A comprehensive two-dimensional liquid chromatographic separation system based on the combination of an immobilized liposome chromatographic (ILC) column and an ODS column was developed for the separation of components in Ginkgo biloba, a traditional Chinese medicine. Two columns were coupled by a two-position, eight-port valve equipped with two storage loops, and the system was controlled by a computer. The effluent was detected both by a diode array detector and by an atmospheric pressure chemical ionization (APCI) mass spectrometer. Under the optimization separation conditions with the separation system, more than 41 components in the methanol extract of Ginkgo biloba were resolved. According to their UV and mass spectra, 13 of them were preliminarily identified as ginkgolide B, ginkgolide C, bilobalide, rutin, quercetin, quercetin-3-O-beta-D-glucosyl (1-2)-alpha-L-rhamnoside, quercetin-3-O-beta-D-glucoside, isorhamnetin, kaempferol-3-O-beta-D-glucosyl (1-2)-alpha-L-rhamnoside, isohamnetin-3-O-beta-D-rutinoside, kaempferol-3-O-beta-D-glucoside, kaempferol, kaempferol-3-O-beta-D-rutinoside.
In order to explore the effects of the local strong stimulation generated by electro-acupuncture for treatment of the upper segment ureterolithiasis, a controlled study was carried out among the treatment group (electro-acupuncture with strong stimulation), the control group I (medication) and the control group II (conventional acupuncture). The results showed that the differences in the cure rate and the total effective rate between the treatment group and the two control groups were significant in statistical analysis (P<0.05 and P<0.01 respectively). This indicates that better therapeutic effects can be obtained by the local strong stimulation generated by electro-acupuncture for treatment of the upper segment ureterolithiasis.
OBJECTIVE: To study the anti-myeloma activity of interleukin-2 activated bone marrow (ABM). METHODS: Bone marrow mononuclear cells (BMMNC) from multiple myeloma and iron-deficiency anemia patients were cultured in the presence of rIL-2. The anti-myeloma activity of ABM against U266 cells, cells expressing surface CD45, CD38, CD138, the levels of TNF-alpha and IFN-gamma in ABM culture supernatant were measured with MTT method, flow cytometry and ELISA method respectively after bone marrow was activated with rIL-2 for 24 and 72 hours. RESULTS: The tumor-killing activities against U266 cells of ABM were significantly increased compared with that of non-activated bone marrow (NBM) at 72 hours [(69.70 +/- 26.57)% vs (43.20 +/- 12.39)%, P < 0.05] and 24 hours [(34.25 +/- 11.93)% vs (26.53 +/- 5.48)%]. The CD45(-)CD38(+)CD138(+) cells of ABM from myeloma group at 72 hours were decreased from (8.46 +/- 3.66)% to (4.79 +/- 1.56)% (P < 0.05). TNF-alpha and IFN-gamma were detectable after cultured for 24 hours in both normal control group and myeloma group and went higher at 72 hours. The level of TNF-alpha and IFN-gamma were significantly increased in ABM compared with that in NBM (P < 0.05). Meanwhile, there was a positive relationship between the level of TNF-alpha, IFN-gamma and cytotoxicity of ABM from normal control group at 24 hours and 72 hours (P < 0.05), and was a negative relationship between TNF-alpha and IFN-gamma levels and the CD45(-)CD38(+)CD138(+) cells in myeloma group at 72 hours (P < 0.05). CONCLUSION: Normal BMMNCs activated with rIL-2 have tumor-killing activities against U266 cells. Myeloma cells and tumor burden were decreased in myeloma bone marrow after the marrow was activated with rIL-2. Production of TNF-alpha and IFN-gamma from bone marrow cells including T cells, monocyte-macrophages and NK cells activated with rIL-2 might be involved in anti-myeloma activity of ABM.
