Ultrastructural studies of medullary synaptic inputs to vasopressin-immunoreactive neurons in the supraoptic nucleus of the rat hypothalamus.

Direct projections from the A1/C1 catecholaminergic cell group in the caudal ventrolateral medulla oblongata to neurons containing vasopressin (VP) in the supraoptic nucleus (SON) were studied electron microscopically by a double-labeling technique which combines anterograde tracing after injection of wheat germ agglutinin-conjugated horseradish peroxidase (WGA-HRP) into the A1/C1 cell group with VP immunocytochemistry. WGA-HRP-labeled axon terminals originating from neurons in the A1/C1 cell group were found to make synaptic contacts with VP-immunoreactive cell bodies and dendrites in the SON, most often forming axo-dendritic synapses. This indicates that VP-containing neurosecretory neurons in the SON receive monosynaptic catecholaminergic input from the A1/C1 cell group.

Preservation of Neisseria gonorrhoeae by the gelatin-disc method.

Studies of Neisseria gonorrhoeae are difficult to perform because of the organism's poor survival in vitro. To solve this problem we tried to preserve the organism by a gelatin-disc method. The rate of survival and changes of variations in some biochemical properties of eight strains of N. gonorrhoeae were followed for three years. These studies proved that preservation was satisfactory with only a 1/10 reduction of the living cells. Another trial showed that the organism survived for over six months after being frozen at -20 degrees C. The colonial types, agglutination against red cells from rabbit and guinea pig, and antibiotic susceptibility to penicillin, chloramphenicol, tetracycline, kanamycin, and streptomycin did not change after three years' preservation.

Effects of GABA on neuronal activities in the distal retina of the carp.

When a transient depolarization was evoked in horizontal cells by a transretinal current pulse, a small hyperpolarizing deflection (probably IPSP) was detected in cones. The IPSP was abolished by superfusion of the retina with GABA-containing Ringer solution, which also hyperpolarized the cones. These results suggest that, in the dark, a GABA-mediated negative feedback was operating from horizontal cells to cones, and terminated in the light. Application of GABA also caused remarkable changes in spectral response curves of horizontal cells, mainly because of suppression of responses to monochromatic lights of longer wavelengths. Therefore, it was concluded that the feedback plays an important role in mechanisms of conversion from the trichromatic process in cones to the opponent color process in horizontal cells.

Animals

Six and 14/sec positive spikes with widespread distribution over the scalp as a seizure pattern.

Five patients with either "diencephalic" or "brainstem" epilepsy or "diencephalosis" were examined EEGcally during the course of their hospitalization. The significant changes were detected, mainly by diphenhydramine activation as to the distribution of the 6-14/sec positive spike pattern on the scalp. When the patients were relatively free from their symptoms, the positive spikes were recorded partially on only one or very fre areas, and when they were experiencing clinical paroxysms, the spikes were detected diffusely in almost all areas of the scalp. Chronological EEG observations paralleling the clinical course of illness of a paroxysmal nature are unpopular. However, the necessity of efforts to record EEG's of patients during clinical paroxysms seems important and has been briefly discussed concerning the question as to whether the 6-14/sec positive spike pattern is an abnormal EEG change or perhaps a "seizure pattern".

Adolescent

A method for provoking EEG abnormalities by administration of nialamide.

Nialamide, a kind of monoamine oxydase inhibitor, was used for provoking centrencephalic EEG abnormalities and 6-14 Hz positive spikes. It was assumed that these EEG abnormalities have close relationship with dysfunctions of the brainstem which is rich in monoamines and monoamine oxydase. The nialamide provocation was carried out on 49 inpatients who had centrencephalic discharges in their EEG reports. These patients consisted of four cases of epilepsy, one case of anorexia nervosa, two cases of narcolepsy and 42 cases of diencephalosis. Another series of 22 patients suffering from other diseases, in whome EEG no centrencephalic EEG abnormallities were detected, were examined with nialamide. Out of the 49 patients, 35 cases (71.4+) showed increased EEG abnormalities following the administration of nialamide. It was noteworthy that this drug had provocative effect not only for EEG abnormalities, but also for the symptoms from which patients were suffering. In 26 out of 49 patients, both EEG abnormalities and clinical symptoms were provoked by nialamide. And the nialamide administration resulted in negative on all 22 patients who did not register centrencephalic EEG abnormalities. The mechanism and characteristics of this provocative procedure by the use of nialamide were evaluated.

Identification of amacrine and ganglion cells in the carp retina.

1. Amacrine and ganglion cells in the carp retina were identified from such criteria as photoresponses, intracellular dye staining, responses to optic nerve stimulation and behaviour to a synapse blocking agent. 2. Responses of ganglion cells were accompanied by spike discharges., either facilitated or suppressed by photic stimulation. The cells were also invaded by antidromic impulses, which survived after chemical synapses had been blocked by application of atomized CoCl2 solution. In subsequent histology of the Procion-stained neurones, the cell bodies were found in the ganglion cell layer and the axons were often traced. 3. Amacrine cells were subdivided into two types. The first type gave rise to transient depolarizations at both on- and offsets of spot and annulus illuminations, usually being associated with spike discharges of which the amplitudes varied in different cells. In histology, the cell bodies of this type were situated in the inner nuclear layer and dendrites ramified in two or more discrete sublayers of the inner plexiform layer (the stratified amacrine cell of Cajal). 4. The second type of amacrine cells produced sustained responses during illumination, being associated with no spike but with small oscillatory wavelets. The cell bodies were situated in the inner nuclear layer and the dendrites ramified in a single sublayer of the inner plexiform layer (the monolayered amacrine cell). 5. An attempt was made to see the effect of activation of centrifugal fibres on amacrine cells, but almost all of about 200 cells examined did not respond to optic nerve stimulation. Only two cells produced, with long latency, a small post-synaptic depolarization which disappeared after chemical synapses in the retina had been blocked. It is considered that the physiological role of the centrifugal system is insignificant in the carp retina.

Innominate lines of the colon. Radiological--histological correlation.

Radiological-histological studies showed that the normal mucosal units of the colon are bounded by the innominate lines. The lines are superficial identations of the mucosa with depths of 0.23 mm. No histological difference was demonstrated according to the three radiographic types found. These lines are demonstrable in about 90% of double-contrast examinations using 60-70% w/v barium. The colon should be prepared with dietary restriction, magnesium citrate, and contact laxative under sufficient hydration. The lines can also be demonstrated at endoscopy with dye scattering.

Colon

EEG findings, rapid ACTH test and autonomic nervous symptoms in patients with alopecia areata.

Patients of multiple alopecia areata were examined in their EEG findings and rapid ACTH test and described the symptoms concerning the neuroautonomic system. Some abnormal EEG findings were seen in 75 cases out of the examined 105 patients. As to the rapid ACTH test, 10 cases out of 45 subjects were abnormal. A positive correlation was considered between abnormal EEG findings and abnormal value in the rapid ACTH test. It is speculated that the regulation centers in the brainstem and hypothalamus play a great role in pathophysiology of multiple areata.
