Identification of a protein-binding site in the promoter of the human thymidine kinase gene required for the G1-S-regulated transcription.

When quiescent cells are stimulated to enter the cell cycle, the thymidine kinase (TK) gene is transcriptionally activated at the border of G1 and S. In this report we show that the human TK promoter contains multiple protein-binding sites. By site-directed mutagenesis, we identified a protein-binding site on the human TK promoter required for conferring G1-S-regulated transcription to a heterologous promoter and dissociated it functionally from an adjacent protein-binding domain containing an inverted CCAAT motif required for high basal level expression. Substitution-mutation of this site results in constitutive expression of the neo reporter gene in serum-stimulated fibroblasts, as well as in cells arrested in mid-G1 by a temperature-sensitive mutation. The regulatory domains for the human TK promoter exhibit interesting symmetrical features, including a set of CCAAT motifs and sites similar to the novel Yi protein-binding site recently discovered in the mouse TK promoter.

AlF4- and vanadate stimulate chloride secretion in rabbit colon by a Ca(2+)-dependent mechanism.

Stripped rabbit descending colon mucosae were studied in vitro in modified Ussing chambers to determine the effects of AlF4- and vanadate on Cl- transport. Serosal additions of AlF4- and vanadate increase short circuit current (Isc) and tissue conductance, while luminal addition of the agents is ineffective. Addition of aluminium potentiates the effect of NaF on Isc. AlF4- and vanadate increase serosal-to-mucosal flux of 36Cl without affecting mucosal-to-serosal flux. The effects of these agents on Isc are markedly inhibited by serosal addition of bumetanide and depend on the presence of Na+ in the serosal bathing solution. The effects of AlF4- and vanadate on Isc are dependent on the presence of Ca2+ in the bathing solution, and are completely inhibited by indomethacin, but the effect of forskolin is not affected by the removal of Ca2+ from the bathing solution and the addition of indomethacin. AlF4- and vanadate significantly increase the level of inositol phosphate metabolites. The results indicate that AlF4- and vanadate increase Cl- secretion in the rabbit colon via an increase in prostaglandin synthesis which is mediated by the increase of intracellular Ca2+ concentrations.

Developmental isolation and subsequent adult behavior of Drosophila paulistorum. I. Survey of the six semispecies.

We present results deriving from the inception of an investigation (involving 4,000 scored matings) into the development of discriminatory behavior in Drosophila paulistorum. We examined two complementary situations for their effects on mating behavior: physical isolation (seclusion) beginning at each of six developmental stages and exposure to heterosemispecifics during all preadult stages (mixed culturing). Preadult seclusion at certain stages decreased sexual isolation between three of four semispecific pairs. Seclusion effect, or lack thereof, varied with semispecies and with the stage at which seclusion was imposed: the earlier the stage at which seclusion was initiated, the more drastic the effect. In contrast, mixed culturing increased sexual isolation between the one pair of semispecies that was tested.

Uptake of uric acid and p-aminohippurate (PAH) by renal cortical slices of various mammals.

1. Accumulation of uric acid and PAH was measured in renal cortical slices of various mammalian species. 2. The slice to medium ratio of uric acid was above unity in the rabbit, guinea pig, pig and cow, suggesting an active accumulation of uric acid, while it was near or below unity in the rat and mongrel dog. 3. Uric acid uptake in the rabbit, guinea pig and cow was significantly inhibited by PAH. 4. Uric acid was a potent inhibitor of PAH uptake in the rabbit, guinea pig, dog and pig, but much less potent in the rat and cow. 5. Kinetic analysis showed that uric acid inhibited PAH uptake in a competitive manner in all species studied except for the cow showing a noncompetitive type. 6. These results indicate that uric acid and PAH share a common transport mechanism at the basolateral membrane of the rabbit, guinea pig and pig.

Urate excretion by the cat kidney.

