[Study on mechanism of yangxincao capsule in regulating lipid metabolism].

OBJECTIVE: To investigate the effect and mechanism of Yangxincao Capsule (YXCC) in regulating lipids. METHODS: Sixty rats were randomly divided into 6 groups, the normal control group (A), the hyperlipidemia model group (B), the high, middle and low dose YXCC treated groups (C, D and E), and the Shanzhajing (SZJ) treated group (F) for positive medicine control. Except for the rats in the normal control group, the other 50 were daily fed with fatty emulsion for 10 days to establish hyperlipidemic model. From the I th day on, in the same time of continually feeding with fatty emulsion they were administered with water, high (1.08 g/kg), middle (0.54 g/kg), low dose (0.27 g/kg) of YXCC and SZJ (5.4 mg/kg) respectively for 10 days, while to rats in Group A equal volume of water was given. At the 21th day, after rats were fasted for 16 h, their blood was extracted from post-orbital vein to detect the level of serum lipids, lipoprotein, apolipoprotein (apo) and lipid metabolic enzyme. RESULTS: Compared with Group A, the levels of serum total cholesterol (TC), triglyceride (TG) and low density lipoprotein-cholesterol (LDL-C) increased remarkably, and the level of high density lipoprotein-cholesterol (HDL-C) dropped obviously in Group B. While in the four treated groups the levels of TC, TG and LDL-C were significantly reduced, HDL-C and its sub-components 2 and 3 (HDL2-C and HDL3-C), as well as the ratio of HDL-C/TC were raised. Besides, the content of apo-Al was increased and apo-B was decreased significantly in Group C and D, activity of lecithin cholesterol acyltransferase (LCAT) and lipoprotein lipase (LPL) increased in the three YXCC treated groups, all showed statistical significance (P < 0.05 or P < 0.01) as compared with those in Group B. CONCLUSION: YXCC could remarkably modulate the lipid metabolic disorder in hyperlipidemic rats, and has a certain bi-directional regulating function on lipoprotein, inferring that it could reduce the risk of occurring coronary artery diseases. The mechanism of regulating lipid metabolism might be related with the increasing activity of LCAT, LPL and eliminating of cholesterol by the elevated level of HDL2-C.

[Analyzing the effect of field frequencies on reconstructive accuracy of rebuilt field with spectroscopy].

The field to be reconstructed was simulated with numerical simulation technique. The effect of a three-dimensional field's characteristics, especially, its frequency components, on the reconstructive accuracy was discussed with spectroscopy. A double-peaked field model was built with Gaussian function, and its frequency components were analyzed by Fourier transform. With algebraic reconstruction technique, the model field was reconstructed. The reconstructed field was analyzed with many error indexes. As a result, the reconstructed field was similar to the model one in respect of the position, pointing and shape of the peaks, but the heights of the peaks were shorter than those of corresponding ones in the model. The border of the reconstructive field showed obvious fluctuation. It was considered that the main causation of the reconstructive result was the filtering inserted in iterating process. To confirm this idea, the convergence factor, represented with A, of Gaussian function was changed from six to thirty in order to change the frequency components of the model field, and the same numerical simulation as the one for the first model field with A=18 was carried out for these new model ones consisting of different frequency. As a result, the idea proved to be right. At the same time, it was found that the characteristics of the field at the edge of the reconstructed zone had a deeper effect on the reconstructive accuracy. When the precondition that the field edge approximated to zero could no longer existed, the reconstructive accuracy declined, and furthermore, it would make iterating process divergent. From all the above, the characteristics of the field at the edge should draw much attention when we reconstruct a field with Algebraic reconstruction technique.

Culture of osteoblasts on bio-derived bones.

