A shift of Phloem unloading from symplasmic to apoplasmic pathway is involved in developmental onset of ripening in grape berry.

It remains unclear whether the phloem unloading pathway alters to adapt to developmental transition in fleshy fruits that accumulate high level of soluble sugars. Using a combination of electron microscopy, transport of the phloem-mobile symplasmic tracer carboxyfluorescein, movement of the companion cell-expressed and the green fluorescent protein-tagged viral movement protein, and assays of the sucrose cleavage enzymes, the pathway of phloem unloading was studied in the berries of a hybrid grape (Vitis vinifera x Vitis labrusca). Structural investigations showed that the sieve element-companion cell complex is apparently symplasmically connected through plasmodesmata with surrounding parenchyma cells throughout fruit development, though a small portion of plasmodesmata are apparently blocked in the ripening stage. Both carboxyfluorescein and the green fluorescent protein-tagged viral movement protein were released from the functional phloem strands during the early and middle stages of fruit development, whereas the two symplasmic tracers were confined to the phloem strands during the late stage. This reveals a shift of phloem unloading from symplasmic to apoplasmic pathway during fruit development. The turning point of the phloem unloading pathways was further shown to be at or just before onset of ripening, an important developmental checkpoint of grape berry. In addition, the levels of both the expression and activities of cell wall acid invertase increased around the onset of ripening and reached a high level in the late stage, providing further evidence for an operation of the apoplasmic unloading pathway after onset of ripening. These data demonstrate clearly the occurrence of an adaptive shift of phloem unloading pathway to developmental transition from growing phase to ripening in grape berry.

[High-dose etoposide with granulocyte colony-stimulating factor for mobilization of autologous peripheral blood stem/progenitor cells in patients with hematologic malignancies].

To explore the efficacy and safety of high-dose of etoposide with granulocyte colony-stimulating factor (G-CSF) for mobilization of peripheral blood stem cells, 10 patients with hematologic malignancies including 6 patients with multiple myeloma and 4 with non Hodgkin' s lymphoma received an etoposide dose of 1.6 g/m2. The total dose of undiluted etoposide was given on day 1 as a continuous intravenous infusion via a central vein for 10 hours. G-CSF 5 microg/kg was used on day 3 and given daily subcutaneously until leukopheresis was completed. The results showed that leukopheresis was started at days 11 (range 9-13 days) following etoposide therapy, the mean number of CD34+ cells collected in all 10 patients was 9.4 x 10(6)/kg (range 4.2 - 17.3 x 10(6)/kg), by an average of 2.6 leukophereses (range 1-4) times. Mobilization procedure that produced yields of greater than 4.0 x 10(6)/kg were achieved in every patient. Toxicity showed oropharyngeal mucositis, faucitis and urethritis respectively in 3 patients. It is concluded that high-dose etoposide with G-CSF is an effective and safe mobilizing regimen for autologous peripheral blood stem progenitor cells in patients with hematologic malignancies.

[Reliability evaluation for thirteen parameters describing anteroposterior apical base relationship in Angle Class II division 1 patients].

OBJECTIVE: To study 13 different cephalometric measurements of the anteroposterior jaw relationship in Angle Class II division 1 statistically and geometrically, and to discuss the effects of various factors on this relationship, and then to choose the most adequate measurements. METHODS: 120 patients with skeletal malocclusion Angle Class II division 1 were selected. The samples consisted of 60 men and 60 women between 20 to 28 years of age. Lateral cephalometric radiograph for each patient was taken in natural head posture by the same operator. A wire plumb line and suspended weight recorded the true vertical on each radiograph. The subject was then asked to determine the self-balanced position of the head. After determining the self-balanced neutral position, the subject was asked to look into his/her own eyes in the mirror. ANB angle, A-B plane angle, Wits appraisal, AF-BF distance, AXB angle, AB/SN4 distance, AB/PP distance, AXD angle, AD/SN distance, SGn/AB angle, APDI, FABA angle, beta angle and AB/HP distance were measured. Coefficient correlations among measurements were tabulated to determine which combination would produce a higher value. Fuzzy grouping analysis was made. RESULTS: Statistically significant and highly correlated relationships was found among many measurements except Wits appraisal and SGn/AB angle. The thirteen measurements could be divided into five clusters. CONCLUSION: The cephalometric measurements except Wits appraisal and SGn/AB angle can be used to evaluate anteroposterior jaw relationship. The AB/SN4 distance is the most adequate measurements. ANB angle, A-B plane angle, AF-BF distance, AXB angle, AB/PP distance, APDI, FABA angle and beta angle are similar in describing anteroposterior apical base relationship in Angle Class II division 1 patients.

