[Effect of extracted liquid from qianlietongyu on the proliferation and apoptosis on prostatic smooth muscle cells in vitro].

OBJECTIVE: To explore the effect of extracted liquid from Qianlietongyu on the proliferation or apoptosis on prostatic smooth muscle cells in vitro. METHODS: After extracted liquid from Qianlietongyu treated the cultured prostatic smooth muscle cells, the anti proliferative and apoptotic indices were assessed by MTT assy and terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL) respectively. RESULTS: There was a significant dose-effect relationship between the concentration of extracted liquid from Qianlietongyu and the antiproliferative index on prostatic smooth muscle cells in vitro (P < 0.01), but there was no markedly difference in the apoptosis index between the group of extracted liquid from Qianlietongyu and control group ( P > 0.05). CONCLUSION: Extracted liquid from Qianlietongyu may show significant antiproliferative effect on prostatic smooth muscle cells in vitro, without inducing apoptosis.

[Dynamic fugacity model for describing the fate of persistent organic pollutants in the river].

Dynamic model depending on temperature with fugacity approach is formulated which describes the fate of Persistent Organic Pollutants (POPs) in a river. The fugacity capacity, the degradation rate and transfer coefficients of pollutants are depending on temperature in the model. The model is illustrated by calculating the fate of gamma-HCH in Liao River. The results show that from 273K to 298K, the fugacity capacities in air, water and sediment of gamma-HCH are respectively reduced in 8.4%, 89.7% and 89.7%. However, in the same range of temperature, the degradation rate coefficients in air, water and sediment, and volatilization and diffusion rate coefficients of gamma-HCH are increased in 0.69, 22.0, 4.5, 7.9 and 1.6 times, respectively. The calculated value agree well with the field observed value in the order of magnitude, which shows that the model is appropriate for simulating the fate of POPs in a long time.

Lysophosphatidylcholine-induced elevation of asymmetric dimethylarginine level by the NADPH oxidase pathway in endothelial cells.

Recent studies suggested that endothelium is a main source of reactive oxygen species (ROS) and the major source was via NADPH oxidase pathway. Various stimuli including lysophosphatidylcholine (LPC), a major component of oxidized low-density lipoprotein (ox-LDL), can enhance the activity of NADPH oxidase and lead to a marked ROS generation. Asymmetric dimethylarginine (ADMA) is an endogenous nitric oxide (NO) synthase (NOS) inhibitor, which is synthesized by protein arginine methyltransferase I (PRMT I) and degraded by dimethylarginine dimethylaminohydrolase (DDAH) in endothelial cells. Much evidence showed that ADMA was closely related to endothelial dysfunction. Our previous study showed that LPC elevated ADMA level in endothelial cells via increasing oxidative stress, but the precise cellular mechanism is not defined yet. The present study was to explore the mechanism of NADPH oxidase in LPC-induced elevation of ADMA. In LPC-treated endothelial cells, the ROS production, cell viability, ADMA and NO levels, the activity of DDAH and expression of PRMT I were detected. Treatment with LPC (10 microg/ml) for 24 h markedly increased intracellular ROS production, the expression of PRMT I, level of ADMA, decreased the concentration of NO and the activity of DDAH. These effects were attenuated by diphenyliodonium, the NADPH oxidase inhibitor. In summary, the present results suggested that LPC-induced elevation of ADMA was due to reduction of DDAH activity and the up-regulation of PRMT expression by stimulation of ROS production via NADPH oxidase pathway.

Electro-optic polymer waveguide grating with fast tuning capability.

A novel fast tunable electro-optic (EO) polymer waveguide grating is proposed and designed. Its resonant wavelength can be linearly tuned via the first-order EO effect with a high sensitivity of 6.1 pm/V. We find that the spectrum characteristics of EO polymer waveguide gratings depend strongly on many grating parameters, such as refractive-index modulation, modulation function, grating period, and period number. Material selection, fabrication technology, EO tuning ability, and polarization dependence of EO polymer waveguide gratings are also discussed. Such a waveguide grating not only overcomes the disadvantages of fiber-optic gratings, such as slow wavelength tuning ability and large-scale integration inconvenience, but also has many advantages, such as high resonant-wavelength tuning sensitivity, the same fabrication technology used for semiconductors, and polarization independence.

New fluorescent chemosensor based on exciplex signaling mechanism.

[reaction: see text]. A novel fluorescent chemosensor (compound 1) containing aminonaphthol, which selectively recognizes fluoride anion with high sensitivity, was synthesized. The fluorescence of compound 1 was quenched rapidly by fluoride ion, and a new peak at a longer wavelength emerged concurrently, which constituted the signature for fluoride detection. The mechanism of exciplex formation was proposed for the interesting observation.

