Regional specificity of ethanol and NMDA action in brain revealed with FOS-like immunohistochemistry and differential routes of drug administration.

BACKGROUND: Inhibition of NMDA receptor function in brain is believed to be an important action of ethanol (EtOH). To investigate EtOH inhibition of NMDA receptor responses in vivo, the interaction of these agents in brain after different routes of administration were investigated by using transcription factor Fos protein expression to follow NMDA receptor activation and EtOH inhibition of this response. METHODS: The induction of Fos-like immunoreactivity (Fos-LI) in 38 regions of the rat brain was measured 2 hr after treatment with NMDA, EtOH, or both. To determine the relative contribution of abdominal drug effects on Fos induction, rats received either intraperitoneal (ip) or intragastric (ig) EtOH and ip or intravenous (iv) NMDA. Rats received EtOH (2.5 g/kg ip or 4 g/kg ig) or vehicle 15 min before NMDA (125 mg/kg ip or 60 mg/kg iv) or vehicle. RESULTS: For the 38 forebrain regions examined, ip and iv NMDA significantly induced Fos-LI in 13 and 32 regions, respectively. These effects occurred without elicitation of tonic-clonic seizure activity and were strong after iv NMDA in the frontal, prefrontal, and cingulate cortices, supraoptic nucleus, anterior lateral septum, and dentate gyrus. For EtOH, prominent Fos-LI induction was found in the central amygdala, dorsolateral bed nucleus of the stria terminalis, Edinger-Westphal nucleus, and paraventricular hypothalamus. Despite ip and ig EtOH induction of Fos-LI in these regions, the major effect of EtOH was to block NMDA-induced Fos-LI in 8 of 13 (ip) and 27 of 32 (ig) of the NMDA-positive regions, respectively, including retrosplenial, cingulate, and medial prefrontal cortices, central amygdala, and taenia tecta. CONCLUSIONS: These results provide new evidence for the regionally specific functional interactions of EtOH on NMDA receptors in vivo. Moreover, these results support efforts to identify brain region-specific targets for EtOH and EtOH-induced changes in gene expression.

[A fusion protein of rotavirus VP6 and cholera toxin B subunit: expression in Escherichia coli and analysis of biological activities].

Rotavirus infection is a major cause of dehydrating diarrhea in infants worldwide. The non-toxic cholera toxin B subunit(CTB), known as an immunomodulatory carrier, might help to stimulate mucosal immune response when coupled to rotavirus antigens in oral immunization. Here we report for the first time the construction of a translational fusion of CTB gene 5' to the VP6 gene of a human rotavirus A(field strain T114), and expression of the CTB-VP6 fusion protein in E. coli BL21(DE3). The expressed fusion protein has a molecular weight of 56 kD, as expected, and accounts for about 15% of the total E. coli protein. Western blottings with the hyperimmune serum against rotavirus strain WA and the antibody against cholera toxin indicated that the fusion protein retains the antigenicity identical to the native CTB and VP6. The GM1-ELISA analysis proves that the renatured CTB-VP6 has strong affinity for GM1 ganglioside.

The effect of electro-acupuncture on motor function recovery in patients with acute cerebral infarction: a randomly controlled trial.

UNLABELLED: The aim of this study is to investigate the effect of electro-acupuncture treatment in acute phase of cerebral infarction on the motor functions. In this randomly controlled trial, 86 patients were allocated to two groups, the experimental group given clinical and electro-acupuncture treatments for a period of 4 weeks, and the control group given clinical treatment plus active and/or passive functional exercise. The result showed that the level of impairment and disability in both groups were improvement according to the Chinese Stroke Scale, Brunnstrom-Fugl-Meyer score, and Barthel Index throughout the study and 3 months after. The motor functions and the activities of daily living (ADL) were improved significantly in the electro-acupuncture group as compared with the control group (P < 0.05). Also, the results showed greater reduction of neurological deficit in the electro-acupuncture group than in the control group. CONCLUSION: Early acupuncture treatment for acute stroke patients may improve motor functions, and consequently the activities of daily living.

[Study of the adsorption properties in chromatographic separation of the citric acid].

