[Separation of thoracopagus conjoined twins].

OBJECTIVE: To sam up the experience in diagnosis and separation of a thoracopagus conjoined twins. METHODS: Thoracopagus twin boys were admitted for evaluating possible separation and repair at age of 21 days. X-rays, CT, MRI, and ultrasound scan of torso showed separate and normal gastrointestinal and biliary tracts, little shared vasculature, but shared pericardium, diaphragm and liver parenchyma. The chest junctions were at the sternums and the most inferior two pairs of the ribs. One twin (twin A) had a protruding heart with VSD and ASD. RESULTS: The infants were separated successfully at age of 45 days. The chest defect of twin A was closed with aid of a polyester pericardial patch graft and a silicon supporter. Managing anesthesia met some difficulty in understanding of the anatomical and physiological consequences of the complex anomaly. They are had brain edema and multiple system organ failures after separation, and were cured under intensive care. They are alive and well 6 months after surgery. CONCLUSIONS: Careful preoperative evaluation proper anesthesia and surgical technique, and postoperative care are essential to the successful separation of the conjoined twins.

Structure of Tetrahymena GCN5 bound to coenzyme A and a histone H3 peptide.

Gene activation is a highly regulated process that requires the coordinated action of proteins to relieve chromatin repression and to promote transcriptional activation. Nuclear histone acetyltransferase (HAT) enzymes provide a mechanistic link between chromatin destabilization and gene activation by acetylating the epsilon-amino group of specific lysine residues within the aminoterminal tails of core histones to facilitate access to DNA by transcriptional activators. Here we report the high-resolution crystal structure of the HAT domain of Tetrahymena GCN5 (tGCN5) bound with both its physiologically relevant ligands, coenzyme A (CoA) and a histone H3 peptide, and the structures of nascent tGCN5 and a tGCN5/acetyl-CoA complex. Our structural data reveal histone-binding specificity for a random-coil structure containing a G-K-X-P recognition sequence, and show that CoA is essential for reorienting the enzyme for histone binding. Catalysis appears to involve water-mediated proton extraction from the substrate lysine by a glutamic acid general base and a backbone amide that stabilizes the transition-state reaction intermediate. Comparison with related N-acetyltransferases indicates a conserved structural framework for CoA binding and catalysis, and structural variability in regions associated with substrate-specific binding.

[Surface enhanced Raman scattering of thiourea in acidic solution].

Surface enhanced Raman scattering of thiourea adsorbed on the plated silver surface in an acidic solution has been studied. The results show that the thiourea molecules are inclined to adsorbed on the silver surface via sulphur. When the concentration of HCl in the solution is increased, the plane of the thiourea molecule deviates more from the normal line of the silver surface. The experimental results also suggest that the rather strong chemical enhanced mechanism would exist in this system.

Structures of SAP-1 bound to DNA targets from the E74 and c-fos promoters: insights into DNA sequence discrimination by Ets proteins.

SAP-1 is a member of the Ets transcription factors and cooperates with SRF protein to activate transcription of the c-fos protooncogene. The crystal structures of the conserved ETS domain of SAP-1 bound to DNA sequences from the E74 and c-fos promoters reveal that a set of conserved residues contact a GGA core DNA sequence. Discrimination for sequences outside this core is mediated by DNA contacts from conserved and nonconserved protein residues and sequence-dependent DNA structural properties characteristic of A-form DNA structure. Comparison with the related PU.1/DNA and GABPalpha/beta/DNA complexes provides general insights into DNA discrimination between Ets proteins. Modeling studies of a SAP-1/SRF/DNA complex suggest that SRF may modulate SAP-1 binding to DNA by interacting with its ETS domain.

[Discovery of a novel reassortant H1N2 influenza virus].

Three strains of influenza A virus were isolated from patients suffering from influenza like disease in Railway Hygiene and Anti-epidemic Station of Taiyuan in January 1996. The identification results by serological method indicated that the isolates were different from H1N2 subtype of influenza A virus isolated from men in 1989 and 1992 and that their HA antigenicity was similar to that of A/RP/8/34 (H1N1) virus, but distinguishable from that of influenza A (H1N1) virus circulating in mans recently. The comparison of migration patterns of the RNA among the isolates, the A/PR//34 (H1N1) and the A/Wuhan/359/95 (H3N2) viruses revealed that the migration patterns of RNA 1-4 segments of the isolates were similar to those of A/PR/8/34 (H1N1) virus, but those of RNA 5-6 segments were indistinguishable from those of A/Wuhan/359/95 (H3N2) strain. However, the migration patterns of RNA 7-8 segments of the isolates were not only different from those of A/PR/8/34 (H1N1) virus, but also distingrasgable from those of A/Wuhan/359/95 (H3N2) strain. Therefore, it could be considered that the isolates were a novel reassortant H1N2 influenza A virus.