An effective fluorescent chemosensor for the detection of copper(II).

The fluorescent compound N-substituted 2,6-bis(benzimidazol-2-yl)pyidine (1) has been synthesized. The fluorescent characteristics of the compound 1 and 2,6-bis(benzimidazol-2-yl)pyidine (2) and the complexes formed between the two compounds and different metal ions have also been investigated. The results show that the compound 1 possesses a specific ability to form complex with Cu(2+) ion, but the compound 2 have not such a property. It is proposed that the specific recognition ability of compound 1 to Cu(2+) may be attributed to the cyclic configuration of this compound in polar solvent.

Large-scale fabrication and characterization of Cd-doped ZnO nanocantilever arrays.

We demonstrate bulk synthesis of highly crystalline Cd-doped ZnO nanocantilever arrays (CZNAs) using Cd and Zn powders at 600 degrees C, which is characterized via scanning electron microscopy, powder X-ray diffraction, X-ray photoelectron spectroscopy, transmission electron microscopy (TEM), energy dispersive X-ray spectroscopy, selected area electron diffraction, and high resolution TEM. The results show that the as-prepared CZNAs have diameters of about 15-50 nm, and lengths up to 400 nm and the corresponding process of growth is suggested for conventional vapor solid mechanism.

[Quantification of human ermap by using real-time FQ-PCR].

To develop a real-time FQ-PCR method for quantifying human ermap, a set of primers and a fluorescent probe were designed by primer express 2.0. pBluescriptSK(+) plasmid contained ermap cDNA was transcribed to generate calibration standards for quantification. A real time FQ-PCR method was established. The results showed that when the concentrations of DNA to be amplified were ranged from 1.725 x 10(7) to 1.725 x 10(10) cps/ml, there was a good correlation between template concentration and cycle threshold, and the correlation coefficient reached to -0.999376. In conclusion, real time FQ-PCR which is specific, sensitive and accurate can be used to further research on human ermap.

[Effects of early weaning on the blood glucose, blood lipid and hormone of rat fed a high fat diet].

OBJECTIVE: To observe the effects of early weaning on the blood glucose, blood lipid and hormone of rat fed a high fat diet. METHODS: Male Wistar rats were randomly divided into two groups after born 17 days according to body weight. One group was early weaned in the 17th day, the other was breastfed until the 24th day. After 4 weeks fed with basic diet, 12 rats were randomly selected from each group and were sacrificed. The other rats of each group were further randomly divided into 2 groups according to body weight. One group was fed a high fat diet, the other was fed the basic diet. The rats were sacrificed respectively at the end of the experiment. The blood glucose, serum lipids, and hormones were measured. RESULTS: Compared with the normal weaning rats, the weight and fat pad of early weaning rats significantly increased and the level of blood glucose, serum lipids, insulin, leptin also increased significantly (P < 0.05), the level of HDL, FT3 decreased (P < 0.05). CONCLUSION: The tendency of obesity in early weaning rats were more obvious than normal weaning rats after fed a high fat diet.

[Study on the expression of human ERMAP gene in erythropoietic and macrophage differentiation of K562 cells].

In order to investigate the potential of human ERMAP gene in erythroid cell differentiation, K562 cells were induced to erythroid lineage by Ara-C and to macrophage lineage by TPA, human ERMAP mRNA was detected by fluorescent quantitative PCR. The results showed that human ERMAP mRNA increased while K562 cells were induced to erythroid lineage after treatment with Ara-C at 2.5 x 10(-6) mmol/L/L and 1.0 x 10(-6) mmol/L/L. Human ERMAP mRNA not changed while K562 cells were induced to macrophage lineage after treatment with TPA at 2.0 x 10(-6) mmol/L/L and 1.0 x 10(-6) mmol/L/L. It is concluded that human ERMAP gene plays an important role in differentiation and proliferation of erythroid cells.

[Effects of different diet composition after weaning on the obesity in rats induced by high-energy diet].

OBJECTIVE: To explore the effects of different diet composition on the obesity in rats induced by high-fat diet after weaning. METHODS: Male Wistar rats weaned on 24 days were randomly divided into groups A, B and C and were assigned to high carbohydrate, i.e. basic diet, high protein, high non-saturated fat diets respectively for 3 weeks. Then all groups were fed with basic diet. After 2 weeks, rats from group A were randomly divided into groups A1 and A2, A1 was assigned to basic diet continuously and groups A2, B, C were fed with high-fat diet for 6 weeks. 8 rats from each group were sacrified at the end of different treatments to detect serum glucose, TG and hormones dynamicly. RESULTS: High nonsaturated fat diet after weaning significantly reduced the body weight, body fat content, increased the sensitiveness to insulin, the levels of glucagons, FT4, FT3, improved leptin resistance in rats fed with high-fat diet. High-protein diet also reduced body weight, body fat content and increase lipolysis in rats fed with high-fat diet, but blood glucose in which group was higher than in group A2. CONCLUSION: High non-saturated fat diet after weaning can significantly reduce the obesity of rats fed with high-fat diet.

