Ribosomal protein L3 mutants alter translational fidelity and promote rapid loss of the yeast killer virus.

Programmed -1 ribosomal frameshifting is utilized by a number of RNA viruses as a means of ensuring the correct ratio of viral structural to enzymatic proteins available for viral particle assembly. Altering frameshifting efficiencies upsets this ratio, interfering with virus propagation. We have previously demonstrated that compounds that alter the kinetics of the peptidyl-transfer reaction affect programmed -1 ribosomal frameshift efficiencies and interfere with viral propagation in yeast. Here, the use of a genetic approach lends further support to the hypothesis that alterations affecting the ribosome's peptidyltransferase activity lead to changes in frameshifting efficiency and virus loss. Mutations in the RPL3 gene, which encodes a ribosomal protein located at the peptidyltransferase center, promote approximately three- to fourfold increases in programmed -1 ribosomal frameshift efficiencies and loss of the M1 killer virus of yeast. The mak8-1 allele of RPL3 contains two adjacent missense mutations which are predicted to structurally alter the Mak8-1p. Furthermore, a second allele that encodes the N-terminal 100 amino acids of L3 (called L3Delta) exerts a trans-dominant effect on programmed -1 ribosomal frameshifting and killer virus maintenance. Taken together, these results support the hypothesis that alterations in the peptidyltransferase center affect programmed -1 ribosomal frameshifting.

Role of C/A polymorphism at -20 on the expression of human angiotensinogen gene.

-Angiotensinogen is the glycoprotein precursor of 1 of the most potent vasoactive hormones, angiotensin II. Human angiotensinogen gene contains a C/A polymorphism at -20 located between the TATA box and transcriptional initiation site. We show here that when nucleoside A is present at -20, this sequence binds to the estrogen receptor. We also show that transcriptional activity of reporter constructs containing human angiotensinogen gene promoter with nucleoside A at -20 is increased on cotransfection of an expression vector containing human estrogen receptor-alpha coding sequence in human hepatoma cells (HepG2) followed by estrogen treatment. On the other hand, adenoviral major late transcription factor binds preferentially to this region of the promoter when nucleoside C is present at -20. We also show that reporter constructs containing human angiotensinogen gene promoter with nucleoside C at -20 have increased basal promoter activity on transient transfection in HepG2 cells as compared with reporter constructs with nucleoside A at -20. Our data suggest that C/A polymorphism at -20 may modulate the expression of human angiotensinogen gene in a sex-specific manner.

Osteopontin expression in fetal and mature human kidney.

Osteopontin is a secreted phosphoprotein that is expressed by normal kidney, and has been associated with a number of functions including cell adhesion, migration, signaling, and biomineralization. Although there is a vast literature detailing osteopontin localization in various rodent models of both development and disease, this article presents the first comprehensive description of osteopontin localization in human kidney. In this study, immunohistochemistry, immunoelectron microscopy, in situ hybridization, and Northern blotting are used to analyze osteopontin protein and mRNA expression in human fetal and normal mature renal tissue. Osteopontin is expressed in the human embryonic renal tubular epithelium beginning on approximately day 75 to 80 of gestation. In the fetal kidney, osteopontin can also be seen occasionally expressed in the ureteric buds and in some interstitial cells. As localized at the protein and mRNA level, the tubular expression of osteopontin increases with increasing gestational age and persists into adulthood. In the normal adult kidney, osteopontin is localized primarily to the distal nephron and is strongly expressed by the thick ascending limb of the loops of Henle. Osteopontin expression can also be observed in some collecting duct epithelium. In cases that exhibit foci of interstitial fibrosis and an associated influx of interstitial macrophages, osteopontin expression is significantly upregulated in all tubular segments, including proximal tubules.

Drug resistance and ATP-dependent conjugate transport mediated by the apical multidrug resistance protein, MRP2, permanently expressed in human and canine cells.