OBJECTIVE: To establish a fast, simple, efficient and minimally invasive method for nuclear transfer (NT) to study the early development of mouse embryos reconstructed with cumulus cell nuclei in vitro. METHODS: With a sharp-tipped enucleation needle, an incision approximately 25 mm in length was made in the zona pellucida of a rat oocyte, through which the first polar body was removed and the metaphase II chromosome-spindle complex was gently aspirated along with a minimal volume of the cytoplasm by slightly pressing and vacuum aspiration of the oocyte. A cumulus cell nuclei from C57BL/6j mouse, 10-12 mm in diameter, was inserted into the perivitelline space of the enucleated oocyte. The fusion of the donor-recipient pair was induced by electrofusion and nuclear formation was observed. The development of 2-cell, 4- to 8-cell and morula-stage embryos was observed after a 72-hour culture of the reconstructed oocytes in vitro. RESULTS: The modified NT method enabled one-step removal of the whole nucleus from the oocyte with confirmed reliability of complete nuclear removal by Hoechst 33342 staining of the removed nuclei examined under UV light. The process of enucleation took an average time of 15 s, and the survival rate of the enucleated oocytes reached 95%. The success rate of 76.7% was achieved for cumulus cell nucleus insertion into the zona pellucida of the enucleated oocytes and pronucleus formation occurred in 62.2% of the reconstructed oocytes with nuclear transfer. After 72 h of culture in vitro of the reconstructed oocytes in CZB medium, the percentage of embryos that developed into 2-cell, 4- to 8-cell and morula (more than 16 cells) stages were 57.5%, 39.1% and 27.6%, respectively. Microsatellite sequences (D7Mit22 and D4Mit204) were amplified from the DNA of the reconstructed embryos for identifying their origin, which was proved to be C57BL/6j mouse. CONCLUSION: The modified NT method is simple, minimally invasive, efficient and practicable to reconstruct mouse embryos with somatic cell nuclei.
Many phytopathogenic bacteria were founded to release the superoxide anion (O2-) by themselves from different sites and the production of O2- was related to the pathogenicity of strains. The presences of O2- in cytomembrane, cell wall and filtrate were confirmed by chemical method and electron spin resonance (ESR). The results show that filtrate may be the primary site of O2- production due to its higher O2- generating activity either in the presence or the absence of DDC. Additional lines of evidence further suggested a regulatory mechanism of O2- production in phytopathogenic bacteria.
OBJECTIVE: To study the influence of Xinqin tablets on guinea-pig nasal hypersensitivity. METHOD: 2,4-Toluene Diisocyanate (TDI) was selected as antigen and used in nose to establish guinea-pig allergic rhinitis. The effects of Xinqin tablets on symptoms of nasal hypersensitivity in guinea-pigs, histamine content of nasal mucosa and activity of nitric oxide synthase (NOS) were examined. RESULT: Xinqin tablets could significantly relieve the pathological symptoms of nasal hypersensitivity in guinea-pig, reduce histamine content of nasal mucosa and inhibit the activity of nitric oxide synthase. CONCLUSION: Xinqin tablets have significant effect on nasal hypersensitivity, and prevent the occurrence of allergic rhinitis.
From the methanol extract of the whole plant of Achillea wilsoniana, 23 compounds were isolated. Their structures were elucidated by spectroscopic methods and chemical transformations. Three of them are new: 4E, 10E-9beta-hydroxy-3-(2-methylbutyroyloxy)-germacra-4,10(1)-diene-12,6alpha-olide (1), 4E,10E-3-(2-methylbutyroyloxy)-germacra-4,10(1)-diene-12,6alpha-olide (2) and 1beta,6a-dihydroxy-10beta-methyl-5alphaH,7alphaH-eudesm-4-one (3). In addition, 1beta-hydroxy-alpha-xyperone (5) and 9beta-acetoxy-3-(2methylbutyroyloxy)-germacra-4,10(1)-diene-12,6alpha-olide (1a) exhibited effective antibacterial activity against Staphylococcus aureus.
