Polo-like kinase interacts with proteasomes and regulates their activity.

The polo-like kinase (Plk) has been shown to be associated with the anaphase-promoting complex at the transition from metaphase to anaphase and to regulate ubiquitination, the process that targets proteins for degradation by proteasomes. In this study, we have identified proteasomal proteins interacting with Plk by mass spectrometry and found that Plk and 20S proteasome subunits could be reversibly immunoprecipitated from both human CA46 cells and HEK 293 cells transfected with HA-Plk. Furthermore, both coprecipitated Plk and baculovirus-expressed Plk were able to phosphorylate proteasome subunits, and metabolic labeling studies indicate that Plk is partially responsible for the phosphorylation of 20S proteasome subunits C9 and C8 in vivo. In addition, phosphorylation of proteasomes by Plk enhanced proteolytic activity toward an artificial substrate Suc-L-L-V-Y-AMC in vitro and in vivo. Finally, we were also able to detect Plk associated with 26S proteasomes under certain conditions. Together our results suggest that Plk is an important mitotic regulator of proteasome activity.

[Inhibition effect in vitro of purified endostatin expressed in Pichia pastoris].

Endostatin is a newly found inhibitor of angiogenesis, which is identified as c-terminal 184 amino acid fragment of collagen XVIII NC1-domain. A 570 bp cDNA fragment of endostatin has been amplified by PCR from a commercial human fetal liver cDNA library. After subcloned into the yeast vector pPIC9 and subsequence to prove its correctness, Pichia pastoris was transformed with the recombinant pPIC9-endostatin. The expressed endostatin in P. pastoris was purified by heparin-sapherose affinity chromatography. It's purity identified by SDS-PAGE thin layer scanning analysis was up to 98.7% and its Mol. Weight measured by MS was 20.34 kD. The expression level was up to 40 mg/L. The first fifteen amino acid sequence of the N-terminal was completely identical with the inner sequence C-terminal fragment of collagen XVIII NC1 domain as has been designed. Bioassay indicated that the recombinant endostatin can inhibit angiogenesis stimulated by bFGF in CAM test and also the proliferation of both HUVEC and ECV304 in an in vitro test.

All-trans-retinoic acid increased the expression of integrin alpha5beta1 and induced "anoikis" in SMMC-7721 hepatocarcinoma cell.

It is extensively shown that integrin can regulate various cellular functions, including apoptosis, probably by contributing to signal transduction processes through interaction with extracellular matrix (ECM) proteins. In the present study, DNA flow cytometric analysis demonstrated that SMMC-7721 hepatocarcinoma cells treated with 80 microM all-trans-retinoic acid (atRA) showed an increased expression of the integrin alpha5beta1, which was associated with the growth inhibition of the cells. We found that atRA treated cells showed obvious apoptosis. Then, it was postulated that the enhanced content of integrin alpha5beta1 in the absence of ligation with fibronectin (Fn) would stop transducing survival signals, and lead to decreased cell growth and apoptosis. To elucidate this hypothesis, we cultured the atRA treated cell in L-poly-lysine-coated and Fn-coated flask, respectively. The results indicated that Fn binding prevented the cells from apoptosis induced by atRA, in contrast to L-poly-lysine binding. When the transfectant with enhanced expression of integrin alpha5beta1 at the same level of atRA treated cell was cultured in L-poly-lysine-coated flask, apoptosis was triggered. However, apoptotic cell was not detected when those cells were cultured in Fn-coated flask. Meanwhile, culturing the transfectant in the antibody (against integrin alpha5 subunit)-coated flask induced 18.33% of the cells into apoptosis, which is far more than the control group. Our study suggests that increased expression of integrin alpha5beta1 on the surface of SMMC 7721 hepatocarcinoma cell treated by atRA, when unbound to Fn, would stop transducing survival signals to lead to "anoikis", and can be reverted by the interaction of integrin alpha5beta1 with Fn.

