Activation of beta2-adrenergic receptor stimulates gamma-secretase activity and accelerates amyloid plaque formation.

Amyloid plaque is the hallmark and primary cause of Alzheimer disease. Mutations of presenilin-1, the gamma-secretase catalytic subunit, can affect amyloid-beta (Abeta) production and Alzheimer disease pathogenesis. However, it is largely unknown whether and how gamma-secretase activity and amyloid plaque formation are regulated by environmental factors such as stress, which is mediated by receptors including beta(2)-adrenergic receptor (beta(2)-AR). Here we report that activation of beta(2)-AR enhanced gamma-secretase activity and thus Abeta production. This enhancement involved the association of beta(2)-AR with presenilin-1 and required agonist-induced endocytosis of beta(2)-AR and subsequent trafficking of gamma-secretase to late endosomes and lysosomes, where Abeta production was elevated. Similar effects were observed after activation of delta-opioid receptor. Furthermore, chronic treatment with beta(2)-AR agonists increased cerebral amyloid plaques in an Alzheimer disease mouse model. Thus, beta(2)-AR activation can stimulate gamma-secretase activity and amyloid plaque formation, which suggests that abnormal activation of beta(2)-AR might contribute to Abeta accumulation in Alzheimer disease pathogenesis.

An improved method of left ventricular catheterization in rats.

Left ventricular catheterization is a key step for hemodynamic studies. At present, the right common carotid artery is a popular entrance, but right common carotid artery punctuation and ligation primarily leads to obstruction and second left ventricular catheterization in the same animal cannot be performed. In this note, we will describe an improved method of catheterization in rats using guide wires of percutaneous transluminal coronary angioplasty (PTCA) through the right external carotid artery and right common carotid artery, respectively. 30 rats anesthetized with sodium pentobarbital (50 mg kg(-1)) underwent the first operation and, 2 weeks later, 29 were successfully catheterized again. One rat died of left ventricle perforation while the hemodynamic monitor was performing. Therefore, our method proves able to be performed two times in one animal. Moreover, it inflicts less injury during the operation, because the tube was advanced through a soft guide wire.

Assembly of a tetrathiafulvalene-anthracene dyad on the surfaces of gold nanoparticles: tuning the excited-state properties of the anthracene unit in the dyad.

Due to the unique features of the tetrathiafulvalene (TTF) unit, such as the electron-donating ability and presence of methylthio groups, dyad 1 can be assembled on the surfaces of gold nanoparticles, as indicated by absorption, electrochemical, and fluorescent-spectral studies. Dyad 1 can also be disassembled by the addition of thiols. Assembly of dyad 1 on the surfaces of gold nanoparticles leads to the formation of a triad (A1-D-A2), which in turn modulates the photoinduced electron-transfer process within dyad 1. Accordingly, the fluorescence intensity of dyad 1, after assembly with gold nanoparticles, increases, and the fluorescence lifetime is prolonged. Furthermore, the assembly of dyad 1 on gold nanoparticles facilitates photodimerization of the anthracene units of dyad 1. Both fluorescence and photodimerization are associated with the excited-state behavior of the anthracene unit, thus it may be concluded that the excited-state properties of the anthracene unit can be tuned upon complexation with gold nanoparticles.

A technique for retrograde intubation in mice.

Endrotracheal intubation is critical for some experimental studies in mice, but the animals' small size makes the procedure difficult. The authors describe a new, easily learned retrograde intubation method using angioplasty guide wire. They twice intubated anesthetized mice successfully with no airway complications caused by puncture of the trachea.

Gastrointestinal stromal tumor colliding with angiosarcoma.

The collision of gastric tumors is rare, and those involving gastrointestinal stromal tumor and angiosarcoma have not been previously reported. This article reports a gastric tumor involving collision of gastrointestinal stromal tumor and angiosarcoma. The patient was an 81-year-old man who presented with dyspepsia and large gastric mass. The mass showed a proliferation of spindle cells expanding the muscularis propria. Most of the tumor consisted of spindle cells with low-grade cytology that were positive for CD117 (c-kit), indicating gastrointestinal stromal tumor. Occasional prominent foci showed dissecting and anastomosing channels of ectatic vascular spaces lined by cytologically malignant cells that were positive focally for CD117 and diffusely positive for multiple vascular markers, indicating angiosarcoma.

