Field electroantennogram and trap assessments of aerosol pheromone dispensers for disrupting mating in Epiphyas postvittana.

An electronically controlled aerosol system for mating disruption was evaluated against Epiphyas postvittana Walker in apple orchards in New Zealand. The area in which male moths were affected by the aerosol system was examined using catches in traps radiating from a central single-point source of either one aerosol can dispenser or 100 polyethylene tubing dispensers, compared with catches in orchard plots without pheromone dispensers. Both pheromone dispensing systems decreased moth catch to similar levels at 5-10 m from the central release point, but there was 5.5-fold more pheromone released from aerosol cans than from polyethylene dispensers over a 24 h period. Trap catches were reduced by about 90% in plots treated with either five aerosol cans per hectare or uniform deployment of polyethylene dispensers. Recordings of electroantennograms in open grassed plots and orchards indicated that the treated cotton pad of an aerosol dispenser and a point source of 100 polyethylene tubing dispensers produced similar electroantennogram recordings. Electroantennogram recordings provided evidence that pheromone plume detection from a single-point source was maintained over a range of 5-40 m downwind in the orchard. On present evidence, aerosol pheromone dispensers could not be recommended for further testing towards control of E. postvittana under New Zealand conditions owing to their higher cost of purchase and operation.

PC cell-derived growth factor overexpression promotes proliferation and survival of laryngeal carcinoma.

PC cell-derived growth factor is a novel growth factor for tumor formation and progression. No comprehensive literature concerning PC cell-derived growth factor expression status and its biological function in squamous cell carcinoma, especially in the larynx, is, however, available. The target of this study is to evaluate the clinical significance of PC cell-derived growth factor and the potential of small interfering RNA-induced genetic silencing of PC cell-derived growth factor as a supplementary therapeutic way for laryngeal squamous cell carcinoma. A total of 146 primary laryngeal cancer, 108 adult laryngeal papilloma and 41 laryngeal leukoplakia samples, as well as 10 normal larynx tissues were investigated. The PC cell-derived growth factor mRNA level was examined by real-time polymerase chain reaction and protein localization by immunohistochemistry. The biological function of PC cell-derived growth factor was assessed by transfection of small interfering RNA PC cell-derived growth factor construction. The PC cell-derived growth factor protein levels and mRNA levels of the laryngeal squamous cell carcinomas were significantly higher than those of normal laryngeal tissues (P<0.001). Simultaneously, the difference in the levels of mRNA and protein between those of laryngeal precancerous lesions (papilloma/leukoplakia) and those of normal tissues was significant (P<0.05, P<0.05), whereas those of laryngeal precancerous lesions (papilloma/leukoplakia) were significantly lower than those of laryngeal squamous cell carcinomas (P<0.05, P<0.05). Strong PC cell-derived growth factor expression was associated with lymph node metastases in laryngeal squamous cell carcinoma (P<0.05). Functional studies on Hep-2 cell lines demonstrated that the attenuation of PC cell-derived growth factor expression levels led to diminished cell proliferation rates (P<0.001), anchorage-independent growth in vitro (P<0.001), tumor forming in vivo (P<0.01) and resistance to apoptosis (P<0.001). PC cell-derived growth factor is a pivotal autocrine growth factor in the tumorigenesis of laryngeal squamous cell carcinoma. Our findings also indicate that PC cell-derived growth factor is a logical and potential target for early diagnosis, specific therapy and prognosis of laryngeal squamous cell carcinoma.

The coupling of acetylcholine-induced BK channel and calcium channel in guinea pig saccular type II vestibular hair cells.

Molecular biological studies and electrophysiological data have demonstrated that acetylcholine (ACh) is the principal cochlear and vestibular efferent neurotransmitter among mammalians. However, the functional roles of ACh in type II vestibular hair cells (VHCs II) among mammalians are still unclear, with the exception of the well-known alpha9-containing nicotinic ACh receptor (alpha9-containing nAChR)-activated small conductance, calcium-dependent potassium current (SK) in cochlear hair cells and frog saccular hair cells. The activation of SK current was necessary for the calcium influx through the alpha9-containing nAChR. Recently, we have demonstrated that ACh-induced big conductance, calcium-dependent potassium current (BK) was present in VHCs II of the vestibular end-organ of guinea pig. In this study, the nature of calcium influx for the activation of ACh-induced BK current in saccular VHCs II of guinea pig was investigated. Following extracellular perfusion of ACh, saccular VHCs II displayed a sustained outward current, which was sensitive to iberiotoxin (IBTX). High concentration of apamin failed to inhibit the current amplitude of ACh-induced outward current. Intracellular application of Cs(+) completely abolished the current evoked by ACh. ACh-induced current was potently inhibited by nifedipine, nimodipine, Cd(2+) and Ni(2+), respectively. The inhibition potency of these four calcium channel antagonists was nimodipine>nifedipine>cadmium>nickel. The L-type Ca(2+) channels agonist, (-)-Bay-K 8644 mimicked the effect of ACh and activated an IBTX-sensitive current. In addition, partial VHCs II displayed a biphasic waveform. In conclusion, the present data showed that in the guinea pig saccular VHCs II, ACh-induced BK channel was coupled with the calcium channel, but not the receptor. The perfusion of ACh will drive the opening of calcium channels; the influx of calcium ions will then activate the BK current.

