Molecular analysis of suppression of interleukin-8 production by rebamipide in Helicobacter pylori-stimulated gastric cancer cell lines.

Interleukin-8 (IL-8) may play an important role in Helicobacter pylori infection-associated chronic active gastritis and peptic ulcer disease in human. We have recently reported that a gastric cancer cell line, MKN45, produced a massive amount of IL-8 upon coculture with live H. pylori. Moreover, H. pylori induced the activation of NF-kappaB as well as AP-1, leading to IL-8 gene transcription. In this study, we evaluated the effect of rebamipide, an antigastritis and antiulcer agent, on H. pylori-induced IL-8 production. Rebamipide inhibited the production of IL-8 in several gastric cancer cell lines infected with H. pylori. In addition, rebamipide suppressed H. pylori-induced IL-8 gene expression at the transcriptional level as revealed by northern blotting analysis and luciferase activity in cells that were transfected with a luciferase expression vector linked with a 5'-flanking region of the IL-8 gene (bp -133 to +44). Furthermore, rebamipide significantly suppressed the NF-kappaB activation by H. pylori infection. These results suggest that rebamipide may protect against the mucosal inflammation associated with H. pylori infection through inhibition of a proinflammatory cytokine, IL-8.

Occurrence of terminal alpha 2-->8-linked disialylated poly-N-acetyllactosamine chains with Le(X) and I antigenic glycotopes in tetraantennary arms of an N-linked glycoprotein isolated from rainbow trout ovarian fluid.

The Pronase digestion of a 54K glycoprotein present in ovarian fluid of rainbow trout yielded a major glycopeptide. Carbohydrate compositional analysis revealed that this glycopeptide was likely to possess a single large N-glycan chain having low molecular weight oligomers of N-acetylneuraminic acid (oligoNeu5Ac). Structural studies of this glycopeptide revealed novel alpha 2-->8-linked disialylated poly-N-acetyllactosamine chains with Le(X) and I antigenic determinants on the N-linked tetraantennary core glycan. In our recent studies (Kitazume,S., Kitajima,K., Inoue,S., Inoue,Y. and Troy,F.A. (1994) J. Biol. Chem. 269, 10330-10340) we presented evidence that synthesis of alpha 2-->8-linked polysialic acid (polySia) chains is a two-step process in which chain initiation is catalyzed by an alpha 2-->8-sialyltransferase (alpha 2-->8-ST; initiase) that catalyzes synthesis of the first Sia alpha 2-->8-linkage, forming the disialic acid (diSia) unit, Sia alpha 2-->8-Sia alpha 2-->6-Gal-. Chain polymerization is then postulated to be catalyzed by a second enzyme, an alpha 2-->8-polyST ("polymerase") that converts the diSia units to polySia chains. The present structural studies leading to the discovery of alpha 2-->8-linked disialylated units that terminate poly-N-acetyllactosamine chains in an N-linked glycoprotein is further evidence in support of our hypothesis that more than one sialyltransferase activity is required for polySia chain synthesis and polymerization.

Neo red cell as an organ preservation solution.

Neo red cell (NRC) was derived from outdated human red cells. The following experimental work has been done in order to investigate if NRC is valid for organ preservation. Hearts were obtained from male Lewis rat (250-350 g body weight). Heart transplantation was performed as Ono-Lindsey's method after 1, 3, 6, 12 and 24 hours simple cold storage. Rats were divided into two groups. Group 1; original NRC as preservation solution, Group 2; modified NRC (NRC suspended with UW solution) as preservation solution. After 1, 3, and 6 hour cold ischemic storage, the transplanted hearts in both groups showed contraction immediately after declamping the aorta and the pulmonary artery. After 12 and 24 hour preservation, the transplanted hearts in group 2 showed contraction. Myocardial ATP levels after 6, 12, 24 hours cold storage compared with 0 hour were 62.3%, 28.3%, 32.8% in group 1 respectively. On the contrary, myocardial ATP levels were 87.2%, 57.6%, 52.2% in group 2 respectively. After 24 hour preservation, pathological change was not remarkable between the two groups. Results of the prolonged preservation time and the myocardial ATP changes suggest that there is a possibility for effective use of NRC not only as blood substitute but also organ preservation solution.

