[The application of artificial neural network in QSAR research of PCDD].

The study of QSAR between molecular structure and retention behavior of solutes is an important topic of chromatographic basic theory. PCDD (polychlorinated dioxin) is a kind of important environmental toxin. The basic skeleton of PCDD is [symbol: see text]. PCDDs are all chlorine substituted, so they are only different from each other in the number and position of chlorine substitutents on the skeleton. There are 76 kinds of homologues (1 to 8 chlorine substituent) for 3, 4, 5 and 6 chlorine substituted compounds including more than ten kinds of isomerides. In this paper, the description of molecular structure and feature selection have been investigated. Artificial neural network (ANN) is used to relate the molecular structure and retention time of PCDD. The predicted results of 49 PCDDs retention time by ANN are listed. It was shown that as to the relative error more than 95% data was less than 10% and more than 80% data was less than 5%.

[A method of following analysis for glutaraldehyde preparation].

A GC method for the products in cyclopentene oxidation to form glutaraldehyde in a liquid-liquid phase transfer reactive extraction process, which was a new process, is presented. A model GC-103 gas chromatograph equipped with FID was used. The column is a SS tube(phi 3 mm x 4 m) packed with 13% SE-30 coated on 101 white silanized support and a SS tube(phi 3 mm x 2 m) packed with 9.1% PEG-20M coated on the same support. Operating parameters: column temperature programming, 70 degrees C(4 min), then 10 degrees C/min to 140 degrees C(6 min); injector temperature, 185 degrees C; detector temperature, 185 degrees C; N2, 45 mL/min; H2, 65 mL/min; air, 300 mL/min. By using this method, cyclopentene, intermediates, solvents, glutaraldehyde, and byproducts were separated properly. The yield of glutaraldehyde determined by this method is more reliable than that determined by the chemical method.

(NH4)8[Ce2(SO4)8].4H2O.

Crystals of the title compound, octaammonium dicerium octasulfate tetrahydrate, suitable for X-ray work, were prepared by a hydrothermal reaction. The structure is made up of [Ce2(SO4)8]8- anions, interacting with the crystallization waters and the NH4+ cations through hydrogen bonds. The CeIV atom in the [Ce2(SO4)8]8- anion is ninefold coordinated by O atoms belonging to sulfate groups. The coordination polyhedron is a somewhat distorted tricapped trigonal prism. The Ce-O distances range from 2.325(2) to 2.478(2) A.

Molecular cloning, characterization, and expression of the chicken heme oxygenase-1 gene in transfected primary cultures of chick embryo liver cells.

Using chick heme oxygenase-1 (cHO-1) cDNA as a probe, three independent clones were identified from screening a lambda FixII chick genomic library. Genomic Southern blots using this cDNA probe or a cHO-1 5' specific probe showed that cHO-1 is a single-copy gene. Based on restriction enzyme analysis, Southern blots, polymerase chain reaction analysis and DNA sequencing, it was confirmed that the three overlapping clones isolated cover the entire cHO-1 gene, as well as approximately 10 kb of the flanking regions on both ends. As with mammalian HO-1x, cHO-1 has five exons and four introns. Computer analysis of the DNA sequence obtained identified consensus sequences corresponding to numerous transcription factor recognition elements. These include AP-1, AP-2, NF-kB, C/EBP, c-Myc and a metal-responding element identified in the promoter region, and two Sp-1 elements in intron 1. Transient expression studies in transfected primary cultures of chick embryo liver cells showed that a CAT reporter gene construct containing 2.8 kb of the cHO-1 promoter region responded to sodium arsenite, H2O2, transition metals and 12-0-tetradecanoylphorbol 13-acetate, but not to heme. Studies with deletion mutants, consisting of various lengths of the cHO-1 promoter region, indicated that there are two regions important for sodium arsenite induction, one located between residues -1642 and -1293, and the second located in the first 263 base pairs of the cHO-1 promoter. DNA binding studies by electrophoretic mobility shift assay showed that nuclear protein isolated from primary cultures of chick embryo liver cells bound to the oligonucleotide probe containing an AP-1 element identified at -1573 to -1580. In addition, such binding was increased by cobalt or sodium arsenite treatment.