Effects of droloxifene on apoptosis and Bax, Bcl-2 protein expression of luteal cells in pseudopregnant rats.

AIM: To study the effects of droloxifene on apoptosis and the expression of Bax and Bcl-2 protein in corpus luteum of pseudopregnant rats. METHODS: HE staining was used to examine the histological changes of ovaries. Apoptosis detection in situ was performed with TUNEL method. Expression of Bax and Bcl-2 protein was observed by immunohistochemistry analysis. RESULTS: Apoptosis of luteal cells during the spontaneous regression of corpus luteum of pseudopregnant rats appeared on d 13 of pseudopregnancy, and the marked increase of apoptotic luteal cells could be observed on d 15. When pseudopregnant rats were treated with droloxifene 20 mg . Kg on d 2, apoptosis of luteal cells could be observed on d 8 and the duration of pseudopregnancy could be shortened from (15.5 +\- 1.1) d to (12.8 +\- 1.6) d. In pseudopregnant rats, the expression of Bax and Bcl-2 protein was found in the cytoplasm of luteal cells. However, no obvious differences in the intensity or localization could be found during various days of the pseudopregnancy, while an increase in Bax and a decrease in Bcl-2 protein expression could be induced by droloxifene treatment. CONCLUSION: Droloxifene could facilitate apoptosis of luteal cells in pseudopregnant rats and shorten the period of pseudopregnancy. An increased Bax/Bcl-2 ratio might be involved in the facilitation of apoptosis induced by droloxifene in corpus luteum of pseudopregnant rats.

[Experimental study on resistance to cisplatin and taxol in human ovarian cancer SKOV3ip1 multicellular aggregates].

OBJECTIVE: To investigate the chemosensitivity of ovarian cancer SKOV3ip1 multicellular aggregates to cisplatin and taxol and to explore the possible mechanisms accounting for the effect. METHODS: Liquid overlay system was employed to obtain multicellular aggregates (MCA). We detected the resistance with trypan blue exclusion testing, clonogenic assay, cell cycle profiles and apoptosis with flow cytometry. RESULTS: MCA cells showed higher cell viability than monolayer cells (P = 0.045 and P = 0.003, respectively). After 40 mumol/L cisplatin exposure for 12 hours, no clone (> or = 50 cells) was formed. After 10 mumol/L taxol exposure for 12 hours, the clone formation showed significant difference in 100-cell group between multicellular aggregates and monolayer cells (P < 0.05). MCA cells in G0 + G1 phase was significantly increased (P = 0.003). After cisplatin exposure, the apoptosis rate of MCA cells were not significantly changed (P = 0.100), and so were cell cycle profiles. Taxol exposure brought about significantly decreased apoptosis rate in MCA cells (P = 0.012). Abrogation of G2 + M arrest was also showed in MCA cells (P = 0.002). CONCLUSIONS: SKOV3ip1 MCA cells show varying degree but increased resistance to cisplatin and taxol, in particular the latter. Cisplatin has nearly equal cytotoxicity to monolayer and MCA cells. Cell cycle redistribution, abrogation of G2 + M arrest and multicellular-mediated inhibition of apoptosis can partially account for the resistance.

[Apolipoprotein E 4 gene is a risk factor for Alzheimer's disease].

OBJECTIVE: To study the association of Alzheimer disease (AD) with apolipoprotein E (ApoE) epsilon 4 allele and to find the biological peripheral markers for the laboratory diagnosis of AD. METHODS: 107 patients with AD, 68 patients with vascular dementia (VD) and 74 sex- and age-matched non-demented healthy individuals (NDC) were collected. DNA from patients and healthy individuals was extracted from peripheral blood samples with the phenol-chloroform procedure and ApoE was investigated with the methods of polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP). RESULTS: The frequency of epsilon 4 was significantly higher in AD than that in VD and NDC and there was no difference in the frequency of epsilon 4 between VD and NDC. The age at onset of the disease in the group carrying two epsilon 4 alleles was significantly younger than that in the group with one epsilon 4 allele in AD. Moreover, in comparison with AD patients without epsilon 4 allele, the age at onset in the group with one epsilon 4 allele was younger. Patients with epsilon 4 or without epsilon 4 allele in VD patients did not differ significantly in age at onset. Meanwhile, 3 familial AD cases were found, all carrying epsilon 4 allele. CONCLUSION: AD and ApoE4 were closely related. ApoE epsilon 4 was a dangerous factor of AD and ApoE 4 allele made contribution to the heterogenicity of AD.

