[Study on relationship between Deficiency Syndrome of TCM and the biological behavior in patients of esophageal carcinoma].

OBJECTIVE: To investigate the relation between Deficiency Syndrome (DS) of TCM and the invading depth, lymphnode metastasis, TNM stage and prognosis prediction of esophageal carcinoma. METHODS: According to Syndrome Differentiation of TCM, 101 patients of esophageal carcinoma were evaluated before operation and divided into two groups, the DS group and the non-DS group. RESULTS: Thirty-eight cases in the 101 patients were DS, among them, 14 were Qi-Deficiency, 10 Yin-Deficiency, 10 Qi-Yin Deficiency, 2 Qi- and blood Deficiency, 1 Yang-Deficiency and 1 Yin-Yang Deficiency. There were significant differences between the DS group and non-DS group in the invading depth (deeper in DS), metastasis of lymphnode (severer in DS) and TNM stage (later in DS) of esphageal carcinoma, (P < 0.05). CONCLUSION: There is a significant relation between DS and the biological behavior of esophageal carcinoma. The prognosis of DS patients is poorer than that of the non-DS patients.

[Frequent loss of fragile histidine triad gene transcripts in lung cancer].

OBJECTIVE: To investigate the significance of the fragile histidine triad (FHIT) gene in evaluation of early diagnosis and metastasis in lung cancer. METHODS: 47 lung cancer samples and 16 matched metastatic hilar lymph nodes of lung cancer patients were examined for abnormalities of the gene by using RT-PCR and RT-PCR-SSCP. RESULTS: 32 of 47 (68%) lung cancer samples and 15 of 16 metastatic hilar lymph nodes showed loss of FHIT gene transcripts (exon5-9 or exon1-4). RT-PCR-SSCP reveal no variants. CONCLUSIONS: These data indicate that loss of FHIT gene transcript is frequent in lung cancer and may be an early event in lung tumorigenesis and FRA3B may be involved in the development and progression of the lung cancer.

[The alteration of microsatellite DNA in lung cancer].

OBJECTIVE: To study the alteration of microsatellite DNA in primary lung cancer and its value of diagnosis for lung cancer. METHODS: Five microsatellite markers on chromosome 3P13-26 have been examined in 50 primary lung cancer tissues and their normal lung tissues. RESULTS: In fifty lung cancer, the incidence of microsatellite instability (MSI) was 52% (26/50) and the loss of heterozygosity (LOH) 38% (19/50). The positive rate of MSI in poorly differentiated lung cancer (13/17, 77%) was significantly higher than that in well, moderately differentiated (10/28, 36%). The sensitivity in the diagnosis of lung cancer was 72% by combining MSI and LOH. CONCLUSIONS: MSI and LOH may play a certain role in the carcinogenesis and progression of lung cancer and they may be valuable molecular markers in diagnosis of lung cancer.

[Primary shortening followed by bone lengthening in situ for the treatment of gunshot bone defect].

OBJECTIVE: To study the treatment of gunshot bone defect and the biological basis of primary shortening followed by callus distraction in situ. METHODS: The canine with gunshot bone defect were treated by two methods, primary shortening followed by callus distraction or delayed bone graft. The local blood flow and pathomorphological change were observed and compared with each other. RESULTS: In the experiment, the bone lengthening was stopped at the 56th day with (2.82 +/-e 0.94) cm (19.3%) gain in the group of bone lengthening, and the time of bone healing was much shorter in the group of bone lengthening than in the group of delayed graft. There was no significant difference between the groups in the time of soft tissue healing and local blood flow. CONCLUSIONS: The technique of primary bone lengthening is simple and has no side effect on tissue healing and local blood flow.

[The apoptosis and proliferation after photorefractive keratectomy].