Ab initio configuration-interaction study of the ground and low-lying electronic states of NiI.

For the first time, we have studied the potential-energy curves, spectroscopic terms, vibrational levels, and the spectroscopic constants of the ground and low-lying excited states of NiI by employing the complete active space self-consistent-field method with relativistic effective core potentials followed by multireference configuration-interaction calculations. We have identified six low-lying electronic states of NiI with doublet spin multiplicities, including three states of Delta symmetry and three states of Pi symmetry of the molecule within 15 000 cm(-1). The lowest (2)Delta state is identified as the ground state of NiI, and the lowest (2)Pi state is found at 2174.56 cm(-1) above it. These results fully support the previous conclusion of the observed spectra although our computational energy separation of the two states is obviously larger than that of the experimental values. The present calculations show that the low-lying excited states [13.9] (2)Pi and [14.6] (2)Delta are 3 (2)Pi and 3 (2)Delta electronic states of NiI, respectively. Our computed spectroscopic terms, vibrational levels, and spectroscopic constants for them are in good agreement with the experimental data available at present. In the present work we have not only suggested assignments for the observed states but also computed more electronic states that are yet to be observed experimentally.

Are polymorphisms of N-acetyltransferase genes susceptible to primary liver cancer in Luoyang, China?

AIM: To identify whether the polymorphisms of the N-acetyltransferase (NAT) genes are susceptible to primary liver cancer (PLC) in Luoyang, a PLC low-incidence area of China. METHODS: The NAT1 and NAT2 genotypes of 96 PLC cases and 173 controls were determined by PCR-RFLP. Both interaction between NAT1 or NAT2 and environmental risk factors were analyzed based on case control study. RESULTS: Compared to the control group, the frequencies of alleles NAT1*3, NAT1*4, NAT1*10, NAT1*14B and alleles NAT2*4, NAT2*6, NAT2*7 in PLC group showed no statistically significant difference (chi(2) = 2.61 and 4.16, respectively, both P>0.05). The frequencies of NAT1 genotypes NAT1*3/*3, NAT1*3/*4, NAT1*3/*10, NAT1*3/*14B, NAT1*4/*4, NAT1*4/*10, NAT1*4/*14B, NAT1*10/*10, NAT1*10/*14B, and NAT2 genotypes NAT2*4/*4, NAT2*4/*6, NAT2*4/*7, NAT2*6/*6, NAT2*6/*7 and NAT2*7/*7 also had no statistically significant difference between the two groups (chi(2) = 11.86 and 2.94 respectively both, P>0.05). Neither the frequencies of rapid and slow NAT1 acetylators nor the frequencies of rapid and slow NAT2 acetylators were significantly different between the two groups (chi(2) = 0.598 and 0.44, respectively, both P>0.05). The interaction between NAT1*10 and occupational exposures was found significant with an odds ratio of 3.40 (chi(2) = 8.42, P = 0.004, OR 95%CI:1.03-11.22). But no interaction was found between NAT2 and any environmental risk factors. CONCLUSION: The polymorphisms of NAT1 and NAT2 are not susceptible to PLC in Luoyang. Allele NAT1*10 interacts with occupational exposures.

Quantitative transcriptional profiling of ATDC5 mouse progenitor cells during chondrogenesis.

During the differentiation of a mouse chondroprogenitor cell line, ATDC5, an analysis of the transcription cartilage-related genes was carried out using real-time RT-PCR in a semiquantitative fashion. A total number of 104 genes both previously linked to chondrogenesis and hitherto not associated with the development of cartilage were analyzed. Parametric statistics, and unsupervised hierarchical and K-medians clustering approaches were used to analyze the gene expression during the sequential processes of proliferation, condensation, differentiation, maturation, and hypertrophic conversion of ATDC5 cells. The obtained data provided a robust determination of expression patterns that make possible an accurate assessment of the molecular events along the chondrogenic differentiation pathway. In addition, time-course expression profiles were described for eight highly regulated genes that have not been associated with chondrogenesis as yet. These included Cryab, Rcor2, Hig1, Bnip3, Mst4, Calml4, Gng2, and Islr.