Liposome-C-erbB2 antisense oligodoxynucleotides in human ovarian cancer cells.

OBJECTIVE: To explore the effects of liposome-C-erbB2 antisense phosphorothioate oligodeoxynucleotides (S-ODNs) on C-erbB2 proto-oncogene expression and cell proliferation in human ovarian cancer cells. METHODS: The effects of liposome-C-erbB2 S-ODNs on C-erbB2 protein expression, cell cycle and cell proliferation in human ovarian cancer cells were studied by means of flow cytometry and 3H-thymidine incorporation. RESULTS: Liposome-C-erbB2 S-ODNs can specifically reduce C-erbB2 protein expression in human ovarian cancer cells, accompanied by a 30% inhibition of cell proliferation. The effectiveness of liposome-C-erbB2 S-ODNs on the expression of C-erbB2 was about 40 times higher than that of C-erbB2 S-ODNs. CONCLUSIONS: The data suggest that antisense therapy might be a useful method of gene therapy in ovarian cancer. The effectiveness of C-erbB2 S-ODNs could be greatly increased by adsorption of S-ODNs by liposomes.

[Application of sequential culture in assisted reproductive technique].

OBJECTIVE: To investigate the effects of sequential culture technique on the outcomes of in vitro fertilization-embryo transfer(IVF-ET). METHODS: Sequential culture media were used in 47 IVF-ET cycles, and conventional medium in 114 cycles. The fertilization rate, cleavage rate and speed, good embryo rate and pregnancy rate were compared between these 2 groups. RESULTS: There were significantly higher embryo cleavage and quality score rate, pregnancy rate (97.9%, 64.4%, 46.7%) in sequential media group as compared those in conventional medium group (89.7%, 40.2%, 28.2%). CONCLUSION: Sequential culture system is more suitable for the development of cultured human oocytes and embryos.

[Diagnosis and treatment of 31 patients with splenic tumors].

OBJECTIVE: Splenic tumor, a rare disease, is scarcely mentioned in the literature. Here, we wish to present a clinical study on 31 such patients treated in the PLA General Hospital with emphasis on diagnosis and treatment. METHODS: Thirty-one splenic tumor patients were treated from January 1990 to December 2000. The clinical manifestations and image characteristics of splenic tumor, diagnosis and treatment were analysed. RESULTS: Male 19, female 12. Their ages ranged from 13 to 80 years with an average of 56. The symptoms and signs were non-specific. All patients were first diagnosed by beta-ultrasonography to have single or multiple space-occupying lesions. Then, 19 patients received CT and 7 MRI scans. The diagnosis of seven patients suffering from splenic cysts were initially made definite by beta-ultrasonography among whom the lesions of 6 were confirmed by operation and pathology. The diagnosis of 9 patients with metastatic tumor was ascertained by combined imaging and clinical history. Finally, the lesions were confirmed by operation and pathology. Fifteen patients with primary splenic tumor were found to have primary space-occupying lesion by imaging: Except one patient who was diagnosed to have splenic lymphoma and not operated, all the other 14 patients underwent splenectomy with their lesions confirmed by pathology. Altogether, 25 of these 31 patients were treated by splenectomy with uneventful recovery without any operative mortality. Final pathologic examination disclosed that 20(65%) of 31 lesions were benign. CONCLUSION: Clinical history and imaging scans are important in the diagnosis of splenic tumors. The differentiation of benignity and malignancy is difficult before operation. Surgery is satisfactory in treating splenic tumors.

