Sclerosing stromal tumor of the ovary: US, MR, and dynamic MR findings.

The US, MR, and dynamic MR findings in four patients with sclerosing stromal tumor of the ovary are reported. US showed a tumor with multilocular cystic components and irregularly thickened septa and tumor walls or a solid tumor including several small cystic components. On T2-weighted MR images, signal intensities of the cystic components were high and those of the solid components were inhomogeneous, ranging from intermediate-high to high. Dynamic MRI demonstrated marked early enhancement of the solid components.

Stimulation of motility of human renal cell carcinoma by SPARC/Osteonectin/BM-40 associated with type IV collagen.

SPARC is known to be important in development and tissue remodelling. Here, we examined the effects of SPARC (secreted protein, acidic and rich in cysteine; osteonectin) derived from a rat osteosarcoma cell line on migration of renal cell carcinoma (RCC) by a Boyden chamber assay. YCR RCC cells migrated through type IV collagen-coated filters without stimuli (basal level). SPARC in the lower compartment stimulated chemotactic activity to 120% of the basal level, whereas premixing of YCR with purified SPARC before inoculation reduced their migration to 72% of the basal level. Furthermore, SPARC mixed with type IV collagen more efficiently stimulated their migration in a concentration-dependent manner (up to 170% of the basal level). This suggests that SPARC bound to type IV collagen plays a role in tumor invasion.

Platelet-derived endothelial cell growth factor is not implicated in progression of cervical cancer.

We examined the expression of platelet-derived endothelial cell growth factor (PD-ECGF) mRNAs in 47 invasive cervical cancer tissues by semi-quantitative RT-PCR and addressed its association with clinicopathological features including microvessel density. Squamous cell carcinomas expressed higher levels of PD-ECGF mRNA than non-squamous cell carcinomas (P=0.014). We found no association between FIGO stage and levels of PD-ECGF mRNA. A subset of 31 patients with stage Ib-II cervical cancer underwent radical hysterectomy and pelvic lymphadenectomy. There were no significant differences in PD-ECGF mRNA levels with respect to tumor size, degree of stromal invasion, lymphvascular space involvement, parametrial involvement, vaginal involvement or lymph node metastasis among these patients. There was no correlation between microvessel density and the levels of PD-ECGF mRNA. These findings suggest that PD-ECGF expression is not associated with progression and metastasis of cervical cancer.

Primary choroid plexus carcinoma producing carbohydrate antigen 19-9.

An autopsy case of primary choroid plexus adenocarcinoma arising in a 40-year-old female, who was associated with a high serum level of a carbohydrate antigen 19-9 (CA19-9), is herein presented. After a subtotal removal of a tumor in the left lateral ventricle, the serum level of CA19-9 decreased rapidly, and immunohistochemical examinations of tumor tissue specimens obtained at surgery revealed intense reactivity for CA19-9. The present case may be the first example in which a primary choroid plexus carcinoma was shown to produce CA19-9.

Macrophage infiltration and angiogenesis in endometrial cancer.

The aim of this study was to determine whether tumor-associated macrophages (TAMs) infiltration correlates with clinicopathological factors including microvessel, counts and clinical outcome in endometrial cancer. Overall 56 out of the 109 endometrial cancers (51.4%) expressed distinct tumor-associated macrophages infiltration in their tumor stroma. Tumor-associated macrophages infiltration was significantly high in tumors with deep myometrial invasion, high grade and elderly patients. Microvessel counts strongly correlated with tumor-associated macrophages infiltration in tumor stroma (p = 0.0002). However, tumor-associated macrophages infiltration was not a prognostic factor. In conclusion, tumor-associated macrophages may play a crucial role in the promotion of angiogenesis, but can not be used to prodict prognosis of patients with endometrial cancer.

Development of a three-dimensional color Doppler system.

An unprecedented assembly of a three-dimensional (3D) color Doppler system was arranged for the purpose of producing and displaying precisely calculated and recorded 3D color flow mapping images of fetal and umbilical vessels. Data accuracy and quality were enhanced by a computer controlled probe positioner. Scanning was across any of three perpendicular planes to collect serial slice images in any position and interval width. The series of parallel images was digitized in a color imaging processor by a 3D reconstruction program. Analyzed data were transferred to work stations for immediate processing into 3D form and display. The resulting 3D display was a solid model image of umbilical vessels which could be rotated in real time. In addition to the clear information about the vessel's structure, hemodynamics were apparent.

[Increased HDL-cholesterol level improves the prognosis in patients with coronary vasospasm].

Cardiovascular events were analyzed in the subset of 49 consecutive patients with vasospastic coronary artery disease who underwent diagnostic coronary arteriography from December, 1987 to March, 1991 to confirm coronary artery spasm during anginal attacks. During the follow-up period (30 +/- 14 months, mean +/- SEM), seven patients had cardiovascular accidents including acute myocardial infarction, unstable angina, and stroke (group A), while the remaining 42 patients were event-free (group B). Current smokers at the end of the follow-up period were more common in group A (71%) than in group B (12%) (p < 0.01). Serum total-cholesterol, low density lipoprotein cholesterol, and triglyceride levels were not significantly different between the two groups before or after the follow-up period. The baseline high-density lipoprotein cholesterol (HDL-C) level also did not differ between group A (32.5 +/- 8.3 mg/dl) and group B (41.0 +/- 12.9 mg/dl). However, the HDL-C level significantly increased during the follow-up period in group B (delta HDL-C 6.1 +/- 9.4 mg/dl) (p < 0.01), but not in group A (delta HDL-C -3.3 +/- 7.2 mg/dl). The HDL-C level at the end of the follow-up period in group A (29.2 +/- 9.0 mg/dl) was significantly lower than in group B (47.1 +/- 11.5 mg/dl) (p < 0.01). Cardiovascular accidents were significantly more common in current smokers (50%) (p < 0.01) than in current nonsmokers (5%) after the follow-up.(ABSTRACT TRUNCATED AT 250 WORDS)

Visualization of the metastatic process by green fluorescent protein expression.

We demonstrate here the visualization of the cancer metastatic process in live tissue in vivo by green fluorescent protein (GFP) expression. The human lung adenocarcinoma cell-line Anip 973 was transfected with the humanized GFP-S65T cDNA and stable high-level GFP-expressing transfectants were established. GFP transfectants were initially inoculated subcutaneously in nude mice. Five weeks after transplantation, when the tumor had reached 1.2 cm in diameter, fragments of subcutaneously tumor were implanted onto the visceral pleura of nude mice by surgical orthtopic implantation (SOI) as a spontenous metastatic model. GFP expressing cells were injected intravenously in nude mice as an experimental hematogenous metastasis model. Mice were sacrificed four and eight weeks after treatment. At eight weeks, SOI-treated mice had lymphogenous (3/4 mice) and direct seeding (3/4) metastasis in the pulmonary hilum, cervical lymph nodes, the mediastinum and contralateral pleural cavity as detected by GFP expression in live tissue. All intravenously injected mice had metastases in the lung (4/4) and some of them had metastases in the brain (2/4) and other organs (1/4) as detected by GFP expression in fresh tissue. Some of the lung metastases produced by intravenous injection remained as dormant small colonies even eight weeks after treatment. These different metastatic patterns after SOI and intravenous injection visualized by GFP expression indicates that initial steps of the metastatic cascade influence the subsequent progression of metastasis.