Female sex pheromone of a carpenter moth, Cossus insularis (Lepidoptera: Cossidae).

This study describes the identification of a sex pheromone component of a cossid moth, Cossus insularis. Coupled gas chromatographic-electroantennographic detection (GC-EAD) analysis of solid-phase microextraction (SPME) collections of volatiles released by live female moths showed that two compounds elicited EAG responses from the antennae of male moths. These compounds were identified as (E)-3-tetradecenyl acetate (E3-14:Ac) and (Z)-3-tetradecenyl acetate (Z3-14:Ac) by mass spectral analysis and retention index comparisons with synthetic standards. The ratio of E3-14:Ac and Z3-14:Ac was 95:5 in the effluvia of a female. In field bioassays, sticky traps baited with blends of E3-14:Ac and Z3-14:Ac showed that E3-14:Ac is an essential component of the pheromone. However, the role of Z3-14:Ac is unclear, because E3-14:Ac as a single component was as attractive to male moths as blends of E3-14:Ac and Z3-14:Ac, including the 95:5 blend released by live female moths.

Effect of methylation-associated silencing of the death-associated protein kinase gene on nasopharyngeal carcinoma.

Death-associated protein kinase (DAPK) is a Ca/calmodulin-regulated serine/threonine kinase and a positive mediator of apoptosis. Loss of expression of the DAPK gene by aberrant promoter methylation may play an important role in cancer development and progression. The aim of this study was to investigate the frequency of gene promoter methylation of DAPK in nasopharyngeal carcinoma (NPC) and the effect of 5-Aza-2'-deoxycytidine (5-Aza-CdR), a demethylating agent, on CNE cells, a human nasopharyngeal carcinoma cell line, and on xenografts of CNE cells. Methylation-specific PCR and RT-PCR were used to determine the promoter methylation status and mRNA expression of the DAPK gene in NPC. Furthermore, CNE cells were treated in vitro and in vivo with 5-Aza-CdR to explore the effect of demethylating agents on DAPK mRNA expression and tumor growth. Hypermethylation of the DAPK gene promoter was found in 35 (76.1%) of 46 NPC samples. There was no significant difference in the promoter hypermethylation rate among samples from patients with different TNM stages. No promoter hypermethylation of the DAPK gene was found in all six chronic inflammatory nasopharyngeal tissue specimens. DAPK mRNA expression was not detected in NPC tumor specimens with promoter hypermethylation. However, DAPK mRNA expression was observed in unmethylated NPC tumors and in the chronic inflammatory nasopharyngeal tissue specimens. Promoter hypermethylation of the DAPK gene was found and no DAPK mRNA expression was detected in CNE cells. DAPK mRNA expression in CNE cells and xenografts could be restored by treatment with 5-Aza-CdR. The CNE cell xenografts of nude mice treated with 5-Aza-CdR were obviously smaller in tumor volume than those of nude mice treated with PBS. These results demonstrate that loss of DAPK expression could be associated with promoter region methylation in NPC. 5-Aza-CdR may slow the growth of CNE cells in vitro and in vivo by reactivating the DAPK gene silenced by de novo methylation.

[Comparative study of uptake and washout of 99mTcn(NOEt)2 with 99mTc-MIBI in human nasopharyngeal carcinoma cell line CNE].

