OBJECTIVE: To assess the feasibility of evaluation of CFR for LAD and right coronary artery (RCA) as well as diagnostic accuracy for patency of each vessel by transthoracic Doppler echocardiography (TTDE). METHODS: 65 consecutive patients (age 58 +/- 14, ejection fraction 49% +/- 8%, 48 men and 17 women) were studied for CFR by TTDE with adenosine infusion (140 microg.kg(-1).min(-1)). LAD flow was interrogated nearby the apex in modified 2-ch apical view. RCA flow was interrogated at mid posterior wall on distal part of the posterior descending branch (PDA) in modified 2-ch apical view for PDA. RESULTS: Flow for LAD was detected in all patients (4 with use of contrast agent) and for RCA in 55 patients (5 with contrast agent). Two patients had an occluded RCA. Feasibility of RCA flow detection was 55/63 (87%). In all patients maximal flow of each branch was detected in less than 2 min of adenosine infusion (140 mg.kg(-1).min(-1)). Total time for both CFR estimation was 7.3 +/- 1.6 min. Selecting 2.0 or 1.8 as CFR cut-off value for diagnosis of coronary stenosis (> or = 50% in diameter) or severe stenosis (> or = 70%) was found fitted for both LAD and RCA with similar diagnostic performance (sensitivity 89%/93%, specificity 86%/84%, area under curve 0.89/0.92 respectively, in stenosis diagnosis and sensitivity 88%/89%, specificity 90%/83%, area under curve 0.94/0.95 respectively in severe stenosis diagnosis). CONCLUSIONS: CFR evaluation of both distal LAD and PDA is of high feasibility and can provide high diagnostic yield for relevant vessel patency.
OBJECTIVE: To study the possibility of inducing refractive errors by opening the mouse (C57BL/6J) eye artificially after birth and explore the difference in pathogenic mechanism in form-deprivation myopia by analyzing the histological changes of the animal eyes. METHODS: 4-day-old mice (n = 13) was undergone 10-day monocular artificially opened period till the opposite eye was natural open. Another group 12-day-old mice (n = 13) were raised with monocular suture of eyelid for 7 days. Age-matched untreated mice were used as control. Retinoscopic refraction was performed subsequently. Animals were sacrificed and the eyes enucleated at the end of the experiment. The axial length and weight of the eyes were measured and the sections of the eyes were also observed by light and electron microscope. RESULTS: All eyes treated with two methods showed significant development of myopia compared with the contralateral eye. The axial length was elongated in form deprived eyes, but it was shortened in artificially opened eyes; no change in the weight of eyeball was found in form deprived eyes, but it was reduced in artificially opened eyes when compared with corresponding fellow eyes. Under light microscope, there is no significant histological difference observed in the eyes treated with two methods. Under electron microscope, rod outer segment in form deprived eyes was inordinate and the phagosomes containing fractions of the membrane disc of outer segment were remarkably decreased. No significant changes of photoreceptor were demonstrated in artificially opened eyes. CONCLUSION: The means of artificially opening eyelid can induce myopia in the mouse and the method may be useful to establish myopia animal model.
OBJECTIVE: To study the effects of periodical vacuum-suction of different pressures on the ultrastructure of rabbits' retina. METHODS: Rabbits involved in the present study were divided into a control group and four treatment groups, which were treated with vacuum-suction of different pressures through scleral suction ring. The suction pressure was increased at a rate of 100 mm Hg/3 sec (1 mm Hg = 0.133 kPa) and reached (200 +/- 20) mm Hg (group I), (300 +/- 20) mm Hg (group II); (400 +/- 20) mm Hg (group III) and (500 +/- 20) mm Hg (group IV). The suction pressure was maintained for 5 seconds before released to zero. The corresponding intraocular pressure examined by Tonopen was 35 mm Hg, 45 mm Hg, 55 mm Hg and 75 mm Hg in these 4 groups, respectively. This circle was repeated for 10 times with 1 minute interval. The test was repeated every other day. The retina was examined by electron microscope after 2 weeks. RESULTS: There was no obvious difference in the retina ultrastructure between the control group and group I. The retina ultrastructure changed slightly in group II which showed more active metabolism. Some significant changes were found in group III and group IV with cell necrosis in group IV. CONCLUSION: Different elevations of intraocular pressure following periodical vacuum-suction have different effects on the retina of rabbit. Pressure less than 45 mm Hg has almost no effect, but some effects appear at 55 mm Hg pressure, and obviously damages are caused by 75 mm Hg pressure.

OBJECTIVE: To investigate the conditions essential for culturing hepatocytes colony in vitro, and to study the growth regulation mechanisms of hepatocyte colony. METHODS: Rat hepatocytes were isolated by two-step in situ preperfusion and collagenase circulatory perfusion. The effects of hepatopoietin (HPN), nicotinamide (NA) and dimethyl sulfoxide (DMSO) on DNA synthesis, mitotic activity, morphology (under inverted microscope) and utrastructure (under TEM) of the hepatocytes were investigated in chemically defined culture medium. RESULTS: The time course of DNA synthesis in cultured hepatocytes showed that 3H-TdR incorporation was dramatically enhanced by 10 mmol/L NA, presenting 2 peaks at 60 and 84 h respectively. The plateau of DNA synthesis was diminished in the presence of DMSO, but a peak occurred again at 132 h upon NA treatment. After cell culture in the presence of HPN, NA, and DMSO for 72 h, the hepatocytes presented sustained regular bipolar mitosis, with considerable mitotic activity at 168 h. The growth characteristics of hepatocyte colony, in addition to its potential for expansion, were captured by both light and electron microscopy on day 28 of cell culture. CONCLUSION: The reciprocal actions of NA and DMSO can control the proliferation of HPN-stimulated hepatocytes, which can be used for studying human hepatocyte metabolism, cytotoxicity, biotransformation and mutagenesis, and may provide experimental evidences for the treatment of liver failure and genetic liver diseases with in vitro hepatocyte clones.
