A novel replicating circular DNAzyme.

10-23 DNAzyme has the potential to suppress gene expressions through sequence-specific mRNA cleavage. However, the dependence on exogenous delivery limits its applications. The objective of this work is to establish a replicating DNAzyme in bacteria using a single-stranded DNA vector. By cloning the 10-23 DNAzyme into the M13mp18 vector, we constructed two circular DNAzymes, C-Dz7 and C-Dz482, targeting the beta-lactamase mRNA. These circular DNAzymes showed in vitro catalytic efficiencies (kcat/K(M)) of 7.82 x 10(6) and 1.36 x 10(7) M(-1) x min(-1), respectively. Their dependence on divalent metal ions is similar to that found with linear 10-23 DNAzyme. Importantly, the circular DNAzymes were not only capable of replicating in bacteria but also exhibited high activities in inhibiting beta-lactamase and bacterial growth. This study thus provides a novel strategy to produce replicating DNAzymes which may find widespread applications.

A calix[4]arene carceplex with four Rh2 4+ fasteners.

It is shown that a spheroidal carceplex can be assembled by linking two bowl-shaped calix[4]arenes via four dimetal units, (DAniF)(2)Rh(2) (DAniF = N,N'-di-p-anisylformamidinate), with a molecule (diethyl ether) or a cation (tetraethylammonium ion) trapped inside. The tetraethylammonium carceplex, 1b, has been characterized by X-ray crystallography, (1)H NMR, IR, and mass spectrometry. The tetraethylammonium ion fits snugly in the interior of the spheroidal carceplex. A two-fold axis of the tetrahedral cation coincides with the idealized four-fold axis of the cage, and the cation is disordered over two rotational orientations. Only one axial position on each dirhodium unit is occupied by a ligand, CH(3)CN or H(2)O. The carceplex is very stable, and the axial ligands can be exchanged in single crystals without disrupting the crystallinity of the samples. In this way, a red crystal of the complex with all axial positions occupied by acetonitrile can be transformed to a green crystal of the complex with two axial positions having acetonitrile and the other two having water by simply putting the crystal in contact with distilled water. The calix[4]arene used to make the carciplex structure is 25,26,27,28-tetra-n-propoxycalix[4]arene-5,11,17,23-tetracarboxylic acid. By employing 25,26,27,28-tetrapropoxy-5,17-dibromo-calix[4]arene-11,23-dicarboxylic acid, two 1:1 dimetal:calixarene compounds have also been made and characterized: 2, cis-Rh(2)(DAniF)(2)(calix)(CH(3)OH), and 3, cis-Mo(2)(DAniF)(2)(calix). The Rh-Rh distances in 1b are in the range of 2.410(2)-2.428(2) A, that in 2 is 2.4383(4) A, and the Mo-Mo distance in 3 is 2.0931(4) A.

Comparative study on the inclusion behaviour of cyclodextrin derivatives with venoruton and rutin by thin layer chromatography.

The interaction of rutin and venoruton (troxerutin), with alpha-, beta- and gamma-cyclodextrin (CD), hydroxypropyl-beta-cyclodextrin (HP-beta-CD) and methyl-beta-cyclodextrin (M-beta-CD) was investigated by reversed-phase thin layer chromatography on polyamide plates. A mobile phase consisted of NH(4)OH; NH(4)Cl buffer solution containing various CD concentrations (pH = 9.7, 20 degrees C) was used as mobile phase. The equilibrium constants (K(f)) and the retention factor (R(f)) were determined and used to study the inclusion process. The in fluence of CDs on the solubility of rutin and venoruton was characterized by R(M) values and the increasing hydrophilicity of drugs. The results show that the inclusion capacity of cyclodextrins follows the order HP-beta-CD > M-beta-CD > beta-CD > gamma-CD, and rutin is more easily included by the studied cyclodextrins than venoruton. In addition, the thermodynamic parameters (Delta H, Delta S) for the formation of complexes were obtained from the van't Hoff equation, displaying the enthalpy-entropy compensation effect.

Diapause induction and clock mechanism in the cabbage beetle, Colaphellus bowringi (Coleoptera: Chrysomelidae).

