Identification of genes differentially expressed in association with acquired cisplatin resistance.

The goal of this study was to identify genes whose mRNA levels are differentially expressed in human cells with acquired cisplatin (cDDP) resistance. Using the parental UMSCC10b head and neck carcinoma cell line and the 5.9-fold cDDP-resistant subline, UMSCC10b/Pt-S15, two suppressive subtraction hybridization (SSH) cDNA libraries were prepared. One library represented mRNAs whose levels were increased in the cDDP resistant variant (the UP library), the other one represented mRNAs whose levels were decreased in the resistant cells (the DOWN library). Arrays constructed with inserts recovered from these libraries were hybridized with SSH products to identify truly differentially expressed elements. A total of 51 cDNA fragments present in the UP library and 16 in the DOWN library met the criteria established for differential expression. The sequences of 87% of these cDNA fragments were identified in Genbank. Among the mRNAs in the UP library that were frequently isolated and that showed high levels of differential expression were cytochrome oxidase I, ribosomal protein 28S, elongation factor 1alpha, alpha-enolase, stathmin, and HSP70. The approach taken in this study permitted identification of many genes never before linked to the cDDP-resistant phenotype.

[Analysis of ultrasonic backscattering microstructure feature of human spleen based on "WD cepstrum"].

A new method "WD cepstrum" for analysing ultrasonic scattered signals based on wavelets decomposition before cepstrum has been developed and then applied to process the backscattered signals of normal and pathological human spleen in vitro. The mean scatterer spacing(MSS) has been estimated. The results show that the MSS data of the two tissues are quite different and "WD cepstrum" is more effective for reflecting the microstructure feature of tissue. The results also illustrate that "WD cepstrum" is an effective method for signal analysis of ultrasonic scattering and characterization of tissue scatterers.

Movement disorders in encephalitis induced by Rhodococcus aurantiacus infection relieved by the administration of L-dopa and anti-T-cell antibodies.

Mice injected with Rhodococcus aurantiacus by the intravenous (i.v.) route show neurological disorders, hemiparesis, vertical headshake and turn-round gait after day 7 postinfection (p.i.). Neurological symptoms caused by i.v. inoculation of R. aurantiacus were relieved by treatment with levodopa (l-dopa). R. aurantiacus was isolated from the brain and was found to be completely eliminated at day 7 p. i. Focal encephalitis was mainly observed in the brain stem, and T cells could be isolated from the brain after day 7 p.i. Administration of both an anti-CD4 monoclonal antibody (mAb) and an anti-CD8 mAb suppressed neurological symptoms. These results suggest that R. aurantiacus induces movement disorders in mice, and that the symptoms are mediated by T cells infiltrating the brain, rather than directly by the bacterium.

[Orbital rim reconstruction with coral porous hydroxyapatite].

OBJECTIVE: To search for the effect and safety of coral porous hydroxyapatite (CHA) implanted material in orbital rim reconstruction. METHODS: 7 cases of orbital rim reconstruction were treated with CHA which is a plate form mass with an interconnected system of canals of approximately 200 microns in size. It has three types: 1.0 cm x 1.5 cm x 5.0 cm, 0.6 cm x 1.5 cm x 5.0 cm and 0.4 cm x 1.5 cm x 5.0 cm. According to the shape of a contralateral healthy orbit, a rubber putty model was made before the operation. An incision was designed at the upper 1/3 of the face for the elimination of wrinkles or a prolonged incision of lower lid bag plastic surgery was made. After the exposure of the defective part of the orbit, a correctly molded CHA tamponade was inserted. RESULTS: The orbital rim reconstruction in 7 cases with orbital defect was successfully treated with CHA. During a follow-up of 3-12 months (average 6.6 months), there was no extrusion or migration of implant. CONCLUSION CHA is a good material for substitution of bone graft and is much better than autogenous tissue of other artificial materials.