A new method studying the change of thermodynamic properties at different temperatures to screen suitable stationary phases for chromatographic separation of the citric acid (CA) on an analytical column has been developed. The applied method is mainly based on using CA with a series of solute concentrations (0 g/L-250 g/L) as mobile phase to seriously overpass the unloaded stationary phase to find the breakthrough curves of CA. The adsorption isotherms were calculated from the retention times of breakthrough curve. The bed porosity of column was calculated from the elution time of a peak of the blue dextran as non-adsorbed tracer. The stationary phase, which has been selected through this method, for the separation of CA from fermentation broth, showed a very high adsorption capacity for CA, even under very low concentrations, so it can even be used in the separation and purification process of minute amounts of CA. The separation effect of this stationary phase for citric acid can be greatly enhanced through the technique of change of temperature. This new separation process can shorten the old process path, improve the yield of finished products more than 10%, and reduce the cost more than 10%. At the same time, the pollution of the production of citric acid can be reduced. It turned out that this method will play an important role in the synthesis of stationary phase for the separation of citric acid and the study of its structure.

Polymorphisms of the DNA repair gene XRCC1 and risk of gastric cancer in a Chinese population.

Gastric cancer remains the leading cause of cancer death in China and other countries in eastern Asia. Studies of gastric cancer have revealed that it is a disease of complex etiology involving dietary, infectious, environmental, occupational and genetic factors. DNA repair capacity has been suggested as a genetic factor contributing to variation in susceptibility to cancer. In the present study, we described an association between 2 polymorphisms of the DNA repair gene XRCC1 and risk of gastric cancer in a Chinese population. We used a polymerase chain reaction-based assay to detect Pvu II and Nci I restriction fragment length polymorphisms (XRCC1 26304 C-->T and XRCC1 28152 G-->A, respectively) in 188 patients with gastric cancer and 166 healthy controls. The XRCC1 26304 T allele (194Trp) frequency (34.6%) was higher and the XRCC1 28152 A allele (399Gln) frequency (25.6%) was lower in healthy Chinese controls than previously reported healthy U.S. Caucasian controls (7.2% and 34.1%, respectively). Multivariate logistic regression analysis revealed that the putative high-risk genotypes XRCC1 26304 CC and XRCC1 28152 GA/AA were associated with a non-significant increased risk for gastric cancer (adjusted odds ratio [OR]=1.45, 95% confidence interval [CI]= 0.93-2.25 and OR=1.53, 95% CI= 0.98-2.39, respectively) compared with other genotypes. However, the XRCC1 26304 CC genotype was associated with a significantly increased risk for gastric cardia cancer (adjusted OR=1.86, 95% CI=1.09-3.20). Individuals with both putative high-risk genotypes (CC and GA/AA) had a significantly higher risk (adjusted OR=1.73, 95% CI=1.12-2.69), particularly for gastric cardia cancer (adjusted OR=2.18, 95% CI=1.21-3.94) than individuals with other genotypes. These findings support the hypothesis that these 2 XRCC1 variants may contribute to the risk of developing gastric cancer, particularly gastric cardia cancer.

Templated Synthesis of CdS/PAN Composite Nanowires under Ambient Conditions.

CdS/PAN (polyacrylonitrile) composite nanowires were prepared by templated synthesis under ambient conditions, using a 4 : 1 (v/v) distilled water-isopropyl alcohol solvent mixture and Na(2)S(2)O(3) as sulfur source. Transmission electron microscopy images showed typical CdS/PAN nanowires with diameters less than 6 nm and lengths from 200 nm to 1 µm. CdS/PAN nanowires were also characterized by FT-infrared spectra (FTIR), X-ray powder diffraction (XRD) patterns, and UV-vis spectra. It is found that the feed mole ratio of CdCl(2) to AN exerts a significant influence on the formation and morphology of CdS/PAN nanowires. In addition, possible mechanism of the reactive process was presented. Copyright 2000 Academic Press.

Molecular characterization and leukocyte distribution of a teleost beta1 integrin molecule.

The beta1 integrin, in combination with the alpha subunit, is responsible for migration of leukocytes into areas of inflammation. Although identified in mammalian species; the beta1 or CD29 molecule has yet to be identified in fish. The present investigation has identified a full-length channel catfish, Ictalurus punctatus, cDNA beta1 molecule composed of 2786 bases and a deduced amino acid sequence of 797 amino acids. The catfish molecule has an amino acid identity ranging from 71.87 to 74.12% with bovine, feline, human, and Xenopus. The channel catfish molecule retains several characteristics of mammalian beta1 molecules, such as four cysteine-rich repeat regions, and eight potential N-linked glycosylation sites. Based on Western blotting the channel catfish beta1 molecule has a molecular mass of approximately 130kDa, essentially the same as that for mammalian species. These results confirm the existence and expression of a beta1 gene in channel catfish, a species phylogenetically distant from mammals.

