)-dca)(mu(1,3)-dca)]n 2, [Cu4(mu2-CH3COO)2(mu-bdmap)2(mu(1,1)-NCS)2(NCS)2] 3, [Cu2(mu2-CH3COO)(mu-bdap)(NCS)2] 4 and [Cu2(mu(1,3)-N3)(mu-bdmap)(N3)2]n 5 in which bdmapH is 1,3-bis(dimethylamino)-2-propanol, bdapH is 1,3-bis(amino)-2-propanol and dca is the anionic dicyanamide ligand, are reported herein. Tetranuclear complex 1 crystallizes in the monoclinic system, space group P2(1)/n, with unit cell parameters a = 8.284(8), b = 21.52(1), c = 11.432(3) A, beta = 105.19(2) degrees , Z = 2. Bi-dimensional complex 2 crystallizes in the triclinic system, space group P1, with unit cell parameters a = 8.184(5), b = 8.792(2), c = 10.887(2) A, alpha = 75.65(2), beta = 76.55(3), gamma = 74.36(3) degrees , Z = 2. Tetranuclear complex 3 crystallizes in the triclinic system, space group P1, with unit cell parameters a = 8.455(4), b = 9.114(9), c = 12.744(8) A, alpha = 104.62(8), beta = 99.86(6), gamma = 106.10(8) degrees, Z = 1. Dinuclear complex 4 crystallizes in the triclinic system, space group P1, with unit cell parameters a = 8.15(1), b = 8.18(2), c = 11.44(1) A, alpha = 69.39(2), beta = 80.36(2), gamma = 80.37(2) degrees , Z = 2. One-dimensional complex 5 crystallizes in the orthorhombic system, space group P2(1)2(1)2(1), with unit cell parameters a = 20.45(4), b = 11.36(3), c = 6.43(1) A, Z = 4. The magnetic behavior of all the complexes has been checked giving a bulk antiferromagnetic coupling in all the cases with |J| values in the range 109-144 cm(-1) for 1-4. Compound 5 is diamagnetic in the 2-300 K range of temperatures. The found J values 1-5 for can be justified from the structural data taking into account the orbital countercomplementarity for 1-4 and the orbital complementarity for 5.

Deamination of the amino acid fragment in imine metallacycles: unexpected synthesis of an NH-aldimine organometallic compound.

We report that the action of Lewis bases, such as triphenylphosphine, pyridine, or trimethylamine, on imine metallacycles derived from amino acids leads to the formation of the first organometallic compound of an NH aldimine, a highly reactive organic species, and the corresponding alpha-ketoester, in a deamination reaction that mimics the metabolism of alpha-amino acids. The synthesis of different cyclopalladated compounds by a reaction between palladium acetate and the Schiff bases 2,4,6-Me(3)C(6)H(2)CH=NCH(R(1))COOR(2) (R(1) = CH(2)Ph, R(2) = Et and R(1) = Ph, R(2) = Me) is also reported.

Slow interconversion of enantiomeric conformers or atropisomers of anilide and urea derivatives of 2-substituted anilines.

N-Acylated 2-substituted anilines undergo slow Ar-N bond rotation, allowing in some cases isolation of enantiomeric or diastereoisomeric atropisomers and in others the determination of the rate of Ar-N bond rotation by NMR. 2-Iodoanilides bearing a branched N-substituent demonstrate sufficient enantiomeric stability to be resolvable, either by HPLC or by formation of diastereoisomeric lactanilide derivatives. For the first time, the rates of Ar-N rotation in 2-substituted N,N'-diarylureas have been established: they mainly fall in the region of 50-70 kJ mol(-1) with a relatively weak dependence on substituent size.

Structural analysis of chiral complexes of palladium(0) with 15-membered triolefinic macrocyclic ligands.

The complete structural analysis of the palladium complexes of the triolefinic macrocycles (E,E,E)-1,6,11-tris(arylsulfonyl)-1,6,11-triazacyclopentadeca-3,8,13-trienes, which featured from three identical to three different aryl groups, was achieved by performing X-ray diffraction studies, NMR spectroscopy, and other calculations. The stereochemical complexity is determined by the different isomers formed through complexation of the metal to one or other face of each of the three olefins involved. The palladacyclopropane formulation of the palladium-olefin interaction offers a clear picture of the stereogenicity of the olefin carbon atoms that are complexed to the metal. The energetically favorable isomers were identified in the solid-state and in solution by performing X-ray diffraction and NMR spectroscopic analysis, respectively.

Direct, two-step synthetic pathway to novel dibenzo[a,c]phenanthridines.

Novel dibenzo[a,c]phenanthridines are prepared regioselectively by the application of a straightforward synthetic pathway, starting from new 3,4-diaryl- and 3,4-dihydro-3,4-diarylisoquinolines prepared via Ritter-type heterocyclization and the more classical two-step reductive amination/Bischler-Napieralski cyclization of triarylethanones, respectively. A comparative study of nonphenolic oxidative coupling methodologies provides a highly efficient procedure, based on the hypervalent iodine reagent phenyliodine(III) bis(trifluoroacetate) (PIFA), to accomplish the final coupling step.

Synthesis of novel palladium OCN-pincer complexes: unprecedented sequential C(sp3)-H activation and aerobic oxidation in the reaction of N,N-dialkyl-3-[(N,N-dimethylamino)methyl]-2-iodoanilines with Pd2(dba)3.

The reaction of N,N-dialkyl-3-[(N,N-dimethylamino)methyl]-2-iodoanilines with Pd2(dba)3 under O2 gives palladium OCN-pincer complexes by means of an unprecedented process that involves the formal aerobic oxidation of C(sp3)-H bonds at the alpha position of the aniline N atom.