OBJECTIVE: To search the correlation between the apoptosis and proliferation of keratocytes and investigate the influence of phototherapeutic keratectomy (PTK) and mechanical epithelial scrape (MES) on keratocyte apoptosis and proliferation. METHODS: Rabbit corneas received photorefractive keratectomy (PRK, -9.9 diopters, 6 mm diameter). Animals were evaluated subsequently up to 6 months after surgery by in vivo confocal microscopy. Corneas were prepared for H.E. staining, corneal cell apoptosis was evaluated by terminal deoxyribonucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) to detect DNA fragmentation. RESULTS: Loss of keratocytes and keratocyte death were found anteriorly in the remaining stroma at 4h after PRK. Activated keratocytes observed by confocal microscopy repopulated within 1 month. A significant elevation of apoptosis was detected in keratocytes and epithelium at 4h, 3d, 1m after PRK. The level of keratocyte apoptosis was parallel to the loss of keratocytes at the early stage and was correlated with keratocyte proliferation in the later stage, PTK-PRK was associated with lower levels of central corneal apoptosis and activated keratocytes than MES-PRK. CONCLUSION: It is suggested that the loss of keratocytes and keratocyte apoptosis be correlated with keratocyte activation and proliferation, which may result in haze and regression after PRK, and PTK-PRK induce lower level of early keratocyte apoptosis and late keratocyte activation than MES-PRK.

[Excimer laser photorefractive keratectomy for lower to moderate myopia: 5 year follow-up].

OBJECTIVE: To evaluate the long-term effectiveness and safety of photorefractive keratectomy (PRK) for patient with lower to moderate myopia. METHODS: 148 myopic eyes (-1.5 to -6.00 D) after PRK follow-up of more than 5 years were analyzed. RESULTS: Uncorrected visual acuity was >or= 1.0 in 136 eyes (91.9%), >or= 0.8 in 4 eyes (2.7%) and >or= 0.6 in 8 eyes (5.4%). The mean spherical equivalent change was from (-4.43 +/- 0.97) D to (-0.12 +/- 0.22) D, and the keratometric power (K value) was reduced from (43.81 +/- 1.10) D to (41.36 +/- 1.30) D in the 5 years of follow-up (P < 0.01). CONCLUSION: The long-term effectiveness of PRK for myopia (-1.5 to -6.00 D) is satisfactory.

[The complications of excimer laser in-situ keratomileusis for myopia].

OBJECTIVE: To analyze the complications of excimer laser in-situ keratomileusis (LASIK) for myopia, investigate the causes and discuss the prevention of these complications. METHODS: The SCMD microkeratome and Schwind Keratom F excimer laser were used to correct myopia on 1,265 eyes in 662 cases. The eyes were divided into 3 groups: A: -10.00 D, 252 eyes. All of them were followed up for a period of 12 months. RESULTS: Intraoperative corneal disk completely detached: 6 eyes (0.5%); corneal disk incomplete sectioned: 3 eyes (0.2%); corneal epithelial disk: 2 eyes (0.2%); corneal interface minute foreign metal bodies: 11 eyes (0.9%); postoperative steroid intraocular hypertension: 21 eyes (1.7%); macular hemorrhage: 2 eyes (0.2%). At postoperative 3 months, there was refractive regression > 1.00 D in 0.0%, 3.1% and 7.9% respectively in the 3 groups; the increase in astigmatism > 1.00 D, 0.4%, 2.8% and 4.4% respectively. CONCLUSION: LASIK is a safe effective method for correcting myopia. Yet attention should be paid on the complications during and after operation. With the improvement in operative technique and better designed procedure, these complications can be decreased and the preoperative correctness in estimation of the postoperative results and the postoperative accuracy in the correction of myopia can be elevated.

[Roles of bifidobacterium on prevention of experimental colorectal carcinoma and induction of apoptosis].

OBJECTIVE: To explore the effect of bifidobacterium adolescence on prevention from experimental colorectal carcinoma and its antitumor mechanism. METHODS: An animal model of nude mouse tumor transplanted from colorectal carcinoma was set up. Bifidobacteria adolescence were intraperitoneally injected into nude mice in advance. The growth rate, ultrastructure, density of apoptotic cells, rate of bcl-2 and bax oncoprotein expression and its positive cell density in transplanted tumor of nude mice from colorectal carcinoma were investigated by employing transmission microscopy, in situ end labeling technique and immunohistochemical staining. RESULTS: The growth rate of transplanted tumors from colorectal carcinoma in the group with bifidobacterium prevention was slower than that in the control group. Many tumor cells at varied apoptotic stages could be observed in transplantation tumor tissue in the bifidobacterium prevention group and their density of apoptotic cells, rate of bcl-2 and bax oncoprotein expression and its positive cell density were significantly higher in the group with bifidobacterium prevention, as compared with the control group (P < 0.01). But, expression rate of bcl-2 oncoprotein turned contrary to that of bax oncoprotein. CONCLUSION: Bifidobacteria adolescence could markedly prevent from occurrence and development of colorectal carcinoma in vivo and induce apoptosis of tumor.

