Hydration and dewetting near fluorinated superhydrophobic plates.

The water dynamics near nanoscale fluorinated (CF(3)(CF(2))(7)(CH(2))(2)SiH(3)) monolayers (plates) as well as possible dewetting transitions in-between two such plates have been studied with molecular dynamics simulations in this paper. A "weak water depletion" is found near the single fluorinated surface, with an average water density in the first solvation shells 6-8% lower than its hydrogenated counterpart. The fluorinated molecules are also found to be water impermeable, consistent with experimental findings. More surprisingly, a dewetting transition is found in the interplate region with a critical distance D(c) of 10 A (3-4 water diameters) for double plates with 8 x 8 molecules each (plate size approximately 4 nm x 4 nm). This transition, although occurring on a microscopic length scale, is reminiscent of a first-order phase transition from liquid to vapor. The unusual superhydrophobicity of fluorocarbons is found to be related to their larger size (or surface area) as compared to hydrocarbons, which "dilutes" their physical interactions with water. The water-plate interaction profile shows that the fluorinated carbons have a 10-12% weaker water-plate interaction than their hydrogenated counterparts in the nearest solvation shell, even though the fluorocarbons do have a stronger electrostatic interaction with water due to their larger partial charges. However, the van der Waals interactions dominate the water-plate interaction within the nearest shell, with up to 90% contributions to the total interaction energy, and fluorocarbons have a noticeably weaker (by 10-15%) van der Waals interaction with water in the nearest shell than do hydrocarbons. Both the slightly weaker water-plate interaction and larger surface area contribute to the stronger dewetting transition in the current fluorinated carbon plates.

Biological fingerprinting analysis of the interactome of a kinase inhibitor in human plasma by a chemiproteomic approach.

In this study, a gel free chemiproteomic method based on chromatography was developed and applied for the biological fingerprinting analysis of complex biological system. p-Aminobenzamidine (ABA), an inhibitor of trypsin-like serine proteases, was immobilized for characterizing their interacting proteins in human plasma. By the proteomic analysis method, 214 proteins were identified with obvious affinity to the immobilized ABA. By searching the sequences of above proteins with consensus patterns of the two active sites, seven proteins belong to trypsin-like serine protease group were found. Based on the Gene Ontology annotation, the identified trypsin-like serine proteases have the function of catalytic activity and calcium ion binding, and are mainly involved in the biological process of blood coagulation. Eight more other proteins related to calcium ion binding and blood coagulation were found. Nearly all of these proteins cannot be identified by directly analyzing the plasma sample demonstrating the chemiproteomics a useful approach to characterize interacting proteins in the low abundance range.

Identification and characterization of a novel gene, Mcpr1, and its possible function in the proliferation of embryonic palatal mesenchymal cells.

We cloned a novel mouse cDNA, Mcpr1 (mouse cleft palate-related gene 1), between retinoic acid (RA)-treated murine embryonic palatal and control shelves by improved subtractive hybridization. Its transcript was identified by Northern blotting. The open reading frame encodes 132 amino acids and shows almost no identity to other genetic products. Mcpr1 expression could be detected extensively in adult mouse tissues and during murine embryonic development. It was identified to be significantly stimulated by RA in murine palatal shelves at embryonic day 12 and in palatal mesenchymal cells in vitro. We demonstrate that MCPR1 protein was localized primarily in the cytoplasm and could be synthesized and secreted by transfected COS-7 cells. Both the secretory and recombinant proteins of Mcpr1 inhibited proliferation of murine embryonic palatal mesenchymal cells and impeded the progression from the G1 to S phase in the cell cycle. The cells were prone to apoptosis after exposure to glutathione S-transferase-MCPR1. Furthermore, knockdown of MCPR1 protein levels by antisense oligodeoxynucleotides promoted progression of cells from the G1 to S phase and completely abolished the RA-induced block of the cell cycle from the G1 to S phase. These findings suggest that Mcpr1 might function as one of the RA-up-regulated genes involved in inhibiting cell proliferation during palatogenesis and RA-induced cleft palate by regulating proliferation and apoptosis of embryonic palatal mesenchymal cells and might even play a role in the development of many other organs.

Medical withdrawal management in Vancouver: service description and evaluation.