Circulating mechanism of the "pure" venous flap: direct observation of microcirculation.

The circulating mechanism of the "pure" venous flap was studied via venography and direct observation of the microcirculation through transparent ear chambers and ear edges in a rabbit model. The flap, without arterial inflow, survived with normal hair growth and wound healing. Renewed blood circulation occurred 3 to 5 hr after transfer There were two typical flow patterns in the venules: one was a shaking movement with high frequency and short range; the other was a pendulum-like movement with low frequency and long range. The former was related to heart rate, while the later was not. The pendulum-like movement was the main flow responsible for perfusion of capillaries. Blood flow in the capillaries occurred 1 to 2 hr later than that in the venules. The pendulum-like flow perfused the capillaries in two ways: one was in the direction of venules to capillaries and then back to venules along the same route; the other was from venules to arterioles through arteriovenous shunts and then to capillaries. The first type of perfusion was gradually replaced by the second 2 days after transfer. There were extensive venous plexus flows in the flap, which lasted more than 4 weeks after transfer. Revascularized vessels ultimately supplied blood to the flap. The dynamic energy for the movement of blood at an early post-transfer stage was probably due to spontaneous venomotion.

[Observations of changes in neurobehavioral functions in workers exposed to high-frequency radiation].

OBJECTIVE: To study the effects of exposure to high-frequency radiation on neurobehavioral function of the exposed workers and its measurement in evaluating occupational hazards caused by it. METHODS: Four neurobehavioral functions were tested for the workers exposed to high-frequency radiation with Neurobehavioral Core Tests Battery recommended by WHO. RESULTS: Scores for various indicators in exposed workers were significantly lower than those in controls, and correlated to the detection of neurasthenia in the exposed workers, to certain extent. CONCLUSION: Changes in neurobehavioral function in workers exposed to high-frequency radiation can reflect its important adverse effects.

[Encephalic morphologic changes due to delayed traumatic intracerebral hematoma].

Pathological changes of brain tissues due to delayed traumatic intracerebral hematoma (DTICH) in 6 cases were studied and the mechanism was discussed. It was observed that DTICH went with old brain contusion, traumatic angionecrosis, vasculitis, calcification, encapsulated old focus, proliferation and reactive hypertrophy in glia. Brain contusions and traumatic vessel lesions are important pathological foundations of cerebral hemorrhage and primary causes of DTICH.

NMDA receptor overstimulation triggers a prolonged wave of immediate early gene expression: relationship to excitotoxicity.

Exposure of the rodent striatum to quinolinic acid (QA, N-methyl-D-aspartate receptor agonist) induces immediate early gene (IEG; c-fos, c-jun, jun-B, zif/268) expression that may extend 12-24 h after injection. In order to determine the specificity of the prolonged IEG response to the QA injection, the temporal pattern of c-fos mRNA expression was examined during the first 4 h after administration of saline or QA (40 micrograms). As early as 30 min after intrastriatal injection, both saline and QA increased c-fos mRNA levels. In the saline group, this increase in IEG expression was only transient and returned to baseline by 1 h. In contrast, c-fos mRNA levels within QA-injected animals continued to rise significantly at 1 and 4 h. In a second experiment, rats received 4 ng to 40-micrograms injections of QA followed by sacrifice at 6 h to determine if increasing QA doses caused the appearance of the prolonged IEG response phase. The prolonged IEG response was evident at 6 h only in animal groups that received higher dose ranges (4-40 micrograms) of QA. A final experiment was undertaken to determine if blockage of NMDA receptor stimulation would also inhibit the prolonged IEG response at 6 h in relationship to neuronal sparing evidenced at 24 h post-QA injection. The NMDA receptor antagonist, MK-801, blocked the prolonged IEG response at 6 h following QA (40 micrograms) injection while also preventing striatal neuropeptide mRNA decline by 24 h. Delaying the MK-801 administration for 1-2 h post-QA injection revealed that the intensity of the prolonged IEG mRNA response may be predictive of neuronal demise within the QA lesion site. These results suggest that prolonged IEG expression is associated with QA excitotoxicity of the rodent striatum and subsequent neuronal degeneration.