[Effect of human inhibin alpha-fragment 1-32-Tyr(P33) on apoptosis of cultured rat corpus luteal cells].

Recently, increasing evidence suggests that alpha inhibin or related proteins may be a functional regulator in the ovary, which is independent of hetero-dimer inhibin. In our previous study, it was demonstrated that human inhibin alpha-N-terminal fragment Tyr-1-32 (P(33)) significantly inhibited progesterone production by rat corpus luteal cells in vitro, and stimulated luteal functional regression and apoptosis in vivo. In the present work, the action of P(33) on apoptosis was further studied in vitro in cultured rat CL cells. Gel electrophoretic analysis for detection of oligonucleosomal DNA fragmentation, AO-EB or PI assays and flow cytometry were used to observe the action of P(33) on the occurrence of spontaneous apoptosis by collagenase-DNase dispersed CL cells, obtained from PMSG-hCG induced pseudopregnant rats. The results showed that P(33) (1 microg/ml) stimulated spontaneous apoptosis of CL cells. The inhibitor of tyrosine protein kinase, genistein (50 microg/ml),inhibited P(33) enhanced spontaneous apoptosis. RNA and protein synthesis inhibitors cycloheximide (Cyx,50 microg/ml) and actinomycin D(Act D,50 microg/ml) did not protect the cells from apoptosis stimulated by P(33). The results suggest that P(33) stimulates spontaneous apoptosis in cultured rat CL cells with the involvement of tyrosine specific protein kinase system. This work provides further evidence for the hypothesis that alpha inhibin or related protein might be a functional regulator in the ovary.

[Study of cleft palate with problems of middle ears and its treatment].

OBJECTIVE: To gain a clear idea of the following: 1. the relationship between the incidence of functional obstruction of middle ear and surgery on cleft palate and patients' age; 2. the bacterial culture of secretion taken from the tympanic cavity with secretory otitis media; 3. the preliminary effects of treatment by inserting a tube into the tympanic cavity of a patient with secretory otitis media. METHODS: 275 middle ears of patients with cleft palate were selected, then acoustic immitence examination and hearing threshold of pure tone were adopted, and bacterial culture was carried out for studying secretion of the middle ear with secretory otitis media. RESULTS: The tympanograms of 75.5% ears of the patients before cleft palate operation were found to be abnormal, among which patients under 3 years were 81%, and 41.2% ears of those after closure of cleft palate were abnormal. Gram's positive and negative germs of bacterial culture were 20.1% and 24% respectively. The hearing of 6 ears with secretory otitis media has recovered from the moderate hearing loss due to tube insertion. And the tympanograms of 4/8 ears were transformed from type B to type A. CONCLUSION: It is beneficial to improve function of Eustachian tube if cleft palate can be repaired at an early stage and beneficial to improve the patients' hearing and devlope their speech if a tube is inserted into the typanic cavity in cases of secretory otitis media at an early stage.

[Immunofluorescence studies on the localization of integrin-like protein in plant pollen tubes].

The Strong fluorescence signals were obtained in pollen tube of Lilium davidii Duch with labeled anti-VnR integrin serum, and anti-beta 3, alpha v integrin subunit cytoplasmic domain serum separately. The highest density of immunolabel was in the tip of pollen tube. There was little or no immunolabel in control experiment using non-immune serum, second antibody alone and anti-FnR, LnR integrin serum separately. In pollen of Prunus persica f. rubro-piena Schneid, fluoresence signals were also obtained in tube using labeled anti-beta 1, beta 3 integrin subunit cytoplasmic domain serum separately and in apertures using anti-beta 1 serum. Preliminary results show that during the germination of pollen and the growth of pollen tube, there may be integrin-like proteins in pollen tube, consisting of alpha v and beta s integria subunits in the pollen tube of Lilium davidii Duch, which is the receptor of vitronectin-like protein.

