Multilevel quality-guided phase unwrapping algorithm for real-time three-dimensional shape reconstruction.

A multilevel quality-guided phase unwrapping algorithm for real-time 3D shape measurement is presented. The quality map is generated from the gradient of the phase map. Multilevel thresholds are used to unwrap the phase level by level. Within the data points in each level, a fast scan-line algorithm is employed. The processing time of this algorithm is approximately 18.3 ms for an image size of 640x480 pixels in an ordinary computer. We demonstrate that this algorithm can be implemented into our real-time 3D shape measurement system for real-time 3D reconstruction. Experiments show that this algorithm improves the previous scan-line phase unwrapping algorithm significantly although it reduces its processing speed slightly.

Effects of salmon calcitonin on fracture healing in ovariectomized rats.

OBJECTIVE: To explore the effects of salmon calcitonin on the healing process of osteoporotic fractures in ovariectomized rats. METHODS: We performed this study in The First Affiliated Hospital of Guangzhou Medical College, Guangzhou, China, during the period March 2002 to December 2004. We used 120 female adult Wistar rats in this experiment, among which 90 underwent ovariectomy (OVX) and the other 30 had sham-operation. All rats had their left tibias fractured 3 months later. The 90 OVX rats were randomly divided into 3 groups with 30 in each, while the 30 sham-operated rats served as control group. After the fracture the rats had subcutaneous injection of normal saline, salmon calcitonin and estrogen, respectively. X-ray film, histological examination, bone mineral density (BMD) measurement and biomechanics testing were carried out to evaluate the fracture healing. RESULTS: Compared with OVX rats treated with normal saline, the rats with salmon calcitonin had significantly higher BMD values in the left tibia, higher max torque, shear stress of the left tibia 8 weeks after fracture (p<0.05), and presented with stronger callus formation, shorter fracture healing time and faster normalization of microstructure of bone trabeculae. CONCLUSION: Salmon calcitonin can, not only increase BMD in osteoporotic bone, but also enhance the bone biomechanical properties and improve the process of fracture healing in fractured osteoporotic bone.

Co-inoculation of DNA and protein vaccines induces antigen-specific T cell suppression.

Immunization can sometimes lead to antigen-specific immune suppression. In this study, we investigated this phenomenon by testing several combinations of DNA and protein vaccines directed against various viruses. We find that co-inoculation of mice with combined DNA and protein vaccines induces immune suppression if the two vaccines are "matched" by targeting the same antigen. Conversely, vaccine combinations never lead to immune suppression if they are derived from different viruses and, thus, mismatched antigenically. We have further identified CD4+CD25- T cells as the type of regulatory T cells induced by and are responsible for suppressing T cell activities in an antigen-specific manner in immunized animals. These regulatory T cells are phenotypically unique in their expression of Foxp3, IL-10, and IFN-gamma. Our study thus shows for the first time that co-administration of antigen-matched DNA and protein vaccines can generate this type of adaptive regulatory T cells.

Selective etching of metallic carbon nanotubes by gas-phase reaction.

Metallic and semiconducting carbon nanotubes generally coexist in as-grown materials. We present a gas-phase plasma hydrocarbonation reaction to selectively etch and gasify metallic nanotubes, retaining the semiconducting nanotubes in near-pristine form. With this process, 100% of purely semiconducting nanotubes were obtained and connected in parallel for high-current transistors. The diameter- and metallicity-dependent "dry" chemical etching approach is scalable and compatible with existing semiconductor processing for future integrated circuits.

Expression of human calcitonin by microencapsulated recombinant myoblasts.

A novel approach to treat post-menopausal osteoporosis is proposed by engineering cell lines for the secretion of human calcitonin (hCT) and which would then be suitable for implantation in different allogeneic hosts. Mouse myoblast were transfected with the cDNA for hCT using the liposome-based gene delivery technique and clones secreting of human calcitonin were isolated. Human calcitonin expression was detected by ELISA. Western blot and immunohistochemical analyses also confirmed the plasmid, pcDNA3-hCT, had been transfected into the cells. Upon enclosure in microcapsules, which are biocompatible membranes that permit exit of therapeutic proteins but not entry of immune mediators, the myoblasts continued to secrete human calcitonin into the culture medium. These results demonstrate the feasibility of using these encapsulated recombinant myoblasts to deliver human calcitonin and the potential of allergenic gene therapy for postmenopausal osteoporosis.