[The effect of leukotriene antagonist on the parameters of dose-response curve obtained after methacholine challenge test].

OBJECTIVE: To investigate the effect of leukotriene antagonist (zafirlucast) on the parameters of dose-response curve (DRC) obtained after methacholine challenge test. METHODS: A therapy trial was conducted with the use of zafirlucast in 20 cases of cough variant asthma (CVA) and 20 cases of asthma. Placebo was used in 4 cases of CVA and 10 cases of asthma for comparison with the two therapy groups respectively. Before and after the zafirlucast treatment a series of parameters of DRC were obtained after the methacholine challenge test and were calculated. These included the response threshold (PC35sGaw), the dose-response slope (DRS), the ratio of the area under the curve to the logarithm of the maximal concentration (AUC/1 g[Cmax]), and the maximal response. RESULTS: Zafirlucast significantly increased the PC35sGaw in the CVA and asthma groups. It conspicuously decreased all the parameters of DRS and AUC/1 g[Cmax] except for maximal response in the cases of CVA, and it had no significant effects on all the parameters of DRS and AUC/1 g[Cmax] except for PC35sGaw in the cases of asthma. CONCLUSION: Short-term use of Zafirlucast in asthmatic patients could improve the parameters of dose-response curve obtained after the methacholine challenge test.

[Measurement of the parameters of dose-response curve obtained after the methacholine challenge test].

OBJECTIVE: To investigate the clinical value in the measurement of parameters of the dose-response curve (DRC) obtained after the methacholine challenge test. METHODS: Twenty-seven cases of cough variant asthma (CVA), 29 of mild asthma, 19 of moderate asthma, and 15 healthy volunteers underwent the methacholine challenge test. The dose-response curves were constructed. The following parameters were calculated: position (PC35sGaw), dose-response curve (DRS), ratio of the area under the curve to the logarithm of the maximal concentration (AUC/lg[Cmax]) and maximal response. RESULTS: A plateau appeared in 11 cases among the healthy subjects, while a plateau appeared only in 2 cases of CVA and 2 of asthma. There were no significant differences among the asthmatic groups in the measurements of PC35sGaw and AUC/lg[Cmax] (geometric mean PC35sGaw: mild asthmatics = 0.369, moderate asthmatics = 0.251, cough variant asthmatics = 0.547), AUC/lg[Cmax] was significantly greater in the asthmatic groups than in the normal group [AUC/lg [Cmax](mean +/- s): mild asthmatics = 24.7 +/- 4.7, moderate asthmatics = 26.6 +/- 4.3, cough variant asthmatics = 25.6 +/- 3.7, normal subjects = 15.5 +/- 4.3, P < 0.01]. DRS of moderate asthmatics was significantly greater as compared with cough variant asthmatics (geometric mean DRS: mild asthmatics = 30.761, moderate asthma-tics = 59.020, cough variant asthmatics = 19.231, P < 0.05), but there was no difference between the other groups. PC35sGaw was negatively correlated with DRS (r = 0.866, P < 0.001) and with AUC/lg[Cmax] (r = 0.502, P < 0.001). CONCLUSION: Four parameters of DRC obtained after the methacholine challenge represent airway sensitivity and reactivity respectively. The parameters of DRC of asthmatic patients are different from those of normal subjects. The sensitivity is related to the reactivity, but they are not completely parallel with each other.

[The dynamic study of cytokines pre- and postoperation in patients with carcinoma of larynx].