This study evaluates a medically managed inpatient detoxification service (Vancouver Detox (VD)) and investigates its clients' characteristics over a 1-year period. We present information on the daily operation at VD, and examine accessibility and efficiency of it from five perspectives: wait time, pretreatment attrition rate, treatment completion rate, length of stay (LOS), and occupancy rate (OR). In addition, we explore the impacts of seasonality and the monthly welfare check on the utilization of the service. Among 2294 referrals, 35% dropped out without engaging in treatment. Among those who were admitted to VD, 78% completed the treatment program. Shorter wait time and lower OR were found during the welfare check issuance period, and lower OR was found during summer. In addition, the patterns of utilization of VD were changed over the welfare check issuance period. Our analysis indicates that operational accessibility and efficiency at VD could be improved by specifically addressing these factors.

Improving the performance of sequencing batch reactor (SBR) by the addition of zeolite powder.

Two types of operation means "SBR reactor alone (control reactor)" and "adding zeolite powder into SBR reactor (test reactor)" were used to treat municipal wastewater. The test results revealed that zeolite powder addition could improve the activity of the activated sludge. It was investigated the specific oxygen utilization rate (SOUR) of the tested zeolite sludge were about double times that of the control activated sludge, and the nitrification rate and settling property of zeolite-activated sludge were both improved. Due to the combination of zeolite adsorption for NH(4)(+)-N and enhanced simultaneous nitrification and de-nitrification (SND), a higher nitrogen removal was observed in test reactor compared to the control reactor, and the addition of zeolite powder is helpful to inhabit sludge bulking. In addition, through long-term parallel shock load test, it was found that the zeolite powder addition could enhance the ability of activated sludge in resisting the shock load of organics and ammonium. Compared to the control activated sludge, zeolite powder added activated sludge could remove COD, NH(4)(+)-N, TN and TP significantly in a shorter cycle time. At the same operational time period, the test SBR could treat wastewater quantity 1.22 times that treated in control SBR.

Influence of electric field on the phase transitions of the hexagonal cylinder phase of diblock copolymers.

We have used the cell dynamic simulations (CDS) method to study the evolution of asymmetric and symmetric diblock copolymers under electric fields. For symmetric diblock copolymers, long-range-ordered lamellar phases form readily under electric fields. For asymmetric diblock copolymers, sphere-to-cylinder phase transitions occur rapidly when strong electric fields are applied, but it takes longer for the system to form hexagonal cylinder structures. The results of these simulations suggest that the sphere phase is stable under weak electric fields, but a threshold electric intensity exists for the sphere-to-cylinder phase transition. In addition, we also studied the kinetic pathways of the transition of the lamellar phase to the hexagonal cylinder phase of the asymmetric diblock copolymers under electric fields. Hexagonal cylinder structures form when the lamellar phase is subjected to a sudden temperature jump. The scattering functions suggest that the hexagonal cylinder structures are very regular and possess very few flaws.

Estrogen regulates epithelial cell deformability by modulation of cortical actomyosin through phosphorylation of nonmuscle myosin heavy-chain II-B filaments.

The objective of the study was to understand how estrogen modulates the rigidity of the cytoskeleton in epithelial cells. Estrogen depletion decreased, and treatment with 17beta-estradiol increased deformability of cervical-vaginal epithelial cells. Estrogen also induced redistribution of nonmuscle myosin II-B (NMM-II-B); lesser interaction of NMM-II-B with actin; increased phosphorylation of NMM-II-B-heavy chains at threonine and serine residues; and decreased filamentation of NMM-II-B in vitro. The effects of 17beta-estradiol were time and dose related and could be mimicked by diethylstilbestrol. The effects of estrogen were blocked by cotreatment with antisense oligonucleotide for the estrogen receptor-alpha and inhibited by ICI-182,780 and tamoxifen; omission of epithelial growth factor (EGF) from the culture medium; and cotreatments with the EGF receptor inhibitor AG1478, the ERK-MAPK inhibitor PD98059, the casein kinase-II (CK2) inhibitor 5,6-dichloro-1-beta-(D)-ribofuranosylbenzimidazole, the Rho-associated kinase inhibitor Y-27632, and the nonspecific phosphatase inhibitor okadaic acid. Coadministration of 5,6-dichloro-1-beta-(D)-ribofuranosylbenzimidazole plus okadaic acid blocked the 17beta-estradiol effect. H-89 or LY294002 did not significantly affect estrogen effects. Treatment with estrogen increased activation of ERK1/2 and CK2 activity. These data suggest a novel pathway of estrogen regulation of the cytoskeleton in epithelial cells. The effect is mediated by estrogen receptor-alpha and involves in part the EGF-EGF receptor and ERK-MAPK cascades as proximal signaling networks and the CK2 and Rho-associated kinase-regulated myosin heavy chain phosphatase as terminal effectors. Augmented phosphorylation of NMM-II-B can block filamentation and induce disassociation of the myosin from the cortical actin, and disruption of the actomyosin ring can increase cell deformability. This mechanism can explain estrogen regulation of paracellular permeability in cervical-vaginal epithelia in vivo.

