Controlled aggregation of heterometallic nanoscale Cu12Ln6 clusters (Ln = Gd(III) or Nd(III)) into 2D coordination polymers.

Discrete dinuclear and polymeric heterometallic copper(II)-lanthanide(III) complexes have been synthesized upon variation of pH and characterized by X-ray diffraction analysis. Reactions of the ligand Htza (tetrazole-1-acetic acid) with copper(II) and lanthanide(III) salts gave dinuclear [CuLn(tza)4(H2O)5Cl] complexes at the low pH of 3.5 and 2D heterometallic coordination polymers with high-nuclearity [{Cu2(OH)2}2{Cu12Ln6(mu3-OH)24(Cl)(1/2)(NO3)(1/2)(tza)12(H2O)18}](NO3)(9).8H2O (Ln = Gd or Nd) at a higher pH of 6.6. The acidity of the reaction solution can cause drastic changes in the structures of the products. In the dinuclear complexes, each pair of adjacent dinuclear molecules is linked through hydrogen bonds and pi-pi stacking interactions, and the whole structure is a hydrogen-bonded three-dimensional cubic net. In the coordination polymers, the connecting nodes are [Cu12Ln6] units, which are interconnected by [Cu2O2] units into two-dimensional structures. Magnetic studies exhibit the existence of weak exchange interactions between the Cu(II) and Ln(III) ions bridged by carboxylate and hydroxy ligands.

Pathogen elicitor-induced changes in the maize extracellular matrix proteome.

The extracellular matrix is a vital compartment in plants with a prominent role in defence against pathogen attack. Using a maize cell suspension culture system and pathogen elicitors, responses to pathogen attack that are localised to the extracellular matrix were examined by a proteomic approach. Elicitor treatment of cell cultures induced a rapid change in the phosphorylation status of extracellular peroxidases, the apparent disappearance of a putative extracellular beta-N-acetylglucosamonidase, and accumulation of a secreted putative xylanase inhibitor protein. Onset of the defence response was attended by an accumulation of glyceraldehyde-3-phosphate dehydrogenase and a fragment of a putative heat shock protein. Several distinct spots of both proteins, which preferentially accumulated in cell wall protein fractions, were identified. These three novel observations, viz. (i) secretion of a new class of putative enzyme inhibitor, (ii) the apparent recruitment of classical cytosolic proteins into the cell wall and (ii) the change in phosphorylation status of extracellular matrix proteins, suggest that the extracellular matrix plays a complex role in defence. We discuss the role of the extracellular matrix in signal modulation during pathogen-induced defence responses.

[High expression of the foot-and-mouth disease's structural protein P1 in Escherichia coli and analysis of its biology activity].

Foot-and-mouth disease virus (FMDV) is the aetiological angent of a highly contagious viral disease. The complete gene encoding the structural protein of FMDV (P1) was subcloned into expression vector pGEX-KG, resulting in the fusion expression plasmid pKG-P1. After transformed into E. coli BL21(DE3) and induced by IPTG, the results of SDS-PAGE showed that the GST-P1 fusion protein was expressed in high level. The molecular weight of the fusion protein wa 110kD and the expressed products were soluble. Western-blotting was performed to confirm that the expressed fusion protein could specifically react with antiserum against FMDV. The fusion proteins were further purified by GST purification kit and an indirect ELISA (P1-ELISA) based on the purified proteins was developed. Comparison between P1-ELISA and the standard indirect haemagglutinin assay showed the two methods had 87 per cent agreement by detecting 864 serum samples, indicating the purified P1 protein was specific as the antigen of indirect P1-ELISA.

[Immunogenicity of DNA vaccine expressing GP5 of porcine reproductive and respiratory syndrome virus fused with VP22 of bovine herpesvirus 1].

