[Construction of the eukaryotic expression vector containing rat transforming growth factor beta type II receptor and interferon gamma fusion genes].

OBJECTIVE: To express a bioactive fusion protein comprising rat soluble transforming growth factor beta type II receptor and interferon gamma(rsTGFbetaR II-IFN gamma) in mammalian cells. METHODS: mRNA was extracted from rat liver and the sTGFbetaR II-IFN gamma genes amplified by RT-PCR, then the two gene segments were cloned into the same pSecTag2A expression vector, and pSecTag2A/rsTGFbetaR II-IFN gamma recombinant plasmid was obtained, which was later transfected into CHO cells using liposomes. The expression of pSecTag2A/rsTGFbetaR II-IFN gamma in the supernatant was detected by ELISA and Western blotting. The bioactivities of the fusion protein were tested by sTGF betaR II-IFN gamma and IFN gamma bioassays. RESULTS: pSecTag2A/rsTGFbetaR II-IFN gamma transfectants expressed rsTGF betaR II-IFN gamma fusion protein. The purified fusion protein exhibited anti-viral activity and antagonized the proliferation-inhibitive effect of TGFbeta1 on CCL-64 cells. It inhibits the HSC activation in vitro. CONCLUSION: The pSecTag2A/rsTGFbetaR II-IFN gamma recombinant plasmid constructed in this study can express bioactive rsTGFbetaR II-IFN gamma fusion protein.

[Two-dimensional electrophoresis and mass spectrometry analysis on effects of trichloroethylene on protein of L-02 liver cells].

OBJECTIVE: To study the effects of trichloroethylene (TCE) on the protein in L-02 cells in vitro. METHODS: Thiazolyl blue and Trypan blue tests were used to investigate the cytotoxicity of TCE to L-02 liver cell. The 2-D electrophoresis was used to analyse the expression of proteins in L-02 liver cells. The differentially expressed protein spots were identified by matrix assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF-TOF-MS). RESULTS: When the concentration of TCE exceeded 30 micromol/L, there was distinct cytotoxicity to L-02 cell (P < 0.05). Selected 40 micromol/L to treat L-02 liver cells and analyze the differential proteome expression, the results showed that the expression level of 37 protein spots was up-regulated and 15 protein spots was down-regulated. And 15 proteins were identified by MALDI-TOF-TOF-MS. CONCLUSION: TCE can change the proteome expression of L-02 liver cell. It should provide the fundamental information to identify proteins related to TCE in further study.

[Common infantile scrotum or testicle disease diagnosed by high frequency ultrasonoscopy].

OBJECTIVE: To diagnose and distinguish the swell in the infantile scrotum or testicle by the high frequency ultrasonoscopy so as to find the acoustic imaging characters and clinical application value. METHODS: With the high frequency linear-array probe used, the multi-section and two-side contrast scanning checks were conducted on 46 cases of the infantile scrotum or testicle swell and position with abnormal physical signs. The structures of scrotum, testicle, epididymis and spermatic cord as well as the positions, sizes, forms and interior echoes were observed. When it was necessary, the position should be changed, bladder filled up and the groin and two sides of bladder scanned. RESULTS: A total of 45 cases of infantile scrotum or testicle disease were diagnosed by ultrasound, which accorded with the clinical behavior and operation result while 1 case of one-side cryptorchid was not found. CONCLUSION: It is quick and accurate to diagnose the infantile scrotum or testicle disease by high frequency ultrasonoscopy.

[Study on the comet-tail system of 12C16O+ in the near-infrared region].

The molecular cation CO+ is a very important transient molecular radical, and the spectra of the ion has been investigated by numerous researchers. Optical-heterodyne-magnetic-rotation-enhanced velocity modulation spectroscopy(OH-MR-VMS) was employed to measure the comet-tail system (A2 pi(i)-X2 sigma+) of 12C16O+ molecular ion. The vibrational rotation absorption spectra wereobserved in the near-infrared region, including three bands of (0, 3), (1, 4) and (3, 6). A set of precise molecular constants for the A2 pi(i) (v' = 0, 1, 3) were determined using the least-squares fit. This experiment has further proved that OH-MR-VMS is very valuable in measuring absorption spectra of molecular ions.