Evaluation of two assay kits for thermostable direct hemolysin (TDH) as an indicator of TDH-related hemolysin (TRH) produced by Vibrio parahaemolyticus.

Reversed passive latex agglutination (RPLA) or enzyme-linked immunosorbent assay kits with beads (Bead-ELISA) are commercially available in Japan to detect the thermostable direct hemolysin (TDH) produced by Vibrio parahaemolyticus isolates. We evaluated whether these kits can be used to assay the pathogenic toxin, TDH-related hemolysin (TRH), produced by some so-called Kanagawa phenomenon-negative V. parahaemolyticus strains isolated from patients with diarrhea. Our results showed that the two kits, RPLA and Bead-ELISA, can detect TRH, although they were originally developed for detection of TDH. This may be due to the use of polyclonal anti-TDH antisera that cross react with TRH. Although the sensitivity for TDH detection by RPLA and Bead-ELISA differed tenfold, that for TRH detection was essentially equal. The minimum concentration of TRH required for detection by the two assay kits was about 10 ng/ml.

Phytogrowth-inhibitory activities of 2-thiophenecarboxylic acid and its related compounds.

2-Thiophenecarboxylic acid (I) exhibited growth-inhibitory activity in five kinds of plants. In particular, I strongly inhibited the growth of the roots of Lactuca sativa L. var. longifolia LAM and Echinochloa utilis OHWI et YABUNO, even at the low concentration of 5.0 x 10(-3) M. Furthermore, all of the I-related compounds (II-V and VII-X) except for VI, showed more or less obvious inhibitory activity on the seeds of Sesamum indicum L. Compounds VII-X, in which the carboxyl group of I was replaced by acetic acid, propionic acid, butyric acid and acrylic acid, and exhibited more potent phytogrowth-inhibitory activity than I. Among these compounds, 2-thiophenebutyric acid (IX) showed the strongest activity. Esterification of the carboxyl group in I increased the inhibitory activity relative to that of I, while amidation and reduction of this group markedly decreased its inhibitory activity. The radicles of the plants treated with each of the compounds except for VI showed negative geotropism, even though germination occurred.

An immunohistochemical study of basic fibroblast growth factor in the developing chick.

An antiserum against basic fibroblast growth factor (bFGF) was characterized by immunoblot experiments and used to investigate immunohistochemically the appearance of bFGF-like immunoreactivity in the developing chick. Crude homogenates of chick embryos at every developmental stage, when subjected to immunoblotting with the use of bFGF antiserum, exhibited a main band with the same molecular weight (18 kDa) as bovine bFGF. With immunohistochemistry, bFGF immunoreactivity (bFGF-IR) was detected exclusively in intracellular components of various tissues at different stages of development; bFGF-IR appeared initially on embryonic (incubation) day 3 (E3) in the myotome, on E12 in the spinal cord and ganglia, on E8 in chondrocytes and osteoblasts of the vertebrae, and on E10 in the esophageal epithelium. Immunoreaction products were present either in the cytoplasm or in the nuclei, depending on the types of individual bFGF-containing cells; developing chondrocytes and cells in the stratum basale of the esophagus exhibited intense immunoreactions exclusively within the nuclei, and the other cells mainly within the cytoplasm. Moreover, bFGF-IR was observed in discrete regions of these tissues at different stages; the epithelium of the esophagus contained bFGF-IR in all layers on E10 to E18 with a superficial-to-basal gradient, but it began to exhibit bFGF-IR only in the stratum basale after E20; and bFGF-IR was more abundant in hypertrophic chondrocytes than in proliferating ones. As chicks aged, bFGF-IR decreased or disappeared in the muscles, vertebrae and esophageal epithelium, but neuronal bFGF increased in intensity until the perinatal period and thereafter remained unchanged.(ABSTRACT TRUNCATED AT 250 WORDS)

Dental findings of patients who underwent liver transplantation.