[Expression of human ERMAP gene in different cell lines].

To investigate the pattern of human ERMAP gene expression in different cell lines, 15 cell lines derived from hematopoietic tumor, somatic tumor and normal tissue were chosen and were cultured; cells were harvested after culture for 12, 24, 36, 48, 60 and 72 hours; the expression of the human ERMAP was detected by using fluorescent quantitative PCR. The results showed that human ERMAP gene expression was positive in K562 cell line at interval of 12 and 24 hours, and the expression levels were (5.092 +/- 2.331) x 10(6) cps/microl RNA, (5.328 +/- 3.916) x 10(6) cps/microl RNA, respectively; ERMAP gene expression was also positive in ECV304 cell line at interval of 24 hours, and the expression level was (0.84 +/- 0.12) x 10(6) cps/microl RNA; and its expression was negative in other 13 cell lines. It is concluded that human ERMAP gene expression in ECV304 cell line was found first, and its expression in K562 cell line was further confirmed.

Morphological study on the megakaryocytes with nuclear extrusion and nucleocytoplasmic separation in four cases.

To investigate the morphological changes of megakaryocytes with nuclear extrusion and nucleocytoplasmic separation, the morphological characteristics of megakaryocytes in peripheral blood films, bone marrow smears, and bone marrow biopsies from 4 newly diagnosed patients with primary myelofibrosis (PMF), myelodysplastic syndrome (MDS), myeloblastic leukemia with maturation (M(2)) and erythroleukemia (M(6)) were studied by using light microscope. The results showed that many kinds of dysmegakaryocytes were observed in bone marrow smears of 4 cases, while in case A (PMF) and case D (M(6)) micromegakaryocytes were ripped apart; in case B (MDS) and case C (M(2)) megakaryocytes were accompanied by nuclear extrusion or nucleocytoplasmic separation, and their bodies were large or giant, the part of nucleus separated from their body and little cytoplasm remained as micromegakaryocytes. The nucleocytoplasmic separation could be displayed by immunocytochemistry stain. It is concluded that the phenomenon of nuclear extrusion and nucleocytoplasmic separation in megakaryocytes suggested the process that dispersed multinuclear releasing towards surround or even totally left the cell body during the megakaryocyte maturation. It also showed that the micromegakaryocytes may be the result of nucleocytoplasmic separation or splittings from multi-separated nucleus.

A novel colorimetric and fluorescent anion chemosensor based on the flavone quasi-crown ether-metal complex.

The metal-ligand complex 1 ([Mg (L)] (2+)) (or 2 ([Ca (L)]( 2+))) was demonstrated to selectively bind HSO(4)(-) (or H(2)PO(4)(-)) over other anions by using UV-vis absorption and fluorescence spectroscopy. The studied complex exhibits the remarkable color change and fluorescence quenching upon introducing HSO(4)(-) (or H(2)PO(4)(-)) anion in acetonitrile. Both the mechanism and structure of the secondary complex of complex 1 with anion were proposed on the basis of theoretical computation.

[Effects of microsome enzyme induced by phenobarbarbital on the stereoselectivity of recemic propranolol glucuronidation metabolism].

OBJECTIVE: To study the stereoselectivity of R-(+) and S-(-)-propranolol glucuronidation and metabolic interaction between R(+)- and S-(-)-propranolol. METHODS: A RP-HPLC analytical method was developed for determination of R-(+)-and S-(-)-propranolol glucuronide (PG) incubated with rat hepatic microsome induced with phenobarbital (PB). The method was applied to investigate the stereoselectivity metabolism of racemic propranolol glucuronidation in vitro. RESULT: In control and PB group, the concentration of R-(+)-PG produced at different substrates was higher than that of S-(-)-PG. Compared with the control, the V(max) and Cl(int) for R(+)-and S-(-)-propranolol increased significantly the K(m) for R(+)-propranolol was elevated, while that for S-(-) propranolol was decreased. CONCLUSION: There is a stereoselectivity in glucuronidation of propranolol in rat hepatic microsome induced with PB and R-(+)-propranolol is preferred. Metabolic interaction between R-(+)-and S-(-)-propranolol exists with a concentration-dependent mode.