1. The renal handling of urate by the cat kidney was investigated during continuous infusion of urate. 2. Fractional urate excretion (FE(UA)) in cats was 0.57 +/- 0.04, indicating net reabsorption of urate. In contrast, FE(UA) in rabbits was 1.76 +/- 0.08, reflecting net secretion of urate. 3. Fractional PAH excretion (FE(PAH)) was 3.94 +/- 0.26 in cats and 4.12 +/- 0.76 in rabbits, showing net secretion in both species. 4. FE(UA) in cats was dependent on urine flow, but was independent of plasma urate concentration. 5. The urate excretion in cats was enhanced by probenecid, but was insensitive to PAH and PZA. 6. The PAH excretion in cats was reduced by probenecid, but was unaltered by urate and PZA. 7. These results indicate that urate is handled in the cat kidney by a unique transport system which is distinct from that for organic anions.

High molecular weight proteins in the nematode C. elegans bind [3H]ryanodine and form a large conductance channel.

Single-channel properties of a polypeptide fraction from the nematode Caenorhabditis elegans highly enriched in binding sites were studied in planar bilayers. [3H]Ryanodine binding sites were purified by sucrose gradient centrifugation of C. elegans microsomes solubilized in CHAPS detergent. The highest [3H]ryanodine binding activity sedimented at approximately 18% sucrose (wt/vol), and was composed of a major polypeptide with a M(r) of 360,000 and a minor polypeptide with a M(r) of 170,000. The ryanodine-binding polypeptide(s) formed a Ca(2+)-permeable channel with a permeability ratio P(divalent)/P(monovalent) = 4 and two conductance states of 215 pS and 78 pS in 0.25 M KCl. Ryanodine locked the channel in the 78 pS state and inhibited transitions between the 215 pS and 78 pS states. These data demonstrated the presence of a ryanodine receptor in C. elegans with functional properties comparable to those described in mammalian muscle.

Acquired tufted angioma associated with pregnancy.

Acquired tufted angioma is a distinctive condition that is different from other types of acquired vascular proliferation. Despite the progressive spread of these angiomas, they appear to be benign, and malignant change has not been encountered. We describe a case of recurrent acquired tufted angioma associated with pregnancy, an association which has not been previously recorded.

Dicarboxylate transport in renal basolateral and brush-border membrane vesicles.

Characteristics of succinate transport were determined in basolateral and brush-border membrane vesicles (BLMV and BBMV, respectively) isolated in parallel from rabbit renal cortex. The uptake of succinate was markedly stimulated by the imposition of an inwardly directed Na+ gradient, showing an "overshoot" phenomenon in both membrane preparations. The stimulation of succinate uptake by an inwardly directed Na+ gradient was not significantly affected by pH clamp or inhibition of Na(+)-H+ exchange. The Na(+)-dependent and -independent succinate uptakes were not stimulated by an outwardly directed pH gradient. The Na dependence of succinate uptake exhibited sigmoidal kinetics, with Hill coefficients of 2.17 and 2.38 in BLMV and BBMV, respectively. The Na(+)-dependent succinate uptake by BLMV and BBMV was stimulated by a valinomycin-induced inside-negative potential. The Na(+)-dependent succinate uptake by BLMV and BBMV followed a simple Michaelis-Menten kinetics, with an apparent Km of 22.20 +/- 4.08 and 71.52 +/- 0.14 microM and a Vmax of 39.0 +/- 3.72 and 70.20 +/- 0.96 nmol/(mg.min), respectively. The substrate specificity and the inhibitor sensitivity of the succinate transport system appeared to be very similar in both membranes. These results indicate that both the renal brush-border and basolateral membranes possess the Na(+)-dependent dicarboxylate transport system with very similar properties but with different substrate affinity and transport capacity.

Effect of probenecid on tetraethyl ammonium (TEA) transport across basolateral membrane of rabbit proximal tubule.