OBJECTIVE: To improve the understanding of the pathogenesis of lupus nephritis (LN) by studying the relationship between apoptosis and proliferation in children with LN. METHODS: Thirty-one children (23 girls, 8 boys, aged 6-16 years) with biopsy-proven LN (21 type IV, 6 type V, 2 type II and 2 type III) and nine controls were included in the study. Apoptosis was detected by in situ nick-end labeling techniques (TUNEL) in renal biopsy samples. Proliferation was shown by PCNA detection. Immunohistochemistry was employed to detect the expressions of PCNA, PDCD5 and Caspase-3. The images of glomeruli (G) and renal tubuli (T) were selected randomly on each biopsy sample, respectively, and transferred into computer by SECOM video camera. The quantity of PDCD5 and Caspase-3 expressions were evaluated according to the average intensity of PDCD5 and Caspase-3. RESULTS: 1. Compared to control group, the patients with LN had increased apoptotic cells (G: 0.14 +/- 0.09 vs. 0.86 +/- 0.26; T: 0.16 +/- 0.07 vs. 1.16 +/- 0.42; P < 0.01) and increased PCNA positive cells (G: 0.47 +/- 0.25 vs. 8.45 +/- 2.83; T: 1.32 +/- 0.15 vs. 13.73 +/- 3.54; P < 0.01) in glomeruli and renal tubuli. The high ratios of PCNA/apoptosis (P/A) were observed in glomeruli (3.37 +/- 1.93 vs. 10.01 +/- 2.96, P < 0.01) and tubuli (8.94 +/- 1.79 vs. 12.81 +/- 3.91, P < 0.05) in LN group. 2. There was no difference in expression of PDCD5 in glomeruli between LN children and controls (0.09 +/- 0.05 vs. 0.08 +/- 0.02, P > 0.05). The expression of PDCD5 in renal tubuli decreased significantly in LN children compared with controls (0.13 +/- 0.05 vs. 0.21 +/- 0.07, P < 0.01). 3. The expression of Caspase-3 in children with LN increased significantly in glomeruli and tubuli compared with that in controls (0.22 +/- 0.07 vs. 0.05 +/- 0.02; 0.08 +/- 0.03 vs. 0.05 +/- 0.01; P < 0.01). 4. The number of glomerular apoptotic cells showed a positive correlation with the expression of Caspase-3 (r = 0.718, P < 0.01), but no correlation with the expression of PDCD5 in all cases (r = 0.054, P > 0.05). The intensity of PDCD5 expression in glomeruli and renal tubuli showed no correlation with that of Caspase-3 expression in all cases (r = 0.061, P > 0.05, r = 0.049, P > 0.05). Stepwise multiple regression revealed that the number of apoptotic cells in renal tubuli correlated negatively with the intensity of PDCD5 expression, whereas correlated positively with intensity of Caspase-3 expression in all cases. CONCLUSIONS: 1. Although the number of apoptotic cells in the kidney of LN was more than that in the kidney of control group, the apoptosis in the kidney of LN was relative insufficient compared with the proliferation. 2. Caspase-3 was involved in the apoptosis of glomeruli and renal tubuli of LN. 3. It was unclear if PDCD5 participated in the apoptosis of glomeruli of LN. While PDCD5 might participate the apoptosis of renal tubuli in LN.