OBJECTIVE: To determine the cellular kinetics of 99mTc-bis {N-ethoxy-N-ethyl dithiocarbamato-nitrido [N(NOEt)2]} in human nasopharyngeal carcinoma CNE cells line and to compare it with the cellular kinetics of 99mTc-methoxyisobutylisonitrile (MIBI) in the same conditions; to define the possible clinical application value of 99mTcN(NOEt)2 in nasopharyngeal carcinoma imaging. METHOD: 99mTcN(NOEt)2 or 99mTc-MIBI were incubated with CNE cells at 37degrees C, 22 degrees C, at different time points after incubation, the uptake and washout characteristics of radiotracers in CNE cells were investigated and compared. RESULT: The maximum uptake ratio of 99mTcN(NOEt)2 in CNE cells was 43.15%, that of 99mTc-MIBI was 11.08% (P < 0.01). The uptake of 99mTcN(NOEt)2 after 5 min incubation in CNE cells was 63% of total uptake, uptake of 99mTc-MIBI was 48% of total uptake (P < 0.05); 1 h after removal of the unbound tracer, 63.92% of the total uptake of 99TcN(NOEt)2 was retained in the CNE cells, 53. 97% of 99mTc-MIBI was retained (P < 0.05); not like 99mTc-MIBI, the cellular kinetics of 99mTcN (NOEt)2 were similar at 37 degrees C and 22 degrees C ( P > 0.05). CONCLUSION: 99mTcN(NOEt)2 may be a gamma imaging agent better than 99mTc-MIBI for nasopharyngeal carcinoma imaging. 99mTcN(NOEt)2 is a potential human nasopharyngeal carcinoma imaging agent.

Identification of effective siRNA against K-ras in human pancreatic cancer cell line MiaPaCa-2 by siRNA expression cassette.

AIM: We shall construct the small interfering RNA (siRNA) expression cassette (SEC) targeting activated K-ras gene sequence, identify more effective siRNA sequence against K-ras gene in human pancreatic cancer cell line MiaPaCa-2 by SEC and reveal the anti-cancer effects of RNA interference (RNAi) and its therapeutic possibilities. METHODS: Three different sites of SECs were constructed by PCR. K1/siRNA, K2/siRNA and K3/siRNA are located at sites 194, 491 and 327, respectively. They were transfected into MiaPaCa-2 cells by liposome to inhibit the expression of activated K-ras. In the interfering groups of sites 194 and 491, we detected the apoptosis in cells by FACS after they were incubated for 48 h, then we tested the alternation of K-ras gene in MiaPaCa-2 cells by RT-PCR immunofluorescence, respectively. RESULTS: Introduction of the K1/siRNA and K2/siRNA against K-ras into MiaPaCa-2 cells leads to increased apoptosis, and the number of apoptotic cells is increased compared with control cells. The tests of RT-PCR immunofluorescence show the effects of inhibiting expression of activated K-ras gene by RNA interference in the K1/siRNA and K2/siRNA groups. We also find that the introduction of K3/siRNA has no effect on MiaPaCa-2 cells. CONCLUSION: K1/siRNA and K2/siRNA can inhibit the expression of activated K-ras but K3/siRNA has no effect, demonstrating that K1/siRNA and K2/siRNA are effective sequences against K-ras gene and K3/siRNA are not. We conclude that specific siRNA against K-ras expression may be a powerful tool to be used therapeutically against human pancreatic cancer.

High yield of poly-gamma-glutamic acid from Bacillus subtilis by solid-state fermentation using swine manure as the basis of a solid substrate.

Solid-state fermentations (SSF), using swine manure as the basis of a solid substrate, were carried out for high yield of poly-gamma-glutamic acid (gamma-PGA) by Bacillus subtilis CCTCC202048. Fermentation medium and process parameters were optimized through three orthogonal array designs. The optimal medium consisted of 62.3% (w/w, dry weight basis) swine manure, 25.0% soybean cake, 5.0% wheat bran, 5.0% glutamic acid, 2.5% citric acid and 0.2% MnSO4.H2O. The optimal process parameters were 15.0 g medium with initial moisture content 60% and initial pH 9.0 in 250 ml flask, inoculation at mid-log phase with a 4% inoculum level and cultivation for 48 h at 37 degrees C. The average-PGA yield (6.0%) in triplicate under optimal conditions was obtained on the laboratory scale while it was 4.5% at compost experiment. These would lay a foundation for lessening the pollution of swine manure, increasing fertilizer efficiency and exploring a late-model organic fertilizer that retains water and nutrients.

[The expressions of survivin in squamous cell carcinoma of larynx and its clinical significance].

OBJECTIVE: To investigate the expressions of Survivin in squamous cell carcinoma of larynx (SCCL) and the correlation between Survivin and survival time. METHOD: The expressions of Survivin in SCCL tissues of 40 cases and normal larynx tissues of 5 cases were studied by immunohistochemical staining and quantitative analysis of image. RESULT: The expressions of Survivin in SCCL tissues were higher than those in mucosa of controls (P < 0.01). The expressions of Survivin in SCCL with the metastasis of lymph nodes were higher than those without metastasis( P < 0.05). Age and sex did not change the expressions obviously( P > 0.05). The expressions of Survivin in SCCL have a positive correlation between T-stages( P < 0.05). There was also a negative correlation in staining intensity between Survivin and survival time ( r = -0.646, P < 0.01). CONCLUSION: A significant correlation between Survivin expression and T-stage( P < 0.05), lymph node metastasis( P < 0.05) and prognosis (P < 0.01) was revealed in squamous cell carcinoma of larynx. The high expression suggests worse prognosis. The up-regulate of Survivin suggests that it play an important role in the formation and metastasis of SCCL.