The multidrug resistance protein MRP1 functions as an ATP-dependent conjugate export pump and confers multidrug resistance. We cloned MRP2 (symbol ABCC2), a MRP family member localized to the apical membrane of polarized cells. Stable expression of MRP2 in transfected human embryonic kidney (HEK-293) and Madin-Darby canine kidney (MDCK) cells was enhanced by inhibitors of histone deacetylase. In polarized MDCK cells, both rat and human MRP2 were sorted to the apical plasma membrane. An antibody raised against the amino terminus of rat MRP2 recognized the recombinant protein on the apical surface of nonpermeabilized cells, providing direct evidence for the extracellular localization of the amino terminus of MRP2. ATP-dependent transport by recombinant human and rat MRP2 was measured with membrane vesicles from stably transfected cells. The Km value of human MRP2 was 1.0 +/- 0.1 microM for leukotriene C4 and 7.2 +/- 0.7 microM for 17beta-glucuronosyl estradiol; the Km values of human MRP1 were 0.1 +/- 0.02 microM for leukotriene C4 and 1.5 +/- 0.3 microM for 17beta-glucoronosyl estradiol. Thus, the conjugate-transporting ATPases MRP2 and MRP1 differ not only by their domain-specific localization but also by their kinetic properties. Drug resistance conferred by recombinant MRP2 was studied in MDCK and HEK-293 cells using cell viability assays. Expression of human and rat MRP2 enhanced the resistance of MDCK cells to etoposide 5.0-fold and 3.8-fold and to vincristine 2.3- and 6.0-fold, respectively. Buthionine sulfoximine reduced resistance to these drugs. Human MRP2 overexpressed in HEK-293 cells enhanced the resistance to etoposide (4-fold), cisplatin (10-fold), doxorubicin (7.8-fold), and epirubicin (5-fold). These results demonstrate that MRP2 confers resistance to cytotoxic drugs.

[Isolating candidate inserted fragment from positive fused phage clones using quantitative PCR].

OBJECTIVE: To isolate quickly and exactly the specific inserted fragment from fused phage clones which were obtained from cDNA library by hybridization. METHODS: According to the amplification principle of quantitative polymerase chain reaction (PCR) and based on the difference of original template quantity,target cDNA fragment was isolated and identified by two PCRs. RESULTS: A positive clone with specific cDNA fragment of HumGT-H1 gene was obtained from a two-phage fused clone by using this method,and the inserted fragment was verified to be the 5' cDNA sequence of HumGT-H1,1.9kb in length. So another hybridization screening is not necessary. CONCLUSION: The method presented is effective and rapid in gene cloning and can greatly save time and materials.

Effects of acupuncture on the levels of endothelin, TXB2, and 6-keto-PGF1 alpha in apoplexy patients.

In order to delve into the mechanism governing the treatment of apoplexy by acupuncture at yangming channel points as main points, we observed the changes in the endothelin (ET) level in plasma, TXB2 and 6-Keto-PGF1 alpha levels in urine in convalescent apoplexy patients during acupuncture treatment. The results showed that the ET level in plasma in convalescent apoplexy patients was significantly higher than that in healthy subjects (P < 0.05), and the ET level in plasma in patients was decreased after one course of acupuncture treatment. It was found that before treatment the TXB2 level in urine in apoplexy patients was significantly higher than in healthy subjects, and the 6-Keto-PGF1 alpha level in urine in the patients was significantly lower than that in healthy subjects, with an increased ratio of TXB2 to 6-Keto-PGF1 alpha. After acupuncture treatment, the TXB2 level in urine was lowered with a decrease in the ratio of TXB2 to 6-Keto-PGF1 alpha. All this indicated that one of the mechanisms governing acupuncture treatment of apoplexy acupuncture at yangming channel points as main points was that acupuncture could produce therapeutic effects by adjusting the imbalance of important vaso-active substances, ET, TXA2, and PGI2.

Comparison of anastomotic suturing techniques in the rat esophagus.

BACKGROUND: Long-gap esophageal atresia continues to be a challenging pediatric thoracic surgical problem. Despite the use of various tension relieving procedures, the esophageal anastomosis is often performed under considerable tension. Excessive tension can cause anastomotic sutures to pull through the esophageal tissue, with resultant early esophageal anastomotic dehiscence. To test the hypothesis that interrupted horizontal mattress sutures would withstand the forces of tension better than interrupted simple sutures, an experimental study of rat esophageal anastomoses was done. METHODS: Twenty rats were killed and their esophagi were excised. The esophagi were divided in the mid portion and end-to-end anastomoses were done using interrupted 6-0 polypropylene sutures. Ten rats had anastomoses done with interrupted simple sutures and ten had interrupted horizontal mattress suturing. Anastomotic breaking strength was tested in a tensiometer. RESULTS: Anastomotic breaking strength was 3.22+/-0.56 N for the interrupted simple sutured anastomoses and 3.51+/-0.61 N for the interrupted horizontal mattress group (p=0.30). The difference was not significant. CONCLUSIONS: In this animal study interrupted simple and horizontal mattress suturing withstood the disruptive forces of anastomotic tension equally well.

Interstitial brachytherapy using iridium-192 for malignant brain tumors: clinical results.

OBJECTIVE: To determine the effects and toxicity of interstitial brachytherapy using iridium-192 on brain malignant gliomas. METHODS: Between January 1992 and January 1995, 56 patients with anaplastic astrocytoma and glioblastoma multiforme were treated with stereotactic brachytherapy using temporary high-activity iridium-192 sources. RESULTS: The median survival for patients receiving brachytherapy was 28 months. The survival rates at 1, 2, 3 years were 92.8%, 83.9% and 71.4% respectively. CONCLUSIONS: Brachytherapy may improve the control of local tumor and prolong the survival, when used in deep malignant brain gliomas, by temporary implanted high doses of iridium-192 sources.