In order to explore what role relative cytokines play in acute GVHD (aGVHD) of mice transplanted with H-2 haploidentical nonmyeloablative bone marrow, a murine model was established by using C57BL/6J mouse as donor and (C57BL/6J x BALB/C) F1 mouse as the recipient. The recipient mice were given CsA and mycophenolate mofetile (MMF) for prophylaxis of aGVHD. The expression levels of IL-2, INFgamma, IL-4 and IL-10 in the spleen were detected by semi-quantitative RT-PCR at different time points after transplantation. The results showed that the expression levels of these cytokines increased slightly in the group only received nonmyeloablative conditioning. The expression levels of IL-2 and INF-gamma elevated significantly after transplantation in group 2 (without aGVHD prophylaxis), peaked at day 21 and day 14 respectively, then dropped rapidly, returned to the basal levels at day 35. The study on kinetic pattern of expression of IL-2 and INF-gamma in group 3 (with aGVHD prophylaxis) gave a similar result to group 2, but the peak value of cytokine expression in group 3 was much lower than that in group 2. The expression of IL-4 in Group 2 and group 3 all peaked at day 14, but the peak value in group 3 was higher than that in group 2, and decreased slowly in group 3. The expression of IL-10 increased gradually after transplantation, peaked at day 14, decreased after day 21, elevated again until day 35. It is concluded that the expression levels of these cytokines in the spleen all increase after H-2 haploidentical nonmyeloablative bone marrow transplantation. CsA and MMF can reduce the incidence and severity of aGVHD by down-regulating the expression levels of IL-2 and INF-gamma.
Phox homology (PX) domains specifically bind to phosphoinositides, and this interaction is crucial for their cellular function. A full-length cDNA of human PX domain containing serine/threonine kinase gene (PXK) that we termed PXK_v1 had previously been cloned. PXK_v1 consists of a S_TKc domain (serine/threonine kinases, catalytic domain), but lacks several residues that are indispensable for intrinsic catalytic activity. Evidence obtained in the present study demonstrated the existence of four other splice isoforms of human PXK in fetal brain, designated as PXK_v2, PXK_v3, PXK_v4 and PXK_v5. The results of RT-PCR indicated that human PXK_v1, PXK_v2 and PXK_v4 transcripts were widely expressed in human adult tissue, except heart tissue. In contrast, PXK_v3 transcripts were only expressed in peripheral blood leukocytes at a low level and PXK_v5 transcripts were not detectable in any of the tissue analyzed. Subcellular localization analysis of EGFP-PXK fusion proteins in COS7 cells indicated that EGFP-PXK_v3 had a different subcellular localization compared to other EGFP-PXK fusion proteins. Mutation analysis of EGFP-PXK_v1 showed that PXK_v1-Tyr56 and Arg92 are essential for subcellular localization of the protein in the cytoplasm.
AIM: To investigate the effect of EP(3) agonist--ONO-AE-248 on neutrophils. METHODS: Electron microscopy, MTS assay, RT-PCR and FACS were used to observe the morphology, viability, EP(3) receptor expression and apoptosis of neutrophils treated with ONO-AE-248. RESULTS: EP(3) receptor was expressed on neutrophils. ONO-AE-248 rapidly caused a unique form of neutrophil death, showing morphological changes of nucleus, including fusion of the lobules, blebbing and rupture of the nuclear membrane, which were different from typical morphological changes of apoptosis and necrosis. The death was dependent on ATP. CONCLUSION: The death of neutrophils caused by ONO-AE-248 is likely to be one of the forms of "non-apoptotic programmed cell death".
OBJECTIVE: To isolate hantavirus and to genotype isolated strains of hantavirus (SY 13) in Liaoning province. METHODS: Hantavirus was isolated by means of cell culture. The G1, G2 fragments of M segments and S segment of the strain of SY 13 were amplified respectively by means of RT-PCR in order to compare and analyze the homology between the sequences of the three isolated gene fragments and the sequences downloaded from GenBank with neighbor-joining through sequence analysis software of DNA Star, Clustal, Phylip, etc. RESULTS: SY 13 and HTN showed higher homology, reached up to 79.3%-97.0%; SY 13 and BAO 10, BAO 14, JIANG 13, BAO 9, Q 33 were on the same branch and their homology reached up to 95.0%-97.0%. CONCLUSION: The isolated hantavirus strain in Liaoning area was the same as the one in Heilongjiang area which belongs to HTN H4.