[The expressions of K-ras in human laryngeal squamous cell carcinoma cell lines (Hep-2) and its significance].

OBJECTIVE: K-ras oncogene activation has been common in pancreatic carcinomas and colon cancer and lung cancer. Some researchers in other countries have reported that human laryngeal squamous cell carcinoma is unrelated to K-ras gene activation. This study aims to present our research on the expression of K-ras in human laryngeal squamous cell carcinoma cell lines (Hep-2) and its significance for establishing a solid foundation for further study of the relativity of human laryngeal squamous cell carcinoma and K-ras gene point mutations. METHOD: We have measured the expression of K-ras in human laryngeal squamous cell carcinoma cell lines (Hep-2) and in human pancreatic carcinoma cell lines (MIAPaCa-2) with RT-PCR technology. RESULT: The expression of K-ras mRNA in Hep-2 and MIAPaCa-2 is strong and positive. CONCLUSION: The expression of K-ras mRNA in human laryngeal squamous cell carcinoma cell lines (Hep-2) is positive. Development of laryngeal carcinoma could be in connection with the activation of K-ras gene point mutation.

Development, survival, body weight, longevity, and reproductive potential of Oemena hirta (Coleoptera: Cerambycidae) under different rearing conditions.

Oemona hirta (F.) is a New Zealand native longicorn beetle, whose larvae bore into the wood of branches and stems of living trees and vines, causing serious damage. To explore effective methods for maintaining laboratory colonies and biology of immature stages of this species we evaluated four laboratory rearing methods with both natural and artificial diets and compared biological parameters of laboratory colonies with those of field-collected insects. On an artificial diet, approximately 40% of neonate and 70% of autumn- and 11% of winter-collected larvae reached adulthood. Neonate larvae could not complete their development in cut poplar (Populus nigra variety italica Koehne) twigs; however, when twigs were standing in water >46% of neonate larvae survived to adulthood. Mean larval development time ranged from approximately150 to almost 300 d, depending on rearing methods. Mean pupal stage ranged from 15 to 19 d. Adult females were significantly heavier than males. Although adult females from field-collected twigs and reared on the artificial diet had similar mean body weight, which was significantly greater than that of other rearing colonies, the potential fecundity was the highest in the former (83.1 +/- 29.4 eggs) and the lowest in the latter (33.5 +/- 9.1 eggs). In all laboratory-reared and field-collected insects, the total number of eggs produced (eggs laid + eggs that remained in dead female body) by females was positively correlated with their body weight. When larvae fed on the artificial diet, there was no positive correlation between the number of eggs successfully laid and female body weight. However, when larvae lived on natural food, a positive correlation was found. In laboratory colonies, mean longevity of females (36 to 52 d) was slightly greater than that of males (30 to 50 d) without significant difference between sexes but in the adults from field-collected twigs, males (52 d) lived significantly longer than females (33 d). In terms of time, labor, and the number of resulting adults, collecting larvae in the field in autumn and then transferring them onto artificial diet is the most effective method for maintaining a laboratory colony.

[Clinical effect and mechanism of nitroglycerin patch on arresting preterm labor].

OBJECTIVE: To observe the effect of nitroglycerin patch on treatment of preterm labour and to identify the mechanism responsible for arrest of preterm labour with nitroglycerin patch. METHODS: Sixty women diagnosed as threatened preterm labour were divided into two groups: group of nitroglycerin patch therapy (n = 30), and group of magnesium sulfate and salbutamol sulfate therapy (n = 30). Nitroglycerin patch was applied to the abdomen of patients. Measurement of cortictrophin-releasing hormone (CRH) levels in maternal plasma was performed by radioimmunoassay. RESULTS: The mean time of pregnancy prolongation was 25 days in the nitroglycerin patch group, and 8 days in the magnesium sulfate and salbutamol sulfate group. The mean level of CRH concentration in nitroglycerin patch group before treatment was (257 +/- 61) ng/L, and it was decreased sharply to (38 +/- 17) ng/L after treatment. Those were (248 +/- 60) ng/L and (56 +/- 22) ng/L respectively in the magnesium sulfate and salbutamol sulfate group. CONCLUSION: Reduction of CRH secretion may be the mechanism of nitroglycerin patch on preterm labour therapy. It may act as an effective, safe, well-tolerated, and non-invasive method for treatment of preterm labour.