[PPAR family and its relationship to metabolic syndrome].

Peroxisome proliferator-activated receptors (PPARs) are members of the nuclear hormone receptor superfamily of ligand--activated transcription factors. Three PPAR isoforms , designated PPARalpha, -beta/delta, and -gamma, have been identified and attracted enormous attention due to the key role these receptors play in regulating adipogenesis, lipid metabolism, insulin sensitivity, inflammation and blood pressure. Growing evidence points to a causative relationship between PPAR activity and the metabolic syndrome, including insulin resistance, glucose intolerance or type II diabetes, obesity, dyslipidemia, hypertension, atherosclerosis, and albuminuria. Importantly, both PPARalpha activators such as fibric acid class of hypolipidemic drugs and PPARgamma agonists including antidiabetic thiazolidinediones (TZDs) have been proved to be effective for improving metabolic syndrome. All three PPAR isoforms appear to play important roles in the development of type II diabetes and diabetic nephropathy. Accumulating data has begun to emerge suggesting PPARs may serve as potential therapeutic targets for treating the metabolic syndrome and its related complications. Here we review the literature pertaining to the action, ligand selectivity and physiological role of PPARs. Particular emphasis is placed on their pathogenic roles in the metabolic syndrome and the therapeutic utility of PPAR modulators in the treatment of type II diabetes.

A distinct role for norepinephrine in memory retrieval.

A role for norepinephrine in learning and memory has been elusive and controversial. A longstanding hypothesis states that the adrenergic nervous system mediates enhanced memory consolidation of emotional events. We tested this hypothesis in several learning tasks using mutant mice conditionally lacking norepinephrine and epinephrine, as well as control mice and rats treated with adrenergic receptor agonists and antagonists. We find that adrenergic signaling is critical for the retrieval of intermediate-term contextual and spatial memories, but is not necessary for the retrieval or consolidation of emotional memories in general. The role of norepinephrine in retrieval requires signaling through the beta(1)-adrenergic receptor in the hippocampus. The results demonstrate that mechanisms of memory retrieval can vary over time and can be different from those required for acquisition or consolidation. These findings may be relevant to symptoms in several neuropsychiatric disorders as well as the treatment of cardiac failure with beta blockers.

Phloem unloading in developing walnut fruit is symplasmic in the seed pericarp and apoplasmic in the fleshy pericarp.

The sieve element-companion cell (SE-CC) complex of the sepal bundles feeding the fleshy pericarp of developing walnut (Juglans regia L.) fruit is structurally symplasmically isolated, but the SE-CC complex of the minor ventral carpellary bundles located in the seed pericarp and feeding the seed is structurally symplasmically connected to its adjacent parenchyma cells. 14C-autoradiography indicated that the phloem of both the sepal and carpellary bundles was functional for unloading. Confocal laser scanning microscopy imaging of carboxyfluorescein unloading showed that the dye is confined to the phloem strands of the sepal bundles in the fleshy pericarp, but released from the phloem strands of the minor ventral carpellary bundles into the surrounding parenchyma cells in the seed pericarp. A 60-kDa acid invertase was immunolocalized to the cell wall of SE-CC complex and parenchyma cells in both the fleshy and seed pericarp. These data provide clear evidence for an apoplasmic phloem unloading pathway in the fleshy pericarp and a predominant symplasmic phloem unloading pathway parallel with a possible apoplasmic path as suggested by the presence of the extracellular invertase in the seed pericarp. A model of complex phloem unloading pathways in developing walnut fruit has been proposed.

[Advanced methods of preparing pachytene bivalents and high resolution multiple bands of zebrafish (Danio rerio)].

