Ongoing chronic strain of living with limb deficiencies and normal daily hassles may interact to produce a negative impact on adjustment. Degree of limb loss, daily hassles/microstressors, and classmate, parent, teacher, and friend social support were investigated as predictors of depressive symptomatology in 27 children with congenital/acquired limb deficiencies. Hierarchical multiple regression analyses were utilized to test the main effects and buffering interactions effects models of the stress-social support-depressive symptomatology relationship. Degree of limb loss was not a significant predictor. Microstressors and classmate, parent, and teacher social support were all significant predictors of depressive symptomatology. None of the Stress X Social Support interaction terms were statistically significant. The overall model of stress and social support predicted 73% of the variance in depressive symptomatology.

CAPP Terminal Device, Size #2 for teenaged and adult amputees offers an alternative to hooks and hands available today. CAPP TD 2 is intended to be pleasing in appearance; it blends with the forearm in shape, colour and material to give a continuous flowing natural look. It was designed to provide secure grip through the combined action of a closing spring, a full frictional, resilient cover and an automatic lock. The lock operates as part of the voluntary opening control system and requires no conscious action by the amputee. CAPP TD 2 is a general purpose terminal device which especially serves unilateral amputees by performing functions usually carried out by the non-dominant hand. CAPP TD 2 comes with a built-in wrist connector which is available in two models; both offer quick disconnect. One wrist unit has adjustable friction and the other attaches to an existing friction wrist unit so CAPP TD 2 can be applied to an existing prosthesis. Ten patients have completed the evaluation programme with promising results. Prototypes of CAPP TD 2 are available for patients interested in evaluating it. The CAPP Terminal Device, Size #2 offers unilateral teenaged or adult amputee patients a unique alternative. It is introduced here in hope of stimulating researchers to explore new mechanical solutions in their component designs. The CAPP research designer, Mr. Carl T. Sumida. CPO, introduced the idea of a non-hand, non-hook TD with the CAPP TD 1 for infants (Sumida and Setoguchi, 1967); that terminal device had more function than a passive hand and it had a softer appearance than a hook.(ABSTRACT TRUNCATED AT 250 WORDS)
Family functioning and child temperament variables were investigated as predictors of psychologic and social adaptation in 42 children with congenital or acquired limb deficiencies. Higher psychologic and social adaptation were seen when there was more family cohesion and moral-religious emphasis and organization, in combination with less family conflict. With regard to child temperament, more emotionality predicted greater internalizing and externalizing behavior problems and less social competence. In addition to the main effects of the family functioning and child temperament predictor variables, the interaction between family cohesion and child emotionality significantly predicted both internalizing and externalizing behavior problems. The findings are discussed in terms of the risk and protective effects of family functioning domains and temperament on the psychologic and social adaptation of children with visible physical handicaps.
Social support, family functioning, self-perceptions of appearance, school, behavior, athletics, and social competence, as well as daily hassles/microstressors, were investigated as determinants of general self-esteem in 41 children with congenital/acquired limb deficiencies. Classmate, parent, teacher, and friend social support all were significant predictors of self-esteem. Family conflict and organization were also significantly associated with self-esteem, as were microstressors. The overall model of significant predictor variables accounted for 66% of the variance in general self-esteem. General self-esteem also was significantly predictive of depressive symptomatology. The findings suggest a number of factors which are potentially modifiable and which might enhance general self-esteem in children with visible physical handicaps.
Integrative mapping with vectors containing ribosomal DNA sequences were used to complete the mapping of the 10 rRNA gene sets in the endospore forming bacterium Bacillus subtilis. Southern hybridizations allowed the assignment of nine operons to distinct BclI restriction fragments and their genetic locus identified by transductional crosses. Nine of the ten rRNA gene sets are located between 0 and 70 degrees on the genomic map. In the region surrounding cysA14, two sets of closely spaced tandem clusters are present. The first (rrnJ and rrnW) is located between purA16 and cysA14 closely linked to the latter; the second (rrnI, rrnH and rrnG) previously mapped within this area is located between attSPO2 and glpT6. The operons at or near the origin of replication (rrnO,rrnA and rrnJ,rrnW) represent "hot spots" of plasmid insertion.