[Construction of human pLXSN-CD80 retrovirus vector and expression in CHO and PA317 cells].

CD80, a molecule on the antigen presenting cells, provide costimulation signals for T cell activation and play a key role in tumor immune. Based on our previous work of human CD80 full length cDNA cloning, a retrovirus expression vector pLXSN-CD80 was constructed. CD80 expression cells were selected by G418 from pLXSN-CD80 transfected PA317 and CHO cells by calcium phosphate. Expression, distribution and molecular weight (MW) of CD80 were measured by RIA, FACs and western blot. pLXSN-CD80 transfected CHO cells expressed relatively high level of CD80 protein (approximately the same as Raji cells) with an apparent MW of 40 kD. In the presence of G418 or not, pLXSN-CD80 transfected PA317 and CHO cells maintained CD80 expression for five months of passage. The results indicate that our construct is potent for experimental use in gene therapy.

[Registration and fusion of multimodality medical images].

This paper introduces the present situation of research and methods of multimodality medical images fusion. Some algorithms and relative techniques are given in detail. A summary of the clinical application of multimodality medical images fusion is also presented.

[Modification of N-terminal cDNA of hGM-CSF and high-expression in E. coli].

OBJECTIVE: N-terminal cDNA of human granulococyte-macrophage colony stimulating factor(GM-CSF) was designed to be modified and highly-expressed in E. coli. METHODS: A pair of oligo-nucleotide primers were used to modify the mature N-terminal cDNA sequence of hGM-CSF with the method of PCR. the modified cDNA of hGM-CSF was cloned into E. coli. expressive vector PBV220 and expressed in E. coli DH5 alpha strain, the biological activities of the recombinant protein was identified by means of cell colony formation and TF-1 cell growth assay in vitro. RESULTS: The expression level of modified hGM-CSF cDNA was higher than that of unmodified native type. SDS-PAGE revealed that expressed protein of hGM-CSF accounted for about 25% of total bacterial cell protein. The biological activity of recombinant protein was about 1.5 x 10(7) U/mg protein. The sequence of 1-16 amino acid of N-terminal of the recombinant protein was same with native hGM-CSF. CONCLUSIONS: Modification of the N-terminal cDNA of hGM-CSF could dramatically enhance the expression level in E. coli system.

[Determination of B(a)p contents of pine tar and its extractives].

In this paper, the B(a)P contents of pine tar and its extractives A, B were determined by PC-Scanning method. After combining extraction by caffeine and formic acid with PC-Scanning method, the B(a)P contents were obtained. The results showed the B(a)P contents of pine tar and its extractives A, B were 6171.3 ppb, 2285.2 ppb, 129.6 ppb, among them, the B(a)P content of B was the lowest.

[Efficient expression of human papillomavirus type 16 (HPV16) capsid proteins L1 and L2 in E. coli].

OBJECTIVE: To study the expression of HPV16 capsid proteins L1 and L2 in E. coli. METHODS: The plasmids pET-30a-L1 and pET-30a-L2 were constructed from the pET-30a vector, and transducted into the BL21. The fusion proteins 6 x His-L1 and 6 x His-L2 were expressed when induced by adding of IPTG. SDS-PAGE and Western blot analyses were used to detect the expressed proteins. RESULTS: Fusion proteins 6 x His-L1 and 6 x His-L2 were expressed efficiently in E. coli, the Mr were about 60,000 and 97,000 respectively, 6 x His-L1 degraded more than 6 x His-L2. CONCLUSION: L1 and L2 proteins were expressed at a high level in prokaryotic expression system, and 12 protein were more stable than L1 protein.

[Photoacoustic spectroscopy investigation of intramolecular energy transfer in Eu-beta-disketones].

The complex crystals of Eu(III) with AA, HFA and TTA have been synthesized. The absorption intensities of Eu-AA, Eu-HFA and Eu-TTA are different while the photoacoustic (PA) signals of the corresponding solid complexes are almost the same, which shows that the probabilities for nonradiative transition of these chelates are different. The results of the fluorescence spectra are identical. The PA signal was obtained by detecting the heat generated through the nonradiative processes after absorption, and the intramolecular energy transfer processes of Eu-beta-diketones were studied from a new point of view.
