Proteolytic cleavage of the amino terminus of the U(L)15 gene product of herpes simplex virus type 1 is coupled with maturation of viral DNA into unit-length genomes.

The U(L)15 gene of herpes simplex virus type 1 (HSV-1), like U(L)6, U(L)17, U(L)28, U(L)32, and U(L)33, is required for cleavage of concatameric DNA into genomic lengths and for packaging of cleaved genomes into preformed capsids. A previous study indicated that the U(L)15 gene encodes minor capsid proteins. In the present study, we have shown that the amino-terminal 509 amino acids of the U(L)15-encoded protein are sufficient to confer capsid association inasmuch as a carboxyl-terminally truncated form of the U(L)15-encoded protein with an M(r) of approximately 55,000 readily associated with capsids. This and previous studies have shown that, whereas three U(L)15-encoded proteins with apparent M(r)s of 83,000, 80,000, and 79,000 associated with wild-type B capsids, only the full-length 83,000-M(r) protein associated with B capsids purified from cells infected with viruses lacking functional U(L)6, U(L)17, U(L)28, U(L)32, and U(L)33 genes (B. Salmon and J. D. Baines, J. Virol. 72:3045-3050, 1998). Thus, all viral mutants that fail to cleave viral DNA into genomic-length molecules also fail to produce capsid-associated U(L)15 80,000- and 79,000-M(r) proteins. In contrast, the 80,000- and 79,000-M(r) proteins were readily detected in capsids purified from cells infected with a U(L)25 null virus that cleaves, but does not package, DNA. The conclusion that the amino terminus of the 83,000-M(r) protein is truncated to produce the 80,000- and/or 79,000-M(r) protein was supported by the following observations. (i) Whereas the C termini of the 83,000-, 80, 000-, and 79,000-M(r) proteins are identical, immunoreactivity dependent on the first 35 amino acids of the U(L)15 83,000-M(r) protein was absent from the 80,000- and 79,000-M(r) proteins. (ii) The 79,000- and 80,000-M(r) proteins were detected in capsids from cells infected with HSV-1(U(L)15M36V), an engineered virus encoding valine rather than methionine at codon 36. Thus, initiation at codon 36 is unlikely to account for production of the 80,000- and/or 79, 000-M(r) protein. Taken together, these data strongly suggest that capsid-associated U(L)15-encoded protein is proteolytically cleaved near the N terminus and indicate that this modification is tightly linked to maturation of genomic DNA.

[Isolating candidate inserted fragment from positive fused phage clones using quantitative PCR].

OBJECTIVE: To isolate quickly and exactly the specific inserted fragment from fused phage clones which were obtained from cDNA library by hybridization. METHODS: According to the amplification principle of quantitative polymerase chain reaction (PCR) and based on the difference of original template quantity,target cDNA fragment was isolated and identified by two PCRs. RESULTS: A positive clone with specific cDNA fragment of HumGT-H1 gene was obtained from a two-phage fused clone by using this method,and the inserted fragment was verified to be the 5' cDNA sequence of HumGT-H1,1.9kb in length. So another hybridization screening is not necessary. CONCLUSION: The method presented is effective and rapid in gene cloning and can greatly save time and materials.

Test of a vertical scan mode in 3-D imaging of residual limbs using ultrasound.

We have developed a new ultrasound system for imaging residual limbs that employs two scan modes: a vertical scan mode (VSM) primarily used for capturing the skin surface, and a horizontal compound scan mode (HCSM) primarily used for imaging the internal tissue structures. Since the HCSM was described previously (J Rehabil Res Dev 1997;34(3):269-78), the main purpose of this article is to elucidate the principle of reconstructing the skin surface using the new VSM and to report the accuracy of volumetric measurement based on this reconstruction method. For a residual limb with a length of up to 26 cm, the vertical scan takes about 80 s. The acquired skin surface has a resolution of 144 points per circumference in the transverse direction and a resolution of 28 points per cm in the longitudinal direction. The measurement accuracy of the system is tested using a cylindrical phantom and a custom-made limb model. The diameter of the phantom measured by the ultrasound method is 113.8 +/- 0.23 mm, compared with a value of 113 mm measured by a caliper. For the limb model, cumulative segment volumes measured by the ultrasound method are compared with those measured by the hydrostatic weighing method. The relative errors are <1%. This study demonstrates the feasibility of using ultrasound to perform limb measurement for prosthetic CAD/CAM applications.

