Collaborative study on rat sperm motion analysis using CellSoft Series 4000 semen analyzer.

A collaborative study was conducted to determine useful and sensitive rat sperm motion parameters in a CellSoft Series 4000 semen analyzer to detect the effects of compounds on sperm motion. The effects on the sperm motion parameters were investigated using alpha-chlorohydrin, boric acid, ethinylestradiol, ethyl methanesulfonate, nitrazepam, nitrobenzene, ornidazole, sulfasalazine or valproic acid which are well known to induce reproductive or testicular toxicities. All compounds used in this study decreased percentage of motile sperm (% motile). Curvilinear velocity (VCL), maximum and mean amplitude of lateral head displacement (ALH max and ALH mean) were decreased by treatment with all compounds except for valproic acid. Treatment with alpha-chlorohydrin, ornidazole or sulfasalazine under mid-dosage regimens decreased only these parameters. Beat cross frequency (BCF) was increased by treatment with sulfasalazine. There were some treatments which caused either decreased or increased changes irrespective of dosage regimen in linearity, average radius, percentage of circular-swimming sperm out of motile sperm (circular/motile) and percentage of circular-swimming sperm out of all sperm (circular/all). Based on these results, we concluded that % motile, VCL, ALH max and ALH mean are considered useful and sensitive parameters for evaluating the effects of compounds on sperm motion. A parameter of BCF can be useful to detect the effects of specific compounds on sperm motion. Linearity, average radius, circular/motile and circular/all are not considered useful or sensitive indicators to detect the effects of compounds on sperm motion.

[Positive rate of anti-mitochondrial antibody in Japanese corporate workers].

Anti-mitochondrial antibody(AMA) has been reported to be detectable in approximately 85% of patients with primary biliary cirrhosis(PBC). Therefore, a test for AMA is acceptable to be essential for diagnosing PBC. However, the positive rate in Japanese general population has not yet been determined. We tested sera from 1,145 corporate workers who took an annual health check and evaluated the liver of AMA-positive subjects. An indirect immunofluorescence method was used for screening AMA. ELISA and immunoblotting method were used for detecting anti-M2 in AMA-positive cases. AMA was detected in 5 of 1,145(0.44%) corporate workers. AMA positive rate was higher in females than in males(0.91% and 0%, respectively) and the AMA-positive people are all females over age 40. All of the AMA-positive sera are also positive for Anti-M2. Liver biopsy was performed in two AMA-positive cases and the histology was compatible with PBC in both cases.

Availability of sperm examination for male reproductive toxicities in rats treated with boric acid.

Sperm morphological examination, computer-assisted sperm analysis (CASA) and histopathologic examination of the testis and epididymis were performed for male rats treated orally with boric acid for 3 weeks at dosage levels of 50, 150 and 500 mg/kg/day, and treated males were mated with untreated females. None of the males treated with 500 mg/kg/day could impregnate untreated females. The fertility index showed a tendency to decrease in males treated with 150 mg/kg/day. At necropsy, the pre-implantation loss rate in females mated with males treated with 150 mg/kg/day was higher than the control values. Upon epididymal sperm analysis using the CASA system, all parameters including the number of sperm and sperm motions were found to be affected in males treated with 500 mg/kg/day, and the number of sperm, percent motile, velocities and amplitude of lateral head displacement (ALH) were affected in males treated with 150 mg/kg/day. Upon sperm morphological examination, head and tail abnormalities were observed in males treated with 150 and 500 mg/kg/day. In the histopathological examination, atrophy of the seminiferous tubules and multinucleated giant cells in the testes were observed in males treated with 500 mg/kg/day.

Sperm abnormalities and histopathological changes in the testes in Crj:CD(SD)IGS rats.

In this study, morphological examination and computer-assisted sperm analysis (CASA) of epididymal spermatozoa in non-treated Crj:CD(SD)IGS rats were performed, and the relationship between the data obtained and the retention of step 19 spermatids in Stage IX to XI seminiferous tubules was examined. Retention of step 19 spermatids in Stage IX to XI seminiferous tubules was observed in all 50 untreated males, and the incidence ranged from 3.3% to 100%. Eighteen animals showed a high incidence of retention (74.7 +/- 14.2%, HIR for short), and the others showed a low incidence (24.9 +/- 11.0%, LIR for short). Although the incidence of retention in Stage X and XI seminiferous tubules was very low in LIR males, it was high in HIR males (1.8 +/- 3.0% vs 58.6 +/- 23.2%). Morphological abnormalities of sperms in the caudal region of the epididymis, mainly amorphous head and no head, were more frequently observed in HIR males than in LIR males (36.2 +/- 28.5% vs 1.8 +/- 1.2%). Sperm analysis also revealed some differences between HIR and LIR males: sperm motility in HIR males was severely lower than that in LIR males, and sperm velocity, beat/cross frequency and amplitude of lateral head displacement in HIR males were lower than the corresponding values in LIR males. In summation, retention of step 19 spermatids frequently occurred in the non-treated Crj:CD(SD)IGS males, and a relationship between the retention of these spermatids and sperm abnormalities, such as morphologically abnormal sperms, low motility and other items revealed by sperm analysis (CASA), was suggested.

Collaborative project to establish the optimal period and parameters for detection of reproductive toxicity in male rats--effects of compound T on male fertility.