OBJECTIVE: To observe the dynamic changes of cytokines pre- and postoperation in patients with carcinoma of the larynx and provide reference for recurrence monitor and prognosis evaluation in postoperative patients with head and neck tumor. METHOD: Thirty-four patients with head and neck squamous cell carcinoma were included in this study. Serum levels of IL-2, sIL-2R and TNF-alpha were measured before surgery, 7 days, 30 days and 90 days after surgery with ELISA and RIA. RESULT: The serum levels of sIL-2R and TNF-alpha before surgery in patients with carcinoma of the larynx were significantly elevated compared with controls(P < 0.01), while the level of IL-2 was not different. Serum concentrations of sIL-2R and TNF-alpha in patients without recurrence found at 7 days after operation were slightly higher than those before operation, but there was not significant difference. Moreover, the levels of sIL-2R and TNF-alpha at 30 days and 90 days postoperation decreased gradually, especially the sIL-2R levels decreased more significantly. No statistical difference were found among serum IL-2 levels in non-recurrence patients. The serum levels of sIL-2R and TNF-alpha in patients with recurrence at 30 days after operation were lower than those before operation and increased at 90 days after operation, while the levels of IL-2 decreased gradually. CONCLUSION: Dynamic study of the cytokines pre and postoperation play reference role in recurrence monitor and prognosis evaluation after the larynx carcinoma were removed.

[Influence of different collection time on yield and constituents of essential oil of herba pogostemonis from Gaoyao County].

The constituents of the volatile oil of Herba Pogostemonis from Gaoyao county collected at various different time by GC/MS combination technology were analysed and compared. The results showed that the average dry weight of single plant(i.e. yield) increased gradually from July to next April, the content of volatile oil was highest in November. The contents of pogostemone in the leaf oil were higher in July and September, then decreased gradually with growth time; pachouli alcohol was higher in September, October and November, lower in other months. The contents of pogostmone in stem oil were higher in July, October and next January, lowest in November. The content change of pachouli alcohol in stem oil were not obvious in all growth time, higher slightly only after November. Other compounds were not showed their variation law. So the quality of Herba Pogostemonis collected in November was higher if the content of volatile oil was used as a standard of quality evaluation, the result was in accordance with traditional collection experience.

[A kinetic study on the relationship between of IL-5, IL-10 and eosinophil apoptosis in asthmatic airway inflammation].

OBJECTIVE: To observe time-course profile of eosinophil apoptosis, in murine asthma models in airway explicate its implication for asthmatic inflammation remission, and investigate the role of IL-5 and IL-10 in modulating cell apoptosis. METHODS: An animal model of asthma was established by OVA sensitizing-challenging BALB/c mice. Histologic study of lung tissue was made with the use of electronmicroscope. At seven points (0 h, 8 h, 24 h, 48 h, 96 h, Day 7 and Day 14) in the time course after challenge, bronchoalveolar lavage (BAL) was performed obtain BAL cells and recover fluid. Levels of IL-5 and IL-10 were detected by ELISA. Cell apoptosis was assayed by PI stain on a flow cytometer. RESULTS: After challenge, apoptosis of eosinophils, lymphocytes, endothelial cells could be seen under the electronmicroscope. IL-5 in BALF elevated significantly whereas no significant difference was found between the levels of IL-10 in BALF before and after challenge. Eosinophil apoptosis was identified and a two-phase elevation of apoptosis percentages was observed, although no significant difference was found between average apoptosis in asthmatic group and that in control. Data analyses revealed a negative correlation between apoptosis percentages and the levels of IL-5, and a closer correlation between the percentages and IL-5/IL-10 ratio. CONCLUSION: IL-5 and IL-10 could up- and down-regulate asthmatic airway inflammation, respectively. EOS underwent apoptosis in situ and this was modulated by IL-5 and IL-10 interaction.

[A multi-centered clinical study an domestic intravenous Turbutalin sulfate in the treatment of asthma and chronic asthmatic bronchitis].