Comparison of ancillary studies in the detection of residual disease in plasma cell myeloma in bone marrow.

Morphologic determination of residual plasma cell myeloma is often difficult. This study was conducted to evaluate the diagnostic value of several ancillary methods. A total of 121 bone marrow specimens from patients with myeloma after bone marrow transplantation or chemotherapy were studied. Of those, 49 cases were studied by conventional cytogenetics (CC), 86 by flow cytometry (FC), 50 by immunohistochemical analysis, 72 by molecular studies (MS) for immunoglobulin gene rearrangements, and 18 by standard fluorescence in situ hybridization (FISH) study. Of the patients, 70 were found to have residual myeloma. Residual disease was detected in 58 of 70 patients by morphologic examination (detection rate, 83%), 25 of 26 by immunohistochemical analysis (96%), 39 of 54 by FC (72%), 6 of 40 by CC (15%), 6 of 12 by FISH (50%), and 24 of 40 by MS (60%). Among the ancillary studies, immunohistochemical analysis and FC were the most helpful ancillary studies for detection of residual disease in myeloma, with immunohistochemical analysis the most effective single method.

Primary vitrectomy for rhegmatogenous retinal detachment associated with choroidal detachment.

PURPOSE: To evaluate the role and the anatomic and visual results of primary pars plana vitrectomy (PPV) in treating cases with rhegmatogenous retinal detachment associated with choroidal detachment. METHODS: All patients were divided into 2 groups. Each group included 23 consecutive eyes with rhegmatogenous retinal detachment and choroidal detachment with proliferative vitreoretinopathy less than grade C. In the study group, controlled removal of vitreous traction was achieved by primary vitrectomy and augmented by scleral buckling if needed. The breaks were treated by focused endolaser coagulation. Postoperative tamponade was done by SF6 or C3F8 gas. In the control group, all patients underwent regular scleral buckling procedure. The cases were followed up for 6 to 12 months. RESULTS: In the study group, retinal reattachment could be achieved in 21 cases (91.30%) after the first operation and in all cases after the second procedure. No occurrence of choroidal detachment occurred after the first procedure. Retinal reattachment rate and visual results tended to be better compared with conventional surgical techniques in the control group. CONCLUSION: Primary vitrectomy represents a safe, effective method in the management of rhegmatogenous retinal detachment associated with choroidal detachment.

Association of beta-arrestin and TRAF6 negatively regulates Toll-like receptor-interleukin 1 receptor signaling.

Tumor necrosis factor receptor-associated factor 6 (TRAF6) is critical for mediating Toll-like receptor (TLR)-interleukin 1 receptor (IL-1R) signaling and subsequent activation of NF-kappaB and AP-1, transcriptional activators of innate immunity. Here we show that beta-arrestins, a family of multifunctional proteins, directly interacted with TRAF6 after TLR-IL-1R activation. Formation of the beta-arrestin-TRAF6 complex prevented autoubiquitination of TRAF6 and activation of NF-kappaB and AP-1. Endotoxin-treated beta-arrestin 2-deficient mice had higher expression of proinflammatory cytokines and were more susceptible to endotoxic shock. Thus, beta-arrestins are essential negative regulators of innate immune activation via TLR-IL-1R signaling.

Changing pattern of AIDS: a bone marrow study.

We compared bone marrow findings in 2 groups of patients with AIDS during 2 different periods: group 1, n = 20; male/female ratio, 19/1; and group 2, n = 120; male/female ratio, 6/1. Bone marrow iron stores were decreased significantly in group 2 (P < .01), and the incidence of AIDS-related lymphomas was higher, with frequent bone marrow involvement. Two group 1 patients had Kaposi sarcoma, and a 21-month-old girl with transfusion-transmitted AIDS had Burkitt-like lymphoma. In group 2, 44 patients had a history of malignant neoplasms, including Kaposi sarcoma (10 cases), hematologic neoplasms (33 cases), and metastatic leiomyosarcoma (1 case). Of the 120 patients, 15 (12.5%) had bone marrow involvement by malignant neoplasms. The majority of the non-Hodgkin lymphomas were high-grade lymphomas. Patients with AIDS-related malignant neoplasms had higher CD4+ cell counts and viral loads than patients without malignant neoplasms (P < .01, P < .05, respectively). The finding of decreased iron stores in patients with AIDS might aid clinical management of their anemia. The increased incidence of malignant neoplasms, especially lymphomas, in recent years might be related to prolonged survival but with incomplete reconstitution of immune function after antiretroviral therapy.