OBJECTIVE: To study the effect of bio-derived bones, as substitutes of autogenous bone grafts and demineralized cadaver bones, on the attachment, spreading and proliferation of isolated osteoblasts. METHODS: Osteoblasts were isolated from the calvaria of a fetal rabbit through sequential collagenase digestion. In the attachment study, the osteoblasts labeled with 3H-leucine were incubated with the bio-derived bone materials in sterile microcentrifugable tubes for 15, 90 and 180 minutes, and 24 hours, respectively. The attached cells were collected and the radioactivity was measured with liquid scintillation spectrometry. In the proliferation study, the osteoblasts were cultured with the bio-derived bone materials for 24 hours and 3H-thymidine was added during the last 2 hours of the incubation. The attached cells were collected and the radioactivity was measured with liquid scintillation spectrometry. Osteoblasts were seeded on the bone graft materials for 60 or 120 minutes, 24 or 48 hours, and 3 or 7 days, then the co-culture was processed for scanning electron microscopy to observe the interaction of osteoblasts and the bio-derived bone materials. RESULTS: Osteoblasts attached to the bio-derived bone materials in a time-dependent manner. There were significantly (P<0.05) more attached cells after 180 minutes than after 15 and 90 minutes of incubations (P<0.05). Osteoblasts were proliferated in a large amount on the surface and in the materials. Osteoblasts seeded onto 100 mg bio-derived bones resulted in significantly (P<0.05) more measurable proliferation than those seeded onto 10 mg bones. Osteoblasts appeared round as they attached to the materials, then flattened and spread over with time passing. CONCLUSIONS: Bio-derived bones can provide a good environment for the attachment and proliferation of osteoblasts.

Oxidative stress related enzymes in response to chromium (VI) toxicity in Oxya chinensis (Orthoptera: Acridoidae).

The toxic effects of Cr(VI) on antioxidant enzymes of Oxya chinensis (Orthoptera: Acridoidae) were determined. Changes in the activities of the antioxidant enzymes superoxide dismutase (SOD), catalase (CAT), and guaiacol peroxidase (GPx) were measured in O. chinensis insects injected with Cr(VI). Fifth-nymphs of O. chinensis insects were injected with Cr(VI) with different concentrations (0, 75, 150, 225, 300, 375, 450 mg/kg of body weight). The results showed that Cr(VI) led to the change of SOD, CAT, and GPx activities at different concentrations, which revealed that: (1) The oxidative stress of SOD increased with the increase of Cr(VI) concentration. (2) With the increase of Cr(VI) concentrations, CAT activities for females increased at lower concentrations, but decreased at higher concentration range, which indicated that antioxidant system of O. chinensis was not influenced by the presence of Cr(VI). A very similar response to Cr(VI) effect for males indicated that Cr(VI) concentrations were not high enough to damage O. chinensis in terms of CAT. (3) The GPx activity for females increased in all treatments, which revealed that the damage power of Cr(VI) was increased with the increase of Cr(VI) concentrations in terms of GPx, but the effect was not so remarkable. There was not a consistent trend of GPx activities for males in all treatments of Cr(VI). Cr (VI)-induced changes in antioxidant enzymes were different for SOD, CAT and GPx, of which the tendency was that activities generally changed with increase of concentrations of Cr(VI) suggesting SOD, CAT, and GPx could serve as indices of oxidative stress to some extent.

[Function of pancreatic islet beta cells and features of TCM symptoms and syndromes in the non-diabetic first-grade relatives of patients with type 2 diabetes mellitus].

OBJECTIVE: To study the function of pancreatic islet beta cells, insulin resistance (IR) and features of TCM symptoms and syndromes in the non-diabetic first-grade relatives (ND1GR) of patients with type 2 diabetes mellitus (DM2). METHODS: A total of 68 ND1GR of DM2 patients were enrolled in the observed group and 45 healthy subjects with matched sex, age and body mass index (BMI) but without family history of DM were selected into the control group. Levels of fasting blood glucose (FBG), 2 hrs postprandial glucose (2hPG), fasting insulin (FINS) and 2 hrs postprandial insulin (2h INS) in all the subjects were measured to calculate and compare the IR and beta-cell function of the homeostatic model analog (HOMA-IR and HOMA-beta), and the insulin sensitive index (ISI). Moreover, the symptoms manifested in the ND1GR were also observed to analyze the features in them. RESULTS: FBG and FINS were obviously higher in the observed group than those in the control group (P < 0.01), while no significant difference was found in 2hPG or 2h INS (P > 0.05). HOMA-IR and HOMA-beta were significantly higher (P < 0.05) and ISI were significantly lower (P < 0.01) in the observed group than those in the control group. Compared with the control group, the main symptoms such as dark purplish tongue, listlessness, thready and thin pulse, lassitude in loin and legs in the observed group were seen more frequently. In the observed group syndrome of deficiency of Qi and Yin accounted for 51.47%, syndrome of deficiency of Yin for 30.88%, subjects with syndrome of blood stasis as the main accompanying syndrome accounts for 61.76%. CONCLUSION: Higher beta cell secretion function and lower insulin sensitivity appear in ND1GR of DM2 patients, suggesting the existence of insulin resistance. The feature of TCM syndrome in them is characterized by deficiency of Qi and Yin with inner obstruction of blood stasis.