OBJECTIVE: To explore the method of treating serious secondary fronto-orbital fracture deformities through intro-extra cranial approach. METHODS: The fronto-orbital fracture was divided into two types according to whether there were any large scale fronto-orbital bone defects: type I: Large scale fronto-orbital bone defect; type II: Concave fronto-orbital fracture deformity without large scale bone defect. Both types were treated through intro-extra cranial approach to expose the fracture site. For type I deformity, the bone defects were repaired and reconstructed with outer table of cranial bone and artificial bone. For type II, the deformity was repaired by osteotomy, bone reposition and internal rigid fixation. RESULTS: 18 cases were treated from June 1998 to October 2000, include type I, 12 cases, and type II, 6 cases. All the patients recovered well and the post-operative appearance were greatly improved. CONCLUSIONS: Intro-extra cranial approach can expose the fractured site better than the simple extrocranio approach, and make the operation more easily done. Combined with the technique of cranio maxillo facial surgery, the treatment can be more complete and the results can be more satisfactory.
OBJECTIVE: To investigate the effect of retrorsine on the proliferation of SD rat immortalized hepatocytes after intrasplenic transplantation. METHODS: Immortalized hepatocytes from SD rats was prepared by transfecting the primary hepatocytes with a retroviral vector SSR69 expressing the genes encoding Simian virus 40 T large antigen. Twenty-four adult SD rats were randomly assigned to 3 equal groups designated respectively as A, B, and C groups. Intraperitoneal retrorsine injections at the dose of 26 mg/kg.b.w. were performed twice in groups A and B at the interval of two weeks before hepatocyte transplantation. After 20% hepatic lobe resection were completed in all the groups, groups A and C received intraplenic transplant with immortalized hepatocytes, while group B received normal hepatocyte transplant. Post-mortem examinations were performed on the spleen samples with HE staining. Albumin expression in the transplanted hepatocytes was examined by immunohistochemical staining, and the ultrastructure of the transplanted cells was observed under transmission electron microscope. RESULTS: Histological examination demonstrated that the rat spleens in all the 3 groups contained cluster of transplanted hepatocytes from 1 week to 3 weeks after the transplantation, and the proliferation of the transplanted cells were comparable between groups A and B, but both of which showed significant difference from group C. Albumin excretion by the immortalized hepatocyte was identified in all the groups, and the transplanted immortalized cells preserved normal cell ultrastructure. CONCLUSION: Immortalized hepatocytes of SD rat have the normal cell ultrastructure and albumin-excretion function, and retrorsine can promote the proliferation of the transplanted hepatocytes.

OBJECTIVE: To study the inhibitory effects of antisense focal adhesion kinase (FAK) oligodeoxynucleotides (ODN) on the invasion of Bel 7402 cells, and investigate the mechanisms. METHODS: LipofecTAMINE-mediated antisense FAK ODN was transfected into Bel 7402 cells. Cell number and viability were evaluated every 24 hours by trypan blue dye exclusion. Cell attachment assay was carried out at intended time points in a microculture well pre-coated with fibronectin (FN). The invasive activity of tumor cells was assayed in a transwell cell culture chamber. Cell cycle and cell apoptosis analysis were performed with flow cytometry (FCM). RESULTS: The expression of p125FAK in the group treated with antisense FAK ODN (6.49%+/-0.10%) significantly decreased, compared with those in the group treated with sense FAK ODN (14.33%+/-1.88%) and control group (16.68%+/-1.62%), F=7.66, P<0.01. Antisense FAK ODN significantly inhibited the growth of Bel 7402 cells by 30%-60%, the attachment by 25%-55%, and the invasion, 15%-25%. The decreased expression of FAK in Bel 7402 cells caused a G2/M cell cycle arrest, and the cells at S phase decreased significantly. The occurrence of apoptosis detected by FCM increased significantly in the group treated with antisense FAK ODN. CONCLUSIONS: Inhibition of FAK expression significantly decreases the attachment between ECM and Bel 7402 cells, and the ability of Bel 7402 cells to invade the reconstituted basement membrane. In addition, FAK suppression significantly inhibits the proliferation of Bel 7402 cells in vitro, and increases their apoptosis.
OBJECTIVE: To study the clinical and imaging features together with diagnosis and management of orbital histocytosis X. METHODS: In a retrospective study from 1973 to 1999, 47 cases of histocytosis X were reviewed related with clinical materials, including eosinophilic granuloma 17 cases and Hand-Schuller- Christian disease 30 cases. RESULTS: Most occur in children and teenagers, mainly in male. Characteristic clinical features, roentgenography, ultrasonography, CT, MRI provide the most diagnostic assistance. CONCLUSIONS: Solitary unifocal eosinophilic granuloma mainly involves teenagers. Hand-Schuller-Christian disease usually take places between birth and 5 years of age, which is characterized by multifocal variant of eosinophilic granuloma, especially the classic triad. Characteristic images contribute to the diagnosis. Multiple management is needed, including surgery, chemotherapy and radiotherapy. The prognosis varies with the three types.