The pachytene bivalents with high-resolution multiple G bands of zebrafish were obtained after the treatment with alkaline hypotonic solution and high concentration of chloroform fixative solution. When comparing six group chromosomes from different pachytene specimens, the characteristic and the number of bands were well matched. In order to systematize this technique and get stable result, we summarize the preparation procedure of the zebrafish bivalents. The 6-month-old to one-year-old zebrafish whose spermary appears ivory-white and opaque, is good material. The whole testis should be treated with hypotonic solution for 1.5 approximately 2 h at room temperature. Then, the testes were fixed for 20 min in high chloroform fixative solution (chloroform: methanol: acetic acid, 3: 6:1), and fixed in Carnoy's solution (methanol: acetic acid, 3:1) for two times. In addition, with the treatment of restriction endonuclease Alu I directed in situ nick translation, we successfully obtained well-resolved restrictive endonuclease banding of zebrafish bivalents, which was considered as G-like band patterns. The aging of the specimen is also important factor, should let them dry at room temperature for one week. The application of these methods in cytogenetics research of zebrafish and other fish can be expected. Construction of the steady technique system to prepare high resolution banding bivalents and idiogram of zebrafish is the basement to found stable and accurate framework for physical map.

K+ and Na+-induced self-assembly of telomeric oligonucleotide d(TTAGGG)n.

The telomeric DNA oligomers, d(TTAGGG)(n), where n=1, 2, 4, could self-associate into the multi-stranded structures in appropriate condition, exhibited different CD spectra. The presense of Na(+) was more advantage to facilitate the formation of anti-parallel conformation, but the presense of K(+) enhanced their thermal stability. Spectroscopic analysis of 3, 3'-diethyloxadicarbocyanine (DODC) showed the formation of hairpin quadruplex structures for d(TTAGGG)(2) and d(TTAGGG)(4), but d(TTAGGG) could not. The four-stranded tetraplexes and branched nanowire formed in the presense of K(+) or Na(+) alone were observed by atomic force microscopy (AFM). The ability of d(TTAGGG)(n) to self-assemble into four-stranded tetraplexes and nanowires depends strongly on the number of repeating units and ionic environment. A model to explain how these structures formed is proposed.

Activation of gp120 of human immunodeficiency virus by their V3 loop-derived peptides.

V3 loop peptides from three different human immunodeficiency virus type 1 (HIV-1) strains were synthesized. BH10, ADA, and 89.6 strains whose infections are dependent on CXCR4, CCR5, and both, respectively, were selected. Co-transfection of luciferase reporter gene and corresponding envelope genes (HXB2, ADA, and 89.6) generate pseudotype viruses (HXB2/Luc, ADA/Luc, and 89.6/Luc). The effects of each peptide on the infection of U87 cells expressing CD4 and one of the coreceptors with all pseudotype viruses were evaluated. V3 loop peptide from BH10 (V3-BH10) alone increased the HXB2/Luc infection by 93% at 10 microM. Both V3-ADA and V3-89.6 enhanced ADA/Luc infection by 38% and by 55% at 10 microM, respectively. For 89.6/Luc infection, only V3-89.6 enhanced the infections on both target cells. V3-BH10 modulated the epitopes of coreceptor binding site and V2 loop of gp120 on HIV-1 IIIB infected H9 cells, indicating that V3 loop peptide activates viral gp120 and enhances infectivity.

High-Valent Manganese in Polyoxotungstates. 3. Dimanganese Complexes of gamma-Keggin Anions.

Dimanganese-substituted gamma-Keggin heteropoly tungstates have been synthesized by reaction of the lacunary species gamma-[(SiO(4))W(10)O(32)](8)(-) with appropriate mixtures of Mn(II) and MnO(4)(-). The crystal structure of [(CH(3))(3)(C(6)H(5))N](4)[(SiO(4))W(10)Mn(III)(2)O(36)H(6)].2CH(3)CN.H(2)O (anion 1) was determined by X-ray diffraction. Crystallographic data: space group P&onemacr;, a = 12.951(3) Å, b = 14.429(3) Å, c = 20.347(4) Å, alpha = 81.95(3) degrees, beta = 88.92(3) degrees, gamma = 67.48(3) degrees, V = 3475.2(13) Å(3), and Z = 2. The final R value is 7.29% for 15861 reflections with I > 2sigma(I). The anion has the anticipated gamma-Keggin structure with virtual C(2)(v)() symmetry. The two Mn cations occupy adjacent, edge-shared octahedra with bridging hydroxo and terminal aqua ligands. Anion 1 can be oxidized and reduced to the corresponding Mn(III)Mn(IV) (2) and Mn(II)(2) (3) species respectively. The magnetic susceptibility of 1 between 2 and 300 K indicates that the Mn(III) cations are antiferromagnetically coupled, with J = -17.0 cm(-)(1) and g = 1.965. No simple magnetic behavior was observed for 2 or 3.