The C-terminal region of proSAAS is a potent inhibitor of prohormone convertase 1.

ProSAAS is a recently discovered 26-kDa neuroendocrine protein that was previously found to inhibit prohormone convertase (PC) 1 and not PC2. In the present study, the specificity of proSAAS toward other members of the prohormone convertase family was determined. Two microm proSAAS selectively inhibits PC1 but not furin, PACE4, PC5A, or PC7. The PC1 inhibitory region of proSAAS was mapped to an 8-12-residue region near the C terminus that includes a critical Lys-Arg sequence. Synthetic peptides corresponding to this region are competitive inhibitors of PC1 with apparent K(i) values of 14-40 nm. The inhibition becomes more effective with incubation time, indicating that the inhibitor is slow binding. A fusion protein containing the inhibitory region of proSAAS linked to the C terminus of glutathione S-transferase binds the 71-kDa form but not the 85-kDa form of PC1. This binding, which occurs at pH 5.5 and not at pH 7.4, is stable to incubation at room temperature for 1 h in the presence or absence of 0.5% Triton X-100 and/or 0.5 m NaCl. The removal of Ca(2+) with chelating agents partially releases the bound PC1. High concentrations of the inhibitory peptide quantitatively release the bound PC1. Taken together, these data support the proposal that proSAAS functions as an endogenous inhibitor of PC1.

Association between attention deficit hyperactivity disorder and the DXS7 locus.

Attention deficit hyperactivity disorder (ADHD) is a prevalent disorder in children. The etiology of this disease is not clear. Genetics studies have suggested the involvement of the dopamine DRD-4 receptor gene and dopamine transporter gene (DAT1). Clinical studies have shown that monoamine oxidase-B (MAO-B) inhibitors are effective in the treatment of ADHD. These findings suggest that monoamine oxidase (MAO) genes might be involved in the origin of ADHD. In the present work, the DXS7 locus of chromosome X, which is closely linked to MAO genes, was selected as a marker to study the possible association between ADHD and MAO genes in the Chinese population. Haplotype-based haplotype relative risk (HHRR) and the transmission disequilibrium test (TDT) methods were employed to analyze the association and the linkage disequilibrium, respectively. Significant association (X(2) = 15.86; 1 df; P < 0.001) and linkage (X(2) = 14.88; 1 df; P < 0.001) were detected between the 157-bp allele of the DXS7 locus and the DSM-III-R-diagnosed ADHD (N = 72) in trios composed of father, mother, and affected offspring. The data suggested that ADHD was associated and in linkage with DXS7 locus.

Solvothermal route to tin monoselenide bulk single crystal with different morphologies.

Orthorhombic SnSe bulk single crystals with different morphologies were conveniently grown through solvothermal process at low temperature (180 degrees C). X-ray diffraction (XRD) patterns, scanning electronic microscope (SEM) images, electronic diffraction (ED), and X-ray photoelectron spectra (XPS) were used to characterize the products. In the solvothermal process, the solvent plays an important role in the growth of the single crystal. The morphology of the crystal is influenced not only by the kind of solvent but also by the concentration of the reactants in solvent. Single crystals with rodlike or platelike morphology can be conveniently obtained under controlled concentration of reactants in solvent.

Supernovae versus Neutron Star Mergers as the Major r-Process Sources.

I show that recent observations of r-process abundances in metal-poor stars are difficult to explain if neutron star mergers (NSMs) are the major r-process sources. In contrast, such observations and meteoritic data on 182Hf and 129I in the early solar system support a self-consistent picture of r-process enrichment by supernovae (SNe). While further theoretical studies of r-process production and enrichment are needed for both SNe and NSMs, I emphasize two possible direct observational tests of the SN r-process model: gamma rays from the decay of r-process nuclei in SN remnants and surface contamination of the companion by SN r-process ejecta in binaries.

Genetic association study between alpha 1-antichymotrypsin polymorphism and Alzheimer disease in Chinese Han population.

We investigated a common signal peptide polymorphism in the alpha 1-antichymotrypsin (ACT) gene in 125 sporadic Alzheimer disease (AD) patients and 141 healthy control subjects in Chinese Han population. We found no significant difference in the distribution of ACT polymorphism between AD cases and controls, and failed to detect any effects of ACT genotypes associated with AD. Thus, our data do not support the involvement of ACT polymorphism in the risk of AD in Chinese Han population. Am. J. Med. Genet. (Neuropsychiatr. Genet.) 96:133-135, 2000.
