Long-term results of reconstructive surgery for femoro-popliteal arterial occlusive lesions.

A retrospective review of 156 reconstructions performed for femoro-popliteal arterial occlusive lesions in 141 patients was carried out in terms of the reconstructive procedure and the degree of out-flow in the distal vessel. The cumulative patency rate for 95 autogenous vein grafts was 60.7 per cent at 12 years and higher than the 27.4% patency rate for 50 endarterectomies (p less than 0.001). All of 11 synthetic bypass prostheses failed with late thrombosis within 3 years after operation. The cumulative patency rate in cases with good distal run-off vessel and those with fair distal run-off vessel were 77.1% at 11 years and 57.3% at 12 years, respectively, and these values were significantly superior to 10.3% in cases with poor distal run-off vessel. The patency rate for 41 in situ grafts was 72.2%, being superior to 65.5% for 42 free grafts.

Effect of 1 alpha-hydroxycholecalciferol on the skeletal mineral metabolism and bone morphogenesis.

Administered 1 alpha-OH-D3 is hydrolyzed in liver and then converted to active 1 alpha, 25-(OH)2-D3. An increased serum calcium content was observed in rats administered 1 alpha-OH-D3. No calcium and magnesium increase was noticed in cortical bone due to 1 alpha-OH-D3 administration. Morphogenesis of bone and cartilage in implanted bone matrix in a group of 1 alpha-OH-D3 administered host rats seemed relatively increased to a slight extent.

Chemical modification of deltamycins. I. 4''-O-acyl analogs of deltamycins.

Using 4''-O-deacyldeltamycin as a starting material, various 4''-O-acyl derivatives were chemically synthesized by the following scheme: 2'-O-acetylation leads to 4''-O-acylation leads to 2'-O-deacetylation. 4''-O-Deacyldeltamycin is produced during the fermentation of deltamycin producing organisms or can be prepared by the biological deacylation of the deltamycins. 4''-O-Phenylacetyl-4''-O-deacyldeltamycin (PAD) showed good activity against various bacteria and mycoplasma. Among para-substituted PAD derivatives, NPAD (p-nitrophenylacetyl-) and SPAD (p-methylsulfonylphenylacetyl-) showed increased potency as compared to PAD against certain bacteria. The PAD was evaluated by in vivo experiments for plasma levels in mice and dogs and curative activity on the infected mice with Staphylococcus aureus Smith by subcutaneous or oral administration.

[Vitamin K and bone metabolism - on the effect of vitamin K deficiency and gamma-carboxyglutamic acid (author's transl)].

Vitamin K was thought to be primarily involved in the production of clotting factors. The recent findings of gamma-carboxyglutamic acid (Gla) - vitamin K-dependent calcium binding amino acid - in various calcified tissues, however, have prompted suggestions that vitamin K may play an important role in calcification mechanism. The present study was designed to investigate the relation between vitamin K and calcification. Morphological, biomechanical and biochemical analyses were undertaken on bones from vitamin K deficient rats raised on vitamin K deficient diet combined with administration of warfarin for three weeks. The Gla-containing proteins were extracted with EDTA from rat or bovine bones, partially isolated with gel filtration, and amino acid analysis was done. Gross roentgenographical and histological abnormalities were not observed in bones from vitamin K deficient rats in which prothrombin level was reduced to about 20 percent. There were no significant differences in values of Young's modulus, which were measured by three-point bending test, between vitamin K deficient and normal bones. No considerable alteration were noted in the protein, calcium or magnesium content of vitamin K deficient bone. Gla content of vitamin K deficient rat bone (per 1,000 residues of glutamic acid) was reduced by approximately 9% compared with that of normal rat bone. This observation supports the hypothesis that Gla biosynthesis in bone may be dependent on vitamin K. Calcium binding property of bovine bone Gla-containing protein was confirmed by the equilibrium dialysis procedure. Moderate amount of hydroxyproline was present in the Gla-containing proteins extracted from rats or bovine bones. It seems possible that biosynthesis of collagen and Gla protein of bone are somehow related, possibly at the level of procollagen. Further investigation is necessary to isolate Gla protein and clarify the correlation between collagen molecule and Gla protein biosynthesis.

Genetic variance in age-related changes in running performance and growth during adolescence: a longitudinal twin study.

The purpose of this study was to assess an effect of genotype on the patterns of age-related changes in running performance and physical growth during adolescence. The total sample was 14 pairs of male and 25 pairs of female monozygotic (MZ) twins and 19 pairs of male and 15 pairs of female dizygotic (DZ) twins. Performance on the 50-m dash and endurance run (1,500 m for boys and 1,000 m for girls, respectively) and stature and body mass were measured at yearly intervals from 12 to 17 years of age. A principal component analysis was applied to the longitudinal data, and within-pair resemblance in the scores on the first, the second, and the third principal components was compared between MZ and DZ twins. The analysis was conducted with both sexes pooled because of the limited number of twin pairs. The first two principal components explained at least 87% of the total variance, while the third principal component (PC3) explained a smaller portion of the total variance (more than 5%) in the dash and the endurance run. However, the total variance was explained almost entirely (more than 96%) by the first two components in height and mass. The first principal component (PC1), which explained 67-84% of the total variance, was a good indicator of average performance/body size of individuals during the period of observation. The second (PC2) and the third components (PC3) could be considered as indicators of the "shape" of developmental/growth curves, which were not related to inter-individual differences in the average performance/body size. Intraclass correlations (ri) of the scores on PC1 were consistently higher for MZ than for DZ twins, and estimated genetic variance was significant in endurance run, stature and body mass. The situation was the same for the scores on PC2 and/or PC3. However, such a between-genotype difference in ri and a significant genetic variance were not observed in the dash. It was concluded that MZ twins are more similar in the patterns of age-related changes in the performance of endurance run and physical growth during adolescence than DZ twins, suggesting a genetic contribution, while there was no evidence of a genetic contribution in the dash.