To assess the optimal dosing period and parameters for measurement of effects on male fertility, Compound T was administered to male Jcl:Wistar rats at dosage levels of 0, 2, 10 and/or 50 mg/kg/day for 4 weeks (Experiment 1) or for 9 weeks (Experiment 2). Experiment 1: In the 50 mg/kg group, the ventral prostate weight was low when compared to the control value, and desquamation of round spermatids was observed in the testes. Experiment 2: When treated males were mated with untreated females after dosing for 9 weeks (the first mating), the fertility index was slightly lowered and preimplantation loss was significantly elevated in the 50 mg/kg group as compared to the control values. In this treatment group, serum testosterone level at 2.5 hours after dosing was significantly decreased after dosing for 12 weeks, and degeneration of spermatids/spermatocytes in the testis and epididymis was observed after dosing for 13 weeks. After a recovery period of for 6 weeks, remating resulted in copulatory and fertility indices and cesarean section data which were comparable in all groups. In conclusion, there were no differences in toxicity relevant to male fertility between 4 and 9 weeks of dosing, and it is considered that the observed changes resulted from decreased function of Sertoli cells due to depressed production/secretion of testosterone.

[Diagnosis of the pancreatic diseases by intraductal ultrasound of the pancreas--with special reference to clinical cases difficult to diagnose differentially pancreatic diseases by the endoscopic retrograde pancreatography].

We examined the usefulness of the intraductal ultrasound (IDUS) examinations by comparing the IDUS images with the endoscopic retrograde (ER) pancreatograms. We inserted an IDUS probe with 30 MHz at frequency transpapillary into the main pancreatic duct (MPD) in 105 cases with pancreatic diseases (30 cancers, 30 mucin-producing tumor, 3 islet cell tumor, 1 pancreatic serous cyst adenoma, 1 pancreatic teratoma, 1 pancreatic solid and cystic tumor and 39 chronic pancreatitis). All 105 cases were performed ER pancreatography. Sixty-four of 105 cases (22 cancers, 20 mucin-producing tumor, 3 islet cell tumor, 1 pancreatic serous cyst adenoma, 1 pancreatic teratoma, 1 pancreatic solid and cystic tumor and 16 chronic pancreatitis) were operated and the IDUS images were compared with the corresponding histopathological sections. The usefulness of the IDUS examinations of the pancreatic diseases were the following cases; differential diagnosis between benign and malignant cases with localized stenosis of the MPD, differential diagnosis of the cases with displacement of the MPD, and the diagnosis of the intraductal extent and of invasion of the tumor in the mucin-producing tumor of the pancreas with the dilation of the MPD. IDUS examinations are performed by the IDUS probe with high frequency, so the indications of the IDUS examinations are to detect the small lesions in the pancreas.

Effect of dextranase on the extracellular polysaccharide synthesis of Streptococcus mutans; chemical and scanning electron microscopy studies.

A dextranase preparation (AD17) partially purified from a culture liquor of Spicaria violacea strain IFO 6120 significantly inhibited the formation of artifcial dental plaque on a steel wire or on an extracted tooth surface. Changes in the surface morphology of Streptococcus mutans cells due to AD17 action were studied using scanning electron microscopy. S. mutans cells grown in 5% sucrose-containing broth were coated with sticky amorphous capsule-like material, whereas cells grown in sucrose in the presence of AD17 or in glucose instead of sucrose did not synthesize such capsular material. AK17 degraded commercially available dextrans of molecular weight 7 X 1(04) and 2 X 10(6) to liberate glucose and various oligosaccharides, including isomaltose. On the other hand, AD17 hydrolyzed the extracellular polysaccharides (mainly glucan in nature) of some strains of S. mutans to a limited degree. Only 15 to 36% of the total polysaccharides were hydrolyzed by AD1M with little release of isomaltose. Prolonged incubation of the polysaccharides from S. mutans with AD17 did not release additional reducing sugars, which indicates that AD17 did not contain alpha-1,3-glucanase activity. These results suggest that glucosidic linkages which are susceptible to AD17 may play an important role in the adherence of S. mutans cells to smooth surfaces.

An olfactory projection area in orbitofrontal cortex of the monkey.

An olfactory projection area was studied in monkeys anesthetized with Nembutal. 1. Evoked potentials were recorded when the olfactory bulb (OB) was electrically stimulated in the lateroposterior portion of the orbitofrontal cortex (LPOF). However, those potentials disappeared when the anterior pyriform cortex (AP) (probably together with the medial portion of the amygdala (MA)) was aspirated or electrically destroyed. 2. In nearly the entire hypothalamic region, evoked potentials were recorded by the same stimulation of the OB. When the hypothalamic region was stimulated, evoked potentials were recorded in the LPOF. 3. The evoked potentials in the LPOF due to the OB stimulation never disappeared even when the thalamus was extensively aspirated or destroyed electrically, but they did disappear when the anterolateral and dorsoposterior portions of the hypothalamus were absorbed or electrocoagulated. 4. Evoked potentials in the mediodorsal nucleus (MD) of the thalamus were recorded when the OB was stimulated. When this nucleus was stimulated, evoked potentials were observed in the broad extent of the orbitofrontal cortex anterior to the LPOF, but never in the LPOF itself. 5. Monkeys were conditioned to discriminate two odors. When the LPOF was removed, such ability strikingly decreased; but when other areas in the prefrontal cortex were removed, the ability decreased only slightly. 6. It was concluded that there exists an olfactory pathway from the OB to the LPOF through the AP (and probably the MA) and the hypothalamus, but none through the thalamus, and that the LPOF plays an important role in the discrimination of odors. 7. It was proved that the entorhinal cortex (ER) is neither located as an intermediate olfactory area nor is it situated as a higher area than the LPOF in the newly found olfactory pathway stated above. It may be a link between the high olfactory area and the limbic system.