Characterization of PM(2.5) in the ambient air of Shanghai City by analyzing individual particles.

PM(2.5) samples were collected simultaneously at three representative areas (central city, industrial area and clean air suburban) of Shanghai City. Their morphologies and elemental compositions were determined by scanning electron microscopy coupled with energy analysis (SEM-EDX). The particles were classified into four groups based on morphology and elemental composition. Soot aggregates and spherical fly ash particles were the two dominant types and they were identified as originating from automobile exhaust, metallurgical industry and coal combustion. The size distribution of the particles showed that most had diameters in the range of 0.2-1.4 microm. Individual particles were measured by synchrotron radiation micro-beam X-ray fluorescence (micro-SXRF) and the micro-SXRF spectra were obtained. Pattern recognition techniques, which took the micro-SXRF spectrum of a single aerosol particle as its fingerprint, were used to identify the origins of the particles. Seven source types were identified. They were: metallurgical industry, vehicle exhaust, soil dust, coal combustion, diesel exhaust, oil combustion and motorcycle exhaust. Metallurgical industry, automobile exhaust, and coal combustion were recognized to be the main pollution sources of PM(2.5) in the air of Shanghai City.

Hydrogenation and hydrocarbonation and etching of single-walled carbon nanotubes.

We present a systematic experimental investigation of the reactions between hydrogen plasma and single-walled carbon nanotubes (SWNTs) at various temperatures. Microscopy, infrared (IR) and Raman spectroscopy, and electrical transport measurements are carried out to investigate the properties of SWNTs after hydrogenation. Structural deformations, drastically reduced electrical conductance, and an increased semiconducting nature of SWNTs upon sidewall hydrogenation are observed. These changes are reversible upon thermal annealing at 500 degrees C via dehydrogenation. Harsh plasma or high temperature reactions lead to etching of nanotubes likely via hydrocarbonation. Smaller SWNTs are markedly less stable against hydrocarbonation than larger tubes. The results are fundamental and may have implications to basic and practical applications including hydrogen storage, sensing, band gap engineering for novel electronics, and new methods of manipulation, functionalization, and etching of nanotubes.

Lipopolysaccharide (LPS) regulates TLR4 signal transduction in nasopharynx epithelial cell line 5-8F via NFkappaB and MAPKs signaling pathways.

The lipopolysaccharide (LPS) of Gram-negative bacteria induces the expression of cytokines and proinflammatory genes via the TLR4 signaling pathway in diverse cell types. The purpose of the present study was to test the hypothesis that the nasopharynx epithelial cells (NECs) could recognize and respond to LPS. The underlying molecular mechanisms were further elucidated in the NEC line 5-8F for its ability to activate the NFkappaB and TNF-alpha reporter genes, in response to LPS. After LPS stimulation, the TNF-alpha promoter activity and the relevant production of TNF-alpha were significantly increased in 5-8F cells. Moreover, LPS activated NFkappaB p65, ERK1/2 and JNK1/2 and induced their translocation to the nucleus. Western blot analysis showed that the expression of NFkappaB p65, MEK1, ERK1/2, JNK1/2, phospho-ERK1/2 and phospho-JNK1/2 proteins also was increased in NEC 5-8F cells, following the LPS stimulation. Additionally, the expression of TLR1-6, MD2 and CD14 was examined by RT-PCR, and the CD14 expression was determined by flow cytometry analysis. We demonstrated that the expression of CD14, TLR4 and MD2 was crucial for the NEC responses to LPS. In conclusion, our results provide novel mechanisms for the response of nasopharnyx epithelial cells to LPS stimulation, through NFkappaB and MAPKs signaling pathways.

Uptake of zinc, cadmium and phosphorus by arbuscular mycorrhizal maize (Zea mays L.) from a low available phosphorus calcareous soil spiked with zinc and cadmium.