Insecticidal activity of transgenic tobacco plants expressing both Bt and CpTI genes on cotton bollworm (Helicoverpa armigera).

Transgenic tobacco plants expressing both Bacillus thuringiensis (Bt) insecticidal protein and cowpea trypsin inhibitor (CpTI) genes was used to evaluate the insecticidal activity on the cotton bollworm (Helicoverpa armigera) compared with transgenic tobacco with Bt insecticidal protein gene alone. Mortality of first to third instar larvae fed on transgenic two genes tobacco for three days was significantly higher than that fed on transgenic Bt tobacco. First to fourth instar larvae fed on transgenic two genes tobacco continuously could not survive until pupation. Second instar larvae were fed on transgenic tobacco plants for three days and then were transferred to an artificial diet. The efficacy of transgenic two genes tobacco on mortality, larvae weight, percentage of pupation, and time to pupation were significantly higher than that of transgenic tobacco with Bt insecticidal protein gene alone. The results with transgenic tobacco as model plant were valuable for other crops both for the enhancement of insecticidal efficacy and for the delay of insect adaptation to transgenic Bt crops.

Transgenic potato plants expressing osmotin gene inhibits fungal development in inoculated leaves.

Osmotin, a 24-KD protein isolated from Nicotiana tabacum 'Xanthi nc' was very toxic to Phytophthora infestans in vitro assay. Transgenic potato plants expressing wild osmotin protein delayed the emergence of symptoms of disease. Two osmotin mutants have been produced by PCR mutagenesis. One mutant, delta C18 osmotin, had a nonsense mutation near the C-terminus that deleted the carboxyl terminal prepropeptide (CTPP) of 18 amino acids. Another mutant, osmotin-P, had amber mutation at 96th amino acid. Both mutant genes driven by CaMV d35S promoter were introduced in potato by Agrobacterium mediated genetic transformation. The expression and osmotin protein accumulation in transgenic potato were detected by Southern, Northern, and Dot-blot ELISA assay. Disease resistance test using P.infestens complex race showed that expression of both osmotin mutants in transgenic potato reduced the lesion growth rate in inoculated leaves. The results showed that accumulation of osmotin protein in intercellular spaces (delta C18osmotin) inhibited the development of fungal disease in inoculated leaves. Based on the results we suggest that antifungal activity was possibly located at N end of osmotin protein (osmotin-P).

[The boundary design of mandibular model by means of the three-dimensional finite element method].

OBJECTIVE: To construct the mandibular model and simulate the boundary conditions by means of the three-dimensional finite element method. METHODS: A juvenile mandible, including TMJ, was scanned transversely by CT, then a three-dimensional finite element model of mandible was constructed. At the same time, the functions of stomatologic muscles and ligaments and the functional actions of temporomandibular joint (TMJ) were simulated by means of cable elements and compressive gap elements respectively. RESULTS: The cable elements were consistent with the functions of stomatologic muscles and ligaments, and the gap elements were similar to the boundary conditions of the TMJ and teeth. CONCLUSION: Cable and gap elements are reasonable boundary conditions of the finite element model of mandible, which will improve the similarity of the model.

[Computer-aided legal medical examination of body surface].

This paper provides a package of automatic and semi-automatic methods for computing the area of different kinds of body surface injuries. Compared with traditional methods, these processes of examination are faster and the conclusions are more precise and objective. Also presented are the authors classify the items into many types by standards which are necessary to let computer draw conclusions automatically. This software is conducive to improvement in work efficiency and convenience for judicial supervision.

[Three-dimensional finite element study on periodontal membrane of central maxillary incisor under physiological load].

This paper presents the development of a three-dimensional finite element model of central maxillary incisor. Stress distribution of the periodontal membrane was obtained by calculation under physiological load exerted to the natural tooth and tooth restored with post respectively. And it was compared with the stress distribution of the implate (contact with bone and without periodontal membrane). The result indicated that, under physiological load, when the natural tooth was compared with tooth restored with post, there were no apparent diferences in stress distribution of periodontal membrane and in stress values on inner surface and outer surface of periodontal membrane. It suggests that periodontal membrane has apparent cushioning action against stress.