Cyclosporin-induced gingival enlargement was presented. Enlargement was noted at several sites in the mouth of five patients taking Cyclosporin following liver transplantation. Papillary gingival enlargement was observed to affect the entire maxilla and mandible. Enlargement was particularly marked in the interdental papillary region on the labial side of the anterior teeth of the maxilla. Discoloration of the teeth was observed. Radiographic findings were unremarkable. Oral management of these patients is the prevention of dental caries and infection originating in the area of gingival enlargement. It is important to begin proper brushing as early as possible for suppressing progression of gingival enlargement. For prevention of dental caries, prophylaxis is more important in the case of liver transplantation than with normal patients. It should be stressed to the patients or their parents that periodic examination, topical application of fluoride and pit and fissure sealants are of special importance.

Biological nature of Cryptosporidium sp. isolated from a cat.

A small type of Cryptosporidium oocysts was isolated from a naturally infected cat and its biological nature was investigated. In cats experimentally inoculated with Cryptosporidium oocysts, long-lasting shedding of the oocysts occurred after a prepatent period of 8-10 days, and a number of peaks of oocyst count appeared at intervals of several days to a few weeks, earlier in the infection course. Cryptosporidium infection in cats is likely to pass from an acute to a chronic stage. During the chronic stage, prednisolone injection into the cats gave rise to a recurrence of proliferation of the parasite along with a marked increase in the number of oocysts shed. None of the infected cats showed clinical symptoms. Infection experiments using Cryptosporidium oocysts were unsuccessful in several species of animals such as mice, rats, guinea pigs, dogs, suckling mice and mice previously injected with prednisolone or hydrocortisone.

[Oral condition of infantile acute lymphatic leukemia with microbisme substitute].

The oral condition of a 7 year old boy at the 12th and 13th remission induction therapy sessions since contracting acute lymphatic leukemia was investigated clinically and microbiologically. The findings obtained were as follows: 1) The appearance of gingivitis coincided with the decrease of white blood cells 7 to 9 days after administration of the anti-leukopenic drugs. 2) Gingivitis appeared on the same site of the oral cavity at both remission induction therapies sessions. 3) The bleeding from the gingival lesions corresponded to the decrease of blood platelet at both remission induction therapies sessions. Conversely, gingivitis disappeared with the improvement of peripheral blood conditions. 4) The total number of oral bacteria decreased after administration of the antibiotics, although the ratio of Candida in the oral microbial flora increased markedly. 5) Typical symptoms of oral candidiasis were not observed after the appearance of the microbisme substitute.

[A case report on the achondroplasia and its dental findings].

Achondroplasia is characterized by an undergrowth of arms and legs bones, low height and undergrowth of the Basis cranil. The name of this disease was called Chondrodystrohia foetalis before Parrot reported the name of this disease as Achondroplasia, distinguished from other similar diseases. We are reporting in this paper on an 11-year, 2-month-old girl with Achondroplasia. The dental findings as follows: 1) Open bite was observed for the front teeth. 2) The size, number, form and growth of the teeth were usual. 3) The tongue was a little bigger than usual. 4) According to X-ray cephalometric analysis, undergrowth of the Basis cranii and relative anterior cross-bite were visible.

[Dental treatment under general anesthesia of a patient with pulmonary atresia with intact ventricular septum].

A case of pulmonary atresia with intact ventricular septum in a 4-year-4-month boy, whose dental treatment was performed under general anesthesia, was reported. For the general anesthesia, the medication Digoxin was stopped 24 hours before the treatment. For the prevention of bacterial endocarditis, medication consisting on antibiotics of the penicillin series was administered 24 hours before the treatment. The anesthesia was administered with the modified N.L.A. method with Ketamine and Diazepam. The dental treatment went smoothly, and was finished within 1-hour-15 mins from the start. Anesthesia was administered for 1-hour-45 mins. The prognosis was good, and the patient was discharged the next day. It should be noted that as for performing dental treatment under anesthesia or not using anesthesia, there are some different opinions. We think that it must be discussed from the point of view of the effect of the dental treatment on the patient versus the danger of the anesthesia. In this case, we found that, under general anesthesia, stress of the patient was reduced, and the dental treatment was able to be performed smoothly and safely.