[Construction and expression of the eukaryotic expression vector containing human soluble transforming growth factor beta receptor II].

OBJECTIVE: To construct a eukaryotic expression vector encoding the gene of extracellular region of type II transforming growth factor beta receptor (sTGFbetaR II), to express the protein in CHO cell line and to determine its biological activity. METHODS: The extracellular region (amino acids 1-159) of the human TGFbetaR II cDNA was amplified by PCR from a TGFbetaR II chimeric plasmid, and the eukaryotic expression plasmid pCDNA3.1/myc-his(-)B-sTGFbetaR II(pCDNA-sTGFbetaR II) was constructed by inserting the sTGFbetaR II cDNA into the EcoR I/Hind III-digested pCDNA. The DNA sequence was confirmed by double digestion and the pCDNA-sTGFbetaR II plasmid was transfected into the CHO cell line. The sTGFbetaR II protein was confirmed by Western blotting analysis and its biological function was determined. RESULTS: The specific protein was observed in western blotting, and the protein abrogated the growth-inhibitory effects of TGF-beta1 on mink lung epithelial cells (Mv1Lu). CONCLUSION: The eukaryotic expression plasmid pCDNA-sTGFbetaR II has been successfully constructed and the sTGFsTGFbetaR II protein with biological activity obtained.

[Clinical significance of soluble selectins and matrix metalloproteinases-9 in patients after successful cardiopulmonary resuscitation].

OBJECTIVE: To investigate whether patients after successful cardiopulmonary resuscitation (CPR) exist elevated levels of soluble P-and E-selectin (sP-selectin, sE-selectin), matrix metalloproteinases-9 (MMP-9) and to evaluate the clinical significance of changes in these cytokine levels. METHODS: Plasma levels of sP-and sE-selectin, MMP-9 of 82 patients who survived >or=48 hours after CPR were determined on the second day after successful CPR. They were classified into different groups according to whether causing systemic inflammatory response syndrome (SIRS) and developing sepsis. Sixty-five non-critically ill patients were served as control group. RESULTS: Incidence of SIRS was 68.3% (56/82 cases) after successful CPR. Plasma levels of sP-selectin were higher in patients with SIRS compared with those in patients without SIRS or control group (all P<0.01). Plasma levels of sE-selectin and MMP-9 were not significantly different between patients with SIRS and without SIRS (all P>0.05), but were higher in both patient groups than those in the control group (all P<0.01) 43.9% of the patients developed sepsis within 1 week after CPR. Plasma levels of sP-selectin were higher in patients developing sepsis than those in patients without sepsis (P<0.05), but plasma levels of sE-selectin and MMP-9 were not significantly different between two groups (all P>0.05). Plasma levels of sE-selectin and MMP-9 were higher in nonsurvivors than those in survivors (both P<0.01), but plasma levels of sP-selectin were not significantly different between two groups (all P>0.05). CONCLUSION: SIRS is a frequent but unspecific finding after CPR with only minor impact on outcome. Determination of sP-and sE-selectin, MMP-9 early after CPR might help to identify patients at a high risk for sepsis or an adverse, respectively.

[The method of wavelength calibration in high resolution absorption spectroscopy].

A new method for the calibration of wavelength in a measurement of high resolution molecular absorption spectra by referring the standard spectrum of iodine molecule is presented in this paper. Segment offset is employed in the calibration of wavelength, while the data smoothing, baseline determining and peak fitting are applied to preprocess the spectral data. A visual software is provided for automatically calibrating wavelength with a friendly style of mutual dialogue between human and computer. The software has been used to calibrate the absorption spectra of N2+ molecular ion in the visible region 16,422-17,896 cm-1, the absolute accuracy is about 0.006 cm-1.