Photoperiodic control of diapause induction was investigated in the short-day species, Colaphellus bowringi, which enters summer and winter diapause as adult in the soil. Photoperiodic responses at 25 and 28 degrees C revealed a critical night length between 10 and 12 h; night lengths > or =12 h prevented diapause, whereas night lengths <12 h induced summer diapause in different degree. Experiments using non-24-h light-dark cycles showed that the duration of scotophase played an essential role in the determination of diapause. Night-interruption experiments with T=24 h showed that diapause was effectively induced by a 2-h light pulse in most scotophases; whereas day-interruption experiments by a 2-h dark break had a little effect on the incidence of diapause. The experiments of alternating short-night cycles (LD 16:8) and long-night cycles (LD 12:12) during the sensitive larval period showed that the information of short nights as well as long nights could be accumulated. Nanda-Hamner experiments showed three declining peaks of diapause at 24 h circadian intervals. Bünsow experiments showed two very weak peaks for diapause induction, one being 8 h after lights-off, and another 8 h before lights-on, but it did not show peaks of diapause at a 24 h interval. These results suggest that the circadian oscillatory system constitutes a part of the photoperiodic clock of this beetle but plays a limited role in its photoperiodic time measurement.

Critical segment of apocytochrome c for its insertion into membrane.

Apocytochrome c has a potent ability to insert spontaneously into membrane. To identify which sequences were critical for this insertion activity, a series of peptides N19, C8, C15 and C21, corresponding to sequences 1-19, 81-88, 74-88 and 68-88 of apocytochrome c, respectively, were synthesized and purified. Insertion ability into phospholipid monolayer, intrinsic fluorescence emission spectra, and the accessibility of peptide C21 to fluorescence quenchers: KI, acrylamide and HB showed that only segment 68-88 could insert into membrane, while other segments did not. CD spectra demonstrated that its interaction with liposomes containing negatively charged phospholipid could induce a partial alpha-helical conformation in peptide C21. It is interesting to note that a cooperation exists between segment 68-88 and 1-19 in the insertion of apocytochrome c and consequently translocation across membrane.

Phytoextraction of weathered p,p'-DDE by zucchini (Cucurbita pepo) and cucumber (Cucumis sativus) under different cultivation conditions.

Previous studies have shown that zucchini (Cucurbita pepo) and cucumber (Cucumis sativus) under field conditions are good and poor accumulators, respectively, of persistent organic pollutants from soil. Here, each species was grown under three cultivation regimes: dense (five plants in 5 kg soil): nondense (one plant in 80 kg soil): and field conditions (two to three plants in approximately 789 kg soil). p,p'-DDE and inorganic element content in roots, stems, leaves, and fruit were determined. In addition. rhizosphere, near-root, and unvegetated soil fractions were analyzed for concentrations of 11 low-molecular-weight organic acids (LMWOA) and 14 water-extractable inorganic elements. Under field conditions, zucchini phytoextracted 1.3% of the weathered p,p'-DDE with 98% of the contaminant in the aerial tissues. Conversely, cucumber removed 0.09% of the p,p'-DDE under field conditions with 83% in the aerial tissues. Under dense cultivation, cucumber produced a fine and fibrous root system not observed in our previous experiments and phytoextracted 0.78% of the contaminant, whereas zucchini removed only 0.59% under similar conditions. However. cucumber roots translocated only 5.7% of the pollutant to the shoot system, while in zucchini 48% of the p,p'-DDE in the plant was present in the aerial tissue. For each species, the concentrations of LMWOA in soil increased with increasing impact by the root system both within a given cultivation regime (i.e., rhizosphere > near-root > unvegetated) and across cultivation regimes (i.e., dense > nondense > field conditions). Under dense cultivation, the rhizosphere concentrations of LMWOAs were significantly greater for cucumber than for zucchini; no species differences were evident in the other two cultivation regimes. To enable direct comparison across cultivation regimes, total in planta p,p'-DDE and inorganic elements were mass normalized or multiplied by the ratio of plant mass to soil mass. For cucumber, differences in total p,p'-DDE and inorganic element content among the cultivation regimes largely disappear upon mass normalization, indicating that greater uptake of both types of constituents in the dense condition is due to greater plant biomass per unit soil. Conversely, for zucchini the mass normalized content of p,p'-DDE and inorganic elements is up to two orders of magnitude greater under field conditions than under dense cultivation, indicating a unique physiological response of C. pepo in the field. The role of cultivation conditions and nutrient availability in controlling root morphology, organic acid exudation, and contaminant uptake is discussed.