The effects of classic antipsychotic haloperidol plus the extract of ginkgo biloba on superoxide dismutase in patients with chronic refractory schizophrenia.

OBJECTIVES: To explore the association between schizophrenic symptoms and superoxide dismutase (SOD), and to investigate the effect of classic antipsychotic haloperidol plus the extract of Ginkgo biloba (EGb) on SOD. METHODS: In 54 patients with chronic refractory schizophrenia, 27 were treated with haloperidol plus EGb (group 1), and the rest received haloperidol plus placebo (group 2). Superoxide dismutase (SOD) levels of these patients were measured before and after treatment and compared with the levels of 25 healthy volunteers. Therapeutic efficacy was equated with a change in clinical rating scores assessed by standardized measurement tools including the Scale for Assessment of Positive Symptoms (SAPS) and the Scale for Assessment of Negative Symptoms (SANS). RESULTS: Patients in group 1 improved significantly as demonstrated by scores from both SAPS and SANS, while those in group 2 only by scores from SANS. Assessed by SAPS, the response of patients receiving haloperidol plus EGb was more significant than those receiving haloperidol only. SOD levels before treatment in all patients were significantly higher than those in normal controls. After treatment, SOD levels decreased significantly in group 1 but not in group 2. In addition, before treatment, SOD levels in all patients correlated significantly with SAPS score. The levels of SOD measured before treatment were also correlated with the improvement of patients as measured by SAPS and SANS after 12 weeks. CONCLUSIONS: EGb may enhance the efficacy of classic antipsychotic haloperidol on schizophrenia, especially on positive symptoms. It may work through an antioxidant efficacy that is involved in the therapeutic mechanism.

[The use of intervention chemotherapy in comprehensive treatment of advanced nasopharyngeal carcinoma].

OBJECTIVE: To study the application of intervention chemotherapy in the comprehensive treatment of advanced nasopharyngeal carcinoma (NPC). METHODS: In 30 patients with advanced NPC, combined intervention chemotherapy with cisplatin (100 mg), 5-fluoro-uracil (1,000 mg) and bleomycin (16 mg) was given by Seldinger's selective unilateral catheterization of the ascending pharyngeal artery or superior maxillary artery. If the lesion had extended over the midline, the intervention chemotherapy encompassed both sides: 2/3 of the dose infused to the side with the major lesion and 1/3 to the other side. Radiotherapy started one week after intervention chemotherapy. A total dose of 60-70 Gy was given to the primary tumor or 50-60 Gy to the cervical metastases in 8 weeks. Fifty patients with advanced NPC treated in the same period of time with radiotherapy alone served as the control. Both groups of patients were treated with 60Co, the duration of treatment and dose splitting were the same. RESULTS: After intervention chemotherapy, the primary tumor mass reduced in size in 16 patients and the enlarged cervical lymph nodes shrank or became loosened in 12 patients. The radiation dose required to achieve complete response of the primary and metastatic lesions was significantly less as compared to that of patients receiving radiotherapy alone (P < 0.05). At four weeks following completion of radiotherapy, complete response rate in the two groups of patients was 83.3% and 72.0%, respectively (P < 0.05). CONCLUSION: Intervention chemotherapy combined with radiotherapy is an effective treatment for patients with advanced NPC.

[Enhancing effect of Chinese herbal medicine mixture on peripheral blood lymphocyte proliferation and interleukin-2 production in patients of burn].

OBJECTIVE: To study the regulatory effect of Chinese herbal medicine mixture (CHMM) on cell-mediated immune function in patients of burn. METHODS: Patients of burn were treated with CHMM and the therapeutic effect was evaluated by their peripheral blood T-lymphocyte (PBL) proliferation and interleukin-2 (IL-2) production determination. RESULTS: The PBL proliferation began to increase after 5 days of medication and the response was significant between the 10th to 20th day, which was in accordance with the change of IL-2 production. CONCLUSION: CHMM has immune regulatory effect on cell-mediated immune function in patients of burn.

[One-time operation in infant cleft lip and palate plus alveolar process].