The effect of probenecid on the transport of tetraethylammonium (TEA) was investigated in rabbit reanal cortical slices in an attempt to ascertain the interaction of organic anion with the organic cation transport system in proximal tubule. Probenecid reversibly inhibited TEA uptake by cortical slices in a dose-dependent manner over the concentration range of 1 and 5 mM. The efflux of TEA was not affected by the presence of 3 mM probenecid. Kinetic analysis indicated that probenecid decreased Vmax without a significant change in Km. Probenecid inhibited significantly tissue oxygen consumption at concentrations of 3 and 5 mM. However, probenecid did not significantly reduce TEA uptake in brush border and basolateral membrane vesicles prepared from renal cortex even at higher concentration of 10 mM. These results indicate that probenecid reduces TEA uptake in cortical slices by inhibiting the tissue metabolism rather than by the interaction with the organic cation transporter.

Glisoprenins, new inhibitors of acyl-CoA: cholesterol acyltransferase produced by Gliocladium sp. FO-1513. I. Production, isolation and physico-chemical and biological properties.

Gliocladium sp. FO-1513 was found to produce novel inhibitors of acyl-CoA: cholesterol acyltransferase (ACAT). Two active compounds, designated glisoprenins A and B, were isolated from the culture broth of the producing strain by a conventional method. The IC50 values of glisoprenins A and B for ACAT activity were 46 and 61 microM in an enzyme assay using rat liver microsomes, and 1.2 and 0.57 microM in a J774 macrophage assay, respectively.

New cyclodepsipeptides, enniatins D, E and F produced by Fusarium sp. FO-1305.

New cyclodepsipeptides named enniatins D, E and F were isolated from the culture broth of Fusarium sp. FO-1305 as inhibitors of acyl-CoA:cholesterol acyltransferase (ACAT). The respective structures of enniatins D, E and F were determined to be cyclo[D-alpha-hydroxyisovaleryl(D-Hiv)-L-N-methylleucinyl(L- Me-Leu)-D-Hiv-L-N-methylvalinyl(L-Me-Val)-D-Hiv-L-Me-Val], a mixture of cyclo-[D-Hiv-L-Me-Leu-D-Hiv-L-N-methylisoleucinyl(L-Me-Ile)-D-Hiv- L-Me-Val] and cyclo(D-Hiv-L-Me-Ile-D-Hiv-L-Me-Leu-D-Hiv-L-Me-Val), and cyclo(D-Hiv-L-Me-Leu-D-Hiv-L-Me-Ile-D-Hiv-L-Me-Ile) by spectral analyses and chemical degradation. The IC50 values of enniatins D, E and F for ACAT activity in an enzyme assay using rat liver microsomes were calculated to be 87, 57 and 40 microM, respectively.

Emphysematous pyelonephritis.

We herein describe 2 cases of emphysematous pyelonephritis, which can be added to the 42 cases previously reported. Both female patients were diabetic and had Escherichia coli infections. Both had ureteral obstruction and underwent nephrectomy to survive septic and/or azotemic crises. Timely surgical intervention after unsuccessful antibiotics and conventional medical measurements is life-saving.

Primary structure of the marmoset (Saguinus fusicollis) hemoglobin. I. Use of tryptic maleylated peptides in the solubilization and sequence elucidation of the alpha- and beta-chains.

The primary structure of adult marmoset hemoglobin has been determined. The alpha- and beta-chains of HbA were separated on a CM23 column in 8 M urea using a sodium phosphate gradient. Tryptic digest of the alpha- and beta-chains were fractionated on a Dowex 50W-X2 column using a pH and pyridine acetate gradient. Large peptide fragments were obtained by the cyanogen bromide cleavage of the alpha- and beta-chains, as well as by tryptic digestion of the maleylated alpha- and beta-chains. The sequence was derived from the amino acid compositions and sequences of the individual tryptic peptide, automated sequence determination of intact alpha- and beta-chains, as well as automated sequence determination of cyanogen bromide fragments and tryptic maleylated peptides derived from the alpha- and beta-chains. The complete structure of marmoset adult hemoglobin is closely homologous to that of other primate hemoglobins. The sequence of the marmoset alpha-chain differs from the alpha-chian of human HbA at positions 8, 19, 23, 68, and 116. The beta-chain from marmoset HbA differs from the beta-chain of human HbA at positions 5, 13, 21, 50, 87, and 125.