Modifying electronic communication in dimolybdenum units by linkage isomers of bridged oxamidate dianions.

Reactions of Mo(2)(O(2)CCH(3))(DAniF)(3), DAniF = N,N'-di-p-anisylformamidinate, with oxamidate dianions [ArNC(O)C(O)NAr](2-), Ar = C(6)H(5) and p-anisyl, give pairs of isomeric compounds where the [Mo(2)] units are bridged by the oxamidate anions. For the alpha isomers, the C-C unit of the dianion is nearly perpendicular to the Mo-Mo bonds, and these are essentially perpendicular to each other. For the beta isomers, the corresponding C-C unit and the Mo-Mo bonds are essentially parallel to each other. Each type of isomer is stable in solution. The electronic communication as measured by the DeltaE(1/2) for the oxidation of each of the Mo(2) units is significantly better for the beta isomers. This is supported also by the appearance of what is conventionally called an intervalence charge-transfer band in the near infrared region upon oxidation of the beta isomers but not the alpha isomers. Molecular mechanics and DFT calculations help explain the relative conformations in the alpha isomers and the relative energy differences between the alpha and beta isomers.

Fully localized mixed-valence oxidation products of molecules containing two linked dimolybdenum units: an effective structural criterion.

Two previously reported compounds [Mo(2)](CH(3)O)(2)M(CH(3)O)(2)[Mo(2)] (Cotton, F. A.; Liu, C. Y.; Murillo, C. A.; Wang, X. Inorg. Chem. 2003, 42, 4619), in which [Mo(2)] is an abbreviation for the quadruply bonded Mo(2)(4+) unit embraced by three (p-anisyl)NC(H)N(p-anisyl) anions and M = Zn (1) or Co (2), have been chemically oxidized. One-electron oxidation products [Mo(2)](CH(3)O)(2)M(CH(3)O)(2)[Mo(2)](PF(6)) (3, M = Zn; 4, M = Co) and the two-electron oxidation product [Mo(2)](CH(3)O)(2)Zn(CH(3)O)(OH)[Mo(2)](PF(6))(2) (5) have been isolated and structurally characterized. As expected, oxidations occur at the dimolybdenum units. The mono-charged cations in 3 and 4 have asymmetric molecular structures with two distinct [Mo(2)] units. In each case, one of the [Mo(2)] units has a lengthened Mo-Mo bond distance of 2.151[1] A, as expected for one-electron oxidation, whereas the other remains unchanged at 2.115[1] A. These correspond to bond orders of 3.5 (sigma(2)pi(4)delta(1)) and 4.0 (sigma(2)pi(4)delta(2)), respectively. The crystallographic results thus show unambiguously that in the crystalline state, the mixed-valence compounds (3 and 4) are electronically localized and the unpaired electron is trapped on one [Mo(2)] unit. These results are supported by the EPR spectra. The doubly oxidized compound 5 has two equivalent [Mo(2)] units, both with a Mo-Mo bond distance of 2.149[1] A. EPR and magnetic susceptibility measurements for 5 indicate that there is no significant ferromagnetic or antiferromagnetic spin coupling and the species is valence-trapped.

REMGa3Ge and RE3Ni3Ga8Ge3 (M = Ni, Co; RE = rare-earth element): new intermetallics synthesized in liquid gallium. X-ray, electron, and neutron structure determination and magnetism.