Non-histone chromatin proteins prepared from a normal rat cell line (No. 7) and the cells transformed with Rous sarcoma virus (RSV) (s7-1) were compared by means of reverse-phase high performance liquid chromatography (reverse-phase HPLC), followed by SDS-polyacrylamide gel electrophoresis (SDS-PAGE). The results revealed that several proteins were specifically present in the transformed cell chromatin. A specific non-histone chromatin protein with a molecular weight of 38,000 daltons, 38K protein, was purified as a single species from s7-1 cells. This 38K protein was only detected in the transformed state of the cells transformed with a temperature-sensitive (ts) mutant of the src gene and the mutant cells which showed temperature sensitivity as to the transformation with wild type RSV.
An investigation of the properties and kinetics of serum cholinesterase (chE) was conducted in familial cases of serum hyper-chE activity. An inhibition study with specific pseudo-chE inhibitor (ethopropazine), or with anti-human pseudo-chE sera, has produced evidence that this genetically variant chE is pseudo-chE. The Km or relative activity of the variant chE against enzyme protein turns out to be the same as that of the normal serum chE. However, the variant serum chE shows more basic pK in addition to the main pK of the normal pseudo-chE on isoelectrofocusing analysis. The serum chE isozyme analysis has revealed two different extrabands in the variant: one demonstrated as a sharp line between bands 3 and 4, which we described previously, and the other as slow migrating broad band in the cathodic region close to the origin. Both prove to be pseudo-chE. Although there have been some similar descriptions about the latter type of extraband, no reports are found about the former type. It is noteworthy that the use of heparin as an anti-coagulant caused the total disappearance of the first type of extraband. It should be remembered that only heparinized plasma was used in the earlier studies. Since the detection of the former type of band seems to be specific and is simple, our findings would probably help clinicians diagnose a familial case of hyper-chE.
Non-histone chromatin proteins prepared from the livers of estrogen-treated and nontreated male chickens were compared by reverse-phase high performance liquid chromatography (RP-HPLC), followed by sodium dodecyl-sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The results revealed that the hormone-treated male liver chromatin contained a specific protein corresponding to the vitellogenin-specific protein previously purified from the liver of egg-laying hens (Nakayama 1985). The chicken protein, purified further by gel-filtration high performance liquid chromatography (GF-HPLC), showed specific binding activity to DNA fragments carrying a part of the vitellogenin gene. On the basis of similarities in the elution patterns from GF-HPLC and RP-HPLC as well as in the mobility on SDS-PAGE, we concluded that this hormone-induced protein in the male chicken liver was identical to the vitellogenin-specific protein identified in the hen liver, and assumed it to be a specific regulatory protein for the vitellogenin gene expression. The amino acid composition of this chicken protein has been determined.

Although Poland's syndrome is classically considered to consist of simple syndactyly, limb hypoplasia, and ipsilateral pectoral muscle agenesis, a rate group of patients with pectoral agenesis has more severe limb deficiencies. In ten of these patients and in similar patients reported in the literature, a broad range of limb deformities was observed. The authors classified the patients according to limb anomalies: type 1, five digits present even if hypoplastic; type 2, functional border digits with the absence of central digits; type 3, more severe absence deformities with no functional digits; and type 4, radial ray defects with absent thumb. The understanding that Poland's syndrome includes a spectrum of limb anomalies is important in searching for associated birth defects, in genetic counseling, and, possibly, in elucidating the pathogenesis of the syndrome.
A case is described which, at birth, had a bizarre pattern of hypopigmentation (incontinentia pigmenti achromians), ectrodactyly involving all four extremities, and unilateral cleft lip and palate. This patient does not have the seizures or other neurological and developmental anomalies previously described as associated with hypopigmentation of Ito. This condition is also clearly different from the syndrome of ectrodactyly, ectodermal dysplasia, and clefting (EEC).
The relative transcription from L and H strands of Bacillus subtilis DNA during consecutive stages of spore outgrowth was determined and compared to the transcription pattern during log-phase growth of vegetative cells. Pulses of [3H] uridine were administered during early, middle and late outgrowth phases of germination and the RNAs isolated. The asymmetry ratio of H/L as determined by hybridization at saturating RNA/DNA inputs showed a gradual decrease. During the period studied (10-90 and 90-160 min post-induction), about 50 and 35%, respectively, of the radioactive RNA consisted of ribosomal RNA transcripts. The decrease in the H/L asymmetry ratio was due predominantly to the appearance and accumulation of L strand transcripts and not to either changes in the quantity of H strand transcripts nor to fluctuation in the rate of rRNA synthesis.
Hybrid-competition analyses were done to estimate the relatedness of 3H-labeled mRNA species synthesized during spore germination and log-phase growth. The competitions showed that early in the germination process 10--15 and 1--3% of the RNA transcribed from the H and from the L strand, respectively, were unique and absent during log-phase growth. At later stages, the amounts of the germination-specific H transcripts decreased more rapidly than the L transcripts. The competitions with pulse-labeled log-phase RNAs showed that vegetative genes were transcribed more rapidly from the H strand than from the L strand. Most of the results could be correlated with the observed decrease in the H/L asymmetry ration during spore germination.
Six cases have been presented with combined limb deficiencies and cranial nerve dysfunctions. The major features are congenital disarticulation and congenital amputation associated with various orofacial deficits. Anatomic variations in limb and facial anomalies in these cases, as well as in other cases previously reported; seem to indicate that they are part of a single dysmorphic syndrome rather than a group of distinct clinical syndromes as reported in the past.