Paralysis caused by acute myelitis in Theiler's murine encephalomyelitis virus strain GD VII infection is induced by CD4+ lymphocytes infiltrating the spinal cord.

Intravenous infection by Theiler's murine encephalomyelitis virus strain GD VII causes acute encephalomyelitis and paralysis in infected mice. However, nude mice and cyclophosphamide-treated ddY mice did not show paralysis when they were able to survive until day 20 post-infection (p.i.). Of ddY mice infected with 5 x 10(7) p.f.u./mouse, 70-80% showed symptoms of paralysis on day 20 p.i. The viral titres in the brain and spinal cord in infected mice were not significantly different between paralytic and non-paralytic mice. In all of the mice infected with the virus, CD4+ lymphocytes and CD8+ lymphocytes had infiltrated the brain on days 10, 12, 14 and 20 p.i. as demonstrated by flow cytometric analysis. In contrast, few T lymphocytes infiltrated the spinal cord in the non-paralytic mice. Administration of an anti-CD4 monoclonal antibody (MAb) or anti-T cell receptor-alpha beta MAb on day 6 p.i. inhibited paralysis until day 20 p.i., though 20% of the MAb-treated mice and 80% of the control mice showed paralysis. Administration of anti-CD8 MAb was not effective in the suppression of paralysis. The MAb treatment did not significantly augment viral replication in the spinal cord, although the viral titres in the brain of the MAb-treated mice increased significantly. After the transfer of spleen cells from infected C3H mice, the recipient mice infected with a small amount of the virus showed paralysis, though uninfected mice did not. This transfer could be blocked by CD4+ lymphocyte depletion of the donor mice. These results indicate that paralysis caused by acute myelitis in Theiler's virus strain GD VII infection is induced by CD4+ lymphocytes infiltrating the spinal cord.

Chemical synthesis, cloning and expression of human epidermal growth factor gene in Saccharomyces cerevisiae.

A gene coding for human epidermal growth factor (hEGF) has been chemically synthesized by solid-phase phosphoramidite method. The 173 base-pair synthetic DNA duplex consists of a structural gene encoding hEGF, a stop codon TGA at 3' end and some convenient restriction sites at both ends of the gene. The synthesis of the gene involved enzymatic joining of 8 oligonucleotides to form a DNA duplex which was cloned into vector M13mp18. The recombinant colonies were identified by dot hybridization and restriction enzyme digestion. Its accuracy was confirmed by DNA sequence analysis. The hEGF DNA was inserted into yeast secretory vector YFD59. The resulting expression plasmid YFD104 was introduced into yeast Saccharomyces cerevisiae. The binding assay showed that the yeast transformants could express and secrete hEGF.

[Blepharocoloboma repaired with preserved scleral homograft].

40 cases of blepharocoloboma of various causes and sizes were repaired with preserved sclera and satisfactory results. The effective rate was 100%. Histological and immunological studies of scleral homografts in rabbits showed that slight cellular immunoreactions occurred in the early postoperative period; however, no graft was rejected, but gradually replaced by connective tissue of the recipient to become a structural and functional part of the eyelid.

[Synthesis and cardiovascular activities of peptidyl dopamine derivatives].

Amino acids and small peptides with different steric configurations and lipophilicities were appended to dopamine. Eighteen compounds have been synthesized. The attachment of D-amino acids or N-methyl amino acids onto the dopamine molecule caused a marked decrease in cardiovascular activity by intravenous injection, while the introduction of lipophilic amino acids caused a marked increase in myocardial contractility and blood pressure in anesthetized dogs. The durations of action were also prolonged.

The use of short-term increments in weight to monitor growth in infancy.

The use of weight measurements at regular intervals of 2-8 weeks in infancy is considered in the context of detecting slowing-down of growth due to nutritional or other causes. Mixed longitudinal data from 427 children in the Sudan covering the first year of life are presented. Using a simple model it is shown that measurements at short intervals are quicker to detect growth deficiencies, but at the cost of a substantial increase in the false alarm rate.