Effects of nimodipine and fructose-1,6-diphosphate on cerebral damage in carbon monoxide poisoning mice.

OBJECTIVE: To study the dose- and time-dependent protective effects and the synergistic effects of nimodipine (NMDP) and fructose-1,6-diphosphate (FDP) against cerebral damage induced by acute carbon monoxide (CO) poisoning in mice. METHODS: Male mice were exposed to CO 170 mL/kg, i.p. After CO intraperitonealy exposure, mortality of mice, change in memory function estimated by passive avoidance test, the pathomorphologic observation of brain tissue slices, as well as changes of activities of monoamine oxidase (MAO)-B and Ca(2+)-Mg(2+)-ATPase in cerebral tissue were studied. In dose-dependent protective effect study, NMDP (10.6, 5.3, 2.7 mg/kg) and FDP (2.6, 1.3, 0.67 g/kg) was injected ip, respectively 15 min after CO exposure. To study the time-effect relationship of drugs, NMDP (5.3 mg/kg) and FDP (1.3 g/kg) were administered ip respectively 15 minutes, 45 minutes and 120 minutes after CO exposure. The combination of NMDP (2.7 mg/kg) and FDP (0.67 g/kg) was administered ip15 minutes, 45 min and 120 minutes after CO exposure to study the synergism of the two drugs. RESULTS: Either NMDP (10.6, 5.3 mg/kg) or FDP (2.6, 1.3 g/kg) administered ip within 15 minutes after CO exposure significantly decreased the impairment of memory function and mortality rate induced by CO, inhibited the decrease of Ca(2+)-Mg(2+)-ATPase activity, blunted the rising of MAO-B activity and prevented the delayed hippocampal neuronal death in poisoning mice. To our surprise, the combined use of NMDP (2.7 mg/kg) and FDP (0.67 g/kg) within 15 minutes after CO exposure had similar effects to that in NMDP (10.6, 5.3 mg/kg) and FDP (2.6, 1.3 g/kg). CONCLUSIONS: These results suggest that the impairment of CO on brain can be attenuated if NMDP or FDP are administered sufficiently and quickly as soon as possible after CO exposure and there exists a synergism of FDP and NMDP against CO poisoning damage.

Vascular dysregulation in the choroid of subjects with acral vasospasm.

OBJECTIVE: To assess the relationship between ocular perfusion pressure and blood flow in the choroid in subjects with acral vasospasm. PATIENTS AND METHODS: Twenty otherwise healthy subjects with acral vascular dysregulation and 55 age-matched nonvasospastic healthy volunteers were recruited. After a 20-minute rest in a sitting position, intraocular pressure and choroidal blood flow were determined by means of applanation tonometry and choroidal laser Doppler flowmetry, respectively. The laser Doppler flowmetry variables velocity, volume, and flux were assessed. The correlations between mean ocular perfusion pressure ([23 x [(23 x diastolic blood pressure) + (13 x systolic blood pressure)]] -intraocular pressure) and blood flow measures were determined by means of the Pearson linear correlation factor. The t test was used to evaluate differences between normal subjects and patients with vasospasm. RESULTS: Apart from a slight difference in systolic blood pressure (mean +/- SD, 113.70 +/- 11.88 mm Hg in the vasospastic group and 121.09 +/- 14.58 mm Hg in the control group; P =.05), the 2 study groups were completely comparable. Velocity and flux correlated significantly with the mean ocular perfusion pressure (r = 0.76, P<.001; r = 0.64, P =.002, respectively) in vasospastic subjects. Such correlations did not occur in the control group, and the difference between vasospastic patients and control subjects with regard to these correlations was statistically significant (P<.001 and P =.003, respectively). CONCLUSIONS: Choroidal blood flow seems, to some degree, to be independent of perfusion pressure, but not in subjects with acral vasospasm.