Intra- and inter-ethnic differences in the allele frequencies of cytochrome P450 2B6 gene in Chinese.

This study aimed to investigate the allele frequencies of CYP2B6 gene in 193 Han Chinese and compared with 91 Uygur Chinese. Five single nucleotide polymorphisms of CYP2B6, 64C > T, 516G > T, 777C > A, 785A > G and 1459C > T, were tested using the polymerase chain reaction-restriction fragment length polymorphism method. The allele frequencies for CYP2B6*2, CYP2B6*3, CYP2B6*4, CYP2B6*5, CYP2B6*6, CYP2B6*7 and CYP2B6*9 in Han and Uygur Chinese were 0.034 and 0.027, 0 and 0.011, 0.091 and 0.033, 0.003 and 0.049, 0.184 and 0.214, 0 and 0.022, and 0.018 and 0.044, respectively, with CYP2B6*4, CYP2B6*5, and CYP2B6*7 being significantly different between these two races (P < 0.05). CYP2B6*6 was the most prevalent allele among all detected variants in Han and Uygur Chinese. The most frequent genotypes were CYP2B6*1/CYP2B6*1 (50.8%), CYP2B6*1/CYP2B6*6 (24.4%), and CYP2B6*1/CYP2B6*4 (7.3%) in Han subjects, whereas the most frequent genotypes in Uygur subjects were CYP2B6*1/CYP2B6*1 (36.3%), CYP2B6*1/CYP2B6*6 (25.3%), CYP2B6*1/CYP2B6*5 (5.5%) and CYP2B6*6/CYP2B6*6 (5.5%). The frequencies of 64C > T mutation in Han and Uygur Chinese were significantly lower than that in American Caucasian (P < 0.05). These results indicate that there were marked ethnic differences in the mutant frequencies of CYP2B6 between Chinese and other ethnic groups. Further studies are warranted to explore the clinical impact of such ethnic differences.

Electromagnetic functionalized and core-shell micro/nanostructured polypyrrole composites.

Core-shell micro/nanostructured and electromagnetic functionalized polypyrrole (PPy) composites were prepared by a self-assembly process associated with the template method in the presence of p-toluenesulfonate acid (p-TSA) as the dopant, in which the spherical hydroxyl iron (Fe[OH], 0.5-5 microm in diameter) functioned not only as the hard template, but also as the "core" of the micro/nanostructure, and the self-assembled PPy-p-TSA nanofibers (20-30 nm in diameter) acted as the "shell" (50-100 nm in thickness) of the microspheres. We found that the core-shell micro/nanostructures exhibit controllable electromagnetic properties by adjusting the mass ratio of Fe[OH] to pyrrole monomer. The micelle model was proposed to interpret the self-assembly of the core-shell micro/nanostructured composites.

Workplace, psychosocial factors, and depressive symptoms among working people with arthritis: a longitudinal study.

OBJECTIVE: To examine the role of demographic, illness-related, workplace support, workplace activity limitations, arthritis-related work changes, and psychosocial factors in predicting subsequent depressive symptoms among employed people with arthritis. METHODS: In a prospective study, 366 employed individuals with arthritis were recruited from Toronto, Canada. Respondents completed a structured questionnaire assessing demographic, disease-related factors, workplace support, and employment-related transitions, as well as psychosocial variables at 2 timepoints 18 months apart. Depression was assessed using the Center for Epidemiologic Studies Depression Scale. Hierarchical multiple regression was used for analyses. RESULTS: Individuals with greater education reported significantly less depression. Lower workplace support and greater workplace activity limitations were significantly associated with future depressive symptoms. No relationship was found between work transitions and later depression, but more work changes were strongly associated with concurrent depressive symptoms. An association was also found between greater pain catastrophizing and future depressive symptoms. CONCLUSION: Our results highlight the need to assess the influence of work-related changes, workplace support, and psychosocial variables on depressive symptoms among people with arthritis. These findings suggest that workplace interventions should address not only ways to reduce workplace activity limitations, but also ways to better manage emotional distress related to working with arthritis.