[Pharmacodynamical study on Danqi capsule].

OBJECTIVE: To assess the pharmacodynamical actions of Danqi capsule which was reported to promote blood circulation by removing blood stasis, regulation Danqi and relieving pain so as to be used to treat thoracic obstruction, headache, menstrual pain in clinic. METHOD: To compare the pharmacologic effects of Danqi capsule Danqi tablet in the rats with acute myocardial ischemia and the mice with the increased oxygen-consumption induced by isoproterenol injected subcutaneously. The pain models were prepared by injection of acetic acid and uterospasm model in the female mouse was induced by diethylstilbestrol and pitocin. A hyperlipidemia model was also made in the rats. RESULT: Danqi capsule could significantly improve ECG in myocardial ischemia of rats induced by isoproterenol and prolong the mice survival time under hypoxic situation. In the experiment to observe the pain response with body twist as a index induced by acetic acid and uterospasm induced by diethylstilbestrol and pitocin, Danqi capsule could significantly shorten the latency and the time course of body twist. The contents of triglyceride(TG) and total cholesterol(TC) were decreased, while the level of high-density liporotein-cholesterol(HDL-c) was increased after treatment of Danqi capsule in in the rats with hyperlipidemia. Hemorheological data showed that the blood viscosity, blood reductive viscosity, erythrocyte rigidity index and electrophoresis time were significantly decreased by Danqi capsule in the animal model mentioned above as compared with control group (P < 0.05 or P < 0.01). In addition, the dose of Danqi capsule is less than that of Danqi tablet for producing equivalent effect. CONCLUSION: Danqi capsule plays a good role in improving myocardial ischemia, increasing the tolerant ability against oxygen-deficiency, alleviating pain and descending the levels of blood fat and blood viscosity.

[Quality of methodology and reporting of randomized controlled trials of acupuncture for obesity].

OBJECTIVE: To evaluate the quality of methodology and reporting of randomized controlled trials of acupuncture for obesity worldwide. METHODS: Collecting all the randomized controlled trials of acupuncture for obesity worldwide by systematically searching without language limitation. The quality of methodology and reporting was evaluated by the quality criteria recommended by Cochrane handbook and CONSORT (consolidate standards of reporting trial) and STRICTA (standard for reporting interventions in controlled trials of acupuncture) checklists. RESULTS: Seven randomized controlled trials of acupuncture for obesity were identified. The quality of methodology was graded as "A" in one trial, "B" in two trials and "C" in the other four trials. The reporting quality of the seven trials was graded as "C". CONCLUSIONS: The methodology quality of the randomized controlled trials of acupuncture for obesity was too poor to provide strong evidence for clinical practice, and the poor reporting quality of the trials resulted in inadequate information from the trials.

[Effect of astragalus injection combined with chemotherapy on quality of life in patients with advanced non-small cell lung cancer].

OBJECTIVE: To observe the effect of Astragalus injection (AI) combined with chemotherapy on quality of life (QOF) in patients with advanced non-small cell lung caner (NSCLC). METHODS: Sixty-NSCLC patients were randomly divided into the treated group (n = 30, treated with AI combined with chemotherapy) and the control group (n = 30, treated with chemotherapy alone). Chemotherapy of MVP protocol was applied to both groups. AI was supplemented to the treated group by intravenous dripping 60 ml per day. Treatment of 21-28 days as one treatment cycle, and 2-3 treatment cycles were applied. RESULTS: The effective rate in the treated group was 40.0% and in the control group was 36.7%, the mean remission rate in the treated and control group was 5.4 months and 3.3 months, the median survival period 11 months and 7 months, and the 1-year survival rate 46.75% and 30.0%, respectively, the differences of these indexes between the two groups were all significant (P < 0.05). Moreover, the clinical improving rate and QOF elevation rate in the treated group was 80.4% and 43.3%, as compared with those in the control group (50.0% and 23.3% respectively), the difference was also significant (P < 0.01). CONCLUSION: AI combined with chemotherapy can significantly improve the QOF in NSCLC patients of advanced stage.