OBJECTIVE: To improve safety and viability of one-time operation in the therapy of cleft lip and palate and cleft alveolar process. METHODS: Under the air-vein anesthesia, one-time operations were performed in 42 patients with complete cleft lip and palate plus cleft alveolar process. After rehabilitation in ICU, all patients were put into wards, and left hospital 4-7 days after operation. RESULTS: The situation during anesthesia and operation was good. There were no statistically significant changes in blood pressure, oxygen saturation of blood, heart rate and cardiac rhythm before, during and after operation. No anesthetic accident and no complication happened. The loss of blood was about 42.7 +/- 6.5 ml and the duration of operation was about 139.2 +/- 18.6 minutes. The rehabilitation was successful after operations. First intention took place in almost all patients' incisions, and palatal fistulas appeared in only 2 cases. CONCLUSION: Under the control of provisioned pediatric anesthesia and rehabilitation, one-time complete cleft lip and palate plus cleft alveolar process closure is safe and viable.

[The value of nasal endoscopy in the resection of upper and posterior nasal septum deviation].

OBJECTIVE: In order to find a safe, reliable and satisfying technique to treat the upper and posterior nasal septum deviation. METHOD: Performing the resection of the upper and posterior nasal septum deviation under nasal endoscopy, comparing this new method with the traditional one and discussing the safety and long-term curative effect of this new method. RESULT: The observation of the outcome of 109 patients undergoing this operation after 1-50 months postoperation shows that this technique can resect the upper and posterior deviation safely and completely, improve the nasal respiratory and olfactory function and reduce the occurrence of nasal cavity adhesion and the recurrence of nasal polyp and sinusitis. CONCLUSION: Performing the resection of the upper and posterior nasal septum deviation under nasal endoscopy is a good method for popularizing to clinician.

[Clinical significance and changes of fibrillation potential amplitude following denervation of laryngeal muscles].

OBJECTIVE: To evaluate the value of clinical application of examination of fibrillation potential amplitude of laryngeal electromyogram. METHOD: Fifty-four patients (65 sides) with recurrent laryngeal nerve paralysis were examined and only the maximum fibrillation potential amplitudes were recorded in the posterior cricoarytenoid muscles and thyroarytenoid muscles. RESULT: There was no significant difference between sexes, ages and sides. The fibrillation potential amplitude was maximum at 3 to 4 months after denervation of the posterior cricoarytenoid muscle or at 5 to 6 months after denervation of the thyroarytenoid muscle and still remained at certain level for years in some patients. Significant difference was showed in complete and incomplete nerve injuries groups, but changes of amplitude with time in incomplete nerve injuries group were small. CONCLUSION: The changes of fibrillation potential amplitude might indicate that the changes from muscle denervation was still reversible and might be more accurate than traditional method of examination.

[Clinical use of nasal airflow measurement in septoplasty].

OBJECTIVE: To discuss the clinical use of nasal airflow measurement in septoplasty. METHOD: After nasal decongestion, 21 patients's nasal airway resistance and nasal airflow were evaluated by anterior rhinomanometry before and after submucous resection of nasal septum. RESULT: The amount of nasal airflow asymmetry could show nasal respiratory function better than nasal airway resistance does, the patients with absolute amount of nasal airflow asymmetry greater than 50% had bad postoperative outcome, patients' satisfication with operation was significantly correlated with the presence of post-operative symmetrical nasal airflow. CONCLUSION: The amount of nasal airflow asymmetry can serve as an indicator of patient selection, outcome evaluation and prognosis in nasal septum deviation.

[Clinical and bacteriology observation on intraoperation usage of tarivid otic solution in treatment of chronic otitis media with cholesteatoma].

OBJECTIVE: To determine the effect and safety of Tarivid (oflorxacin) otic solution used during the operation in treatment for chronic otitis media with cholesteatoma. METHOD: 42 patients were divided into two groups. In Tarivid group, 23 patients' operative cavities were soaked with 0.3% Tarivid, while other 19 patients' operative cavities in boric acid group were soaked with 4% boric acid during the open mastoidectomy with tympanoplasty. RESULT: The recovery rate was 82.6% and 50% respectively. There was significant difference between two groups (P < 0.05). No side effect was observed. CONCLUSION: It shows that using Tarivid otic solution in the operation can improve the outcome of the operation and has high security.

[Microwave therapy of radiotherapeutical choanal atresia under nasal endoscope].

OBJECTIVE: In order to know efficacy of microwave to treat the secondary choanal atresia under nasal endoscope. METHOD: Three patients suffering from membranous choanal atresia due to radiotherapy of nasopharyngeal carcinoma (NPC) were treated by microwave under nasal endoscope. RESULT: Satisfing recoveries were accomplished in all patients and no relapse was found within the 7-13 months' follow-up. CONCLUSION: This method has many advantages, such as: processing available under local anesthesia without incision, mild damage to normal tissues, clear operation visual field, less hemorrhage and postoperative pain, complete coagulation and gasification of atresic membrane, no need of postoperative dilation and stable therapeutical effect.