OBJECTIVE: To investigate the expressions of telomerase reverse transcriptase (TRT) and vascular endothelial growth factor (VEGF) and their correlation in prostate cancer (PCa). METHODS: TRT and VEGF expressions were assayed in 30 cases of PCa and 30 cases of benign prostatic hyperplasia (BPH) by means of immunohistochemistry (SP) combined with computer assisted image analysis. RESULTS: The expression of TRT was detected in 19 of the 30 cases of PCa and 5 of 30 cases of BPH, and that of VEGF in 23 of the 30 PCa and 14 of the 30 BPH patients. TRT and VEGF expressions were significantly higher in cancer tissues than in BPH (P < 0.05). A significant correlation was observed between TRT and VEGF expressions (r = 0.8333, P < 0.05). CONCLUSION: The expression of TRT or VEGF might be a malignant phenotype in PCa. The expression of TRT is significantly correlated with that of VEGF, but the mechanisms are yet to be further studied.
Several bacteria, Bacillus brevis R-6, Pseudomonas delafleldii R-8, Nocardia globerula R-9, Bacillus sphaericus R-16, Rhodococcus erythropolis LSSE8-1 and Gordonia nitida LSSEJ-1, which can convert dibenzothiophene into 2-hydroxybiphenyl and sulfate, were investigated. Desulfurization products were quantitively determined by HPLC. Result revealed that each of these bacteria desulfurize DBT at a different rate. In order to obtain more information, the fragments encoding desulfurizing enzymes were studied. Desulfurization genes of R-6 and R-8 were separately amplified via PCR with specific primers based on the related sequences of Rhodococcus sp. IGTS8. Both sequences areminimally 99% related to IGTS8 sequence. Afterwards, dsz operon of LSSEJ-1 and R-9 were amplified and cloned. Sequences are also highly conservative. Data shows that identity of dszA between R-9 and IGTS8 is 99.6%, and identity of dszA between LSSEJ-1 and IGTS8 is 99.9%; dszB sequence of R-9 and LSSEJ-1 is 99.6% similarity to their counterpart sequence from IGTS8;Identity of dszC between R-9 and IGTS8 is 99.9%, and identity of dszC between LSSEJ-1 and IGTS8 is 99.1% . It may be deduced that the origins of desulfurization genes from mesophilic bacteria are the same.
Quercetin is a herbal flavonoid derived from various foods of plant origin and plays a role in anti-inflammation. Although a number of researches in the field have been done, the mechanism of anti-inflammatory effect of quercetin should be further clarified. In the present study, we investigated the effects of quercetin on IL-6 production by LPS-stimulated neutrophils in human. Neutrophils were were pre-treated with quercetin at the final concentrations of ranging from 0-80 microM for 30 min, or not treated, and then incubated in the presence or absence of lipopolysaccharide (LPS) at a final concentration of 100 ng/ml for indicated time. The secretion level of IL-6 in the culture supernatants was assayed by ELISA, the intracellular level of IL-6 was detected by flow cytometry and the expression of IL-6 mRNA was analyzed by RT-PCR. The experiment results showed that neutrophils cultured with medium or quercetin alone did not express IL-6, but LPS (100 ng/ml) induced IL-6 expression of neutrophils. However, after pre-treatment of neutrophils with quercetin (40 microM) for 30 min, the inducible effects of LPS on the increase of IL-6 secretion, intracellular IL-6 level and IL-6 mRNA expression by neutrophils were abrogated. IL-6 is one of the important pro-inflammatory factors, especially in early phage of inflammation. Thus, our data suggested that quercetin might exert its anti-inflammatory effect through negatively modulating pro-inflammatory factors, such as IL-6. The inhibitory effects of quercetin on IL-6 production by neutrophils may provide a theoretical basis on future therapy of inflammation.
OBJECTIVE: To study the optimal extraction process of polysaccharide from Baphicacanthus cuaia (Nees) Bremek. METHODS: The optimal extraction process was selected with uniform design and statistical analysis system (SAS). The content of polysaccharide in the extract was determined to investigate the influence of temperature of extraction, the time of extraction and the solid-liquid ratio on it.
