The HLA-B8 antigen is one of the characteristic antigens associated with organ specific autoimmune diseases among Caucasians. Among the Japanese, HLA-B8 positives have been assumed to be extremely rare. By using some HLA-B8 typing sera, a new HLA-B antigen, HLA-Bw4 positive, was found in the Japanese. This new HLA-B antigen, tentatively called HOK-1, showed a phenotype frequency of 3.7% and a gene frequency of 1.9 +/- 0.6%. This antigen has a strong linkage disequilibrium with HLA-Cw1 among the Japanese.
61 patients suffering from chronic pancreatitis were studied in order to clarify the psychosomatic aspects of this disease. They were classified into 5 types, out of which 3 types were examined in detail according to their personality traits and life habits. It was found that the mind-body relationship took the primary form in the psychosomatic and neurotic types, while in the organic type it took the secondary form such as distorted life habits. This finding indicated that the distortion in the personality structure and that of life habits were closely interrelated. The authors also discussed the clinical features of chronic pancreatitis from the standpoint of alexithymia. The patients with the organic type were most alexithymic, which were followed by the psychosomatic type. It was learned that alexithymic problems were reflected in the patients' life habits.
A comparative study was conducted on the psychological features of chronic pancreatitis, peptic ulcer, ulcerative colitis and the irritable colon syndrome by means of psychological tests and interviews. The patients with irritable colon syndrome were found to be the most neurotic of all, whereas those with the definite type of chronic pancreatitis and ulcerative colitis presented prominent alexithymic characteristics. About one half of the patients having peptic ulcer showed alexithymic features. Despite their neurotic tendencies shown on the psychological tests, the patients with the suspicious type of chronic pancreatitis were socially better adjusted than those with irritable colon syndrome. It was also noted that the concept of alexithymia needs to be elucidated by including such factors as over-adaptation to the environment, certain distorted life habits and lack of bodily sensations.
Bilateral radiofrequency lesions were stereotaxically placed in the rostral hypothalamus of four adult female rhesus monkeys. These lesions resulted in extensive destruction of the ventromedial preoptic-anterior hypothalamic area (POA-AHA) and included the suprachiasmatic nucleus as well as, with the exception of one animal, the organum vasculosum of the lamina terminalis. In three of these four animals, gonadotropin surges similar to those observed before surgery were elicited in response to either a spontaneous increment in serum estrogen concentration or an estradiol benzoate injection. This stimulatory action of estradiol on LH and FSH release was not demonstratable in the remaining lesioned animal, but estradiol benzoate injections also failed to elicit a gonadotropin discharge in one of a series of five normal control animals. These findings fail to support the view that destruction of the ventromedial POA-AHA in this species compromises the ability of the hypothalamicohypophysial apparatus to respond to the positive feedback action of estradiol. The diurnal variation in serum cortisol concentration was not interrupted by placement of the lesions in the ventromedial POA-AHA.
To elucidate whether insulin-induced hypoglycemia enhances the release of beta-endorphin in man, plasma extracts obtained from healthy subjects and patients with Graves' disease before and 45 min after insulin injection were subjected to gel chromatography, and the fractions obtained were measured by RIA for beta-endorphin. In four healthy subjects, basal plasma beta-endorphin levels were less than 3 to 3.1 pg/ml, and the levels rose substantially to 47.5 +/- 12.4 pg/ml (mean +/- SE) 45 min after insulin injection. Basal plasma beta-endorphin levels in three hyperthyroid patinets (less than 3 to 3.8 pg/ml) did not seem to be different from those in healthy subjects; however, the rise after insulin injection tended to be higher in cases of hyperthyroidism, with a peak value of 68.5 +/- 9.7 pg/ml. Plasma beta-lipotropin and ACTH levels also rose in parallel with beta-endorphin in response to insulin-induced hypoglycemia in both healthy subjects and hyperthyroid patients. It would thus appear that beta-endorphin, like ACTH or beta-lipotropin, is released in human subjects by hypoglycemic stress.