Screening and analysis of an antineoplastic compound in Rhizoma Chuanxiong by means of in vitro metabolism and HPLC-MS.

A new screening and analysis method that combines in vitro metabolism with high-performance liquid chromatography-mass spectrometry (HPLC-MS) was developed for the screening and analysis of an antineoplastic compound, coniferyl ferulate, which is present in the rhizome of Rhizoma Chuanxiong. Infrared (IR), ultraviolet visible spectroscopy (UV-Vis), nuclear magnetic resonance (NMR) and element analysis were used to identify the molecular structure of coniferyl ferulate. The quantitative analysis of coniferyl ferulate in different extracts of Rhizoma Chuanxiong was carried out, and the metabolism of coniferyl ferulate was investigated by in vitro incubation with rat liver homogenate. The metabolite of coniferyl ferulate, ferulic acid ethyl ester, was identified by HPLC-MS, UV-Vis and IR. In addition, antineoplastic activities of coniferyl ferulate and ferulic acid ethyl ester were detected by the MTT assay. The observed inhibition rate of coniferyl ferulate on the activity of HeLa cells was over 80% at 5.4 ng microl(-1). However, its metabolite, ferulic acid ethyl ester, showed no antineoplastic activity in vitro.

A framework of integrating gene relations from heterogeneous data sources: an experiment on Arabidopsis thaliana.

One of the most important goals of biological investigation is to uncover gene functional relations. In this study we propose a framework for extraction and integration of gene functional relations from diverse biological data sources, including gene expression data, biological literature and genomic sequence information. We introduce a two-layered Bayesian network approach to integrate relations from multiple sources into a genome-wide functional network. An experimental study was conducted on a test-bed of Arabidopsis thaliana. Evaluation of the integrated network demonstrated that relation integration could improve the reliability of relations by combining evidence from different data sources. Domain expert judgments on the gene functional clusters in the network confirmed the validity of our approach for relation integration and network inference.

Effect of solids concentration on removal of heavy metals from mine tailings via bioleaching.

Mining of mineral ore and disposal of resulting waste tailings pose a significant risk to the surrounding environment. The objective of this work is to demonstrate the feasibility to remove heavy metals from mine tailings with the use of bioleaching and meanwhile to investigate the effect of solids concentration on removal of heavy metals from mine tailings by indigenous sulfur-oxidizing bacteria and the transformation of heavy metal forms after the bioleaching process. This work showed the laboratory results of bioleaching experiments on Pb-Zn-Cu mine tailings. The results showed that 98.08% Zn, 96.44% Cu, and 43.52% Pb could be removed from mine tailings by the bioleaching experiment after 13 days at 1% (w/v) solids concentration and the rates of pH reduction, ORP rise and sulfate production were reduced with the increase of solids concentration, due to the buffering capacity of mine tailing solids. The results also indicated that solid concentration 1% was found to be best to bacterial activity and metal solubilization of the five solids concentration tested (1%, 2%, 5%, 8% and 10%) under the chosen experimental conditions. In addition, the bioleaching had a significant impact on changes in partitioning of heavy metals.

[Effects of testosterone on insulin receptor substrate-1 and glucose transporter 4 expression in cells sensitive to insulin].