In a multifactorial pot experiment, maize (Zea mays L.) with or without inoculation with the arbuscular mycorrhizal (AM) fungus Glomus mosseae BEG167 was grown in a sterilized soil spiked with three levels of zinc (0, 300 and 900 mg Zn kg(-1) soil) and three levels of cadmium (0, 25 and 100 mg Cd kg(-1) soil). At harvest after 8 weeks of growth, the proportion of root length of inoculated plants colonized decreased with increasing Zn or Cd addition, and was 56% in the absence of both metals and was reduced significantly to 27% in the presence of the higher levels of both metals. Mycorrhizal plants had higher biomass than non-mycorrhizal controls except at the highest soil level of Cd. Cadmium had more pronounced effects on plant biomass than did Zn at the levels studied and the two metals showed a significant interaction. The data suggest that mycorrhizal inoculation increased plant growth with enhancement of P nutrition, perhaps increasing plant tolerance to Zn and Cd by a dilution effect. AM inoculation also led to higher soil solution pH after harvest, possibly reducing the availability of the metals for plant uptake, and lowered the concentrations of soluble Zn and Cd in the soil solution, perhaps by adsorption onto the extrametrical mycelium.

Influence of scale-breaking phenomena on turbulent mixing rates.

Simulations not seen before compare turbulent mixing rates for ideal fluids and for real immiscible fluids with experimental values for the surface tension. The simulated real fluid mixing rates lie near the center of the range of experimental values. A comparison to theoretical predictions relating the mixing rate, the bubble width, and the bubble height fluctuations based on bubble merger models shows good agreement with experiment. The ideal fluid mixing rate is some 50% larger, providing an example of the sensitivity of the mixing rate to physical scale breaking interfacial phenomena; we also observe this sensitivity to numerical scale-breaking artifacts.

Efficacy of modified levamisole adjuvant on inactivated virus vaccine.

To improve efficacy, especially for the cell-mediated response to inactivated viral vaccines, a modified levamisole (LMS) adjuvant formulation, designated LMS+, was evaluated for its efficacy in mice and chickens, using Newcastle Disease Virus (NDV) as a model pathogen. Compared with oil adjuvant, the killed NDV in LMS+ induced a significantly higher helper T cell type 1 response, as shown by higher levels of interleukin-2, interferon-gamma, T cell proliferation, and delayed-type hypersensitivity responses, without sacrificing the level of IgG production in mice. In addition, vaccine in LMS+ formulation increased the expression of MHC and costimulatory molecules as well as the number of CD11c+ dendritic cells, suggesting that the better response to the LMS+ formulation occurs partly via the maturation of dendritic cells and activation of MHC-antigen presentation and costimulation. Furthermore, this formulation provides 100% protection in chickens after challenge with a lethal dose of virulent NDV strain F48E9 at 1000 ELD50 (50% egg lethal dose). These results demonstrated that modified LMS+ adjuvant could be used to improve both humoral and cell-mediated responses for inactivated viral vaccines and its development as an effective inactivated viral vaccine is warranted.

Origins of PM10 determined by the micro-proton induced X-ray emission spectra of single aerosol particles.

The micro-proton induced X-ray emission (micro-PIXE) spectrum of a single aerosol particle (SAP) was considered as its fingerprint for tracing its origin. A proton microprobe was used to extract fingerprints of SAPs. Environmental monitoring samples of PM(10) were collected from a heavy industrial area of Shanghai and were analyzed by proton microprobe for finding their pollution sources. In order to find the sources of SAPs collected from environmental monitoring sites, a fingerprint database of SAPs collected from various pollution sources was established. The origins of samples collected through environmental monitoring were identified by comparison of the micro-PIXE spectra of SAPs with those of SAPs in the fingerprint database using a pattern recognition technique. The results of this study show that most of the measured PM(10) is derived from metallurgic industry, soil dust, coal combustion, automobile exhaust, and motorcycle exhaust. The study also shows that the proton microprobe is an ideal tool for the analysis of SAPs. The unidentified particles of PM(10) are classified into seven classes by hierarchical cluster analysis based on the element peak intensity in the spectra.

Surface enhanced Raman scattering effects of silver colloids with different shapes.