In the preoptic, infundibular and posterior recesses of the lamprey, Lampetra japonica cerebrospinal fluid (CSF)-contacting neurons are distributed much more abundantly than in higher vertebrates. They are classified into three different types on the basis of their granules size or electron density: type 1 containing round dense granules of large size (180--230 nm in diameter), type 2 containing variously shaped dense granules of medium size (150--200 nm) and type 3 containing variously shaped granules of small size (100--150 nm). The neurons of the third type may be further classified into two subtypes: type 3a possessing dense granules and type 3b showing less dense granules. The intraventricular process of CSF-contacting neurons contains Golgi cisternae that are forming new secretory granules and well developed granular endoplasmic reticulum with dilated cisternae. It is suggested that secretory granules might be formed in the intraventricular process in addition to the perikaryon. Besides the small cytoplasmic protrusions presumably involved in microapocrine secretion, intraventricular processes occupied with many empty granules are detected which might represent a process of diacrine secretion. The intraventricular processes further contain multivesicular bodies which incorporate old or overproduced secretory granules, suggesting a process (crinophagy) and large lipid droplets likely corresponding to the final digestive product. The CSF-contacting neurons of all types are usually supplied with axo-somatic synapses on the perikaryon and subsurface cisternae are sometimes observed beneath the postsynaptic membrane. Axon terminals are also found forming synapses on the intraventricular process of type 2 and 3 neurons.
The pituitary thyrotrophs and thyroid follicular cells of rats were studied by electron microscopy and morphometric analysis at 5, 10, 20, 30 and 60 min after intravenous injection of 200 microgram of thyrotropin releasing hormone (TRH) and at 10 min after injection of 10 microgram TRH. Multiple granule extrusions in a group were often observed around the pituitary thyrotrophs at 5, 10 and 20 min after administration of 200 microgram TRH, as well as at 10 min after injection of 10 microgram TRH. The number of released granules reached its maximum at 10 min after 200 microgram TRH injection, but the total number of secretory granules in the cytoplasm of thyrotrophs did not show any significant variation throughout the experimental period. Percent area of the rough endoplasmic reticulum indicated the maximum value at 20 min and that of the Golgi apparatus at 10 min. These findings suggest that TRH stimulates the thyrotroph and accelerates synchronously the secretion as well as the synthesis of TSH. In the thyroid follicular cells after 200 microgram TRH injection, numerous small vesicles which might be secretory granules were found in the apical cytoplasm at 5 and 10 min. Pseudopod formation on the luminal surface and accumulation of colloid droplets and lysosome-like granules in the apical cytoplasm were most frequently observed at 20 and 30 min. The value of the mean ratio of the diameter of the colloid lumen to the cell height of follicular cells in the thyroid follicles of all experimental animals stimulated with TRH decreased much more than that of the control animals. This value may indicate the increased activity of follicular cells to reabsorb the colloid.
1. [14C]Ethyltetrazolylchromone ([14C]ETC) was promptly absorbed from the rat small intestine by the portal route. 2. The maximum plasma concn. of unchanged drug after oral administration (10 mg/kg) was highest in dogs (456 microgram/ml), followed by monkeys (287 microgram/ml), guinea-pigs (146 microgram/ml) and rats (55 microgram/ml), and lowest in rabbits (09 microgram/ml). The half-life of the drug in plasma varied with the species, ranging from 13 to 133 h. The drug was highly bound to plasma protein. In dogs and rats, the plasma 14C was predominantly the unchanged drug, whereas in guinea-pigs, rabbits and monkeys it was mainly metabolites. 3. At 10 min after oral administration of the drug to rats there was a wide distribution of the 14C in the tissues. At this time, the 14C concn. were the highest in stomach, followed by kidney, liver, plasma, heart and lung, and lowest in brain. 4. Almost all administered 14C was eliminated from the body in 72 h. The major route of excretion was via the urine except with guinea-pigs, in which animal the 14C was almost equally divided between urine and faeces. 5. only trace amounts of the unchanged drug were found in urine and bile. The major urinary metabolites were as follows: I (1-hydroxyethyl ETC), II (acetyl ETC), III (IIIa, 2-hydroxyethyl ETC) and IV (1,2-dihydroxyethyl ETC) in rats, I and VI (5-carboxymethylsalicylic acid) in guinea-pigs, I, III (IIIb, carboxymethyl ETC) and VII (ETC-N-1-glucuronide) in rabbits, I and VII in dogs, and I and IV in monkeys.
