Synthesis and antitumor activity of ester-modified analogues of bengamide B.

Bengamide B, a novel sponge-derived marine natural product with broad spectrum antitumor activity, was not suitable for further preclinical development because of its difficult synthesis and very poor water solubility. Bengamide B produced a 31% T/C at its solubility-limited maximum intravenous dose of 33 micromol/kg in MDA-MB-435 breast carcinoma implanted subcutaneously as a xenograft in nude mice. Compound 8a, a bengamide B analogue with three structural changes (t-Bu alkene substituent, unsubstituted lactam nitrogen, and inverted lactam 5'-myristoyloxy group), was as potent as bengamide B in vitro and more efficacious than bengamide B in vivo. A series of ester-modified analogues based on 8a were synthesized and tested in vitro and in vivo (MDA-MB-435). The cyclohexyl- and phenethyl-substituted esters, 8c and 8g, respectively, had in vitro and in vivo activities similar to that of 8a and enhanced water solubility (ca. 1 mg/mL). Consequently, 8c and 8g were tested in the MDA-MB-435 xenograft model at 100 micromol/kg and produced 29% and 57% tumor regression, respectively.

[The development of a computer-based visual field analyzer].

Visual field is one of the important visual functions; it is the extent of the visual field defect that can be employed in judging whether the visual function is impaired. The rapid achievements in computer technologies do provide an impulse for improvement of visual field detection, making possible the automatic, rapid, accurate, detailed and large-scaled visual field detection. This paper gives a thorough description about development of the visual field analyzer, model TEC-2A, which is based on PC windows platform, Visual Basic software developing tool, ISA peripheral circuits, standard Goldmann visual field half-ball and standard stimulus.

[Resection of caudate lobe of liver: report of 26 cases].

OBJECTIVE: To summarize the experience in caudate lobectomy for 26 cases. METHODS: Caudate lobectomy was performed in 26 cases by Peng's multifunctional operative dissector (PMOD) to transect the liver parenchyma (Curettage and Aspiration technique). Primary liver cancer was noted in 18 cases, hilar cholangiocarcinoma in 4, gallbladder carcinoma in 1, right adrenocortical adenocarcinoma in 1, hepatoangioma in 1 and hepatoangiosarcoma in 1. Five cases underwent isolated caudate lobectomy, 1 combined caudate lobectomy with right trisegmentectomy, 6 combined caudate lobectomy with right hemihepatectomy, 9 combined caudate lobectomy with left hemihepatectomy, and 5 combined caudate lobectomy with segmentectomy. RESULTS: The operations were carried out successfully. Follow up for 2 - 23 months, 1 case died from pulmonary infraction 2 weeks after operation, 1 died from lung metastasis 2 months after operation, 10 died from tumor recurrence or metastasis 3, 11, 12, 15, 15, 15, 15, 19, 30, 30 months after operation respectively. The other 14 cases are still alive for 2 to 18 months. CONCLUSIONS: With PMOD, it is possible to dissect the structure precisely. Therefore the caudate lobe can be isolated from the three porta hepatis without much difficulty. Caudectomy can be performed safely.

The selective inhibition of beta 1 and beta 7 integrin-mediated lymphocyte adhesion by bacitracin.

Integrins play an important role in lymphocyte adhesion to cellular and extracellular components of their microenvironment. The regulation of such adhesion often involves changes in the functional state of the integrins rather than alterations in their expression levels. Although the functional basis for such transitions is unknown, a possible role for disulfide exchange might be postulated based on the observations that integrin function can be activated by bifunctional reducing agents or by Abs that react with areas adjacent to predicted long-range disulfide bonds in integrins. Recently, it has been reported that enzymes that catalyze disulfide exchanges such as protein disulfide isomerase (PDI) are present on the surface of lymphoid cells, raising the possibility that such enzymes might be involved in the control of lymphocyte adhesion. A number of inhibitors of PDI function were examined for their effects on integrin-mediated adherence of T cells. The results did not support role for PDI in the regulation of integrin function, as the inhibitors somatostatin A, tocinoic acid, dithiobisnitrobenzoic acid, and anti-PDI mAb did not interfere with adherence. However, one of the PDI inhibitors, bacitracin, selectively interfered with the beta1 integrin-mediated adherence of lymphoid cells to collagen, fibronectin, laminin, and VCAM-1, and with alpha4beta7-dependent adherence to fibronectin and to VCAM-1. In contrast, alpha(v)beta3- and alpha(L)beta2-mediated adherence were not inhibited. Thus, it appears that bacitracin may be a selective inhibitor of beta1 and beta7 integrin functions by an as yet unknown mechanism.

Expression of N-acetyl-D-galactosamine associated epitope in synovium: a potential marker of glycoprotein production.

OBJECTIVE: To investigate synovial glycoprotein production in situ, a novel monoclonal antibody (Mab), A13D8, was used to evaluate the expression of an epitope containing N-acetyl-D-galactosamine (GalNAc) in normal and pathological synovium. METHODS: Immunohistological and cytochemical analysis of synovial tissue samples was undertaken with single and double staining techniques using the A13D8 Mab, anti-CD68, vascular cell adhesion molecule-1 (VCAM-1), the hyaluronan associated enzyme uridine diphosphoglucose dehydrogenase (UDPGD), and the anti-Golgi Mab SSN/HR-1992. The specificity of the A13D8 Mab was established through blocking studies using carbohydrate residues, including GalNAc and N-acetylglucosamine (GlcNAc). RESULTS: A13D8 is expressed intensely in the cytoplasm of normal type B lining cells, which coexpress VCAM-1 and UDPGD, and is not expressed by CD68+ type A lining cells. In the lining layer of RA synovium, there is a negative correlation between A13D8 expression and the level of lymphocytic infiltration in the sublining areas (r = -0.43, p < 0.001). The endothelium of a subset of venules, typically in lymphocyte-rich aggregates, also stains intensely for A13D8. Pretreatment of the Mab with GalNAc completely eliminates the tissue staining, as well as the 110 kDa band seen on immunoblot, whereas pretreatment of A13D8 with GlcNAc and lactose has no effect. Double staining of HEp-2 cells with A13D8 and the anti-Golgi Mab SSN/HR-1992 reveals co-localization of the A13D8 epitope to the Golgi apparatus. CONCLUSION: Type B synovial lining cells and selected synovial endothelium express GalNAc containing epitope identified by Mab A13D8. Marked reduction in the expression of this epitope in the lining layer of inflamed RA synovium suggests that the synovial production of GalNAc containing glycoproteins, such as mucins, may be altered in this disorder.

[Surgical treatment for iatrogenic damage at the confluence of hepatic duct].

Since 1989, 4 patients with iatrogenic bile duct injuries at the level of bifurcation were treated. Among them, the bifurcation together with both right and left hepatic duct had had wrongly been resected in 2 cases. The bifurcation was partially resected in the other 2 cases. The establishment of a permanent hepaticoentertostomy is essential. Abnormal bile flow would impede surgical effect, hence in the author's opinion, peripheral hepaticoenterostomy such as Longmire's procedure is unsuitable for iatrogenic bile duct injuries. The author's special technique for adequate exposure and accurate dissection of intrahepatic bile ducts is described. Four cases have been followed up for 7 months to 5 years with good result.