By solution-based method, three kinds of silver colloids, self-assembled nanowires, triangular nanoplates and quasispherical nanoparticles, have been synthesized. TEM studies revealed that they exposed different crystal planes, such as {111} crystal planes to triangular nanoplates, mainly {100} and {111} planes to self-assembly nanowires. Hereby, do the distinct shapes and crystal planes have an impact on the surface enhanced Raman scattering (SERS)? The great differences of the SERS spectra of rhodamine B at these Ag colloids confirmed that the shapes and crystal planes of silver have great effect on Raman enhancement, especially the crystal planes.

Pc 4 photodynamic therapy of U87-derived human glioma in the nude rat.

BACKGROUND AND OBJECTIVES: As a potential therapy for malignant glioma, we tested the phthalocyanine photosensitizer Pc 4 for: (1) rapid clearance from the vasculature, (2) specificity for glioma, and (3) tumoricidal photosensitizing capability. STUDY DESIGN/MATERIALS AND METHODS: Parenchymal injection of U87 cells into athymic rat brains (N = 100) was followed after 12 days by tail vein injection of 0.5 mg/kg Pc 4. After 1 day, the tumor was illuminated with either 5 (N = 11) or 30 (N = 16) J/cm(2) red light at 672 nm. Sacrifice was 1 day later. The brains from these 27 animals underwent H&E (necrosis) and TUNEL assay (apoptosis) histology. Pc 4 concentration of explanted brains and tumors (N = 16), and all blood samples (N = 52) were determined by HPLC-MS 1 day post Pc 4 administration. RESULTS: Tumor-specific apoptosis was almost uniformly seen; however, necrosis was found mostly in the high-light-dose group. Pc 4 concentration in bulk tumor averaged 3.8 times greater than in normal brain. CONCLUSIONS: These results warrant expanding this pre-clinical study to seek effective baseline Pc 4 drug- and light-doses and infusion-to-photoirradiation timing that would be necessary for a Pc 4-mediated PDT clinical trial for glioma patients.

[Field distribution pattern and metabolic activity of AM fungi and their effects on Ipomoea batatas growth].

The study showed that after eight weeks of inoculating sweet potato seedlings with AM fungus Glomus intraradices BEG141 under field condition, the shoot dry weight, individuals of tuber and their fresh weight, as well as the P uptake by the plant increased significantly. Compared with the control, the root colonization rate, extraradical hyphal density and intraradical alkaline phosphatase (ALP) activity were greatly increased. After inoculation with BEG141, the total and succinate dehydrogenase (SDH)- active hyphae paralleling with ridge were much more than those in underside direction, while there was no significant difference in different directions in the control. The increase of intra- and extraradical active hyphae density and the change of AM fungus distribution pattern after inoculation with mycorrhizal fungi may be the main reasons for the growth improvement of sweet potato.

Source identification of PM10, collected at a heavy-traffic roadside, by analyzing individual particles using synchrotron radiation.

Synchrotron radiation microbeam X-ray fluorescence (micro-SXRF) was used to analyze individual aerosol particles collected at a height of 2 m above a heavy-traffic roadside in a heavy-industrial area of Shanghai. A pattern recognition technique, which took micro-SXRF spectra of single aerosol particles as its fingerprint, was used to identify the origins of the particles. The particles collected from the environmental monitoring site are mainly from metallurgic industry (26%), unleaded gasoline automobile exhaust (15%), coal combustion (10%), cement dust (10%) and motorcycle exhaust (8%).

[Effect of sodium phytate on alkaline phosphatase (ALP) activity in intraradical hyphae of AM fungi and development of its extraradical hyphae].

In this paper, a pot experiment with three compartments was installed to study the effect of Na-phytate on the development and metabolic activity of AM fungi. Maize was selected as host plant and inoculated with arbuscular mycorrhizal fungus (Glomus intraradices), and different amounts of Na-phytate were applied into hyphal compartments. The results showed that the absorbed P in the shoots and roots of maize inoculated with AM fungi was 1-2 times higher than that of non-inoculated maize. The ALP activity in intraradical hyphae of AM fungi was significantly decreased, and the total hyphal density of AM fungi increased with applied Na-phytate. Observations suggested that the ALP activity in intraradical hyphae of AM fungi and the development of its extraradical hyphae could be regulated by Na-phytate. Additionally, AM fungi could enhance the plant's P absorption and utilization from native soil P and additive Na-phytate.