Prevalence of Cryptosporidium infection among domestic cats in the Tokyo Metropolitan District, Japan.

From February, 1988 to February, 1989, an epidemiological survey for feline cryptosporidiosis was carried out on domestic cats that had been committed to the Tokyo Metropolitan Animal Protection Center by owners residing in the Tokyo Metropolitan District. Of a total of 608 cats, 23 (3.8%) were found to have Cryptosporidium oocysts. The percentage of cats positive for the infection ranged from 0 to 13.6% by month. The infection was found to be more common in "baby" and "young" cats than in "middle" and "old" ones. No significant difference in incidence was shown between the sexes. In each positive cat, no apparent clinical symptom such as diarrhea was found, and the size of the oocysts measured from 4.3 to 4.7 microns in diameter (small type).

[Case report on the Ehlers-Danlos syndrome].

The Ehlers-Danlos syndrome is a hereditary disease of the connective tissue, characterized by tendencies of hyperextensibility and fragility of the skin, and hypermobility of the joints. We are reporting in this paper on a 4 year, 4 month old boy with the Ehlers-Danlos syndrome, Type III. The following findings were obtained: 1) In addition to the three main symptoms the complications of exotropia, low position of the auricle, defect of the right musculuspectoralis major, right double thumbs, dislocation of hip joint, right inguinal hernia, right undescended testis and pes planovalgus were found. 2) Based on histopathological tissue examination, hypoplasia of the collagen fibers and the elastic fibers was found. 3) The occlusion was edge to edge, the plate was high arched and the upper and the lower dental arches were contracted. 4) The mesiodistal lengths of all primary teeth and the lengths and widths of the dental arches were smaller than the standard size. 5) The orthopantomo and dental x-rays revealed a tendency toward late calcification of the deciduous teeth and permanent teeth germs, and the stricture of the root canals. 6) According to x-ray cephalometric analysis, retardation of the growth of the upper face as well as the maxilla and mandibula was found.

[A case of Prader-Willi syndrome].

Prader-Willi syndrome is characterized by hypotonia, hypomentia, hypogonadism and obesity. A case of a 10-year-5-month-old girl who was diagnosed to have the typical symptoms associated with the Prader-Willi syndrome is described in the present report. The following are the dental findings. 1) Enamel hypoplasia, crowding over the anterior teeth and narrow dental arch were found. However there was no high palate in the maxilla. 2) The mesio-distal width of the present teeth were small compared with the national average. 3) According to X-ray cephalometric analysis, a retardation of the growth of the maxilla and mandible was found. 4) Because of hypomentia and the difficult management of the patient, dental treatment was performed under general anesthesia.

(Pyridoxylated hemoglobin)-(polyoxyethylene) conjugate solution as blood substitute for normothermic whole body rinse-out.

In order to investigate a new possibility for artificial blood with oxygen-carrying capability to be applied to other than mere supplementation, normothermic whole body rinse-out in which artificial blood deriving from perfluorochemical emulsion, Fluosol-DA 20% (Green Cross Co., Ltd., Osaka, Japan) or stabilized hemoglobin solution, (pyridoxylated hemoglobin)-(polyoxyethylene) conjugate solution (Ajinomoto Co., Ltd., Tokyo, Japan) were used as rinsing fluid for a blood purification experiment. Replacement either with approximately 150 ml/kg of Fluosol-DA or stabilized hemoglobin solution showed effective removal of digoxin at a reduction rate of 96.3% or 92.2%, respectively. However, when Fluosol-DA was used, a certain amount of perfluorochemical should be retrieved by centrifugation to avoid a possible toxic effect on the reticulo-endothelial system. Even though 3 out of 6, and 3 out of 8 dogs, respectively, survived for a long period after the procedure, the experimental dogs were very susceptible to infection.