New quaternary intermetallic phases REMGa(3)Ge (1) (RE = Y, Sm, Tb, Gd, Er, Tm; M = Ni, Co) and RE(3)Ni(3)Ga(8)Ge(3) (2) (RE = Sm, Gd) were obtained from exploratory reactions involving rare-earth elements (RE), transition metal (M), Ge, and excess liquid Ga the reactive solvent. The crystal structures were solved with single-crystal X-ray and electron diffraction. The crystals of 1 and 2 are tetragonal. Single-crystal X-ray data: YNiGa(3)Ge, a = 4.1748(10) A, c = 23.710(8) A, V = 413.24(2) A(3), I4/mmm, Z = 4; Gd(3)Ni(3)Ga(8)Ge(3), a = 4.1809(18) A, c = 17.035(11) A, V = 297.8(3) A(3), P4/mmm, Z = 1. Both compounds feature square nets of Ga atoms. The distribution of Ga and Ge atoms in the REMGa(3)Ge was determined with neutron diffraction. The neutron experiments revealed that in 1 the Ge atoms are specifically located at the 4e crystallographic site, while Ga atoms are at 4d and 8g. The crystal structures of these compounds are related and could be derived from the consecutive stacking of disordered [MGa](2) puckered layers, monatomic RE-Ge planes and [MGa(4)Ge(2)] slabs. Complex superstructures with modulations occurring in the ab-plane and believed to be associated with the square nets of Ga atoms were found by electron diffraction. The magnetic measurements show antiferromagnetic ordering of the moments located on the RE atoms at low temperature, and Curie-Weiss behavior at higher temperatures with the values of mu(eff) close to those expected for RE(3+) free ions.

Subspecies-level variation in the phytoextraction of weathered p,p'-DDE by Cucurbita pepo.

Previous studies indicate that Cucurbita pepo can phytoextract highly weathered persistent organic pollutants (POPs) from soil and translocate large quantities to aerial tissues. To investigate intraspecific variability in uptake potential, a field study was conducted to quantify the phytoextraction of weathered p,p'-DDE by 21 cultivar varieties of summer squash from two distinct subspecies, C. pepo ssp texana and C. pepo ssp pepo. Significant differences exist between the two subspecies, with average root and stem to soil bioconcentration factors (BCF, dry weight ratio of contaminant concentration in the vegetation to that in the soil) of 7.22 and 5.40 for ssp pepo and of 2.37 and 0.454 for spp texana, respectively. The amounts of weathered p,p-DDE extracted from the soil by ssp pepo and ssp texana were 0.301 and 0.065%, respectively, with maximum values within each subspecies of 0.780 and 0.182%, respectively. The quantities of 14 inorganic elements were determined in both the soil and tissues (roots, stems, leaves, and fruit) of all 21 cultivar varieties. Phosphorus concentrations in the tissues of ssp pepo were 14 (fruit)-73% (stems) greater than those of ssp texana. These data support our hypothesis that the unique ability of certain cultivars of C. pepo to phytoextract highly weathered POPs from soil is the result of low molecular weight organic acid exudation as a unique phosphorus acquisition mechanism.

New chemistry of the triply bonded divanadium (V2 4+) unit and reduction to an unprecedented V2 3+ core.

We report here the synthesis and characterization of seven new divanadium compounds with the paddlewheel geometry bridged by nitrogen-donating ligands. Five of these contain the diamagnetic V(2)(4+) core with short V-V distances of less than 2.0 A, consistent with a formal triple bond. The V-V distances vary with the basicity of the bridging ligands; more basic ligands such as those of the guanidinate type have the shorter metal-metal separations, and those with the less basic formamidinate groups have longer separations. One compound, V(2)(DPhF)(4) (DPhF = the anion of N,N'-diphenylformamidine), has been reduced by one electron, and two structures, [K(THF)(3)]V(2)(DPhF)(4) and [K(18-crown-6)(THF)(2)]V(2)(DPhF)(4), have been obtained. These have an unprecedented V(2)(3+) core where the formal oxidation state of each vanadium atom is +1.5. The decrease in V-V bond distance and the multiline EPR spectrum in the reduced species provide evidence that these two molecules contain a bond of order 3.5.

A mixed-valence compound with one unpaired electron delocalized over four molybdenum atoms in a cyclic tetranuclear ion.