OBJECTIVE: This multi-centered clinical trial was designed to assess the efficacy and safety of domestic intravenous Turbutalin Sulfate in the treatment of asthma and chronic asthmatic bronchitis. METHODS: Bricanyl was used as control. A total of 120 patients were included in this randomized controlled study. The two medications, both at a dose of 0.25 mg intravenous drip, were given three times a day for 3-5 days. RESULTS: The two drugs' clinical excellent rates were 66.7% and 73.3%; overall efficacy rates were 98.3% and 100.0%, respectively. Their pulmonary functional excellent rates were 78.4% and 76.6%; overall effect rates were 88.3% and 90.0%, respectively. Composite curative evaluation showed that the two drug's composite excellent rates were 61.6% both; their overall efficacy rates were 88.3% and 90.0%, respectively. All of these showed no significant difference (P > 0.05). The incidence rates of adverse side effects of the two groups were 13.5% and 15.0%, respectively; most of them were mild and tolerable. CONCLUSION: Domestic intravenous Terbutalin Sulfate is effective and safe in treating asthma and chronic asthmatic bronchitis.

[Subtotal laryngectomy reconstructed by platysma myocutaneous flaps].

OBJECTIVE: To study the feasibility, surgical techniques and the results of laryngeal function preservation during subtotal laryngectomy in laryngeal carcinoma. METHODS: A retrospective review was made of 13 patients treated by subtotal laryngectomy and laryngeal reconstruction from 1991 to 1996. Eleven patients were male and 2 were female. The age ranged from 46 to 72 years, with a mean of 53.25. All cases were of stage IV. The essence of this operation is to reconstruct the new larynx with two platysma myocutaneous flaps following subtotal laryngectomy. RESULTS: The success rate of voice rehabilitation was 100%. Among them, the acceptability rating scores in 11 cases were 7 and the intelligibility scores ranged from 95% to 98%. The other two cases had acceptability 5 and intelligibility form 90% to 94%. The three-year survival rate was 84.6% (11/13), 5-year survival rate was 76.9% (10/13). CONCLUSION: This form of laryngeal reconstruction in subtotal laryngectomy is helpful to restore laryngeal function and decrease the rate of total laryngectomy. The procedure is easy, safe and applicable to patients treated by subtotal laryngectomy.

[Relationship among lymphatic metastasis, pericancerous lymphocytic reaction and dendritic cell infiltration in laryngeal carcinoma cells].

OBJECTIVE: To study the role and the clinical significance of dendritic cell infiltration against tumor cells in patients with laryngeal squamous cell carcinoma. METHODS: Immunohistochemical method using S-100 protein antibody was employed to detect dendritic cells (DC) on paraffin-embedded tissue sections from 23 patients with laryngeal squamous cell carcinomas. The relationships among the density of dendritic cell infiltration in laryngeal carcinomas and cervical lymphatic metastasis, pericancerous lymphocytic reaction and other clinicopathologic parameters were observed. RESULTS: The numbers of dendritic cells infiltrating among laryngeal carcinoma cells in patients with no cervical lymph metastasis was significantly larger than those with cervical lymphatic metastasis (t = 4.889, P < 0.01). Significant increase DC infiltration among laryngeal carcinoma cells was found in the group with intensively positive reaction of pericancerous lymphocyte than in the medium and weakly positive groups. The number of infiltrating dendritic cells in patients who had survived more than 5 years was significantly larger than those survived less than 5 years (t = 4.423, P < 0.01). The numbers of the infiltrating dendritic cells in patients with well-differentiated squamous cell carcinoma and poorly differentiated squamous cells carcinoma were 2.2 +/- 1.07 and 14.6 +/- 7.14 respectively, which were significantly different (q = 4.532, P < 0.05). CONCLUSION: The study showed that DC infiltration among laryngeal squamous carcinoma cells played an important role in the host immune reaction against tumor. It indicated that when the density of infiltrating DC is higher, the patients had less chance of cervical lymph metastasis and may have a longer survival time. There was a coordinative effect between DC infiltration in tumor cells and pericancerous lymphocyte reaction. Thus, the DC infiltration among laryngeal squamous cells could be used as an index of prognosis.