[A study on the counteracting effect of hypoxia and Qigong on simulated weightlessness and orthostatic tolerance after bedrest].

-6 degrees head down bed rest (-6 degrees HDBR) 21d were used to simulate weightlessness. 15 healthy males, aged 19-22, were divided into 3 groups with 5 people in each group: control group (-6 degrees HDBR), hypoxia group (-6 degrees HDBR + Low Oxygen), Qigong group (-6 degrees HDBR + Qigong). The results showed that the reduction of weight, perimeter of shank, daily average cardic output, basal cardic output and the pulse pressure of the Qigong group were lower than those of the control group (P<0.05). The basal cardic output and the perimeter of shank of the Hypoxia group reduced significantly than those of the control group (P<0.05). However, the reduction of the other physiological functions of the Hypoxia group were not significantly lower than that of the control group (P>0.05). During HUT +75 degrees 20 min orthostatic tolerance test after HDBR, the parameters in the Hypoxia group were better than those in the other groups. The average standing time was also longer, and no presyncopal symptom occured.

[Effect of body position changes and circadian rhythm on cerebral blood flow velocity].

To study the effects of -6 degrees head down tilt (HDT), upright posture and 24 h circadian rhythm on cerebral blood flow (CBF) velocity in humans, the CBF velocity was measured by use of transcranial Doppler technique in the middle cerebral artery (MCA) of 12 healthy male subjects. The results showed that the mean CBF velocity increased significantly during -6 degrees HDT and decreased during upright posture, and CBF velocity was higher in day time than at night.

[Preliminary studies of auditory fusion frequency in noise effects].

Auditory fusion frequency (AFF) of impulse sound and paired pitch in 80 workers exposed to noise and 60 non-exposed ones were measured to study the relationship between AFF and auditory fatigue. AFF test software was made by the authors themselves, and a stimulating sound signal was evoked in a personal computer. Relationship between AFF in non-exposed workers and their ages was explored, and AFF in exposed ones before and after their exposure was compared. Results showed that no relationship between AFF and their sex and ears in non-exposed group was found. There was a significant difference in AFF between groups of 50-59-year and less than or equal to 49-year. Stimulating sound was more sensitive in its carrier wave or center frequency of 1,000 Hz, and paired pitch has higher value in screening for subjects with auditory threshold shift at about 1/40 octave band. There was significant consistency in transient auditory threshold shift, an important indicator of AFF and auditory fatigue. Measurement of AFF is more stable, repeatable and less deviated, and needs only simple acoustic conditions. Hence, it is a practical superthreshold audiometry.

The individual and combined effects of ozone and simulated acid rain on growth, gas exchange rate and water-use efficiency of Pinus armandi Franch.

The seedlings of Pinus armandi Franch. were exposed to ozone (O(3)) at 300 ppb for 8 h a day, 6 days a week, and simulated acid rain of pH 3.0 or 2.3, 6 times a week, alone or in combination, for 14 weeks from 15 June to 20 September 1993. The control seedlings were exposed to charcoal-filtered air and simulated rain of pH 6.8 during the same period. Significant interactive effects of O(3) and simulated acid rain on whole plant net photosynthetic rate were observed, but not on other determined parameters. The exposure of the seedlings to O(3) caused the reductions in the dry weight growth, root dry weight relative to the whole plant dry weight, net photosynthetic rate, transpiration rate in light, water-use efficiency and root respiration activity, and increases in shoot/root ratio, and leaf dry weight relative to the whole plant dry weight without an appearance of acute visible foliar injury, but did not affect the dark respiration rate and transpiration rate in the darkness. The decreased net photosynthetic rate was considered to be the major cause for the growth reduction of the seedlings exposed to O(3). On the other hand, the exposure of the seedlings to simulated acid rain reduced the net photosynthetic rate per unit chlorophyll a + b content, but did not induce the significant change in other determined parameters.

Modulation of multidrug resistance gene (mdr-1) with antisense oligodeoxynucleotides.