The first oxidation of a species derived from a compound having two linked, quadruply-bonded Mo2 units has been performed and [[cis-Mo2(DAniF)2]2(mu-Cl)4]PF6, 2, has been isolated and characterized in many ways; it has one unpaired electron and a fully delocalized structure where the Mo-Mo distances increase from 2.1191(4) A in the reduced species to 2.1453(3) A in 2 and the Kc of 1.3 x 10(9) is three orders of magnitude larger than that of the Creutz-Taube ion.

Molecular squares with paramagnetic diruthenium corners: synthetic and crystallographic challenges.

The reaction of Ru(2)(mu-O(2)CMe)(4)Cl with approximately 2.5 equiv of HDAniF (HDAniF = N,N'-di(p-anisyl)formamidine) in refluxing THF resulted in the synthesis of cis-Ru(2)(mu-DAniF)(2)(mu-O(2)CMe)(2)Cl (1). This Ru(2)(5+)complex, which has two labile acetate groups occupying two equatorial cisoid positions, was used for the construction of discrete paramagnetic supramolecular arrays. Subsequent reactions of 1 with HO(2)C-CO(2)H (oxalic acid) and 1,4-HO(2)C-C(6)H(4)-CO(2)H (terephthalic acid) followed by recrystallization from 1,2-C(2)H(4)Cl(2)/hexanes solutions containing wet MeCN or 4-Bu(t)py (4-tert-butylpyridine) gave crystals of [[is-Ru(2)(mu-DAniF)(2)Cl(H(2)O)]mu-oxalate)](4).MeCN.2(C(6)H(14)).12(H(2)O) (2) and [[is-Ru(2)(mu-DAniF)(2)Cl(0.646)(4-Bu(t)py)(1.354)]mu-terephthlate)](4)Cl(1.414).17(1,2-C(2)H(4)Cl(2)).C(6)H(14) (3), respectively. These are the first polygonal supramolecular assemblies consisting of paramagnetic M(2) units at each apex, where M(2) = Ru(2)(5+). Compounds 2 and 3 have been structurally characterized, and their electrochemistry and magnetic properties have been studied. These studies have confirmed that oxalate linkers are better than the terephthalate linkers in effecting electronic and magnetic coupling.

Trapping tetramethoxyzincate and -cobaltate(II) between Mo24+ units.

Two compounds of a new type, [Mo(2)](CH(3)O)(2)M(CH(3)O)(2)[Mo(2)] where [Mo(2)] is an abbreviation for Mo(2)[(p-MeOC(6)H(4))NCHN(p-MeOC(6)H(4))](3) and M = Zn (1) and Co (2), are reported. Discrete [M(OR)(4)](2-) ions, either as such or in the mu(2),eta(4) role, have not heretofore been described. In these compounds they have distorted tetrahedral structures and bridge two [Mo(2)] groups in much the same way as did SO(4)(2-), MoO(4)(2-), and WO(4)(2-) ions in other recently reported compounds (Cotton, F. A.; Donahue, J. P.; Murillo, C. A. Inorg. Chem. 2001, 40, 2229). The (1)H NMR spectrum of 1 and the visible spectrum and magnetic properties of 2 are consistent with these structures. The M(OCH(3))(4) bridges are moderately effective in coupling the two [Mo(2)] redox centers. Compounds 1 and 2 may also be viewed as having Zn(II) and Co(II) centers tetrahedrally coordinated by the bidentate ligand [Mo(2)[(p-MeOC(6)H(4))NCHN(p-MeOC(6)H(4))](3)(OMe)(2)](-). From that point of view they may be compared with Zn(DPM)(2) and Co(DPM)(2) (3), where DPM is the anion of dipivaloylmethane. For purposes of comparison, 3 has been fully characterized structurally, spectroscopically, and magnetically. Close analogies between 2 and 3 are shown to exist.

Enhancing the stability of trinickel molecular wires and switches: Ni(3)(6+)/Ni(3)(7+).