[Studies on epidemic hemorrhagic fever among rats in Jianghan Plian, Hubei Province, China].

Capture of rats by digging their cavities, identification of rat strains, sex and age, examination of their skin cut or damage, and detection of antigen and antibody of epidemic hemorrhagic fever virus (EHFV) were conducted in epidemic areas of epidemic hemorrhagic fever (EHF) in Jinaghan Plain, Hubei Province, to study prevalence of EHFV infection in rats. Results showed that Apodemus agarius and Rattus norvegius accounted for 69.7% and 23.6% of the total rats captured, respectively, their infection rates of EHFV were 21.8% and 30.9%, respectively. Infection rate of EHFV was significantly higher in adult rats than in sub-adult and young ones, with prevalence of 40.8%, 17.0% and 12.4%, respectively. Infection rate of EHFV was 45.9% in rats with skin cut, significantly higher than in those without it (13.9%). There was an obvious litter-clustering phenomenon in infected rats, but their litters distributed in space randomly. Serotyping of infected rats showed that Apodemus agarius mainly was in a wild rat pattern, Rattus norvegius in a domestic rat pattern, and a small number of rats were in undefined patterns. It indicated that Jinaghan Plain was a mixed epidemic area of EHF with domestic and wild rat patterns and with Apodemus agarius and Rattus norvegius as main reservoir host, and age, skin cut, and close contact with them were important factors contributing to transmit it.

[Differential expression of HSU17714 gene in colorectal cancer and normal colonic mucosa].

HSU17714 is a new gene discovered by subtractive hybridization. Its cDNA is 2,932 bp long and the gene is located at human chromosome 22 as defined by fluorescence in- situ hybridization combined fluorescence R-banding technique. Northern blot and RNA dot blot analyses showed that the expression level of HSU17714 gene was significantly lower in 15 cases of colorectal cancer than that in adjacent normal colonic mucosa. The defective expression of HSU17714 in colorectal cancer suggests that it is a new candidate tumor suppressor gene.

Biochemical complementation of chalcone synthase mutants defines a role for flavonols in functional pollen.

Chalcone synthase catalyzes the initial step of that branch of the phenylpropanoid pathway that leads to flavonoids. A lack of chalcone synthase activity has a pleiotropic effect in maize and petunia mutants: pollen fertility as well as flavonoid synthesis is disrupted. Both maize and petunia mutants are self-sterile due to a failure to produce a functional pollen tube. The finding that the mutant pollen is partially functional on wild-type stigmas led to the isolation and identification of kaempferol as a pollen germination-inducing constituent in wild-type petunia stigma extracts. We show that adding micromolar quantities of kaempferol to the germination medium or to the stigma at pollination is sufficient to restore normal pollen germination and tube growth in vitro and full seed set in vivo. Further we show that the rescue ability resides in particular structural features of a single class of compounds, the flavonol aglycones. This finding identifies another constituent of plant reproduction and suggests that addition or removal of the flavonol signal during pollen germination and tube growth provides a feasible way to control plant fertility.

[Preliminary report of 10 cases of anterior segment eyeball reconstruction].

We performed anterior segment reconstruction by total penetrating Keratoplasty with scleral rim in 10 patients who had infectious ulceration with total corneal necrosis and perforation and prolapse of intraocular contents of the severely damaged 10 eyes, 9 were salvaged and retained a semi-transparent corneal graft with improvement in vision for 6 months or longer. It suggested that, for eyes with severe corneal damage that would otherwise be enucleated, anterior segment reconstruction by corneoscleral transplantation is of value.

Recording pH method of characterizing composition and monitoring dissolution profile of an anhydride-acid copolymer and its salt derivatives.

A sensitive potentiometric monitoring method was developed that permits the continuous measurement of the disolution profiles of methyl vinyl ether-maleic anhydride-acid copolymers and salt derivatives. Three distinct rate periods were observed in the dissolution rate of the anhydride copolymer, expressed as percent anhydride dissolved, was independent of sample weight over the weight range studied. The acid form of the copolymer showed only one dissolution rate period, with dissolution being very rapid. The rapid initial pH decrease observed during the first stage of dissolution for a series of anhydride-acid copolymer powder samples correlated closely with the anhydride-acid ratio, permitting chemical characterization of the copolymer functionality simultaneously with the analysis of dissolution profiles. Similarly, the extent of copolymer alkaline salt conversion was inversely proportional to the initial maximum pH increase observed during the first stage of dissolution of these salts. Mechanisms of dissolution of copolymer powder materials are discussed and compared to the dissolution of compressed disks and films reported previously.
