A new method of collection of nasal secretion, by dilution with condensed mositure of the exhaled air, is described. This method may be defined as a sefl-administered nasal lavage. The method permits quantitative determination of pH, relative viscosity, and electrolyte and protein composition. Concentrations of sodium, potassium, calcium, total protein, IgA, IgG, IgM, secretory compoenent and polysaccharide are reported for normal subjects. Significant correlations were noted between the relative viscosity and variables: dry weight, total protein and calcium.

The incidence of conjugative R plasmids in multiple drug-resistant strains of gram-negative bacteria isolated in 1973 from patients in a 700-bed general hospital in Tokyo and some properties of the R plasmids isolated are described. Conjugative R plasmids were found in 52 of the 96 strains (54%), from which 74 R plasmids were demonstrated. It is remarkable that the isolation frequency of R plasmids mediating quadruple- or five-drug resistance was rather low, and the complete pattern of multiple resistance of the original isolates was only rarely transferred by conjugation. These results revealed the existing state of the distribution of R plasmids among hospital strains with multiple drug-resistance.

Spectinomycin-resistant (Spcr) mutants of Escherichia coli were isolated from nutrient agar plates containing 20% sucrose and 100 mug of spectinomycin per ml. About one-third of the Spcr mutants thus obtained were sucrose dependent (Sucd) and were classified into two types: I, those unable to grow on sucrose-free medium in the presence of spectinomycin; and II, those unable to grow on sucrose-free medium irrespective of the presence of spectinomycin. Most of these mutants were hypersensitive to antibiotics, dyes, and detergents and were abnormal in cell morphology, suggesting changes in cell envelopes. Reversion experiments indicated that the sucrose-dependent spectinomycin resistance and hypersensitivity to various chemicals were not independently induced properties. The Sucd-Spcr mutations of type I mutants were transducible by phage P1 and were mapped at the strA-aroE region.

Six representative strains of staphylococci were selected on the basis of DNase activity and inoculated into the mice to study the correlation of the presence of this enzyme to the virulence of the staphylococci. Two strains of staphylococci among them produced DNase but not the free coagulase, and one of these two strains had an obvious virulence against the mice. The results of experiments suggest that there are at least certain strains which may be designated as Staphylococcus aureus among these DNase-producing staphylococci producing no free coagulase. This is an additional evidence that this enzyme activity should be adopted as a criterion to distinguish from the other S. aureus which has lost the free coagulase activity. In this sense, the staphylococci producing neither free coagulase nor DNase are after all Staphylococcus epidermidis, regardless of their ability to ferment mannitol or not.