This paper describes in detail four new compounds that contain extended metal atom chains (EMACs) of three nickel atoms wrapped by either di(2-pyridyl)amide (dpa) or the new homologous ligand with an ethyl group at the para position of each pyridyl group, depa, and compares them to the precursor Ni(3)(dpa)(4)Cl(2) (1) and the oxidized and rather unstable Ni(3)(dpa)(4)(PF(6))(3) (2). The new molecules are Ni(3)(depa)(4)Cl(2) (3), Ni(3)(depa)(4)(PF(6))(3) (4), [Ni(3)(dpa)(4)(CH(3)CN)(2)](PF(6))(2) (5), and [Ni(3)(depa)(4)(CH(3)CN)(2)](PF(6))(2) (6). These compounds are fully described as to preparation, elemental composition, structure, infrared spectra, (1)H NMR spectra (where possible), electrochemistry, magnetic susceptibility, and an EPR spectrum for 4. The effects of (a) introducing the ethyl substituents on the ligands, (b) replacing axial anions by neutral axial ligands, and (c) oxidizing the Ni(3) chains are reported and discussed. The point of major interest is how oxidation profoundly alters the electronic structure of the EMAC.

Oxidation of Ni3(dpa)4Cl2 and Cu3(dpa)4Cl2: nickel-nickel bonding interaction, but no copper-copper bonds.

Of the known trinuclear dipyridylamido complexes of the first-row transition metals, M(3)(dpa)(4)Cl(2) (dpa is the anion of di(2-pyridyl)amine, M = Cr, Co, Ni, Cu), the one-electron-oxidation products of only Cr(3)(dpa)(4)Cl(2) and Co(3)(dpa)(4)Cl(2) have been isolated previously. Here we report one-electron-oxidation products of Ni(3)(dpa)(4)Cl(2) (1) and Cu(3)(dpa)(4)Cl(2) (3): Ni(3)(dpa)(4)(PF(6))(3) (2) and [Cu(3)(dpa)(4)Cl(2)]SbCl(6) (4). While there are no Ni-Ni bonds in 1, the Ni-Ni distances in 2 are 0.15 A shorter than those in 1, very suggestive of metal-metal bonding interactions. In contrast, the oxidation of 3 to 4 is accompanied by a lengthening of the Cu-Cu distances, as expected for an increase in electrostatic charge between positively charged nonbonded metal ions, which is further evidence against Cu-Cu bonding in either 3 or 4. A qualitative model of the electronic structures of all [M(3)(dpa)(4)Cl(2)](n+) (n = 0, 1) compounds is presented and discussed.

Effects of antidepressants on the exploration, spontaneous motor activity and isolation-induced aggressiveness in mice.

OBJECTIVE: To study the role of different antidepressants on exploration, spontaneous motor activity and isolation-induced aggressiveness in mice, further to discuss different mechanisms of their anti-aggression. METHODS: With an aggressive model induced by isolation housing in mice, antagonism of different antidepressants against isolation-induced aggression was evaluated. In the group-housed mice given the same treatment as aggressive test, exploration and spontaneous motor activity were measured. RESULTS: (1) Mianserin (0.5-5 mg/kg-1), buspirone (2.5-10 mg.kg-1) and meclobemide (2.5-10 mg.kg-1) significantly inhibited the exploration in the group-housed mice, but not fluoxetine (2.5-10 mg.kg-1), imipramine (2.5-10 mg.kg-1) and DOI (0.5-2 mg.kg-1); (2) Both mianserin and buspirone, but not fluoxetine, imipramine, meclobemide and DOI, obviously reduced spontaneous motor activity; (3) Fluoxetine, miaserin, imipramine and buspirone significantly and dose-dependently antagonized isolation-induced aggressive behavior, whereas meclobemide failed to attenuate aggression. DOI dual-regulated aggressiveness in isolation mice. CONCLUSION: Our findings suggest that the effects of fluoxetine, mianserin, buspirone, imipramine, meclobemide and DOI on exploration, spontaneous motor activity and isolation-induced aggression in mice are different, which may involve different pharmacological mechanisms underlying their anti-aggression in isolation mice. 5-HT1A and 5-HT2A/2C receptors may mediate isolation-induced aggressive behavior in mice. The involvement of 5-HT receptor subtypes needs further clarification.