[One-stage reconstruction with the pedicle skin flap of forehead for defective tissue of head-neck tumors dissection].

OBJECTIVE: To discuss the feasibility on reconstructing the defective tissue with the pedicle skin flap of forhead after head-neck tumors dissection. METHOD: 14 patients with advanced head-neck tumors were studied, whose defective tissues were reconstructed by their forehead skin flaps. The flap area ranged from 5 cm x 11 cm to 7 cm x 16 cm. RESULT: 2 cases of them were partial necrosis, 3 years and 5 years survival rates were 76.9% and 63.6% respectively. The function of breath through nose or mouth and the function of swallow were satisfactory. CONCLUSION: The forehead skin flap should be a good selection for the reconstruction of defective tissue after basicranial and nasalfacial dissection.

[79 cases: a clinical analysis of hoarseness in children].

OBJECTIVE: To research the clinical characters of the hoarseness in children. METHOD: 79 cases are analysed retrospectively. RESULT AND CONCLUSION: Age for this disease is usually 3 years old. People with irritable character are more easy to get it. Vocal nodule is the main cause to chronic hoarseness in children. Conservative treatment is important.

[Prevention of complications of endoscopic sinus surgery].

OBJECTIVE: To investigate the causes and prevention of the complications of endoscopic sinus surgery (ESS). METHOD: We summurized the complications of 26 cases among 100 patients who were performed the ESS operations. RESULT: The complications included hemorrhage, cerebrospinal fluid leak, orbital hematoma, subcutaneous orbital emphysema, synechia and antrotomy closure. CONCLUSION: We concluded the factors which probably result in complications and the prevention of complications.

[A new adaptive filtering method for micro-particle image processing].

Based on the local gray level histogram of macro-particle image which is more special than other images, a new adaptive noise filtering method for macro-particle image processing is developed and described in this paper. It is made up of median filter and special order statistic (OS) filters (alpha-mean filter and inverse alpha-mean filter). This adaptive filter not only has satisfactory performance in both of noise-removal and edge-preservation of macro-particle image, but also is easy and quick to implement. An example is presented in this paper.

[3-channels data acquisition system based on single-chip-microcomputer used in the measurement of coronary sinus blood flow].

The coronary sinus blood flow can be figured out, which based on the principle of thermodilution, so long as gets the temperature of blood, indicator and mixture of blood and indicator respectively. This system is a smart slave module with single-chip-microcomputer. The structure and principles of hardware and the flow chart of software are described in detail.

[Interferences in analysis of mineral elements in salt by FAAS and their correction].

This paper studies interference factors in the analysis of mineral elements in salt by FAAS and discusses the problems of background interference, Cl- interference, Na+ interference and other ion interferences. K, Na, Mg, Cu, Mn, Zn, Fe, Pb, Cr and Cd in commercial salt were determined. The results showed that this method did not suit Cr,background absorption only interfered with the determination of K, and Mg, 1% Cl- interfered with the determination of K, and 10% Cl- or 2% Na+ interfered with the other elements. After technical treatment and eliminating all sorts of interferences, the results of analysis for salt are satisfactory.

Reversion of the neoplastic phenotype of human glioblastoma cells by connexin 43 (cx43).

Connexins (cx), structural components of gap junction, are believed to play a role in the regulation of cell proliferation and suppression of the neoplastic phenotype. We used human brain glioblastoma tumor cells as a model system to test this hypothesis. Western blot and reverse transcription-PCR analysis indicate that the expression levels of the gap junction protein connexin 43 (cx43) are profoundly decreased in several human brain tumor cell lines examined. Transfection of human cx43 into human glioblastoma cell lines U251 and T98G profoundly reduces cell proliferation in monolayer culture, in soft agar, and in athymic nude mice. Surprisingly, these effects are not associated with the establishment of gap junction communication in cx43 transfected cells. We conclude that the loss of cx43 expression may play a role in the development of human gliomas and that cx43 acts as a tumor suppressor gene to human glioblastoma.