1. Multidrug resistance is the major obstacle to successful cancer chemotherapy. Circumventing multidrug resistance therefore represents a high priority for clinical anti-cancer treatment. Among many reversal strategies, antisense oligodeoxynucleotides may offer a molecular targeting tool for overcoming cellular multidrug resistance. 2. Two 17-mer phosphorothioate antisense oligomers, complementary to the 5' end of the ATG initiator codon-containing region and loop-forming site (located at nucleotides 991-1007 from the first ATG codon) in mdr-1 cDNA sequence, were synthesized. The purpose was to study their effects on the function and expression of P-glycoprotein and mdr-1 gene. 3. The results showed that 10 mumol/l antisense oligomers could significantly inhibit the growth of multidrug resistant K562/Adm cells cultured in adriamycin-containing medium. No such effect was observed for parental (sensitive) K562/S cells. Intracellular daunorubicin accumulation increased greatly in the K562/Adm cells after they were treated with oligomers for 48 h and P-glycoprotein synthesis was strikingly reduced. 4. Further investigation with [alpha-32P]dCTP incorporation by the reverse transcriptase-polymerase chain reaction method revealed that antisense oligomers could result in a reduction in the level of mdr-1 mRNA, probably through hindering mdr-1 gene transcription. 5. The high reversal efficiency and specificity of antisense oligomers in regulating mdr-1 gene expression suggest a potential clinical application in gene therapy for drug resistant malignancies.

The peak latency of orbital presaccadic spike potential with horizontal eye movements.

PURPOSE: To investigate the peak latency of the orbital presaccadic spike potential (SP) with horizontal eye movement in normals. METHODS: Orbital SP was recorded in 28 normal subjects from 8 electrodes around the eyes with Pz as the reference while performing 5 degrees, 10 degrees, 20 degrees, 30 degrees and 40 degrees horizontal saccades to visual targets. SP peak latency was measured from SP onset to SP peak on averaged data aligned on SP peak. RESULTS: Significant main effects on SP peak latency are found for saccade size (P < 0.01), saccade direction (P < 0.01) and electrode site (P < 0.05). No significant main effect on SP peak latency is found for eye (P > 0.05). SP peak latency increases with increasing saccade size from 5 degrees to 40 degrees. SP peak latency is longer with saccades back to center than away from center, and with abducting saccades than with adducting saccades. SP peak latency differs at the electrode sites with an order from shorter to longer as follows: inner canthus (IC); inferior orbit (IO); outer canthus (OC); superior orbit (SO). CONCLUSIONS: The effects on the peak latency of orbital SP can be explained by the saccade dynamic property, volume conduction as well as physiologic and anatomic factors of the eyes and orbits. The peak latency of orbital SP can be used to reflect the temporal characteristics of ocular motor units controlling saccadic eye movement.

Transcription of the Huntington disease gene during the quinolinic acid excitotoxic cascade.

Although Huntington disease (HD) is characterized by the selective neurodegeneration of the basal ganglia and cerebral cortex, efforts to define the disease pathology have been complicated by the widespread expression of the disease gene (IT15) throughout the body. In this study, we examined IT15 mRNA levels during the quinolinic acid (QA) excitotoxic cascade to determine whether neuronal and/or glial expression is regulated by neurodegeneration. Following an initial increase between 1 h and 6 h, IT15 mRNA levels declined in a pattern homologous to a group of neuron-specific genes. Decreased mRNA levels after 24 h demonstrated that glial transcription is not activated by neurodegeneration or gliosis. The 1 h and 24 h mRNA levels strongly suggest that IT15 transcription preferentially localizes to degenerating neurons.

The effects of time point alignment on the amplitude of averaged orbital presaccadic spike potential (SP).

Effects of two time-point selection averaging techniques on orbital SP amplitude were studied on 10 normal subjects performing horizontal saccades. One technique involved averaging from the SP peak, another technique involved averaging from the saccadic onset. The time-point selection methods only affected the magnitude of SP amplitude but did not interact with experimental conditions. Repeated measures analysis of variance showed that SP amplitudes were higher in data aligned on SP peak than those aligned on saccadic onset (P < 0.01). No significant second order (saccade by degree) or third order (saccade by direction) interactions involving averaging method were found (P > 0.05).