OBJECTIVE: To investigate the molecular mechanism of the influence of testosterone (T) on insulin sensitivity. METHODS: Preadipocytes of the line 3T3-L1 and myoblasts of the line C2C12 were cultured to develop into mature adipocytes and skeleton muscle cells. Testosterone of the concentration of 10(-9) mol/l was added into the culture fluids for 0, 4, 8, 12, 24 and 40 hours. And testosterone of the concentrations increased by 10 times from 10(-12) mol/L, 10(-5) mol/L was added for 24 hours. Then the cell protein was extracted and the expression of insulin receptor substrate-1 (IRS-1) and glucose transporter 4 (GLUT4) in were measured by Western blotting. RESULT: (1) Treated with T of the concentration of 10(-9) mol/L, the IRS-1 expression in the 3T3-L1 adipocytes increased along with the treatment time, peaked 12 hours later with a peak value 1.42 +/- 0.42 times that at the 0 h, and the values 4, 8, and 24 hours later were 1.13 +/- 0.03, 1.19 +/- 0.05, and 1.08 +/- 0.02 times that at the 0 h (all P < 0.05). The expression of IRS-1 in the C2C12 cells increased along with the treatment time of the testosterone of the concentration of 10(-9) mol/L too, peaked 24 hours later, with the values 8, 12, and 24 hours later 1.41 +/- 0.18, 1.53 +/- 0.14, and 1.50 +/- 0.14 times that at the 0 h (all P < 0.05). The GLUT4 expression value in the 3T3-L1 adipocytes increased since 4 hours after the treatment, peaked 24 hours after the treatment (3.22 +/- 0.10 that at the 0 h) (all P < 0.05). The GLUT4 expression value in the C2C12 cells increased since 4 hours after the treatment, peaked 24 hours after the treatment (5.17 +/- 1.06 that at the 0 h) (all P < 0.05). (2) The IRS-1 expression in the 3T3-L1 adipocytes and C2C12 cells increased dose-dependently along with the increase of the T concentration and peaked when the T concentration was 10(-9) mol/l (4.23 +/- 0.27 and 3.16 +/- 0.15 times that of blank controls respectively, both P < 0.05), and then turned to decrease. (3) The GLUT4 expression in the C2C12 cells increased along with the increase of the T concentration, peaked when the T concentration was 10(-7) mol/L (2.99 +/- 0.15 times that of the blank control, P < 0.05), and then remained at the similar level. (4) The GLUT4 expression in the 3T3-L1 adipocytes was highest when treated with 10(-11) mol/L testosterone (2.58 +/- 0.02 times that of the blank control, P < 0.05), and then decreased along with the increase of the T concentration (P < 0.05). CONCLUSION: Testosterone has bidirectional dose- and time-dependent effects on the cellular expression of IRS-1 and GLUT4, which increase with low dose and short time testosterone treatment and decrease with higher dose and longer time treatment.

Cox7a2 mediates steroidogenesis in TM3 mouse Leydig cells.

AIM: To investigate the regulatory function of Cox7a2 on steroidogenesis and the mechanism involved in TM3 mouse Leydig cells. METHODS: The cDNA of Cox7a2 was cloned from TM3 mouse Leydig cells. It was subcloned to pDsRed-Express-N1 and transfected back into TM3 mouse Leydig cells for Cox7a2 overexpression by transient gene transfection. Steroidogenesis affected by overexpressed Cox7a2 was studied by ELISA. To elicit the mechanism of this effect, expression of steroidogenic acute regulatory (StAR) protein and reactive oxygen species (ROS) were examined by Western blot and fluorometer, respectively. RESULTS: The cDNA of Cox7a2 (249 bp) was cloned from Leydig cells and confirmed by DNA sequencing. After constructed pDsRed-Express-N1-Cox7a2 was transfected back into TM3 mouse Leydig cells, Cox7a2 inhibited not only luteinizing hormone (LH)-induced secretion of testosterone but also the expression of StAR protein. At the same time, Cox7a2 increased the activity of ROS in TM3 mouse Leydig cells. CONCLUSION: Cox7a2 inhibited LH-induced StAR protein expression, and consequent testosterone production, at least in part, by increasing ROS activity in TM3 mouse Leydig cells.

Genetic variants of human UGT1A3: functional characterization and frequency distribution in a Chinese Han population.

UDP-glucuronosyltransferase 1A3 (UGT1A3) contributes to glucuronidation of many important endogenous compounds and xenobiotics, including some flavonoids. Recently, a total of six single nucleotide polymorphisms (SNPs) have been identified in the human UGT1A3 gene. Among them, four SNPs (A17G, Q6R; T31C, W11R; C133T, R45W; and T140C, V47A) cause amino acid substitutions. Variants caused by these SNPs showed an activity change in estrone metabolism, whereas their activities toward other substrates were not examined. In the present study, three common flavonoids, quercetin, luteolin, and kaempferol, were used as substrates for glucuronidation by wild-type and variant UGT1A3s. Our results demonstrated that the activities of three variants, UGT1A3.2, UGT1A3.3, and UGT1A3.5, were remarkably lower than that of UGT1A3.1. In contrast, UGT1A3.4 exhibited an increase in glucuronidation efficiency of approximately 4 times and a clear preference to quercetin 7- and 3-hydroxyl groups. The frequency distributions of UGT1A3 alleles and SNPs in UGT1A3 in a Chinese Han population were statistically different from the reported value in German-Caucasians (p < 0.05). UGT1A3 variants have an altered glucuronidation activity toward quercetin, luteolin, and kaempferol and may alter human susceptibility to flavonoid exposure.