To enhance the immuogenicity of DNA vaccines expressing the GP5 protein of Porcine Reproductive and Respiratory Syndrome Virus (PRRSV), the tegument protein VP22 (encoded by VP22 gene) of Bovine Herpesvirus 1 (BHV-1), which has been demonstrated to exhibit the unusual protein transduction property, was fused to N-terminus of GP5 of DNA vaccine construct pCI-ORF5M, resulting in pCI-VP22-ORF5M expressing VP22-GP5 fusion protein. The expression of VP22-GP5 fusion protein was confirmed by both indirect immunofluorescence assay (IFA) and Western blot. To investigate its immunogenicity, BALB/c mice were immunized with the fusion expression plasmid pCI-VP22-ORF5M and non-fusion expression plasmid pCI-ORF5M, respectively. The GP5-specific ELISA antibodies, neutralizing antibodies and lymphocyte proliferative responses were evaluated at various time points after primary immunization. The results showed that GP5-specific ELISA antibodies, neutralizing antibodies, and lymphocyte proliferative responses induced by DNA vaccine pCI-VP22-ORF5M were higher significantly than those of DNA vaccine pCI-ORF5M, indicating that fusion expression with BHV-1 VP22 significantly enhances the immuogenicity of DNA vaccine expressing the PRRSV GP5 protein, and that this strategy may also be useful to develop more efficient DNA vaccines against other pathogens.

Syntheses, structures, photoluminescence, and theoretical studies of d(10) metal complexes of 2,2'-dihydroxy-[1,1']binaphthalenyl-3,3'-dicarboxylate.

The free solvated ligand, H(2)bna.CH(3)OH.H(2)O (1), and its dimeric complex, [Cd(2)(bna)(2)(H(2)O)(6)] (2) (bna = 2,2'-dihydroxy-[1,1']-binaphthalene-3,3'-dicarboxylate), were obtained by evaporation of the solutions, while two new d(10) metal-hydroxy cluster-based coordination polymers, namely [Cd(8)(OH)(4)(H(2)O)(10)(bna)(6)].17H(2)O (3) and [Hpy](2)[Zn(4)(OH)(2)(H(2)O)(2)(bna)(4)].2H(2)O.2CH(3)CN (4), were obtained by a hydrothermal route. All the compounds have been characterized by X-ray crystallography and photoluminescence measurements. Compound 1 consists of a three-dimensional, hydrogen-bonded supramolecular array, 2 exhibits a dimeric molecule featuring a square motif organized by two Cd(II) atoms and two bna ligands each at the corner, and 3 contains unprecedented [Cd(8)(micro(3)-OH)(2)(micro-OH)(2)(micro-H(2)O)(2)](12+) octanuclear metallacrown cores which are interlinked through bna to afford a two-dimensional structure, while 4 features layers with butterfly-shaped [Zn(4)(micro(3)-OH)(2)](6+) clusters. All the complexes display photoluminescent properties in the blue/green range. The manifestation of photoluminescence, as probed by molecular orbital calculations performed on the complexes and also on hypothetical multinuclear complexes, is attributed to a ligand-to-metal charge-transfer mechanism. In addition to presenting a new approach for the study of the photoluminescent properties of metal-cluster-based coordination polymers by using simple model compounds, the study also reveals the dominant role of the structure of the ligand over that of the d(10) metal-hydroxy (or oxy) cluster and the presence of the cluster significantly increasing the emission lifetime.

Proteomic analysis of the Arabidopsis thaliana cell wall.

With the completion of the Arabidopsis genome, many hypothetical proteins have been predicted without any information on their expression, subcellular localisation and function. We have performed proteomic analysis of proteins sequentially extracted from enriched Arabidopsis cell wall fractions and separated by two-dimensional gel electrophoresis (2-DE). The proteins were identified by peptide mass fingerprinting using matrix-assisted laser desorption/ionisation-time of flight (MALDI-TOF) mass spectrometry and genomic database searches. This is part of a targeted exercise to establish the entire Arabidopsis secretome database. We report evidence for new proteins of unknown function whose existence had been predicted from genomic sequences and, furthermore, localise them to the cell wall. In addition, we observed an unexpected presence in the cell wall preparations of proteins whose known biochemical activity has never been associated with this compartment hitherto. We discuss the implications of these findings and present results suggesting a possible involvement of cell wall kinases in plant responses to pathogen attack.
